Eine Acyloxygruppe kann sich von einer gesättigten oder ungesättigten aliphatischen Carbonsäure, einer cycloaliphatischen, araliphatischen oder einer aromatischen Carbonsäure mit vorzugsweise bis zu 15 C-Atomen ableiten. Beispiele solcher Säuren sind, Ameisensäure, Essigsäure, Pivalinsäure, Pro pionsäur Buttersäure, Capronsäure, Oenanthsäure, Undecylensäure, Olsäure, Cyclopentylpropionsäure, Cyclohexylpropionsäure, Phenylessigsäure und Benzoesäure. Besonders bevorzugt sind Cs 7-Alkanoyloxygruppen. Alkyloxygruppen können gerad- oder verzweigtkettig sein, vorzugsweise mit bis zu 15 C-Atomen. Besonders bevorzugt sind Niederalkoxygruppen. Die: Hydrolyse des 3ständigen Substituenten in einem D-Homosteroid der Formel II kann in an sich bekannter Weise mit Säuren, z. B. Mineralsäuren wie Salzsäure; oder mit Carbonsäuren, wie Oxalsäure bewerkstelligt werden. Als Reaktionsmedium eignen sich insbesondere alkoholische oder wässrig-alkoholische Lösungen, z. B. Methanol oder Methanol/Wasser, das gegebenenfalls weitere Lösungsmittel, z. B. Chloroform, enthält.
Beispiele von D-Homosteroiden der Formel II sind solche, in denen X zusammen mit dem Doppelbindungssystem in den Ringen A und/oder B eine 3-Alkoxy- (z. B. Methoxy-)-h 25 < '0,-3- Alkylthio (z. B. 3-Methylthio-A 2I0); 3-sek-Amino (z. B. Pyrroli dino-)t Z5c10); 3-Alkylendioxy- (z. B. Äthylendioxy-)-d 0), A 4oder A 5-; oder eine 3-Alkylendithio- (z. B. Äthylendithio-S A Ylo oL 4- oder A 5-Gruppierung darstellt
Erwünschtenfalls kann eine erhaltene Verbindung der Formel I, worin Rl7a Hydroxy bedeutet, acyliert werden.
Die Acylierung kann in an sich bekannter Weise durch Behandlung mit einem reaktionsfähigen Säurederivat, wie z. B.
einem Acylhalogenid oder -anhydrid in Gegenwart von p-N,N Dimethylaminopyridin erreicht werden.
Beispiele von Verbindungen, die unter die allgemeine Formel I fallen sind: 17aa-Methyl-D-homo-1 9-norpregna4,16-dien-3,20-dion, 17aa-Äthyl-D-homo-19-norpregna4,16-dien-3,20-dion, 1 7aa-Methoxy-D-homo- 19-norpregna-4, 16-dien-3,20-dion.
Die erfindungsgemäss als Ausgangsmaterial verwendeten D-Homosteroide können soweit ihre Herstellung noch nicht beschrieben wurde, in Analogie zu den in den Beispielen angegebenen Methoden hergestellt werden.
Die Verbindungen der Formel I wirken hormonal, insbesondere auf das endokrine System und sind durch Selektivität der Wirkung gekennzeichnet. Sie können demgemäss als hormonal wirksame Mittel, z. B. als Progestativa verwendet werden und oral oder parenteral verabreicht werden. Als Dosierungsrichtlinie kommen 0,005 mg/kg bis 0,15 mg/kg pro die in Betracht.
Die Verbindungen der Formel I können als Heilmittel z. B.
in Form pharmazeutischer Präparate Verwendung finden, welche sie in Mischung mit einem für die enterale, perkutane oder parenterale Applikation geeigneten organischen oder anorganischen inerten Trägermaterial, wie z. B. Wasser, Gelatine, Gummi arabicum, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Polyalkylenglykole, Vaseline usw. enthalten. Die pharmazeutischen Präparate können in fester Form, z. B. als Tabletten, Dragees, Suppositorien, Kapseln; in halbfester Form, z. B. als Salben; oder in flüssiger Form, z. B. als Lösungen, Suspensionen oder Emulsionen, vorliegen. Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Beispiel 1
Eine Lösung von 4,9 g 3,3-(Äthylendioxy)-17a-hydroxy-D- homo-19-norpregna-5 (10),16-dien-20-on in 100 ml Methanol wurde mit 1,5 ml konz. Salzsäure in 10 ml Wasser versetzt und während 18 Stunden bei 25 gerührt. Es wurde durch Zugabe von wässrigem Bicarbonat neutralisiert, am Rollverdampfer von Methanol befreit und zwischen Dichlormethan und Wasser verteilt. Nach Waschen mit Wasser, Trocknen über Natriumsulfat und Eindampfen am Rollverdampfer lieferten die organischen Phasen 5 g Rohprodukt, woraus aus Dichlormethan-Äther in zwei Kristallisaten 3,4 g 17a-Hydroxy-D-homo-l9- norpregna4,16-dien-3,20-dion erhalten wurden; Smp. 219-220"; Ca]289 (Dioxan; c = 0,0099).
Herstellung des Ausgangsmaterials:
Zu einer Lösung von 27,7 ml N-Cyclohexyl-isopropylamin in
100 ml Tetrahydrofuran wurden bei -78" unter Argon 76,5 ml einer ca. 20-250/cigen Lösung von n-Butyllithium in Hexan zugerührt. 150 ml dieser Lösung wurden innert 2 Stunden unter Argon zu einer auf gekühlten Lösung von 20 g D-Homoöstronmethyläther in 100 ml Dichlormethan gerührt Nach weiteren 2 Stunden Rühren bei -78" wurde mit einer Lösung von 10 g Ammoniumchlorid in 50 ml Wasser versetzt, die wässrige Phase durch Zugabe von 1 N Salzsäure sauer gestellt und es wurde mit Dichlormethan ausgezogen. Nach Waschen mit ges.
Bicarbonatlösung und mit Wasser und Trocknen über Natriumsulfat wurde am Rollverdampfer zur Trockene eingedampft Das Rohprodukt wurde in 100 ml Toluol gelöst und während 4 Stunden am Rückfluss erwärmt, und der Eindampfrückstand an 600 g Kieselgel 0,06-0,2 mm adsorbiert; Eluierung mit Hexan-Essigester 9 1 lieferte 20,8 g (900/0)1 17a-Chloro-3methoxy-D-homoöstra-1,3,5(10) -trien-l 7ap-carboxaldehyd, Smp. 116-117" (Dichlormethan-Hexan); [a]2589 -5 (Dioxan; c = 0,101).
Eine Lösung von 20,8 g 17a-Chloro-3-methoxy-D-homoö stra-1,3,5(10)-trien-17ap- carboxaldehyd in 25 ml Hexamethylphosphorsäuretriamid wurde mit 2,5 g Lithiumchlorid versetzt und während 12 Stunden bei 50" gerührt unter portionenweiser, allmählicher Zugabe von 5 g Natriumbicarbonat. Nach Stehen über Nacht bei 25 wurde mit Äther versetzt, dreimal mit Wasser und einmal mit ges. Bicarbonat gewaschen, über Natriumsulfat getrocknet und am Rollverdampfer eingeengt. Der Rückstand wurde an 1 kg Kieselgel 0,06-0,2 mm adsorbiert; Eluierung mit Dichlormethan lieferte 10,1 g (50%) 3-Methoxy D-homoästra-1,3,5( 1 01 7- tetraen-1 7a-carboxaldehyd; Smp.
126-127" (Aceton-Äther);[a]2589 +207 (Dioxan; c = 0,100).
Zu einer Lösung von 18 g 3-Methoxy-D-homoöstra 1,3,5,(10),17-tetraen-17a-carboxaldehyd in 250 ml abs. Tetrahydrofuran wurden bei 0 unter Argon mit Rühren 45 ml einer ca.
2 molaren Lösung von Methyllithium in Äther zugetropft. Nach 2 Stunden wurde mit wässrigem Ammoniumchlorid versetzt und viermal mit Äther ausgezogen. Die organischen Phasen wurden zweimal mit Soda gewaschen, über Natriumsulfat getrocknet und am Rollverdampfer eingeengt. Das Rohprodukt wurde in 400 ml tert-Butanol und 400 ml Tetrahydrofuran gelöst und zu 1,21 wasserfreiem Ammoniak zugetropft. Die siedende Lösung wurde portionenweise mit 6,1 g Natrium versetzt. Nach 21/2 Stunden wurde der Ammoniak abdestilliert, das Reaktionsgemisch am Rollverdampfer eingeengt und zwischen Äther und Wasser verteilt. Nach Trocknen über Natriumsulfat wurde am Rollverdampfer eingeengt. Der Eindampfungsrück stand wurde in 50 ml Dichlormethan gelöst, mit 100 ml Äthylenglykol und 25 ml Eisessig versetzt und während 18 Stunden bei 25" gerührt.
Es wurde auf eiskalte 3 N Natronlauge gegossen, mit Äther ausgezogen und nach Waschen mit Wasser und Trocknen über Natriumsulfat am Rollverdampfer eingeengt.
Das Rohprodukt wurde in 100 ml Pyridin gelöst und bei 0-5" mit 200 ml einer ca. 1 molaren Lösung von Chromtrioxyd in Pyridin-Wasser 10 1 versetzt. Nach Rühren bei 25" während 4 Stunden wurde mit 5 ml Äthanol versetzt und am Rollverdampfer eingeengt. Der Rückstand wurde mit 500 ml Äther und 300 ml Wasser versetzt und über Speedex genutscht. Die organische Phase des Filtrats wurde nach Waschen mit Wasser, Trocknen über Natriumsulfat und Einengen am Rollverdampfer an 500 g Kieselgel 0,06-0,2 mm adsorbiert, wobei mit Dichlor methan +0,5-1 % Methanol 10,5 g reines 3,3-(ÄthylendioxyS D-homo-1 9-norpregna-5(1 0),1 7- dien-20-on eluiert wurden; Smp.
147-148" (Äther-Hexan); [a125589 +238" (Dioxan; c = 0,101).
Eine Lösung von 5,5 g 3,3-(Äthylendioxy)-D-homo-19- norpregna-5(10),17-dien-20-on in 25 ml Tetrahydrofuran wurde zu einem Gemisch von 15 ml tert.-Butanol,25 ml Dimethylformamid und 2,5 ml Trimethylphosphi* zugetropft. Die Lösung wurde auf gekühlt, mit 1,3 g Kalium tert-Butylat versetzt und in einer Sauerstoffatmosphäre kräftig gerührt. Innert 25 Minuten waren 360 ml Sauerstoff verbraucht. Es wurde auf Eiswasser gegossen und mit Äther ausgezogen.
Nach Waschen mit Wasser, Trocknen über Natriumsulfat und Eindampfen am Rollverdampfer wurde das Rohprodukt mit 2,3 g eines identischen Rohproduktes vereinigt und an 250 g Kieselgel 0,060,2 mm adsorbiert; Eluierung mit Hexan-Essigester 4: lieferte 4,18 g amorphes 3,3(Äthylendioxy > 1 7a-hydroxy-1 9-norpregna- 5(10), 1 6-dien-20-on.
Beispiel 2
Eine Lösung von 4 g 17a-Hydroxy-D-homo-19-norpregna- 4,16-dien-3,20-dion in 40 ml Acetanhydrid und 40 ml Triäthylamin wurde mit 0,5 g 4-Dimenthylaminopyridin versetzt und bei 25" gerührt. Nach 24 Stunden wurde auf eiskalte 1 N Salzsäure gegossen und mit Äther ausgezogen. Nach Waschen mit Wasser (zweimal), Trocknen über Natriumsulfat und Einengen am Rollverdampfer, verblieben 7 g Rohprodukt, die an 200 g Kieselgel adsorbiert wurden. Eluierung mit Dichlormethan lieferte 2,5 g 1 7a-Acetoxy-D-homo-1 9-norpregna4, 16- dien-3,20-dion; Smp. 210-212" (aus Dichlormethan-Ather-Hexan); [a]2589 -196" (Dioxan; c = 0,100).
Beispiel 3
Eine Lösung von 2,5 g 1 7a-Hydroxy-D-homo-1 9-norpregna- 4,16-dien-3,20-dion in 30 ml Capronsäureanhydrid und 30 ml Triäthylamin wurde mit 0,5 g 4-Dimethylaminopyridin versetzt und bei 25" gerührt. Nach 21 Stunden wurde auf eiskalte 1 N Salzsäure gegossen und mit Äther ausgezogen. Nach Waschen mit Wasser (dreimal), Trocknen über Natriumsulfat, Eindampfen am Rollverdampfer und Erwärmen am Vakuum (0,1 Torr) verblieben 3,3 g Rohprodukt, die an 100 g Kieselgel 0,060,2 mm adsorbiert wurden. Eluierung mit Dichlormethan lieferte 1,7 g 17a-Caproyloxy-D-homo-19-norpregna4,16- dien- 3,20-dion; UVmax (EtOH): 238/9 nm (e 17 000).

The crude product was dissolved in 400 ml of tert-butanol and 400 ml of tetrahydrofuran and added dropwise to 1.21 anhydrous ammonia. The boiling solution was mixed in portions with 6.1 g of sodium. After 21/2 hours, the ammonia was distilled off, the reaction mixture was concentrated on a roller evaporator and distributed between ether and water. After drying over sodium sulfate, the mixture was concentrated on a roller evaporator. The evaporation residue was dissolved in 50 ml of dichloromethane, mixed with 100 ml of ethylene glycol and 25 ml of glacial acetic acid and stirred at 25 "for 18 hours.
It was poured onto ice-cold 3 N sodium hydroxide solution, extracted with ether and, after washing with water and drying over sodium sulfate, concentrated on a roller evaporator.
The crude product was dissolved in 100 ml of pyridine and 200 ml of an approximately 1 molar solution of chromium trioxide in pyridine-water 10 l was added at 0-5 ". After stirring at 25" for 4 hours, 5 ml of ethanol were added and the mixture was used on a rotary evaporator constricted. The residue was mixed with 500 ml of ether and 300 ml of water and sucked over Speedex. After washing with water, drying over sodium sulfate and concentration on a rotary evaporator, the organic phase of the filtrate was adsorbed onto 500 g of silica gel 0.06-0.2 mm, 10.5 g of pure 3. A solution of 5.5 g of 3,3- (ethylenedioxy) -D-homo-19-norpregna-5 (10), 17-dien-20-one in 25 ml of tetrahydrofuran was added to a mixture of 15 ml of tert-butanol , 25 ml of dimethylformamide and 2.5 ml of trimethylphosphi * were added dropwise. The solution was cooled to, 1.3 g of potassium tert-butoxide was added and the mixture was stirred vigorously in an oxygen atmosphere. 360 ml of oxygen were consumed within 25 minutes. It was poured onto ice water and extracted with ether.
After washing with water, drying over sodium sulfate and evaporation on a rotary evaporator, the crude product was combined with 2.3 g of an identical crude product and adsorbed onto 250 g of 0.060.2 mm silica gel; Elution with hexane-ethyl acetate 4: gave 4.18 g of amorphous 3,3 (ethylenedioxy> 1 7a-hydroxy-1 9-norpregna-5 (10), 1 6-dien-20-one.
Example 2
A solution of 4 g of 17a-hydroxy-D-homo-19-norpregna-4,16-diene-3,20-dione in 40 ml of acetic anhydride and 40 ml of triethylamine was mixed with 0.5 g of 4-dimenthylaminopyridine and at 25 " After 24 hours, the mixture was poured onto ice-cold 1 N hydrochloric acid and extracted with ether After washing with water (twice), drying over sodium sulfate and concentration on a rotary evaporator, 7 g of crude product remained, which were adsorbed on 200 g of silica gel, elution with dichloromethane provided 2.5 g 1 7a-acetoxy-D-homo-1 9-norpregna4, 16-diene-3,20-dione; mp 210-212 "(from dichloromethane-ether-hexane); [a] 2589 -196 "(dioxane; c = 0.100).
Example 3
A solution of 2.5 g of 1 7a-hydroxy-D-homo-1 9-norpregna-4,16-diene-3,20-dione in 30 ml of caproic anhydride and 30 ml of triethylamine was mixed with 0.5 g of 4-dimethylaminopyridine and stirred at 25 ". After 21 hours, the mixture was poured onto ice-cold 1N hydrochloric acid and extracted with ether. After washing with water (three times), drying over sodium sulfate, evaporation on a rotary evaporator and heating in vacuo (0.1 torr) remained 3.3 g of crude product adsorbed on 100 g of silica gel 0.060.2 mm, elution with dichloromethane yielded 1.7 g of 17a-caproyloxy-D-homo-19-norpregna4,16-diene-3,20-dione; UVmax (EtOH): 238/9 nm (e 17,000).
