3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man D-Homosteroide der Formel II, worin R1 Hydroxy oder C1.7-Alkanoyioxy und R2 Wasserstoff oder Chlor darstellen, als Ausgangsmaterial verwendet.
4. Verfahren nach Anspruch 1; dadurch gekennzeichnet, dass man D-Homosteroide der Formel II, worin R2 Wasser stoff und R1 C1 7-Alkanoyloxy darstellen; als Ausgangsmate- rial verwendet.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen D-Homosteroiden der Formel

Eine Acyloxygruppe kann sich von einer gesättigten oder ungesättigten aliphatischen Carbonsäure, einer cycloaliphatischen, araliphatischen oder einer aromatischen Carbonsäure mit vorzugsweise bis zu 15 C-Atomen ableiten. Beispiele sol cher Säuren sind, Ameisensäure, Essigsäure, Pivalinsäure, Propionsäure, Buttersäure, Capronsäure, Onanthsäure, Undecylensäure, ölsäure, Cyclopentylpropionsäure, Cyclohexylpro- pionsäure, Phenylessigsäure und Benzoesäure. Besonders be vorzugt sind C1.7-Alkanoyloxygruppen. Alkoxygruppen können gerad- oder verzweigtkettig sein, vorzugsweise enthalten sie bis zu 15 C-Atome. Besonders bevorzugt sind Niederalkoxygruppen. Niederalkylgruppen sind solche mit vorzugsweise 1-4 Kohlenstoffatomen, insbesondere Methyl und Äthyl bzw. Methoxy und Äthoxy.
Ein 6ständiger Substituent kann α- oder ss-Konfiguration aufweisen, wobei die a-Isomeren bevorzugt sind.
Die D-Homosteroide der Formel I werden erfindungsgemäss dadurch erhalten, dass man in einen D-Homosteroid der Formel

worin Rt, R2 und Ra dasselbe wie oben bedeuten, die 3-Hydroxyd5-Gruppierung zur 3-Keto-A4-Gruppierung oxydiert.
Die Oxydation einer Verbindung der Formel II kann nach Oppenauer, z.B. mittels Aluminiumisopropylat, oder mit Oxydationsmitteln wie Chromtrioxid (z.B. Jones'Reagens), oder nach Pfitzner-Moffatt mittels Dimethylsulfoxid/Dicyclohexylcarbodiimid. (wobei das primär erhaltene A5-3-Keton anschliessend zum A4-3-Keton isomerisiert werden muss); oder mittels Pyridin/SO3 vorgenommen werden.
Eine bevorzugte Gruppe von D-Homosteroiden der For
mel I sind die, worin R1 Hydroxy, Acyloxy oder Alkoxy bedeutet.
Weiterhin sind solche D-Homosteroide der Formel I bevorzugt, worin R1 Hydroxy oder Cl 7-Alkanoyloxy und R2 Wasserstoff oder Chlor darstellen und insbesondere solche worin R2 Wasserstoff und R1 C1 C1-7-Alkanoyloxy darstellen.
Beispiele von Verbindungen, die unter die allgemeine Formel I fallen, sind: 1 7ax-Acetoxy-6a-methy1D-homo-4,1 6-pregnadien-3,20- -dion,
1 7asc-Acetoxy-D-homo-4, 16-pregnadien-3 20-dion,
Q,1 7aa-Dimethyl-D-homo-4, 1 o-pregnadien9 ,20-dion,
17aα-Methyl-D-homo-4,16-pregnadien-3,20-dion.
Die erfindungsgemäss als Ausgangsmaterial verwendeten D-Homosteroide können, soweit ihre Herstellung noch nicht beschrieben wurde, in Analogie zu den in den Beispielen angegebenen Methoden hergestellt werden; vgl. auch die deutsche Offenlöegungsschrift 2 544 701.
Die Verbindungen der Formel I wirken hormonal, insbesondere auf das endokrine System und sind durch Selektivität der Wirkung gekennzeichnet. Sie können demgemäss als hormonal wirksame Mittel, z.B. als Progestativa verwendet werden und oral oder parenteral verabreicht werden. Als Dosie rungsrichtlinie kommen 0,005 mg/kg bis 0,15 mg/kg pro die in Betracht.
Beispiel I
1,15 g rohes 3ss-Hydroxy-17aα-methyl-D-homo-5,16-preg- nadien-20-on werden in 22 ml Toluol mit 1,1 ml Cyclohexanon zum Sieden erhitzt und mit einer Lösung von 550 mg Aluminiumisopropyiat in 2 ml Toluol versetzt. Nach 45 Minuten langsamem Abdestillieren wird mit Äther versetzt, mit verdünnter Schwefelsäure und Wasser gewaschen, getrocknet und eingedampft. Der Rückstand wird an Silicagel chromatographiert und aus Methanol umkristallisiert. Man erhielt 17aα- -Methyl-D-homo-4,16-pregnadien-3,20-dion vom Schmelzpunkt 181,5-185,5 C. W: #242 = 16 100.
Herstellung des Ausgangsmaterials:
10 g 3ss-Hydroxy-D-homo-5,17(17a)-pregnadien-20-on werden in 100 ml Tetrahydrofuran und 10 ml 2,3-Dihydro-4H -pyran gelöst, mit 0,1 ml Phosphoroxychlorid versetzt und 1 Stunde bei Raumtemperatur nachgerührt. Es wird dann in Eiswasser mit Zusatz von Natriumhydrogencarbonat eingerührt, der ausgefallene Niederschlag abgesaugt und ausgewaschen. Der Rückstand wird in Methylenchlorid aufgenommen, getrocknet und nach dem Eindampfen aus Diisopwpyläther umkristallisiert. Man erhielt 3ss-(Tetrahydropyran-2'-yloxy)- -D-homo-5,17(17a)-pregnadien-20-on vom Schmelzpunkt 153/154-1550C.
1,62 g Kalium-t.-butylat werden in 30 ml abs. Tetrahydrofuran und 45 ml abs. tertiär Butanol gelöst. Zu der im Eisbad gekühlten Lösung werden unter einem Stickstoffstrom 3,6 g 3fi-(Tetrahydropyran-2'-yloxy)-D-homo-5,17(17a)Epreg- nadien-20-on zugegeben. In diese jetzt dunkelbraune Lösung werden 1,65 ml Methyljodid getropft und 30 Minuten bei OOC und Überleiten von Stickstoff nachgerührt. Anschliessend wird in Eiswasser eingerührt, der Niederschlag abfiltriert, in Methylenchlorid aufgenommen, mit Wasser gewaschen und getrocknet. Der nach dem Eindampfen erhaltene Rückstand wird an Silicagel chromatographiert und liefert 3ss-(Tetra- hydropyran-2'-yloxy)-17aα-methyl-D-homo-5,16-pregnadien- -20-on.
Eine aus Diisopropyläther umkristallisierte Probe schmitzt bei 169,5-170,5 C.
1.4 g 3,8-(Tetrahydropyran-2'-ylaxy)-17ax-methyl-D-homo- -5,16-pregnadien-2O-on werden in 27 ml Methanol mit 650 mg Oxalsäure, gelöst in 6,5 mi Wasser, versetzt und 1,5 Stunden am Rückfluss erhitzt. Es wird dann in Eiswasser eingerührt, der ausgefallene Niederschlag abgesaugt, in Chloroform aufgenommen und getrocknet. Nach dem Eindampfen erhielt man rohes 3ss-Hydroxy-17aα-methyl-D-homo-5,16-pregnadien- -20-on.
Beispiel 2
440 mg 17aα-Acetoxy-3ss-hydroxy-D-homopregna-5,16- -dien-20-on werden in 30 ml Aceton mit 0,55 ml Jones'Reagens versetzt, 5 Minuten gerührt, auf Wasser gegossen und mit Methylenchlorid extrahiert. Die Methylenchloridlösungen werden mit Wasser gewaschen, getrocknet und eingedampft.
Das erhaltene Öl wird in 8 ml Dioxan und 0,8 ml 2N Salzsäure 1 Stunde bei Raumtemperatur gerührt. Das Gemisch wird eingedampft und der Rückstand an Kieselgel chromatographiert. Das erhaltene 17aα-Acetoxy-D-homo-pregna-4,16- -dien-3,20-dion schmilzt bei 222-223 C (Aceton-Hexan).
Herstellung des Ausgangsmaterials:
Das 3ss,17aα-Dihydroxy-D-homopregna-5,16-dien-20-on wird in Essigsäureanhydrid und Triäthylamin in Gegenwart von p-N,N-Dimethylaminopyridin zu 3ss;17aα-Diacetoxy-D- --homopregna-5,16-dien-20-on (Smp. 179-180 C, [α]D = 2730, c = 0,1% in Dioxan) umgesetzt und dieses anschliessend mit Pottasche in Methanol partiell zu 17aα-Acetoxy-3ss- -hydroxy-D-homopregna-5,16-dien-20-on verseift.
Beispiel 3 a) Es wird 3ss-Hydroxy-D-homopregna-5,17-dien-20-on in DMF und THF in Gegenwart von Kalium-t.-butylat und Trimethylphosphit mit Sauerstoff zu 3ss,17aα-Dihydroxy-D- -homopregna-5,16-dien-20-on oxydiert. Smp. 241-243 C, [α]D = 1870 (c = 0,1% in Dioxan). Durch Oxydation nach Pfitzner-Moffatt erhält man daraus das 17az-Hydroxy-D- -homopregna-4,16-dien-3,20-dion vom Smp. 177-178 C, UV: ±240 = 16 800. [ L = +180 (c = 0,1% in Dioxan).
b > 4,65 g 17a-Hydroxy-D-homopregna-4,16-dien-3,20 -dion und 1,55 g p-N,N-Dimethylaminopyridin werden in 23,5 ml Triäthylamin und 14,5 mi Essigsäureanhydrid 16 Stunden bei Raumtemperatur unter Argon gerührt. Das Gemisch wird auf eiskalte verdünnte Salzsäure gegossen und mit Methylenchlorid extrahiert. Die Methylenchlon.diösungen werden mit verdünnter Natriumbicarbonatlösung und verdünnter Kochsalzlösung neutral gewaschen, getrocknet und eingedampft. Chromatographie des Rohproduktes an Kieselgel und zweimalige Kristallisation aus Aceton-Hexan gibt 17a0c-Acet- oxy-D-homopregna-4,16-dien-3,20-dion vom Smp. 223 -2240C.
UV: E240 = 16 800. [D = -134 (c = 0,1% in Dioxan).
Beispiel 4
In zu Beispiel 1, 2 oder 3 analoger Weise wird 21-Chlor -17a ,-hydroxy-D-homopregna-4,16-dien-3,20-dion erhalten, Smp. 243-245 C (aus Aceton-Hexan), UV: #240 = 17 200, [α]D = = +860 (c = 0,1% in Dioxan).
Beispiel 5
In zu Beispiel 1, 2 oder 3 analoger Weise erhält man 17ax- -Acetoxy-6α-methyl-D-homopregna-4,16-dien-3,20-dion vom Smp. 216-2180C, [ L = -136 (Dioxan, c = 0,1 > , W: #241 = 14 600.
Beispiel 6
In zu Beispiel 1, 2 oder 3 analoger Weise erhält man 17aa -Acetoxy-6aα-methyl-D-homopregna-4,16-dien-3,20-dion, Smp. 216-2180C, UV: ±241 = 14600, [ L = 1360 (c = 0,1% in Dioxan).
Beispiel 7
1,35 g 3ss-Hydroxy-17aα-äthyl-D-homo-5,16-pregnadien- -20-on werden in 27 ml Toluol mit 1,4 ml Cyclohexanon zum Sieden erhitzt und mit einer Lösung von 650 mg Aluminiumisopropylat in 2 ml Toluol versetzt. Das Reaktionsgemisch wird dann 45 min unter langsamem Abdestillieren erhitzt, anschliessend mit Äther verdünnt, mit 2n Schwefelsäure und Wasser gewaschen und der nach dem Eindampfen erhaltene Rückstand an Silicagel chromatographiert. Nach Umkristalli- sation aus Diisopropyläther erhält man 485 mg 17aα-Äthyl- -D-homo-4,16-pregnadien-3,20-dion vom Smp. 153-154 C.
UV: ±24s = 16 800.
Herstellung des Ausgangsmaterials:
1,9 g Kalium-t.-butylat werden in 24,5 ml Dimethylformamid und -3,75 ml tert.-Butanol gelöst. In der im Eisbad gekühlten Lösung werden unter einem Stickstoffstrom 3,0 g 3ss(Tetrahydropyran-2-yloxy)-D-homo-5,17(17a)-pregnadien -20-on gelöst Innerhalb 15 min werden dann 1,38 mi Äthyljodid zugetropft und 40 min bei Eisbadtemperatur nachgerührt. Das Reaktionsgemisch wird dann in Eiswasser eingerührt, der Niederschlag abfiltriert, gut mit Wasser ausgewaschen und in Methylenchlorid aufgenommen. Nach dem Eindampfen wird an Silicagel chromatographiert und es werden aus Methanol umkristallisiert 1,8 g 30(Tetrahydropyran-2- -yloxy > -17a5C-äthyl-D-homo-5,16-pregnadien-20-on vom Smp.
1364380C erhalten.
1,8 g 3ss(Tetrahydropyran-2-yloxy)-17aα-äthyl-D-homo- -5,16-pregnadien-20-on werden in 36 mi Methanol und 9 ml Wasser mit 300 mg Oxalsäure versetzt und 45 min am Rückfluss erhitzt. Das Reaktionsgemisch wird in Eiswasser eingerührt, der Niederschlag abfiltriert, gewaschen und getrocknet.
Es werden 1,35 g rohes 3ss-Hydroxy-17aα-äthyl-D-homo-5,16- -pregnadien-20-on erhalten.
Beispiel 8
570 mg 3!p-Hydroxy-'17as;-butyll-D-homo-5,16-pregnadien- -20-on werden in 1!1,5 ml Toluol und 0,57 ml Cyclohexanon mit einer Lösung von 280 mg Aluminiumisopropylat in 1 ml Toluol versetzt und wie in Beispiel 11 beschrieben umgesetzt und aufgearbeitet. Nach Chromatographie an Silicagel werden 355 mg 17aα-Butyl-D-homo-4,16-pregnadien-3,20-dion vom Smp. 82-850C erhalten. UV: e24l = 15 600.
Herstellung des Ausgangsmaterials:
2,5 g Kalium-t.-butylat werden in 40 ml Dimethylformamid und 8 ml tert.-Butanol gelöst. Zu der im Eisbad gekühlten Lösung werden unter einem Stickstoffstrom 2,5 g 3s(Te- trahydropyran-2-yloxy)-D-homo-5,17(17a)-pregnadien-20-on gelöst. Innerhalb 15 min werden dann 1,25 ml l-Brombutan zugetropft und 2,5 Stunden bei Eisbadtemperatur nachgerührt.
Das Reaktionsgemisch wird aufgearbeitet und der Rückstand an Silicagel chromatographiert, wobei 780 mg rohes 3,B(Tetra- hydropyran-2-yloxy)-17a-butyl-D-homo-5,16-pregnadien-20-on erhalten werden. Eine aus Methanol umkristallisierte Probe schmilzt bei 139-140 C.
750 mg rohes 3ss(Tetrahydropyran-2-yloxy)-17aα-butyl- -D-homo-5,16-pregnadien-20-on werden in 15 ml Methanol und 3,75 ml Wasser mit 375 mg Oxalsäure wie im Beispiel 11 beschrieben umgesetzt und aufgearbeitet. Es werden 570 mg rohes 3N-Hydroxy- 1 7a-buWi-D-homo-5,1 6-pregnadien-20-on erhalten.
Beispiel 9
In zu Beispiel 1, 2 oder 3 analoger Weise erhält man 17a -Äthoxy-D-homo-pregna-4,16-dien-3,20-dion, Smp. 183-185 (aus Aceton-Hexan), UV: E241 = 16 000, [α]D25 = (c = 0,1 in Dioxan).
Beispiel 10
In zu Beispiel 1, 2 oder 3 analoger Weise erhält man 21 -Fluor-17a-hydroxy-D-homo-pregna-4,16-dien-3,20-dion.

The 3ss, 17aα-dihydroxy-D-homopregna-5,16-dien-20-one is converted in acetic anhydride and triethylamine in the presence of pN, N-dimethylaminopyridine to 3ss; 17aα-diacetoxy-D- --homopregna-5, 16-dien-20-one (m.p. 179-180 C, [α] D = 2730, c = 0.1% in dioxane) and this is then partially converted with potash in methanol to 17a α-acetoxy-3ss- saponified hydroxy-D-homopregna-5,16-dien-20-one.
Example 3 a) 3ss-Hydroxy-D-homopregna-5,17-dien-20-one in DMF and THF in the presence of potassium t.-butoxide and trimethyl phosphite with oxygen to give 3ss, 17aα-dihydroxy-D- -homopregna-5,16-dien-20-one oxidized. Mp 241-243 C, [α] D = 1870 (c = 0.1% in dioxane). Oxidation according to Pfitzner-Moffatt gives 17az-hydroxy-D- homopregna-4,16-diene-3,20-dione with a melting point of 177-178 C, UV: ± 240 = 16,800. [L = + 180 (c = 0.1% in dioxane).
b> 4.65 g of 17a-hydroxy-D-homopregna-4,16-diene-3,20 -dione and 1.55 g of pN, N-dimethylaminopyridine are dissolved in 23.5 ml of triethylamine and 14.5 ml of acetic anhydride for 16 hours stirred at room temperature under argon. The mixture is poured onto ice-cold dilute hydrochloric acid and extracted with methylene chloride. The Methylenchlon.diösungen are washed neutral with dilute sodium bicarbonate solution and dilute sodium chloride solution, dried and evaporated. Chromatography of the crude product on silica gel and crystallization twice from acetone-hexane gives 17a0c-acetoxy-D-homopregna-4,16-diene-3,20-dione of melting point 223-2240C.
UV: E240 = 16,800. [D = -134 (c = 0.1% in dioxane).

In a manner analogous to Example 1, 2 or 3, 21-chloro-17a, -hydroxy-D-homopregna-4,16-diene-3,20-dione is obtained, m.p. 243-245 C (from acetone-hexane), UV: # 240 = 17,200, [α] D = = +860 (c = 0.1% in dioxane).

1.35 g of 3ss-hydroxy-17a-α-ethyl-D-homo-5,16-pregnadien-20-one are heated to boiling with 1.4 ml of cyclohexanone in 27 ml of toluene and a solution of 650 mg of aluminum isopropoxide in 2 ml of toluene are added. The reaction mixture is then heated for 45 min while slowly distilling off, then diluted with ether, washed with 2N sulfuric acid and water and the residue obtained after evaporation is chromatographed on silica gel. After recrystallization from diisopropyl ether, 485 mg of 17aα-ethyl- -D-homo-4,16-pregnadiene-3,20-dione with a melting point of 153-154 ° C. are obtained.

Production of the raw material:
1.9 g of potassium t-butoxide are dissolved in 24.5 ml of dimethylformamide and -3.75 ml of tert-butanol. In the solution cooled in an ice bath, 3.0 g of 3ss (tetrahydropyran-2-yloxy) -D-homo-5,17 (17a) -pregnadien -20-one are dissolved under a stream of nitrogen. 1.38 ml of ethyl iodide are then dissolved in the course of 15 minutes added dropwise and stirred for 40 min at ice bath temperature. Production of the raw material:
2.5 g of potassium t-butoxide are dissolved in 40 ml of dimethylformamide and 8 ml of tert-butanol. 2.5 g of 3s (tetrahydropyran-2-yloxy) -D-homo-5,17 (17a) -pregnadien-20-one are dissolved in the solution, cooled in an ice bath, under a stream of nitrogen. 1.25 ml of 1-bromobutane are then added dropwise over the course of 15 minutes and the mixture is subsequently stirred at ice bath temperature for 2.5 hours.
The reaction mixture is worked up and the residue is chromatographed on silica gel, 780 mg of crude 3, B (tetrahydropyran-2-yloxy) -17a-butyl-D-homo-5,16-pregnadien-20-one being obtained. A sample recrystallized from methanol melts at 139-140 C.
750 mg of crude 3ss (tetrahydropyran-2-yloxy) -17aα-butyl- -D-homo-5,16-pregnadien-20-one are dissolved in 15 ml of methanol and 3.75 ml of water with 375 mg of oxalic acid as in Example 11 described implemented and processed. 570 mg of crude 3N-hydroxy-17a-buWi-D-homo-5,1 6-pregnadien-20-one are obtained.
