Zweck der vorliegenden Erfindung ist es, neue pharmakologisch aktive Derivate des Vincamins herzustellen. Vincamin ist das Hauptalkaloid des Immergrüns, Vinca minor L. Familie Apocynaceae. 1964 wurde Vincamin erstmals synthetisch dargestellt. Zweck der vorliegenden Erfindung ist es, neue pharmakologisch aktive Derivate des Vincamins herzustellen. Die erfindungsgemäss hergestellten neuen Verbindungen unterscheiden sich von Vincamin dadurch, dass sie im aromatischen Ring und in Stellung 14 die in der Formel I angegebenen Substituenten aufweisen. Die neuen Verbindungen bewirken eine Vigilanzerhöhung und Psychostimulation.
Erfindungsgemäss gelangt man zu den Verbindungen der Formel 1, indem man Verbindungen der Formel II,

Das Verfahren kann analog zu bekannten Methoden durchgeführt werden.
Das Verfahren ist eine Additionsreaktion. Sie wird im allgemeinen unter sauren Bedingungen durchgeführt, zweckmässig unter Ausschluss von Wasser. Man verwendet z. B. 1,0 bis 2,0 Mol Säure, beispielsweise Schwefel- oder Methansulfonsäure, pro Mol Verbindung III.
Man kann in einem unter den Reaktionsbedingungen inerten Lösungsmittel wie Chloroform oder in einem Überschuss der Verbindung der Formel II arbeiten.
Die Reaktionstemperatur kann variieren; im allgemeinen wird die Reaktion zweckmässig bei erhöhter Temperatur, z. B.
zwischen 40 bis 80", durchgeführt.
Bei den Umsetzungen gemäss obigem Verfahren entsteht jeweils ein Gemisch von Verbindungen der Formel 1, in denen R in trans-Stellung zum Wasserstoffatom in Stellung C3 steht (Epivincanolderivate), mit Verbindungen der Formel 1, in denen R in cis-Stellung zum Wasserstoffatom in Stellung C3 steht (Vincanolderivate).
Die nach dem erfindungsgemässen Verfahren erhältlichen Verbindungen der Formel I können in freier Form, als Base, oder in Form ihrer Additionssalze mit Säuren vorliegen. Aus den freien Basen lassen sich in bekannter Weise Säureadditionssalze, z. B. das Hydrogenfumarat, herstellen und umgekehrt.
Die Verbindungen der Formel I in freier Form oder in Forn
von physiologisch verträglichen Additionssalzen mit Säuren zeichnen sich durch interessante pharmakodynamische Eigenschaften aus. Sie können als Heilmittel verwendet werden.
So sind sie nützlich, da sie vigilanzerhöhende und psychostimulierende Eigenschaften besitzen.
Folgende Eigenschaften lassen auf die erwähnten Eigenschaften schliessen:
Am Beobachtungstest an der Maus konnte eine Erregbarkeitssteigerung bei Dosen von 10 bis 100 mgikg p.o. beobachtet werden. Die Schlafphasen im Elektroenzephalogramm der Ratte zeigten eine Abnahme des Schlafes und eine Zunahme des Wachzustandes bei Dosen von 10 bis 30 mglkg i.p. oder p.o.
Die erfindungsgemäss hergestellten Verbindungen sind deshalb indiziert bei zerebralen Gefässschädigungen, bei der Zerebralsklerose, bei Zerebralinsuffizienz oder bei Bewusstseinsverlusten aufgrund von Schädeltraumata.
Die zu verwendenden Dosen variieren naturgemäss je nach eingesetzter Substanz, Art der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate erreicht mit einer täglichen Dosis von 0,01 bis 100 mg/kg Tierkörpergewicht; diese Dosis wird zweckmässig in Teildosen, z. B. 2 bis 4 mal täglich, oder in Retardform verabreicht. Für grössere Säugetiere liegt die Tagesdosis bei ungefähr 1 bis ungefähr 500 mg. Geeignete Dosierungsformen für z. B. orale Administration enthalten beispielsweise ungefähr 0,25 bis ungefähr 250 mg der erfindungsgemäss hergestellten Verbindungen neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Das nachfolgende Beispiel erläutert die Erfindung.
Beispiel: 14-Deoxy-10-fluor-14-methylthiovincanol
Zu 29,64 g (100 mmol) (3S,16S)-10-Fluoreburnamenin wurde bei trockenes Bromwasserstoffgas geleitet und kondensiert, bis ein Lösungsvolumen von 100 ml erreicht war. In einem Trockeneis/Äthanol-Bad wurde dann bei 200 Torr unter Rühren bis zum erstarrenden Brei eingeengt, der Rückstand anschliessend 16 Stunden bei 20 Torr und getrocknet, das Vakuum mit Stickstoff entlastet und der spröde Rückstand (Verbindungen der Formel IV; X = 10-Fluor, Z = Brom) unter Stickstoff bei -78" pulverisiert.
Zum pulverisierten Rückstand gibt man bei 780 unter
Rühren 100 ml Methanthiol hinzu, lässt unter Rühren während einer Stunde auf 0" aufwärmen und eine Stunde bei 0" weiter rühren. Hierauf verteilt man das Reaktionsgemisch zwischen 500 ml 2N Ammoniak und 200 ml Methylenchlorid, dampft die
Methylenchloridlösung ein und kristallisiert die Titelverbin dung aus Methanol (Smp. 155").
(3S,16S)-10-Fluoreburnamenin erhält man z. B. wie folgt:
Ein Gemisch von 70,9 g (20bmmol) 10-Fluorapovincamin, 28,96 ml (300 mmol) absolutem Diäthanolamin und 100 ml absolutem Dimethylsulfoxid wurde unter Stickstoff 4 Stunden auf 1800 erhitzt, bis die Gasentwicklung nachgelassen hatte und eine dunkelbraune Lösung vorlag. Verteilung zwischen Toluol und 10/obiger wässriger Natriumchlorid-Lösung, Eindampfen der organischen Phase und Chromatographie an Kieselgel mit Methylenchlorid/Methanol 99 1 ergaben (3S,16S > 10-Fluore- burnamenin in öliger Form.
