PATENTANSPRÜCHE
1. Verfahren zur Herstellung von 6¯ 1,6¯ oder 1,2 Methylen-A6-Derivaten von D-Homosteroiden der Formel

worin R6 Wasserstoff, Fluor, Chlor oder Methyl; R17a Hydroxy, Acyloxy, Alkoxy oder nieder-Alkyl, R10 Methyl und R21 Wasserstoff, Fluor oder Chlor darstellen, oder zur Herstellung von A6-Derivaten von D-Homosteroiden der Formel I, worin Rl Wasserstoff ist, dadurch gekennzeichnet, dass man ein D-Homosteroid der Formel I, worin Rl Methyl darstellt in 1,2- und/oder 6,7 Stellung dehydriert oder ein Afi-Derivat davon in 1,2-Stellung oder ein 1,2-Methylen- oder ein Al-Derivat davon in 6,7 Stellung dehydriert, oder dass man ein D-Homosteroid der Formel I,
worin Rl Wasserstoff darstellt, in 6,7-Stellung dehydriert.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man ein A6- oder Al 6-Derivat von D-Homosteroiden der Formel I herstellt, worin Rl7a Hydroxy oder Acyloxy, R10 Methyl und R21 Wasserstoff darstellen.
3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man ein A6-Den.vat von D-Homosteroiden der Formel I herstellt, worin R6 Wasserstoff, Chlor oder Methyl, R17a Hydroxy oder C1-7-Alkanoyloxy und R21 Wasserstoff darstellen.
4. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man ein A6-Derivat von D-Homosteroiden der Formel I herstellt, worin R21 Wasserstoff, R6 Wasserstoff, Chlor oder Methyl, Rlo Methyl und R17a Cl-7-Alkanoyloxy darstellen.
5. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man 17a n-Acetoxy-6-chlor-D-homopregna-4,6, 16-hien-3,20-dion, 17aa-Hydroxy-D-homo-1 ,4,6,16-pregnatetraen-3,20-dion, 17aa-Acetoxy-6-fluor-D-homo-pregna-4,6, 16- trien-3 ,20-dion 17aa-Acetoxy-6-methyl-D-homopregna-4,6, 16 trien-3,20-dion, oder 17aa-Acetoxy-6-chlor-D-homopregna- 1,4,6, 16-tetraen-dion herstellt.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Al-, A6-, A1- oder 1,2-Methylen-A6-Derivaten von D-Homosteroiden der Formel

Eine Acyloxygruppe kann sich von einer gesättigten oder ungesättigten aliphatischen Carbonsäure, einer cycloaliphatischen, araliphatischen oder einer aromatischen Carbonsäure mit vorzugsweise bis zu 15 C-Atomen ableiten. Beispiele solcher Säuren sind, Ameisensäure, Essigsäure, Pivalinsäure, Propionsäure, Buttersäure, Capronsäure, Önanthsäure, Undecylensäure, Ölsäure, Cyclopentylpropionsäure, Cyclohexylpropionsäure, Phenylessigsäure und Benzoesäure. Besonders bevorzugt sind Cl-7-Alkanoyloxygruppen. Alkoxygruppen können gerad- oder verzweigtkettig sein, vorzugsweise enthalten sie bis zu 15 C-Atome. Besonders bevorzugt sind Niederalkoxygruppen. Niederalkylgruppen sind solche mit vorzugsweise 1-4 Kohlenstoffatomen, insbesondere Methyl und Äthyl bzw. Methoxy und Äthoxy.
Bei 6,7-gesättigten Verbindungen kann ein 6ständiger Substituent a- oder ,13-Konfiguration aufweisen, wobei die a Isomeren bevorzugt sind.
Die D-Homosteroide der Formel I können erfindungsgemäss dadurch erhalten werden, dass man ein D-Homosteroid der Formel I, worin Rl Methyl darstellt, in 1,2- und/oder 6,7 Stellung dehydriert oder ein A6-Derivat davon in 1,2-Stellung oder ein 1,2-Methylen- oder ein A1-Derivat davon in 6,7 Stellung dehydriert, oder dass man ein D-Homosteroid der Formel I, worin R10 Wasserstoff darstellt, in 6,7-Stellung dehydriert.
Die 1,2-Dehydrierung eines D-Homosteroids der Formel I kann in an sich bekannter Weise z. B. auf mikrobiologischem Wege oder mittels Dehydrierungsmitteln wie Selendioxyd, 2,3 Dichlor-5,6-dicyanobenzochinon, Chloranil, Thalliumtriazetat oder Bleitetraacetat vorgenommen werden. Geeignete Mikroorganismen für die 1,2-Dehydrierung sind beispielsweise Schizomyceten, insbesondere solche der Genera Arthrobacter, z. B. A. simplex ATCC 6946; Bacillus, z. B. B. lentus ATCC 13805 und B sphaericus ATCC 7055; Pseudomonas, z. B. P.
aeruginosa IFO 3505; Flavobacterium, z. B. F. flavescens IFO 3058; Lactobacillus, z. B. L. brevis IFO 3345 und Nocardia; z. B. N. opaca ATCC 4276.
Die Einführung einer A6-Doppelbindung in ein D-Homosteroid der Formel I kann z. B. mit einem substituierten Benzochinon, wie Chloranil [vgl. J. Am. Chem. Soc. 82, 4293 (1960); 81, 5951(1959)] oder mit 2,3-Dichlor-5,6-dicyano
benzochinon oder mit Mangandioxid [vgl. J. Am. Chem. Soc.
75, 5932 (1953)] erfolgen.
Durch Behandlung mit 2,3-Dichlor-5,6-dicyanobenzochinon oder Chloranil kann ein D-Homosteroid der Formel I direkt in die entsprechende 1 ,4,6-Trisdehydro-Verbindung übergeführt werden.
Eine bevorzugte Gruppe von A6- oder A1s6-Derivaten von D-Homosteroiden der Formel I sind die, worin Rl7a Hydroxy oder Acyloxy, R10 Methyl, R21 Wasserstoff bedeuten.
Weiterhin sind solche A6-Derivate von D-Homosteroiden der Formel I bevorzugt, worin R6 Wasserstoff, Chlor oder Methyl, R17a Hydroxy oder C1-7-Alkanoyloxy und R21 Wasserstoff darstellen und insbesondere solche worin R21 Wasserstoff, R6 Wasserstoff, Chlor oder Methyl, R10 Methyl und R17a C1-7 Alkanoyloxy darstellen.

Die erfindungsgemäss als Ausgangsmaterial verwendeten D Homosteroide können, soweit ihre Herstellung noch nicht beschrieben wurde, in Analogie zu den in den Beispielen angegebenen Methoden hergestellt werden.
Die Verfahrensprodukte wirken hormonal, insbesondere auf das endokrine System und sind durch Selektivität der Wirkung gekennzeichnet. Sie können demgemäss als hormonal wirksame Mittel, z. B. als Progestativa verwendet werden und oral oder parenteral verabreicht werden. Als Dosierungsrichtlinie kommen 0,005 mg/kg bis 0,15 mg/kg pro die in Betracht.
Die Verfahrensprodukte können als Heilmittel in Form pharmazeutischer Präparate Verwendung finden, welche sie in Mischung mit einem für die enterale, perkutane oder parenterale Applikation geeigneten organischen oder anorganischen inerten Trägermaterial, wie z. B. Wasser, Gelatine, Gummi arabicum, Milchzucker, Stärke, Magensiumsterat, Talk, pflanzliche Öle, Polyalkylenglykole, Vaseline usw. enthalten. Die pharmazeutischen Präparate können in fester Form, z. B. als Tabletten, Dragees, Suppositorien, Kapseln; in halbfester Form, z. B. als Salben; oder in flüssiger Form, z. B. als Lösungen, Suspensionen oder Emulsionen, vorliegen. Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer.
Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Beispiel 1 a) Herstellung des Ausgangsmaterials: 3ss-Hydroxy-D-homopregna-5,17-dien-20-on wird in DMF und THF in Gegenwart von Kalium-t.-butylat und Trimethylphosphit mit Sauerstoff zu 3p,17aci-Dihydroxy-D-homo- pregna-5,16-dien-20-on oxydiert. Smp. 241-2430C, [a]D = -1870 (c = 0,1% in Dioxan). Durch Oxydation nach Pfitzner-Moffatt erhält man daraus das 17aa-Hydroxy-D homopregna-4, 16-dien-3,20-dion vom Smp. 177-178 C.
UV: E240 = 16800. [cr]D = +180 (c = 0,1% in Dioxan).
4,65 g 17a a-Hydroxy-D-homopregna-4, 16-dien-3,20-dion und 1,55 g p-N,N-Dimethylaminopyridin werden in 23,5 ml Triäthylamin und 14,5 ml Essigsäureanhydrid 16 Stunden bei Raumtemperatur unter Argon gerührt. Das Gemisch wird auf eiskalte verdünnte Salzsäure gegossen und mit Methylenchlorid extrahiert. Die Methylenchloridlösungen werden mit verdünnter Natriumbicarbonatlösung und verdünnter Kochsalzlösung neutral gewaschen, getrocknet und eingedampft. Chromatographie des Rohproduktes an Kieselgel und zweimalige Kristallisation aus Aceton-Hexan gibt 17aa-Acetoxy-D-homopregna-4,16-dien-3,20-dion vom Smp. 223-2240C. UV: E240 = 16800. [a]D = -1340 (c = 0,1 % in Dioxan).
17aa-Acetoxy-D-homopregna-4, 1 6-dien-3,20-dion wird in Äther mit Essigsäureanhydrid und Perchlorsäure in 3,17aa Diacetoxy-D-homopregna-3,5,16-trien-20-on überführt und dieses anschliessend in wässeriger Essigsäure in Gegenwart von Kaliumacetat mit Chlor zu 17aa-Acetoxy-6-chlor D-homopregna-4,16-dien-3,20-dion. Smp. 199-2000C (Äther) umgesetzt.
b) Dehydrierung
620 mg 17aa-Acetoxy-6-chlor-D-homopregna-4, 16-dien- 3,20-dion und 30 mg p-Toluolsulfonsäure werden in 10 ml Dioxan und 0,6 ml o-Ameisensäureäthylester 18 Stunden bei Raumtemperatur stehengelassen. Danach wird diese Lösung innert 10 Minuten zu einer Suspension von 3,0 g Braunstein in 30 ml Eisessig und 2,4 ml Wasser getropft und 60 Minuten bei Raumtemperatur unter Argon gerührt. Das Gemisch wird filtriert und das Filtrat mit Wasser und Methylenchlorid aufgearbeitet. Nach Chromatographie an Kieselgel wird 17aa Acetoxy-6-chlor-D-homopregna-4,6,16-trien-3,20-dion vom Smp. 225-226"C (Äther) erhalten.
UV: e2ss = 22300. [a]n v -t83' (c = 0,1% in Dioxan).
Beispiel 2
4,5 g 17aa-Methyl-D-homo-4, 16-pregnadien-3,20-dion werden in 90 ml tert.-Butanol mit 5,4 g Chloranil 18 Stunden unter Rühren am Rückfluss erhitzt. Das Reaktionsgemisch wird dann im Vakuum weitgehend eingeengt, der Rückstand in Äther aufgenommen, vom nicht-umgesetzten Chloranil abfiltriert und das Filtrat mit Natriumhydrogencarbonatlösung und Wasser gewaschen. Der nach dem Eindampfen erhaltene Rückstand wird am Silicagel chromatographiert und liefert aus Diisopropyläther/Aceton umkristallisiert 2,25 g 17aa-Methyl D-homo-4,6,16-pregnatrien-3,20-dion vom Schmp. 180181,50C.
UV: ±2S4 = 26900.
Beispiel 3
10,0 g 17aa-Methyl-D-homo-4, 16-pregnadien-3,20-dion werden in 200 ml Dioxan mit 20 g 2,3-Dichlor-5,6-dicyanobenzochinon und 1 g p-Toluolsulfonsäure 18 Stunden am Rückfluss erhitzt. Das Reaktionsgemisch wird dann im Vakuum weitgehend eingeengt, der Rückstand in Äther aufgenommen, vom ungelösten Hydrochinon abfiltriert und das Filtrat mit Natriumhydrogencarbonatlösung und Wasser gewaschen. Der nach dem Eindampfen erhaltene Rückstand wird an Silicagel chromatographiert. Nach Umkristallisieren aus Diisopropyläther/Aceton werden 5,6 g 17aa-Methyl-D-homo 1,4,6,16-pregnatetraen-3,20-dion vom Schmp. 172-174,5 C erhalten.
UV: E223 = 11700, E253 = 9880, E300 = 11100.
Beispiel 4
7,6 g 17aa-Hydroxy-D-homo-4,6,16-pregnatrien-3,20-dion werden in 190 ml abs. Dioxan mit 7,6 g 2,3-Dichlor5,6-dicyano-benzochinon 18 Stunden bei 100oC gerührt. Es wird dann vom ausgeschiedenen Hydrochinon abfiltriert, gut mit Äther nachgewaschen und das Filtrat mit Natriumhydrogencarbonatlösung und Wasser gewaschen. Nach dem Eindampfen wird der Rückstand an Silicagel chromatographiert und es werden nach Umkristallisieren aus Diisopropyläther/ Aceton 4,4 g 17aa-Hydroxy-D-homo-1 4,6, 16-pregnatetraen- 3,20-dion vom Schmp. 196-198 C erhalten.

Beispiel 5
1 g 17a-Acetoxy-6-chloro-D-homopregna-4,6, 1 6-trien- 3,20-dion und 1 g Dichlorodicyanobenzochinon wurden in 60 ml Dioxan unter Argon am Rückfluss gekocht. Nach 48 Stunden wurden 500 mg Dichlorodicyanobenzochinon zugegeben und nach weiteren 10 Stunden nochmals 500 mg. Nach insgesamt 70 Stunden wurde das Reaktionsgemisch an 20facher Menge Alox II, neutral, mit Methylenchlorid-Aceton filtriert. Aus Aceton-Hexan wurden 500 mg 17a-Acetoxy-6 chloro-D-homopregna-1,4,6,16-tetraen-3,20-dion erhalten.
Smp. 198-2000Ö [a]D = -2400 (c = 0,100% in Dioxan).

An acyloxy group can be derived from a saturated or unsaturated aliphatic carboxylic acid, a cycloaliphatic, araliphatic or an aromatic carboxylic acid with preferably up to 15 C atoms. Examples of such acids are formic acid, acetic acid, pivalic acid, propionic acid, butyric acid, caproic acid, oenanthic acid, undecylenic acid, oleic acid, cyclopentylpropionic acid, cyclohexylpropionic acid, phenylacetic acid and benzoic acid. Cl-7-alkanoyloxy groups are particularly preferred. Alkoxy groups can be straight or branched chain, preferably they contain up to 15 carbon atoms. Lower alkoxy groups are particularly preferred.
