Das Verfahren kann analog zu bekannten Methoden durchgeführt werden.
Das Verfahren ist eine Additionsreaktion. Sie wird im allgemeinen unter sauren Bedingungen durchgeführt, zweckmässig unter Ausschluss von Wasser. Man verwendet zum Beispiel 1,0 bis 2,0 Mol Säure, beispielsweise Schwefel- oder Methansulfonsäure, pro Mol Verbindung III.
Man kann in einem unter den Reaktionsbedingungen inerten Lösungsmittel wie Chloroform oder in einem Überschuss der Verbindung der Formel II arbeiten.
Die Reaktionstemperatur kann variieren; im allgemeinen wird die Reaktion zweckmässig bei erhöhter Temperatur, zum Beispiel zwischen 40 bis 80 , durchgeführt.
Bei den Umsetzungen gemäss obigem Verfahren entsteht jeweils ein Gemisch von Verbindungen der Formel I, in denen R in trans-Stellung zum Wasserstoffatom in Stellung C3 steht (Epivincanolderivate), mit Verbindungen der Formel I, in denen R in cis-Stellung zum Wasserstoffatom in Stellung C3 steht (Vincanolderivate).
Die nach dem erfindungsgemässen Verfahren erhältlichen Verbindungen der Formel I können in freier Form, als Base, oder in Form ihrer Additionssalze mit Säuren vorliegen. Aus den freien Basen lassen sich in bekannter Weise Säureadditionssalze, zum Beispiel das Hydrogenfumarat, herstellen und umgekehrt.
Die Verbindungen der Formel I in freier Form oder in Form von physiologisch verträglichen Additionssalzen mit Säuren zeichnen sich durch interessante pharmakodynamische Eigenschaften aus. Sie können als Heilmittel verwendet werden.
So sind sie nützlich, da sie vigilanzerhöhende und psychostimulierende Eigenschaften besitzen.
Folgende Eigenschaften lassen auf die erwähnten Eigenschaften schliessen:
Am Beobachtungstest an der Maus konnte eine Erregbarkeitssteigerung bei Dosen von 10 bis 100 mg/kg p.o. beobachtet werden. Die Schlafphasen im Elektroenzephalogramm der Ratte zeigten eine Abnahme des Schlafes und eine Zunahme des Wachzustandes bei Dosen von 10 bis 30 mg/kg i.p. oder p.o.
Die erfindungsgemäss hergestellten Verbindungen sind deshalb indiziert bei zerebralen Gefässschädigungen, bei der Zerebralsklerose, bei Zerebralinsuffizienz oder bei Bewusstseinsverlusten aufgrund von Schädeltraumata.
Die zu verwendenden Dosen variieren naturgemäss je nach eingesetzter Substanz, Art der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate erreicht mit einer täglichen Dosis von 0,01 bis 100 mglkgTierkörpergewicht; diese Dosis wird zweckmässig in Teildosen, zum Beispiel 2 bis 4 mal täglich, oder in Retardform verabreicht. Für grössere Säugetiere liegt die Tagesdosis bei ungefähr 1 bis ungefähr 500 mg. Geeignete Dosierungsformen für zum Beispiel orale Administration enthalten beispielsweise ungefähr 0,25 bis ungefähr 250 mg der erfindungsgemäss hergestellten Verbindungen neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Die nachfolgenden Beispiele erläutern die Erfindung.
Beispiel 1:
O-Methylvincanol und O-Methylepivincanol
27,84 g (100 mmol) (3S,16S > Eburnamenin wurden in 100 ml 1 M absolut-methanolischer Schwefelsäure gelöst und 30 Minuten unter Stickstoff gesiedet. Nach Verteilung zwischen 200 ml 2N Ammoniak und Methylenchlorid, Trocknen und Eindampfen erhielt man amorphes Material, bestehend aus einem Gemisch der beiden epimeren Basen. Fraktionierte Kristallisation der Hydrogenfumarate aus 2-Propanol bei 0 lieferte zuerst Kristalle von O-Methylvincanolhydrogenfumarat, Smp.
1610 (Zers.), dann Kristalle von O-Methylepivincanolhydrogenfumarat, Smp. 1100 (Zers.).
Freisetzung der Basen erfolgte jeweils durch Verteilen zwischen 2N wässrigem Ammoniak und Methylenchlorid, worauf die erhaltenen öligen Rückstände aus 2-Propanol kristallisiert werden konnten.
O-Methylvincanol: Smp. 106.
O-Methylepivincanol: Smp. 163".
(3S,16S > Eburnamenin erhält man zum Beispiel wie folgt:
Ein Gemisch von 67,29 g (200 mmol) Apovincamin, 28,96 ml (300 mmol) absolutem Diäthanolamin und 100 ml absolutem Dimethylsulfoxid wurde unter Stickstoff 4 Stunden auf 1800 erhitzt, bis die Gasentwicklung nachgelassen hatte und eine dunkelbraune Lösung vorlag. Verteilung zwischen Toluol und 1 /Oiger wässriger Natriumchlorid-Lösung, Eindampfen der organischen Phase und Chromatographie an Kieselgel mit Methylenchlorid/Methanol 99:1 ergaben (3S,16SpEburnamenin in Form eines hellgelben Honigs.
Beispiel 2: 14-Desoxy-1 4(hydroxyäthylthiovincanol
13,92 g (50 mmol) (3S,16SpEburnamenin und 4,69 g (60 mmol) Mercaptoäthanol wurden in 60 ml 1 M Methansulfonsäurelösung in Chloroform gelöst und 2 Stunden bei 20" stehengelassen. Nach Verteilen zwischen 50 ml 2N wässrigem Ammoniak und Methylenchlorid, Trocknen und Eindampfen erhielt man einen harzigen Rückstand, welcher aus 2-Propanol bei 0 kristallisierte. Die Titelverbindung schmilzt bei 175 ".
Beispiel 3: 14-Desoxy-14-isopropylthiovincanol
13,92 g (50 mmol) (3S,16SpEburnamenin und 4,57 g Isopropylmercaptan wurden in 60 ml 1 M Methansulfonsäurelösung in Chloroform gelöst, 2 Stunden gesiedet und 48 Stunden bei 20 stehengelassen. Aufarbeitung und Kristallisation erfolgten wie unter Beispiel 2 erwähnt. Die Titelverbindung schmilzt bei 95o.
Beispiel 4: 1 4-Desoxy-1 4-n-butylthiovincanol
Analog zu Beispiel 3 erhält man unter Verwendung von n-Butylmercaptan statt Isopropylmercaptan die Titelverbindung. Smp. des p-Toluolsulfonats 1670 (aus 2-Propanol).
