Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer 170c;Propadienyl-A5(l )-steroide der 19 -nor-Androstan- oder östranreihe, welches dadurch gekennzeichnet ist, dass man ein entsprechendes 17,o -Pro- padienyl-3 -oxo-Ä4- 1 9-nor-steroid mit einem Niederalkylorthoester in Gegenwart eines sauren Katalysators umsetzt und den entstandenen Enoläther mit einer Säure hydrolysiert.
Im Schweizer Patent Nr. 492689 wird ein Verfahren zur Herstellung von neuen 17X-1Dihaloggencyclopro- peny!- oder 17s-Dihalogencyclopropyl-steroiden der Oestra-1,3,5(10)-trien-, Androst-4-en- oder 19-nor-Androst-4-en-Reihe beschrieben, welches in der Umsetzung eines entsprechenden 17a;-äthinyl- oder 17,os-äthenylsub- stituierten Steroid mit einem Alkalimetallsalz einer Trihalogenessigsäure in einem wasserfreien inerten polaren organischen Lösungsmittel bei einer Temperatur oberhalb derjenigen, die die Zersetzung des Salzes verursacht, besteht.
Aus den 17ss-Dihalogencyclopropylsteroiden lassen sich die im vorliegenden Verfahren als Ausgangsprodukte verwendeten Steroide herstellen, indem man ein 172-Di- halogencyclopropylsteroid der 1 9-nor-Androstan- oder Oestranreihe in einem in bezug auf die Reaktion inerten organischen Lösungsmittel mit Magnesium, Kalium-tert.
butylat, Butyllithium oder Natriummethylsulfinylcarbanion umsetzt.

Das Verfahren wird so ausgeführt, dass ein 4-19- -nor-Steroid durch Behandlung mit einem Niederalkylorthoester, beispielsweise Methylorthoformiat und dergleichen, in Gegenwart eines sauren Katalysators in den Enoläther übergeführt wird. Der Enoläther wird dann mit Säure, beispielsweise Oxalsäure, behandelt, wobei ein Gemisch des gewünschten A5'l0)-Steroides der Formel II und des A4-19-nor-Ausgangsmaterials entsteht, das durch bekannte Verfahrensweisen, wie beispielsweise Chromatographie, getrennt werden kann.

darstellt, zeigen östrogene Wirkung und sind brauchbar zur Behandlung verschiedener Zustände, bei denen solche Mittel angezeigt sind, wie beispielsweise Östrogenmangelzustände, Klimakterium und dergleichen. Diese Verbindungen können auch in der Veterinärmedizin in gleicher Weise wie bekannte Östrogene und zur Empfängnisverhütung und Regelung der Fruchtbarkeit verwendet werden. Ausserdem zeigen diese Mittel eine Antiandrogenwirkung. Sie können in den üblichen pharmazeutischen Formen bei Dosierungen, die für den behandelten Zustand geeignet sind, verabreicht werden.

darstellt, zeigen Gelbkörperhormonwirkung und sind brauchbar zur Behandlung verschiedener Menstruationsstörungen und zur Empfängnisverhütung und Regelung der Fruchtbarkeit. Sie können in den üblichen pharmazeutischen Präparaten bei Dosierungen, die für den behandelten Zustand geeignet sind, verabreicht werden.
Herstelllmg der Aztsgaslg.vprodlette a) Zu einer Suspension von 1 g l7z-(2'2'-Dichlor- cyclopropyl)-17P-(tetrahydropyran - 2'-yloxy)-östr-4-en-3 -on in 7,5 cm3 wasserfreiem, peroxydfreiem Dioxan werden 1,2 cm3 frisch destilliertes Äthylorthoformiat und 0,8 g p-Toluolsulfonsäure gegeben. Das Gemisch wird 15 Minuten bei Raumtemperatur gerührt und weitere 30 Minuten stehen gelassen. Dann werden 0,8 cm3 Pyridin und darauf Wasser, bis Erstarrung eintritt, zugesetzt.
Der erhaltene Feststoff wird durch Filtration gesammelt, mit Wasser gewaschen und an der Luft getrocknet und ergibt 3-Äthoxy- 1 7-(2'-2'-dichlorcyclopropyl)- 17A-(tetra- hydropyran-2'-yloxy) -östra-3,5-dien.
Ein Gemisch von 1 g des oben hergestellten Enol äthers, 4 molaren Äquivalenten Natriummethylsulfinylcarbanion und 40 cm3 Dimethylsulfoxyd wird unter Stickstoff 24 Stunden bei Raumtemperatur stehen gelassen. Danach wird das Reaktionsgemisch in Wasser gegossen und mit Methylenchlorid extrahiert. Die Extrakte werden mit Wasser gewaschen, getrocknet und eingedampft und ergeben 3-Äthoxy-17x-propadienyl-17,3-(te- trahydropyran-2' -yloxy)-östra-3,5-dien.
Zu einer Lösung von 1 g der oben hergestellten 3 -Athoxy-17,x-propadienylverbindung in 10 cm3 Aceton werden einige Tropfen 36Cl0ige Salzsäure zugegeben. Das Gemisch wird einige Minuten auf Dampfbadtemperatur erhitzt, mit Wasser verdünnt und filtriert. Die so gesammelte Festsubstanz wird getrocknet und aus einem Gemisch von Aceton und Hexan umkristallisiert, wobei 17o - Propadienyl - 17p - hydroxy - östr-4-en-3-on erhalten wird.
Durch Wiederholung der vorstehenden Verfahrensweise unter Verwendung von 17α-(2',2'-Dichlorcyclopropyl)- 170-(tetrahydropyran-2'- -yloxy)-1 8-äthylöstr-4-en-3-on, 17,o:-f(2',2'-Oichlor-3'-methylcyclopropyl)-17p-(tetrahydro- pyran-2' -yloxy) -östr-4-en-3-on, 17α-(2',2'-)Dichlorcyclopropyl)- 170-(tetrahydropyran-2'- -yloxy)-18-methylöstr-4-en-3-on, 17α-(2',2'-Dichlorcyclopropyl)- 17p (tetrahydropyran-2'- -yloxy)- 18-methyl-19-norandrost-4-en-3-on oder 171o:
:-(2',2'-Dichlorcyclopropyl)-17JB-(tetrahydropyran-2'- -yloxy)-l9-nor-androst-4-en-3-on als Ausgangsmaterial wird 17-Propadienyl-178-hydroxy-18-äthylöstr-4-en-3-on, 171z(3'-lMethylpropadienyl)-17ss-hydroxyöstr-4-en-3-on, 17,sc-Propadienyl- 17,8-hydroxy- 18-methylöstr-4-en-3-on, 17α-Propadienyl-17ss-hydroxy- 1 8-methyl- l9-nor-androst- -4-en-3-on bzw.
17α-Propadienyl- 1 7p-hydroxy- 19 9-nor-androst-4-en-3-on erhalten.
b) Ein Gemisch aus 1 g 17α-(2',2'-Dichlorcyclopro- pyl) - 17 - (tetrahydropyran -2'-yloxy)-östr-4-en-3-on, 25 cm3 trockenem Benzol, 5 cm3 Äthylenglycol und 50 mg p-Toluolsulfonsäure-monohydrat wird unter Verwendung eines Wasserabscheiders 4 Stunden zum Rückfluss erhitzt. Das Reaktionsgemisch wird dann mit wässriger Natriumbicarbonatlösung und Wasser gewaschen, getrocknet und zur Trockene eingedampft, wobei 3-Äthylendioxy -17α - (2',2'-dichlorcyclopropyl)- 173-(tetrahydro- pyran-2'-yloxy)-östr-5-en erhalten wird, das aus einem Gemisch von Aceton und Hexan umkristallisiert werden kann.
Zu einer Lösung von 1 g der oben hergestellten Verbindung in 40 cm3 wasserfreiem Äther werden innerhalb eines Zeitraumes von 10 Minuten unter Rühren tropfenweise 3 molare Äquivalente Butyllithium in wasserfreiem Äther zugesetzt, während die Reaktionstemperatur auf - 350C gehalten wird. Man lässt das Reaktionsgemisch 1 Stunde bei einer Temperatur von - 350C stehen. Danach lässt man das Reaktionsgemisch auf Raumtemperatur erwärmen, giesst es in Wasser und extrahiert mit Methylenchlorid. Die Extrakte werden mit Wasser gewaschen, getrocknet und zur Trockene eingedampft und ergeben 3-Äthylendioxy-17α-propadienyl- 17p-(tetrahydro- pyran-2'-yloxy)-östr-5-en.
Ein Gemisch aus 0,5 g der oben hergestellten 17α- -Propadienylverbindung in 30 cm3 Aceton und 5û mg p-Toluolsulfonsäure lässt man 15 Stunden bei Raumtemperatur stehen. Es wird dann in Eiswasser gegossen und mit Äthylacetat extrahiert. Diese Extrakte werden mit Wasser neutral gewaschen, über Natriumsulfat getrocknet und zur Trockene eingedampft. Der Rückstand wird mit Äther verrieben und ergibt 17α-Propadienyl-17ss-hydr- oxyöstr-4-en-3-on, das aus einem Gemisch von Aceton und Hexan umkristallisiert wird.
c) Zu einer am Rüclffluss kochenden Lösung von 1 g 17 2',2'-llichlorcyclopropyl)-östr-4-en-17p-ol-3-on, 1,3 g Magnesium spänen und 25 cm3 wasserfreiem Äther werden innerhalb eines Zeitraumes von 1 Stunde 5,7 g Äthylbromid gegeben. Wenn die Zugabe beendet ist, wird das Reaktionsgemisch während einer weiteren Stunde zum Rückfluss erhitzt und dann vorsichtig durch tropfenweise Zugabe von Wasser und anschliessend von 10%iger Salzsäure hydrolysiert. Die Ätherschicht wird abgetrennt, getrocknet und eingedampft und ergibt 17α- -Propadienylöstr-4-en-17ss-ol-3-on.
In ähnlicher Weise kann das Verfahren dieses Abschnittes angewandt werden, um andere, oben beschriebene 417-Dichlorcydopropylverbindungen in die entsprechenden 17a-Propadienylderivate überzuführen.
Beispiel g 17a-lPropadienyl- 17ss-hydroxy- 1 8-methylöstr-4-en- -3-on in 50 cm3 Dioxan, die 50 cm3 Methylorthoformiat und 50 mg p-Toluolsulfonsäure enthalten, wird 1 Stunde gerührt. Dann wird 1 cm3 Pyridin zugesetzt und das Reaktionsgemisch mit Wasser auf 500 cm3 verdünnt. Der Feststoff wird gesammelt, gewaschen und getrocknet und ergibt 0,7 g gemischte Enoläther.
Eine Lösung von û,5 g der gemischten Enoläther in 30 cm3 Aceton wird mit einer Lösung von 0,2 g Oxalsäure in 10 cm3 Wasser behandelt. Nach der Reaktion wird eine Dünnschichtchromatographie vorgenommen, und wenn die Hydrolyse der Enolätherfunktion beendet ist, werden 300 cm3 Wasser, die 1 Äquivalent Natriumcarbonat enthalten, zugesetzt, und das Produkt wird durch Extraktion mit Äther isoliert. Das rohe Produkt wird durch präparative Dünnschichtchromatographie gereinigt und ergibt 17α-Propadienyl-17ss-hydroxy-18-methylöstr- -5(1 0)-en-3-on und 17α-Propadienyl-17ss-hydroxy-18-me- thylöstr-4-en-3-on.
Durch Wiederholung der vorstehenden Verfahrensweise unter Verwendung von 17α-Propadienyl-17ss-hy- droxyöstr-4-en-3-on als Ausgangsprodukt wird 17cs-Pro- padienyl-17ss-hydroxyöstr-5(10)-en-3-on erhalten.

4. Verfahren nach Patentanspruch oder einem der Unteransprüche 1 bis 3, dadurch gekennzeichnet, dass man von 17azPropadienyl- 17ss-hydroxy-18-methylöstr-4 -en-3-on ausgeht und das entsprechende A5(l0'-Steroid herstellt.
5. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass ein erhaltenes Gemisch von Verfahrensprodukt und Ausgangsmaterial durch Chromatographie aufgetrennt wird.

Production of the Aztsgaslg.Vprodlette a) To a suspension of 1 g of 17Z- (2'2'-dichlorocyclopropyl) -17P- (tetrahydropyran-2'-yloxy) -ostr-4-en-3-one in 7.5 cm3 of anhydrous, peroxide-free dioxane are added to 1.2 cm3 of freshly distilled ethyl orthoformate and 0.8 g of p-toluenesulfonic acid. The mixture is stirred for 15 minutes at room temperature and left to stand for a further 30 minutes. Then 0.8 cm3 of pyridine and then water are added until solidification occurs.
The solid obtained is collected by filtration, washed with water and air-dried to give 3-ethoxy- 1 7- (2'-2'-dichlorocyclopropyl) - 17A- (tetrahydropyran-2'-yloxy) -östra- 3,5-diene.
A mixture of 1 g of the enol ether prepared above, 4 molar equivalents of sodium methylsulfinylcarbanion and 40 cm3 of dimethylsulfoxide is left to stand under nitrogen at room temperature for 24 hours. The reaction mixture is then poured into water and extracted with methylene chloride. The extracts are washed with water, dried and evaporated and give 3-ethoxy-17x-propadienyl-17,3- (tetrahydropyran-2'-yloxy) -estra-3,5-diene.
A few drops of 36Cl0 hydrochloric acid are added to a solution of 1 g of the 3-ethoxy-17, x-propadienyl compound prepared above in 10 cm 3 of acetone. The mixture is heated to steam bath temperature for a few minutes, diluted with water and filtered. The solid substance collected in this way is dried and recrystallized from a mixture of acetone and hexane, 17o-propadienyl-17p-hydroxy-oestr-4-en-3-one being obtained.

A mixture of 0.5 g of the 17α- propadienyl compound prepared above in 30 cm3 of acetone and 5û mg of p-toluenesulfonic acid is left to stand at room temperature for 15 hours. It is then poured into ice water and extracted with ethyl acetate. These extracts are washed neutral with water, dried over sodium sulfate and evaporated to dryness. The residue is triturated with ether to give 17α-propadienyl-17ss-hydroxyostr-4-en-3-one, which is recrystallized from a mixture of acetone and hexane.
c) To a solution of 1 g of 17 2 ', 2'-llichlorcyclopropyl) oestr-4-en-17p-ol-3-one, 1.3 g of magnesium shavings and 25 cm3 of anhydrous ether are added within a period of time given 5.7 g of ethyl bromide for 1 hour. When the addition is complete, the reaction mixture is refluxed for a further hour and then carefully hydrolyzed by the dropwise addition of water and then 10% hydrochloric acid. The ether layer is separated, dried and evaporated to give 17α - -propadienylestr-4-en-17ss-ol-3-one.
Similarly, the procedure of this section can be used to convert other 417-dichlorocydopropyl compounds described above to the corresponding 17α-propadienyl derivatives.
Example g of 17a-propadienyl-17ss-hydroxy-1 8-methylestr-4-en-3-one in 50 cm 3 of dioxane, which contains 50 cm 3 of methyl orthoformate and 50 mg of p-toluenesulfonic acid, is stirred for 1 hour. Then 1 cm3 of pyridine is added and the reaction mixture is diluted to 500 cm3 with water. The solid is collected, washed and dried to give 0.7 g of mixed enol ethers.
A solution of û.5 g of the mixed enol ethers in 30 cm3 acetone is treated with a solution of 0.2 g oxalic acid in 10 cm3 water. After the reaction, thin layer chromatography is carried out, and when the hydrolysis of the enol ether function is complete, 300 cm3 of water containing 1 equivalent of sodium carbonate are added and the product is isolated by extraction with ether. The crude product is purified by preparative thin layer chromatography to give 17α-propadienyl-17ss-hydroxy-18-methylestr--5 (10) -en-3-one and 17α-propadienyl-17ss-hydroxy-18-methylestr -4-en-3-one.
By repeating the above procedure using 17α-propadienyl-17ss-hydroxyestr-4-en-3-one as the starting product, 17cs-propadienyl-17ss-hydroxyestr-5 (10) -en-3-one is obtained .
