Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen 4H-s-Tnazolo[1,5-a}[1 ,4jbenzodi- azepinen der allgemeinen Formel I,

in welcher Rl Wasserstoff oder eine Alkylgruppe mit 1 bis 3 Kohlenstoffatomen und R2 und R3 Wasserstoff, Alkylgruppen mit 1 bis 6 Kohlenstoffatomen oder Hydroxyalkylgruppen mit 2 bis 6 Kohlenstoffatomen, oder Aralkylgruppen mit 7 bis 9 Kohlenstoffatomen bedeuten, wobei niedere Alkylgruppen R2 und R3 unter sich in ss- oder -Stellung auch direkt oder über ein Sauerstoffatom, die Iminogruppe,
eine niedere Alkylamino- oder Hydroxyalkylaminogruppe mit höchstens 4 Kohlenstoffatomen zu einem zweiwertigen Rest mit insgesamt höchstens 10 Kohlenstoffatomen verbunden sein können und die Ringe A und B durch Halogen bis Atomnummer 35, Alkyl- oder Alkoxygruppen mit je 1 bis 6 Kohlenstoffatomen, Trifluormethyl- oder Nitrogruppen substituiert sein können.
Zum Gegenstand der Erfindung gehört auch die Herstellung der Additionssalze der Verbindungen der allgemeinen Formel I mit anorganischen und organischen Säuren.
In den Verbindungen der allgemeinen Formel list R1 als Alkylgruppe z. B. die Methyl-, Äthyl- oder Propylgruppe; R2 und R3 sind als Alkylgruppen mit 1 bis 6 Kohlenstoffatomen beispielsweise Äthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, Pentyl-, Isopentyl-, oder Hexylgruppen und vorzugsweise Methylgruppen, als Hydroxyalkylgruppen mit höchstens 6 Kohlenstoffatomen z. B. 2-Hydroxypropyl-, 3-Hydroxypropyl-, 2-Hydroxybutyl-, 3-Hydroxybutyl-, 2-Hydroxy- 1 -methyl- propyl-, 2-Hydroxypentyl-, 2-Hydroxyhexyl- und insbesondere 2-Hydroxyäthylgruppen, und als Aralkylgruppen mit höchstens 7 bis 9 Kohlenstoffatomen z. B. Benzyl-, Phenäthyl-, a- o-, m- oder p-Methylbenzyl-, 3-Phenylpropyl- oder o!,-Methyl- phenäthylgruppen.
Unter sich in ss- oder y-Stellung in der oben definierten Weise verbundene Alkylgruppen R2 und R3 bilden zusammen mit dem anliegenden Stickstoffatom, d. h. als Gruppierung NR2R3 z. B. die 1-Pyrrolidinyl-, Piperidino-, Hexahydro-1Hazepin-1-yl-, Morpholino-, 1-Piperazinyl- oder Hexahydro-1H 1,4-diazepin-1-ylgruppe. Die beiden letzteren Gruppen können in 4-Stellung, d. h. in der Iminogruppe z. B. durch eine Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, 2 Hydroxyäthyl-, 2-Hydroxypropyl-, 3-Hydroxypropyl- oder 3 Hydroxybutylgruppe substituiert sein, während alle vorgenannten Ringe an Kohlenstoffatomen noch durch Äthyl-, Propyl- oder insbesondere Methylgruppen substituiert sein können. Als Beispiele von tC-alkylsubstituierten bzw.
C- und N-substituierten Resten NR2R3 seien die 2,5-Dimethyl- -pyrrolidinyl-, 2-Methyl-, 3-Methyl- und 4-Methyl-piperidino-, 2,6-Dimethyl-piperidino-, 2,4,6-Trimethyl-piperidino-, 2,2,6,6-Tetramethyl-piperidino-, 2,5-Dimethyl-1-piperazinyl-, 2,4,5-Trimethyl-1-piperazinyl-, 2,4,6-Trimethyl-1-piperazinyl- und 3,4,5-Trimethyl-1-piperazinyl-gruppe genannt.
Halogenatome als Substituenten der Ringe A und B sind Fluor-, Chlor- oder Bromatome, während als Alkylgruppen bzw. Alkoxygruppen mit 1 bis 6 Kohlenstoffatomen z. B.
Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, Tert.butyl-, Pentyl-, Isopentyl-, 2,2-Dimethyl-propyl-, Hexyl- oder Isohexylgruppen bzw. Methoxy-, Äthoxy-, Propoxy-, Isopropoxy-, Butoxy-, Isobutoxy-, Pentyloxy-, Isopentyloxy-, 2,2 Dimethylpropoxy-, Hexyloxy- oder Isohexyloxy-gruppen in Betracht kommen. Ein Substituent des Ringes A befindet sich insbesondere in 8-Stellung und ist vorzugsweise Fluor, Brom, die Nitrogruppe, die Trifluormethylgruppe und vor allem Chlor. Der Ring B ist vorzugsweise unsubstituiert oder durch Fluor, Chlor oder Brom in beliebiger Stellung, insbesondere jedoch durch Fluor oder Chlor in o-Stellung, substituiert.
Die Verbindungen der allgemeinen Formel I und ihre Additionssalze mit anorganischen und organischen Säuren besitzen wertvolle pharmakologische Eigenschaften. Sie wirken zentraldämpfend, insbesondere antikonvulsiv, antiaggressiv und narkosepotenzierend. Die antikonvulsive Wirksamkeit lässt sich z. B. im Pentetrazolkrampf-Test an der Maus mit Dosen ab ca. 0,3 mg/kg per os und im Strychninkrampf-Test an der Maus mit Dosen ab ca. 1,5 mg/kg per os feststellen. Die zentral-dämpfenden, insbesondere antikonvulsiven Eigenschaften sowie weitere Wirkungsqualitäten, welche durch ausgewählte
Standardversuche [vgl. W. Theobald und H. A. Kunz, Arzneimittelforsch., 13, 122 (1963) sowie W.
Theobald et al., Arz neimittelforsch 17, 561(1967)] erfasst werden können, cha rakterisieren die Verbindungen der allgemeinen Formel I und ihre pharmazeutisch annehmbaren Additionssalze mit anorga nischen und organischen Säuren als Wirkstoffe für Psychoseda tiva (Tranquilizers) und Antikonvulsiva, die z. B. zur Behand lung von Spannungs- und Erregungszuständen sowie zur
Behandlung der Epilepsie anwendbar sind.
Die Verbindungen der allgemeinen Formel I können auch als Ausgangsstoffe für die Herstellung der entsprechenden 5
Oxide mit ähnlichen pharmakologischen Eigenschaften Ver wendung finden.
Von besonderer Bedeutung sind Verbindungen der allge meinen Formel I, in denen R1 Wasserstoff ist, R2 und R3
Wassserstoff und/oder Methyl- oder Äthylgruppen darstellen, der Ring A unsubstituiert oder vorzugsweise durch ein Halo genatom bis Atomnummer 35, die Nitro- oder Trifluormethyl gruppe substituiert ist und der Ring B entweder unsubstituiert oder durch einen der für den Ring A genannten Substituenten substituiert ist.
Besonders wertvoll sind innerhalb dieser Ver bindungsgruppe einerseits Verbindungen mit einem der oben genannten Substituenten, insbesondere einem Chloratom, im
Ring A in 8-Stellung, und anderseits solche mit unsubstituier tem oder in ortho-Stellung durch Fluor oder Chlor substituier tem Ring B und vor allem jene Verbindungen, welche die genannten Substitutionsmerkmale für die Ringe A und B auf sich vereinigen, wie das N,N-Dimethyl- und das
N,N-Diäthyl-6-phenyl-8-chlor-4H-s triazolo[1,5-a][1,4]benzodiazepin-2-carboxamid und die entsprechenden Verbindungen mit der 6-(o-fluorphe nyl)- und 6-(o-Chlorphenyl)-gruppe anstelle der 6-Phenyl gruppe,
wie das N,N-Diäthyl-6- (o-chlorphenyl)-8 -chlor-4H-s- triazolo[1 ,5-a] [1 ,4]benzodiazepin-2-carbbxamid und das N,N-Dimethyl-6-(o-fluorphenyl) -8 -chlor-4H-s- triazolo[1 ,5-a] [1 ,4]benzodiazepin-2-carboxamid.
Das erfindungsgemässe Verfahren zur Herstellung der Verbindungen der allgemeinen Formel I und ihrer Säureadditionssalze ist dadurch gekennzeichnet, dass man einen reak tionsfähigen Ester einer Verbindung der allgemeinen Formel II,

in welcher Rl, R2 und R3 die unter der Formel I angegebene Bedeutung haben, und die Ringe A und B, wie dort angegeben, substituiert sein können, mit Ammoniak oder Hexamethylentetramin umsetzt und das erhaltene Reaktionsprodukt der allgemeinen Formel I als freie Base oder als Additionssalz mit einer anorganischen oder organischen Säure isoliert. Bei der verfahrensgemässen Umsetzung wird die reaktionsfähige Estergruppe durch die Aminogruppe ersetzt; zugleich tritt unter Wasserabspaltung Ringschluss ein.
Die Umsetzung wird bei Raumtemperatur oder mässig erhöhten Temperaturen, d. h. vorzugsweise zwischen 20 und 1000C in einem inerten organischen Lösungsmittel durchgeführt. Das Ammoniak kann als solches oder in Form der konzentrierten wässrigen Lösung, vorzugsweise in grösserem Überschuss, eingesetzt und als inertes organisches Lösungsmittel z. B. Dioxan, Tetrahydrofuran, Äthanol, Butanol oder bei Verwendung von trockenem Ammoniak z. B. auch Benzol oder Toluol verwendet werden.
Als reaktionsfähige Ester von Verbindungen der allgemeinen Formel II, deren Herstellung weiter unten erläutert ist, kommen beispielsweise die Chloride und insbesondere die aus diesen vorzugsweise unmittelbar vor der Umsetzung mit Ammoniak in situ hergestellten Jodide in Betracht.
Die Bildung von Säureadditionssalzen ist weiter unten näher erläutert.
Die bereits erwähnte Verwendung der nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen der allgemeinen Formel III, in welcher R1, R2 und R3 die unter der Formel I angegebene Bedeutung haben und die Ringe A und B wie dort angegeben substituiert sein können, zur Herstellung ihrer 5-Oxide ist dadurch gekennzeichnet, dass man diese Verbindungen oxidiert. Die Oxidation vorzugsweise mit Wasserstoffperoxid oder mit einer Persäure bei einer Temperatur von ca. 0 bis 700C. Geeignete Persäuren sind z. B. Peressigsäure oder Benzopersäuren, wie Benzopersäure oder insbesondere m-Chlorbenzopersäure. Die Oxidationsmittel werden vorzugsweise in einem Lösungsmittel eingesetzt, z. B. Peressigsäure in Essigsäure und Benzopersäure in Halogenkohlenwasserstoffen, wie Methylenchlorid oder Chloroform.
Die erfindungsgemässen Verfahren zur Herstellung der Verbindungen der allgemeinen Formel I werden z. B. wie folgt erhalten:

in welcher die Ringe A und B, wie unter der Formel I angegeben, substituiert sein können. Solche Verbindungen sind in der Literatur beschrieben, z. B. das 2-Amino-5-chlorbenzophenon [vgl. F. D. Chattaway, J. Chem. Soc. 85, 344 (1904)], das 2-Amino-2',5-dichloracetophenon [vgl. L. H. Sternbach et al., J. Org. Chem. 26, 4488(1961)] sowie das 2-Amino-5chlor-21-fluorbenzophenon und weitere [vgl. L. H. Sternbach et al., J. Org. Chem. 27, 3781-3788 (1962)].
Die Verbindungen der allgemeinen Formel III werden diazotiert und anschliessend die erhaltenen Diazoniumsalze mit (2-Chloralkan amido)-malonsäure-diäthylestern, deren Chloralkanamidogruppe höchstens 5 Kohlenstoffatome aufweist, wie z. B. (2 Chlor-acetamido)-malonsäure-diäthylester [vgl. Ajay Kumar Bose, J. Indian Chem. Soc. 31, 108-110 (1954)], zu den entsprechenden (2-Chloralkanamido)-(2-benzoyl-phenylazo)malonsäure-diäthylestern gekuppelt. Dann führt man die Kupplungsprodukte durch aufeinanderfolgende Einwirkung von Natriumhydroxid und Salzsäure in die Verbindungen der allgemeinen Formel IV,

Chloroform oder Methylenchlorid, umgesetzt und nachher wird gewünschtenfalls das anstelle der Hydroxylgruppe befindliche Chloratom z. B. durch Umsetzung mit Methansulfonsäure- oder p-Toluolsulfonsäure-Natriumsalz durch eine Methansulfonyloxy- bzw. p-Toluolsulfonyloxygruppe, ferner z. B. mittels Kaliumbromid durch Brom oder, vorzugsweise unmittelbar vor der erfindungsgemässen Reaktion mit Ammoniak, mittels Kaliumjodid durch Jod ersetzt.
Die nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen der allgemeinen Formel I werden gewünschtenfalls in üblicher Weise in ihre Additionssalze mit anorganischen und organischen Säuren übergeführt. Beispielsweise verwendet man zur Salzbildung die Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Per chlorsäure, Methansulfonsäure, Äthansulfonsäure oder Citronensäure, vorzugsweise in Anwesenheit eines Lösungsmittels, wie z. B. von Aceton, Methanol, Äthanol, Äther oder deren
Gemischen.
Die Verbindungen der allgemeinen Formel I und ihre pharmazeutisch annehmbaren Säureadditionssalze werden vorzugsweise peroral oder rektal verabreicht. Die täglichen Dosen bewegen sich zwischen 0,02 und 2 mg/kg für Warmblüter.
Geeignete Doseneinheitsformen, wie Dragees, Tabletten oder
Suppositorien, enthalten vorzugsweise 0,5-50 mg eines erfin dungsgemäss herstellbaren Wirkstoffes, d. h. einer Verbindung der allgemeinen Formel I oder eines pharmazeutisch annehmbaren Säureadditionssalzes einer solchen.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht bekannten Ausgangsstoffen sowie ihre Verwendung zur Herstellung von entsprechenden 5-Oxiden näher, sollen jedoch den Umfang der Erfindung in keiner Weise beschrän ken. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
1,99 g (0,012 Mol) Kaliumjodid werden in 6 ml Wasser gelöst. Die erhaltene Lösung verdünnt man mit 20 ml Dioxan, fügt bei 250 unter Rühren eine Lösung von 4,3 g (0,01 Mol) N,N-Diäthyl-1-(2-benzoyl-4-chlorphenyl)-5 (chlormethyl)- 1H-1,2,4-triazol-3-carboxamid in 40 ml Dioxan zu und erwärmt die Reaktionslösung eine
Stunde auf 500. Dann fügt man 35 ml konz. wässrige Ammo niaklösung zu, erwärmt das Gemisch weitere 2 Stunden auf
500 und dampft es anschliessend im Vakuum ein. Zum Rück stand gibt man Wasser und extrahiert zweimal mit Methylen chlorid. Die organische Phase wäscht man mit Wasser und gesättigter Natriumchlloridlösung, trocknet sie über Natrium sulfat und dampft sie im Vakuum zur Trockne ein.
Der Rück stand wird aus Benzol-Cyclohexan umkristallisiert, wobei das reine N,N-Diäthyl-6-phenyl-8-chlor-4H-s- triazolo[1 ,5-a] [1,4] benzodiazepin-2-carboxamidlvom Smp. 175-1770 erhalten wird
In analoger Weise erhält man ausgehend von 4,65 (0,01 Mol) N,N-Diäthyl-1-[2-(o-chlor benzoyl)-4-chlorphenyl]-5-(chlormethyl)-1H- 1,2,4-triazol-
3-carboxamid das N,N-Diäthyl-6-(o-chlorphenyl)-8-chlor 4H-s-triazolo[1 ,5-aj[1 ,4]benzodiazepin-2-carboxamid vom Smp. 156-1580 (aus Isopropanol),
und ausgehend von 4,2 g (0,01 Mol) N,N-Dimethyl-1-[2- (o -fluorbenzoyl) -4-chlorphenyl] -5 - (chlormethyl) -
1H-1,2,4-triazol-3-carboxamid das N,N-Dimethyl-6 (o-fluorphenyl)-8-chlor-4H-s-triazolo[1,5-aj[1 ,4jbenzo- diazepin-2-carboxamid vom Smp. 1801820 (aus Isopropanol).
Ausgehend von 4,03 g (0,01 Mol) N,N-Dimethyl-1 [2-benzoyl-4-chlorphenyl] -5-(chlormethyl) -1H-1,2,4- triazol-3-carboxamid das N,N-Dimethyl-6-phenyl-8-chlor- 4H-s-triazolo[1,5-a][1,4]benzodiazepin-2-carboxamid vom Smp. 135-1370 (aus Methylenchlorid-Hexan).
Ausgehend von 4,38 g (0,01 Mol) N,N-Dimethyl-1-[2- (o-chlorbenzoyl) -4-chlorphenyl]-5 -(chlormethyl) - 1H- 1,2,4-triazol-3-carboxamid das N,N-Dimethyl-6-(o-chlor phenyl)-8-chlor-4H-s-triazolo[1,5-a][1,4]benzodiazepin- 2-carboxamid vom Smp. 142I450 (aus Isopropanol).
Die Ausgangsstoffe werden wie folgt hergestellt: a) Eine Lösung von 58,0 g (0,25 Mol) 2-Amino-5-chlorbenzophenon [vgl. F. D. Chattaway, J. Chem. Soc. 85, 344 (1904)] in 310 ml Eisessig-konz. Salzsäure (4:1) wird bei Raumtemperatur unter Rühren mit 50 ml (0,25 Mol) wässriger Natriumnitritlösung diazotiert. Die erhaltene Diazoniumsalzlösung versetzt man mit 150 g Eis und tropfenweise rasch mit einer Lösung von 52,4 g (0,208 Mol) (2-Chloracetamido)malonsäurediäthylester [vgl. Ajay Kumar Bose, J. Indian Chem. Soc. 31, 108-110 (1954)] in 600 ml Aceton. Anschliessend tropft man bei 5-10o im Laufe von 20 Minuten eine Lösung von 276,0 g (2 Mol) Kaliumcarbonat in 500 ml Wasser zu, rührt noch eine Stunde weiter und fügt dann Benzol und gesättigte Natriumchloridlösung zu.
Die Benzollösung wird abgetrennt, mit gesättigter Natriumchloridlösung gewaschen, über Natriumsulfat getrocknet und eingedampft. Man erhält 121 g rohen 2'-Chloracetamido-(2-benzoyl-4-chlorphenylazo)-malonsäurediäthylester, den man in 1,5 Liter Dioxan löst. Zur erhaltenen Dioxanlösung fügt man 36 g (0,9 Mol) Natriumhydroxid, gelöst in 2 Liter Wasser, rührt das Gemisch 30 Minuten und dampft dann das Dioxan im Vakuum ab. Der Rückstand wird mit 500 ml Wasser verdünnt, 20 g Aktivkohle zugefügt, das Gemisch gut umgerührt und über gereinigte Diatomeenerde filtriert. Man fügt zum Filtrat unter gutem Rühren 2-n. Salzsäure bis zur kongosauren Reaktion, saugt die ausgefallene Carbonsäure ab, wäscht mit Wasser nach und kristallisiert sie aus heissem Methanol um.
Die erhaltene 1-(2-Benzoyl-4-chlorphenyl)-5-(chlormethyl)-1H-1,2,4-triazol-3-carbonsäure sintert bei 137-1380 und schmilzt unter Zersetzung bei 169-1710. Die Kristalle enthalten eine äquimolare Menge Methanol.
In analoger Weise erhält man unter Verwendung von 66,5 g (0,25 Mol) 2-Amino-2',5dichlorbenzophenon die 1-[2-(o-Chlorbenzoyl)-4-chlor- phenyl]-5-(chlormethyl)-lH-1,2,4-triazol-3-carbonsäure, Smp. 1701750 (Zersetzung; Substanz aus Lösung in wässrigem Ammoniak ausgefällt mit 2-n. Salzsäure); unter Verwendung von 62,5 g (0,25 Mol) 2-Amino-5-chlor2'-fluorbenzophenon die 1-[2-(o-Fluorbenzoyl)-4-chlor- phenyl]-5-(chlormethyl)-lH-1,2,4-triazol-3-carbonsäure, (erstarrter Schaum); b) 11,7 g (0,031 Mol) 1-(2-Benzoyl-4-chlorphenyl) 5-(chlormethyl)-lH-1,2,4-triazol-3-carbonsäure [vgl.
Beispiel 1a)] werden mit 60 ml Oxalylchlorid übergossen und eine Stunde unter Rückfluss gekocht. Die klare, gelbe Lösung wird bei 400 im Vakuum eingedampft und der Rückstand zur totalen Entfernung des Oxalylchlorids mit 100 ml Benzol versetzt und nochmals eingedampft.
Das erhaltene, rohe 1-(2-Benzoyl-4-chlorphenyl)5-(chlormethyl)-1H-1 ,2,4-triazol-3-carbonylchlorid wird in 400 ml Dioxan gelöst und bei Raumtemperatur tropfenweise innerhalb 90 Minuten mit einer Lösung von 4,55 g (0,062 Mol) Diäthylamin in 100 ml Dioxan versetzt. Dabei fällt allmählich das Diäthylamin-hydrochlorid aus. Das Reaktionsgemisch wird im Vakuum zur Trockne eingedampft. Den Rückstand versetzt man mit Eiswasser und Äther, trennt die organische Phase ab und wäscht sie nacheinander mit kalter 1 n. Salzsäure, mit kalter 1-n. Natronlauge und mit gesättigter Natriumchloridlösung. Nach Trocknen über Natriumsulfat und Eindampfen in Vakuum erhält man das rohe N,N-Diäthyl-1-(2-benzoyl-4-chlorphenyl)-5-(chlormethyl)- 1H-1,2,4-triazol-3-carboxamid.
In analoger Weise erhält man unter Verwendung von 12,8 g (0,031 Mol) 1-[2-(o-Chlor benzoyl)-4-chlorphenyl]-5-(chlormethyl)-1H- 1,2,4-triazol- 3-carbonsäure und 4,55 g (0,062 Mol) Diäthylamin das N,N-Diäthyl-1-[2-(o-chlorbenzoyl)-4-chlorphenyl]-5- (chlormethyl)-lH-1,2,4-triazol-3-carboxamid, und unter Verwendung von 12,26 g (0,031 Mol) 1-[2-(o-Fluor benzoyl) -4-chlorphenyl]-5 -(chlormethyl)-1H-l ,2,4-triazol-
3-carbonsäure und 2,8 g (0,062 Mol) Dimethylamin das N,N-Dimethyl-1-[2-(o-fluorbenzoyl)-4-chlorphenyl]-5- (chlormethyl)-lH-1,2,4-triazol-3-carboxamid, unter Verwendung von 11,66 g (0,031 Mol) 1-[2-Benzoyl 4-chlorphenyl]-5-(chlormethyl)-lH-1,2,4-triazol-3- carbonsäure und 2,8 g (0,062 Mol) Dimethylamin das

Beispiel 2
Eine Lösung von 4,31 g (0,01 Mol) N,N-Diäthyl-1 (2-benzoyl-4-chlorphenyl)-5-(chlormethyl)-lH-1,2,4-triazol
3-carboxamid [vgl. Beispiel la) und b)] und 2,80 g (0,02 Mol)
Hexamethylentetramin in 120 ml Athanol wird 10 Stunden unter Rückfluss gekocht. Dann wird die Lösung bei 40O im
Vakuum eingedampft, der Rückstand mit 100 ml Eiswasser versetzt und zweimal mit Methylenchlorid extrahiert. Die organische Phase wird zweimal mit kalter 1-n. Salzsäure und zweimal mit Wasser gewaschen, über Natriumsulfat getrock net und im Vakuum eingedampft. Den Rückstand kristallisiert man aus Benzol-Cyclohexan um, wonach das reine N,N-Diäthyl-6-phenyl-8-chlor-4H-s- triazolo[1 ,5-a][1 ,4]benzodiazepin-2-carboxamid bei 175-1770 schmilzt.
In analoger Weise erhält man ausgehend von 2,32 g (0,005 Mol) N,N-Diäthyl-1-[2-(o-chlorbenzoyl)-4-chlor- phenyl-5-(chlormethyl)-1H- 1,2,4-triazol-3-carboxamid [vgl. Beispiel la und b)] das N,N-Diäthyl-6-(o-chlor phenyl)-8-chlor-4H-s-triazolo[1 ,5-a][ 1 ,4]benzodiazepin-
2-carboxamid vom Smp. 156-1580 (Isopropanol) und ausgehend von 2,1 g (0,005 Mol) N,N-Dimethyl-1-[2-(o- fluorbenzoyl)-4-chlorphenyl]-5-(chlormethyl)-lH-1,2,4- triazol-3-carboxamid [vgl. Beispiel la) und b)] das N,N-Dimethyl-6-(o-fluorphenyl)-8 -chlor-4H-s-triazolo- [1,5-a][1,4jbenzodiazepin-2-carboxamid vom Smp. 180-1820 (aus Isopropanol).
Beispiel 3
Zu einer Lösung von 1,0 g (0,0026 Mol) N,N-Dimethyl 6-(o-fluorphenyl)-8-chlor-4H-s-triazolo[1,5-a][1,4]-benzo- diazepin-2-carboxamid in 40 ml Methylenchlorid wird bei 0O eine Lösung von 0,68 g (0,0039 Mol) 75%ige m-Chlorperbenzoesäure in 20 ml Methylenchlorid innerhalb 5 Minuten zugetropft. Das klare Reaktionsgemisch wird 3 Stunden bei 0 5 und danach ca. 14 Stunden bei Raumtemperatur gerührt.
Dann dampft man die Reaktionslösung im Vakuum zur Trockne ein, löst den Rückstand in wenig Methylenchlorid und gibt Äther zu. Die ausgefallenen Kristalle werden abfiltriert und aus Isopropanol umkristallisiert. Man erhält das N,N-Dimethyl-6-(o-fluorphenyl)-8-chlor-4H-s- triazolo[1 ,5-a] [1,4]benzodiazepin-2-carboxamid-5-oxid vom Smp. 160-1640 (unzer Zersetzung).
In ganz analoger Weise können die 5-Oxide der weiteren Endstoffe der Beispiele 1-2 hergestellt werden.

The starting materials are prepared as follows: a) A solution of 58.0 g (0.25 mol) of 2-amino-5-chlorobenzophenone [cf. F. D. Chattaway, J. Chem. In an analogous manner, using 66.5 g (0.25 mol) of 2-amino-2 ', 5-dichlorobenzophenone, the 1- [2- (o-chlorobenzoyl) -4-chlorophenyl] -5- (chloromethyl) -lH-1,2,4-triazole-3-carboxylic acid, melting point 1701750 (decomposition; substance from solution in aqueous ammonia precipitated with 2N hydrochloric acid); using 62.5 g (0.25 mol) of 2-amino-5-chloro-2'-fluorobenzophenone 1- [2- (o-fluorobenzoyl) -4-chlorophenyl] -5- (chloromethyl) -lH- 1,2,4-triazole-3-carboxylic acid, (solidified foam); b) 11.7 g (0.031 mol) of 1- (2-benzoyl-4-chlorophenyl) 5- (chloromethyl) -1H-1,2,4-triazole-3-carboxylic acid [cf.
Example 1a)] are poured over with 60 ml of oxalyl chloride and refluxed for one hour. The clear, yellow solution is evaporated at 400 in a vacuum and 100 ml of benzene are added to the residue to completely remove the oxalyl chloride and the mixture is evaporated again.

Evaporated in vacuo, the residue mixed with 100 ml of ice water and extracted twice with methylene chloride. The organic phase is washed twice with cold 1-n. Hydrochloric acid and washed twice with water, getrock net over sodium sulfate and evaporated in vacuo. The residue is recrystallized from benzene-cyclohexane, after which the pure N, N-diethyl-6-phenyl-8-chloro-4H-s-triazolo [1,5-a] [1,4] benzodiazepine-2-carboxamide is added 175-1770 melts.
In an analogous manner, starting from 2.32 g (0.005 mol) of N, N-diethyl-1- [2- (o-chlorobenzoyl) -4-chlorophenyl-5- (chloromethyl) -1H-1,2, 4-triazole-3-carboxamide [cf. Example la and b)] the N, N-diethyl-6- (o-chlorophenyl) -8-chloro-4H-s-triazolo [1, 5-a] [1, 4] benzodiazepine
2-carboxamide with a melting point of 156-1580 (isopropanol) and starting from 2.1 g (0.005 mol) of N, N-dimethyl-1- [2- (o-fluorobenzoyl) -4-chlorophenyl] -5- (chloromethyl) -lH-1,2,4-triazole-3-carboxamide [cf. Example la) and b)] the N, N-dimethyl-6- (o-fluorophenyl) -8-chloro-4H-s-triazolo- [1,5-a] [1,4-benzodiazepine-2-carboxamide of mp. 180-1820 (from isopropanol).
Example 3
To a solution of 1.0 g (0.0026 mol) of N, N-dimethyl 6- (o-fluorophenyl) -8-chloro-4H-s-triazolo [1,5-a] [1,4] -benzo - Diazepine-2-carboxamide in 40 ml of methylene chloride is added dropwise at 0O a solution of 0.68 g (0.0039 mol) of 75% m-chloroperbenzoic acid in 20 ml of methylene chloride within 5 minutes. The clear reaction mixture is stirred for 3 hours at 0.5 and then for about 14 hours at room temperature.
The reaction solution is then evaporated to dryness in vacuo, the residue is dissolved in a little methylene chloride and ether is added. The precipitated crystals are filtered off and recrystallized from isopropanol. The N, N-dimethyl-6- (o-fluorophenyl) -8-chloro-4H-s-triazolo [1, 5-a] [1,4] benzodiazepine-2-carboxamide-5-oxide of mp. 160-1640 (undecomposition).
The 5-oxides of the other end products of Examples 1-2 can be prepared in a completely analogous manner.
