Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Aryloxy- und Arylthio-alkansäu- ren, deren Salzen und funktionellen Derivaten.

Die Verbindungen der allgemeinen Formel I, wie z.B.
die 2-(6,7,8,9-Tetrahydro-dibenzofuran-2-yloxy)-octansäure, die 2-(6,7,8,9-Tetrahydro-dibenzofuran-2-yloxy)-dodecansäure, die 2-(6,7,8,9-Tetrahydro-dibenzofuran-3 -yloxy)-octansäure, die 2-(6,7,8,9-Tetrahydro-dibenzofuran-3-yloxy)-dodecan- säure, die 2-(6,7,8,9-Tetrahydro-dibenzofuran-3-ylthio)-octan- säure, die 2-(6,7,8,9-Tetrahydro-dibenzofuran-3 -ylthio)-dodecansäure, die 2-(6,7,8,9-Tetrahydro-dibenzofuran-2-ylthio)-octansäure, die 246,7,8,9-Tetrahydro-dibenzothiophen-3-yloxy)-octan- säure, die 2-(6,7,8,9-Tetrahydro-dibenzothiophen -3-ylthio)-octan- säure und die 2-(6,7,8,9-Tetrahydro-dibenzothiophen-2-ylthio)-octan- säure,
besitzen wertvolle pharmakologische Eigenschaften und einen hohen therapeutischen Index. Sie zeigen bei perora ler Verabreichung insbesondere hypolipämische Wirksamkeit, die sich z.B. an der Senkung des Cholesterin- und Triglyceridspiegels in Blut und Leber bei mehrmaliger Verabreichung in Dosen von 2mal 10 mg/kg pro die an männlichen Ratten nach Standardmethoden nachweisen lässt. Das Gesamtcholesterin wird nach R. Richterich und K. Lauber [vgl. Klin. Wochenschrift 40, 1252-1256 (1962)] direkt im Serum bestimmt. Ferner werden nach J. Folch et al. tvgl.J.Biol.Chem.226, 497 (1957)] Serumsowie Leberlipide extrahiert und Triglyceride und Gesamtcholesterin mit dem Autoanalyser nach G. Kessler und H. Lederer [vgl.
Automation in der Analytischen Chemie (1965), Technicon GmbH, Frankfurt/Main, Seite 863-872, bzw. W.D. Block et al., ibid. Seite 970-971] bestimmt.
Die neuen Verbindungen zeichnen sich durch eine im Vergleich zur hypolipämischen Wirksamkeit nur geringe hepatomegale Wirkung aus.
In den Verbindungen der allgemeinen Formel list R1 als Alkylgruppe mit höchstens 14 Kohlenstoffatomen z.B.
die Methyl-, Äthyl-, Propyl-, Butyl-, Isobutyl-, Pentyl-, Isopentyl-, 2,2-Dimethyl-propyl-, Hexyl-, Isohexyl-, 3,3 -Dimethyl-butyl-, Heptyl-, Nonyl-, Decyl-, Undecyl-, Dodecyl-, Tridecyl- oder Tetradecylgruppe und als Cycloal Alkylgruppe mit 5-7 Kohlenstoffatomen z.B. die Cyclopentyl-, Cyclohexyl- oder Cycloheptylgruppe.
Verbindungen der allgemeinen Formel I werden erfindungsgemäss hergestellt, indem man eine Verbindung der allgemeinen Formel II

in welcher Rl, R3, X und Y die unter der allgemeinen Formel I angegebene Bedeutung haben, bis zur Abspaltung der äquimolaren Menge Kohlendioxyd erhitzt, und gewünschtenfalls eine erhaltene unter die allgemeine Formel I fallende freie Carbonsäure in ein Alkali- oder Erdalkalimetallsalz überführt.
Die Verbindungen der allgemeinen Formel II können zur Abspaltung von Kohlendioxyd entweder in Substanz oder aber auch in einem inerten Lösungsmittel, beispielsweise Toluol oder Xylol, auf Temperaturen von 100-1500 erhitzt werden. Die Verbindungen der allgemeinen Formel II können z.B. dargestellt werden, indem man Ester der Malonsäure oder der Malonamidsäure vollständig oder partiell verseift. Die Malonamidsäure kann durch Anlagerung von Wasser an die Nitrilgruppe der entsprechenden Cyanessigsäureester und unmittelbar anschliessende Verseifung der Estergruppe erhalten werden.
Zur Herstellung von Alkalisalzen der Carbonsäuren der allgemeinen Formel I geht man vorzugsweise von sauren Salzen der unter die allgemeine Formel II fallenden Ma lonsäuren aus, erhitzt diese bis zur Abspaltung der äquimolaren Menge Kohlendioxyd und setzt gewünschtenfalls aus dem erhaltenen Salz die unter die allgemeine Formel I fallende Carbonsäure frei.
Die als Zwischenprodukte benötigten 6,7,8,9-Tetrahy dro-dibenzofuran-2-yloxy-, 6,7,8,9-Tetrahydro-dibenzofuran-3-yloxy-, 6,7, 8,9-Tetrahydro-dibenzofuran-2-ylthio-, 6,7,8,9-Tetrahydro-dibenzofuran-3 - ylthio-, 6,7,8,9-Tetra hydro-dibenzothiophen-2-yloxy-, 6,7,8,9- Tetrahydro-dibenzothiophen-3-yloxy-, 6,7,8,9-Tetrahydro-dibenzothiophen-2-ylthio- und 6,7,8,9-Tetrahydro-dibenzothiophen -3 -ylthio-malonsäureester, -malonsäurenitrile und -cyanessigsäurealkylester sind ihrerseits neue Verbindungen.
Sie können beispielsweise durch Umsetzung von Alkalisalzen von Verbindungen der allgemeinen Formel III, nämlich des 6,7,8,9-Tetrahydro-dibenzofuran-2-ols, des 6,7,8,9-Te trahydro-dibenzofuran-3 -ols, des 6,7,8,9-Tetrahydro-dibenzofuran-2-thiols, des 6,7,8,9-Tetrahydro-dibenzofuran-3-thiols, des 6,7,8,9-Tetrahydrodibenzothiophen-2- -ols, des 6,7,8,9-Tetrahydro-dibenzothiophen-3-ols, des 6,7,8,9-Tetrahydro-dibenzothiophen-2-thiols bzw. des 6, 7,8,9-Tetrahydro-dibenzothiophen-3-thiols, mit Rl-substi- tuierten Brommalonsäure-dialkylestern, Brom-cyanessigsäurealkylestern oder Brommalonsäurenitrilen erhalten werden. Von den genannten Bromverbindungen sind einige bekannt, zum Beispiel der Brompropyl-malonsäure diäthylester [vgl. A. W.
Dox und L. Joder, J. Am. Chem.
Soc. 44, 1578-1581(1922)].
Weitere Verbindungen von diesem Typus können analog hergestellt werden.
Die als Ausgangsmaterialien zu verwendenden Phenole und Thiophenole, nämlich das 6,7,8,9-Tetrahydro-dibenzofuran-2-ol, das 6,7,8,9-Tetrahydro-dibenzofuran3ol, das 6,7,8,9-Tetrahydro-dibenzofuran-2,,thiol, das 6,7, 8,9-Tetrahydro-dibenzofuran3thiol, das 6,7,8,9-Tetrahydro-dibenzothiophen-2-ol, das 6,7,8,9-Tetrahydro-dibenzothiophen-3-ol, das 6,7,8,9-Tetrahydro-dibenzothiophen-2-thiol und das 6,7,8,9-Tetrahydro-dibenzothiophen-3-thiol der allgemeinen Formel III

können nach verschiedenen Methoden erhalten werden.
Beispielsweise lässt sich das 6,7,8,9-Tetrahydro-dibenzofuran-2-ol in einfacher Weise darstellen, indem man 1 -Morpholino-cyclohexen-(1) und p-Benzochinon bei Raumtemperatur in Methylenchiorid umsetzt und das zunächst erhaltene 5a, 6, 7, 8, 9, 9a-Hexahydro-5a-mor- pholino-dibenzo-furan-2-ol durch Kochen in wässriger Salzsäure zum 6,7,8,9-Tetrahydro--dibenzofuran-2-ol und Morpholin-hydrochlorid spaltet. [Vgl. G. Domschke, J.
Prakt. Chem. 32, 144-157 (1966)].
Eine weitere Möglichkeit zur Herstellung des 6,7,8,9 -Tetrahydro-dibenzofuran-2-ols, die gleichzeitig die Herstellung des 6,7,8,9-Tetrahydro- dibenzofuran-3 -ols einschliesst, besteht darin, dass man 2-Chlor-cyclohexanon bzw. 2-Bromcyclohexanon mit einem Alkalimetallsalz des Hydrochinon-monomethyläthers bzw. Resorcin-monomethyläthers umsetzt und das zunächst erhaltene 2-(4-Meth oxy-,phenoxy)-cyclolhexanon bzw. das 2-(3-Methoxy -phenoxy)-cyclohexanon anschliessend in Gegenwart eines 'sauren Katalysators, wie z.B.
Phosphorsäure oder Schwefelsäure, in das 2-Methoxy-6,7,8 ,9-tetrahydro - bzw. das 3 -Methoxy- 6,7, 8,9-tetrahydro-dibenzofuran überführt und anschliessend die Methylgruppe abspaltet. Die Abspaltung der Methylgruppe kann z.B. durch Kochen der Substanzen in einer Mischung von konz. Bromwasserstoffsäure und Eisessig oder durch Erhitzen mit Pyridin-hydrochlorid vorgenommen werden.
Das 6,7,8,9-Tetrahydro-dibenzofuran-2-thiol sowie das 6,7,8,9-Tetrahydro-dibenzofuran-3-thiol können in einfacher Weise ausgehend von den entsprechenden 2- bzw.
3-Hydroxyverbindungen erhalten werden, indem man diese mit einem N,N-Dialkyl4hiocarbaminsäurechlorid umsetzt, und die in 2- bzw. 3-Stellung vorliegende N,N-Dialkyl-thiocarbamoyloxygruppe zur N > N-Dialkyl-carbamoyl- thiogruppe umlagert und anschliessend hydrolysiert. Die Umlagerung wird zweclcnässig durch mehrstündiges Erhitzen der Substanzen auf Temperaturen von 250-3000 vorgenommen [Vgl. auch M.S. Newman und H.A. Karnes J. Org. Chem. 31, 3980-3984, (1966)].
Zur.Darstellung des 6,7,8,9-Tetrahydro-dibenzothiophen-2-ols und des 6,7,8,9-Tetrahydro-dibenzothiophen- -3-ols setzt man beispielsweise 2-Chlor- bzw. 2-Brom-cy- clohexanon mit einem Alkalisalz des SMethoxy- bzw. 3 -Methoxy-thiophenols zum 2z(4-Methoxyphenylthio)-cy- clohexanon bzw. 2-(3 -Methoxy-phenylthio) cyclohexanon um und führt diese Verbindungen durch anschliessenden Ringschiuss mit Phosphorsäure und Ätherspaltung mit Pyridinhydrochlorid in das 6,7,8 ,9-Tetrahydro-dibenzo- thiophen-2- bzw. -3-ol über.
Aus diesen Verbindungen können schliesslich das 6,7,8,9-Tetrahydro-dibenzothiophen-2-thiol und das 6,7,8,9-Tetrahydro-dibenzo--thio- phen-3-thiol wieder durch Umsetzung mit N,N-Dialkyl -thiocarbaminsäurechlorid, Umlagerung der in 2- bzw. 3 Stellung vorliegenden N,N-Dialkyl-tbiocarbamoyioxy- gruppe in die N,N-Dialkyl-carbamoyithiogruppe und anschliessende Hydrolyse erhalten werden. Die von der allgemeinen Formel III mitumfassten, als Ausgangs stoffe benötigten Phenole und Thiophenole der allgemeinen Formel lila und Imid,

in welchen X und Y die unter der allgemeinen Formel I angegebene Bedeutung haben, sowie das 6,7,8,9-Tetrahydro-dibenzofuran-2-thiol sind bisher nicht bekannt geworden.
Als Alkali- und Erdalkalimetallsalze von unter die allgemeine Formel I fallenden Carbonsäuren kommen beispielsweise deren Natrium-, Kalium-, Lithium-, Magnesium- und Calciumsalze in Frage. Die Herstellung dieser Salze erfolgt beispielsweise durch Zusammengeben von Säure und Base in einem geeigneten Lösungsmittel, wie z.B. Methanol, Äthanol oder Aceton-Wasser. Entstandene, relativ schwer lösliche Salze können durch Abfiltrieren isoliert werden, leichtlösliche durch Eindampfen des Lösungsmittels. Ferner lassen sich Salze, die im verwendeten Lösungsmittel relativ schwer löslich sind, auch durch doppelte Umsetzung eines andern Salzes der Säure mit der Base oder einem geeigneten Salz derselben herstellen.
Die Verbindungen der allgemeinen Formel I und die Alkali- und Erdalkalimetallsalze der unter diese Formel fallenden freien Garbonsäuren werden, wie weiter vorne erwähnt, peroral oder parenteral verabreicht. Die täglichen Dosen bewegen sich zwischen 0,5-10 mg/kg für Warmblüter. Geeignete Doseneinheitsformen, wie Dragees, Tabletten, Suppositorien und Kapseln, enthalten als Wirkstoff vorzugsweise 10-250 mg, zum Beispiel 50 oder 10(} mg einer Verbindung der allgemeinen Formel I oder eines Alkali- oder Erdalkalimetallsalzes einer von der allgemeinen Formel I umfassten freien Carbonsäure.
Die nachfolgenden Beispiele erläutern die Herstellung der Verbindungen der allgemeinen Formel I und von Salzen derselben näher, sollen jedoch den Umfang der Erfindung in keiner Weise beschränken. Die Temperaturen sind in Celsiusgraden angegeben. Der Ausdruck mMol bedeutet Millimol = 0,001 Mol. Bei der Benennung der hergestellten Verbindungen werden Alkylreste, die von der normalen, unverzweigten Kette abweichen, durch Be zeichnungen wie sek.-, tertiär- oder iso-A+lky! gekennzeichnet. Fehlen diese Bezeichnungen, so ist stets der normale, unverzweigte Rest gemeint.
Beispiel I
3,4 g (9,0 mMol) 2-(6,7,8,9-Tetrahydrodibenzofuran -2-yloxy)-2-hexyl-malonsäure werden in 34 ml Xylol 1% Stunde unter Rückfluss gekocht. Nach dem Abkühlen wird das Reaktionsgemisch im Vakuum vollständig enge- dampft. Der so erhaltene, ölige Rückstand wird zweimal aus Hexan umkristallisiert. Man erhält die reine 2-(6,7,8.9 -Tetrahydroibenzofuran-2-yloxy)-octansäure in Form weisser Nadeln vom Smp. 99-100 .
Analog erhält man aus 1,20 g (ca. 4,0 mMol) 2-(6,7,8,9-Tetrahydro-dibenzo furan-2-yloxy)-2-methyl-malonsäure die 2-(6,7.8,9-Tetra- hydro-dibenzofuran-2-yloxy) -propionsäure vom Smp.
128-129 (aus Methanol-Wasser); aus 1,45 g (ca. 3,0 mMol) 2-(6,7,8,9-Tetrahydro-dibenzofuran-2-yloxy)-2-tetradecyl-malonsäure die 2-(6.7,8,9-Te- trahydro - dibenzofuran - 2 - yloxy) - hexadecansäure vom Smp. 55-56 (aus Hexan); aus 2,25 g (6,0 mMol) 2-(6,7,8,9-Tetrahydro-dibenzofu- ran-3-yloxy)-2-hexyl-malonsäure die 2-(6,7,8,9-Tetrahydro-dibenzofuran-3-yloxy)-octansäure vom Smp. 78-79 (aus Hexan); aus 1,95 g (5,0 mMol) 2-(6,7,8,9-Tetrahydrodibenzofu- ran-2-ylthio)-2-hexyl-malonsäure die 2-(6,7,8,9-Tetrahydro-dibenzofuran-2-ylthio)-octansäure vom Smp. 92-93 (aus Hexan);
aus 1,95 g (5,0 mMol) 2-(6,7,8,9-Tetrahydro-dibenzofu- ran3-ylthio)-2-hexyl-malonsäure die 2-(6,7,8,9-Tetrahy dro-dibenzofuran-3-ylthio)-octansäure vom Smp. 62-63 (aus Hexan); aus 2,23 g (5,0 mMol) 2-(6,7,8,9-Tetrahydro-dibenzofu- ran-3-ylthio)-2-decyl-malonsäure die 2-(6,7,8,9-Tetrahy- dro-dibenzofuran-3-ylthio)-dodecansäure vom Smp. 73,574,50 (aus Hexan).
Die als Ausgangsmaterial verwendete 2-(6,7, 8,9-Te- trahydro-dibenzofuran-2-yloxy)-2-hexyl-malonsäure kann wie folgt erhalten werden: a) 4,0 g (9,3 mMol) 2-(6,7,8,9-Tetrahydroqiibenzofu- ran-2-yloxy)-2-hexyl-malonsäure-diäthylester werden in einer Lösung von 2,5 g Kaliumhydroxid in 10 ml Methanol und 1 ml Wasser 20 Stunden unter Rückfluss gekocht.
Nun wird das Reaktionsgemisch mit 100 ml Wasser verdünnt, zweimal mit wenig Äther gewaschen, die wässrige Phase mit eiskalter konzentrierter Salzsäure angesäuert und das dabei ausfallende öl mit Äther extrahiert. Die so erhaltene ätherische Lösung wird mit Wasser neutralgewaschen, über Magnesiumsulfat getrocknet und im Vakuum schonend eingedampft. Nach Trocknen im Hochvakuum erhält man die rohe 2-(6,7,8,9-Tetrahydro-dibenzofuran-2-yloxy)-2-hexylmalonsäure, die von sehr wenig (ca. 2%) 2-(6,7,8,9-Tetrahydrodibenzofuran-2-yloxy)-oc- tansäure verunreinigt ist. Die Malonsäure wird als zähes, gelbes öl erhalten, das nicht kristallisiert und ohne weitere Reinigung umgesetzt wird.
Analog erhält man aus den entsprechenden Diäthyll- esten die 2-(6,7,8,9-Tetrahydro-dibenzofuran-2-yloxy)-2-me- thyl-malonsäure, die 2-(6,7,8,9-Tetrahydro-dibenzofuran-2-yloxy)-2-tetra decyl-malonsäure, die 2-(6,7,8,9-Tetrahydro-dibenzofuran-3-yloxy)-2-hexyl- -malonsäure, die 2-(6,7,8,9-Tetrahydro-dibenzofuran-2-ylthio)-2-hexyl- smalonsäure, die 2-(6,7,8,9-Tetrahydro-dibenzofuran-3-ylthio)-2-hexyl- -malonsäure und die 246,7,8,9-Tetrahydro-dibenzofuran-3-ylthio)-2-decyl- -malonsäure, welche als Rohprodukte weiter verwendet werden können.
Der als Ausgangsstoff verwendete 2-(6,7,8,9-Tetrahy dro < 1ibenzofuran- 2 - yloxy) - 2- hexyl-malonsäure-d äthyl ester kann, ausgehend von 2-Hexyl-malonsäure-diäthyl ester, wie folgt dargestellt werden: ib) aus 152,7 g (0,6 Mol) 2-Hexyl-malonsäure-diäthyl ester erhält man durch Bromierung mit 96.0 g (1,2 Mol)
Brom den 2-Brom-2-hexyl-malonsäure-diäthylester als farbloses öl. Kp.o,s 107,5-108 ; nD20 : 1,4552.
Analog wird der 2-Brom-2-methyl-malonsäure-di äthylester, der 2-Brom-2-decyl-malonsäure-diäthylester und der 2-Brom-2-tetradecyl-malonsäure-diäthylester her gestellt.
c) In einem Rundkolben mit Rückflusskühler, Tropf trichter, Kaliumhydroxid-Trockenrohr, Rührer und Gas einleitungsrohr fügt man 18,83 g (0,1 Mol) 6,7,8,9-Tetra hydrodibenzofuan-2-ol zu einer Lösung von 2,3 g (0,1
Mol) Natrium in 200 ml abs. Äthanol unter Stickstoff. Zu der so erhaltenen heilbraunen Lösung des Natnumpheno lats tropft man unter Rühren 32,3 g (0,1 Mol) 2-Brom-2 -hexyl-malonsäure < Iiäthylester und kocht 12 Stunden un ter Rückfluss. Nach dem Abkühlen nutscht man das aus gefallene Natriumbromid ab und dampft das Filtrat im
Vakuum ein.
Der so erhaltene braune ölige Rückstand wird in Äther aufgenommen, die ätherische Lösung mit
Wasser neutral gewaschen, mit Magnesiumsulfat getrock net und erneut eingedampft, wobei man ein gelbes öl er hält. Dieses öl reinigt man durch Säulenchromatographie (Kieselgel 0,05-0,2 mm, Merck, Lösungsmittel Benzol).
Die den gewünschten Ester enthaltenden Fraktionen wer den vereinigt und eingedampft. Nach Trocknen im Hoch vakuum erhält man den reinen 246,7,8,9-Tetrahydro-di- benofuran-2-yloxy)-2-hexyl-malonsäure-diäthylester als blassgelbes öl; nD22 :1,51l4.
Analog erhält man aus 1,88 g (10,0 mMol) 6,7,8,9-Tetrahydro-dsbenzofuran- -2 < > 1 und 2,53 g (10,0 mMol) 2-Brom-2-methyl-malonsäu rewdiäthylester den 2-(6,7,8,9-Tetrahydro-dibenzofuran-2- -yloxy)-2-methyl-malonsäure-diäthylester, nD20 :
1,5285; aus 1,88 g (10,0 mMol) 6,7,8,9-Tetrahydrbdibenzofuran- -2-ol und 4,35 g (10.0 mMol) 2-Brom-2-tetradecyl-malon säure-diäthylester den 2-(6,7,8,9-Tetrahydro-dibenzofu- ran-2-yloxy)-2-tetradecyl-malonsäure-diäthylester, nD20t:
1,5033;
aus 3,76 g (20,0 mMol) 6,7,8,9-Tetrahydro-dibenzofuran- -3 oq und 6,46 g (0,02 Mol) 2 Brom-2-hexyl-malonsäure- -diäthylester den 2-(6,7,8,9-Tetrahydro-dibenzofuran-3- -yloxy)-2-hexyl-malonsäure-diäthylester, flD20, :1,5181; aus 1,02 g (5,0 mMol) 6,7,8,9-Tetrahydro-dibenzofuran -2-thiol und 1,61 g (5,0 mMol) 2-Brom-2-hexyl-malonsäu- rediäthylester den 2-(6,7,8,9-Tetrahydrodibenzofuran-2- -ylthio)-2-hexyl-malonsäure-diäthylester. nD20: :
1,5359; aus 2,04 g (10,0 mMol) 6,7,8,9-Tetrahydro-dibenzofuran- -3-thiod und 3,22 g (10,0 mMol) 2-Brom-2-hexyl-malon säure-diathylester den 2-(6,7,8,9-Tetrahydro-dibenzofuran- -ylthio)-2-hexyl-malonsäure-diäthylester, nD20 : 1,5412; aus 2,04 g (10 mMd) 6,7,8,9-Tetrahydro-dibenzofuran-3 -tiliol und 3,79 g (10 mMol) 2-Brom-2-decyl-malonsäure -diäthylester den 2-(6,7,8,9-Tetrahydro-dibenzofuran-3- -ylthio) -2-decylfflralonsäure-diäthylester.
Das als Ausgangsmaterial verwendete 6,7,8,9-Tetrahy dro-dibenzofuran-2-ol kann folgendermassen hergestellt werden: d) In einem Dreihalsrundkolben mit Thermometer, Rührer und Rückflusskühler werden 36,7 g (0,25 Mol) des Natriumsalzes des Hydrochinon-monomethyläthers portionsweise in eine Lösung von 46,6 g (0,264 Mol) 2 -Brom-cyclohexanon in 130 ml absolutem Toluol eingetragen, wobei die Temperatur von 250 auf 500 steigt. Der so erhaltene gelbe Brei wird nun 2 Stunden unter Rückfluss erhitzt, wobei sich das Natriumsalz des Hydrochinon-monomethyläthers allmählich löst und gleichzeitig Natriumbromid ausfällt.
Nach dem Abkühlen nimmt man in 700 ml Äther auf und wäscht die ätherische Lösung viermal mit insgesamt 200 ml 15'70iger Kalilauge und Wasser, trocknet über Magnesiumsulfat und dampft im Vakuum ein, Man erhält so das 2- (4-Methoxy-phenoxy) -cyclohe- xanon als gelbes öl. Nach zweimaligem Umkristallisieren aus Äther-Hexan erhält man das reine 2-(4-Methoxy -phenoxy)-cyclohexanon in Form blassgelber Nadeln vom Smp. 77-790.
e) In einem Rundkolben tmit Rührer werden 4,0 g (18,0 mMol) 2-(4-Methoxy-phenoxy)-cyclohexanon portionsweise in 40 ml fPhosphorsäure < d = 1,71) eingetragen, wobei eine grüne Lösung entsteht, die anschliessend 21/2 Stunden auf 1050 erhitzt wird. Dabei wechselt die Farbe der Lösung von grün nach rotbraun, und es scheidet sich gleichzeitig ein fast farbloses Öl ab. Nach dem Abkühlen glesst man das Reaktionsgemisch auf Eis und extrahiert zweimal mit insgesamt 200 ml Äther. Man wäscht die ätherische Lösung mit l-n. Natronlauge und Wasser, trocknet sie über Magnesiumsulfat und dampft sie im Vakuum ein.
Man erhält das 2-Methoxy-6,7,8,9 -tetrahydro-dibenzofuran als braunes öl, das zweimal im Kugelrohr bei 0,005 Torr zwischen 80-1000 destilliert wird. Das so erhaltene, reine 2-Methoxy-6.7,8,9-tetrahy dro-dibenzofuran ist ein farbloses öl, nD20 : 1,5783.
f) In einem Rundkolben mit Rückflusskühler und Ka liumhydroxid-Trockenrohr werden 3,0 g (14,85 mMol) 2 -Methoxy-6,7,8,9-tetrahydro-dibenzofuran 23/, Stunden unter Rühren mit 20,0 g Pyridinhydrochlorid auf 1700 erhitzt. Dann wird das noch heisse Reaktionsgemisch auf eine Mischung von 200 g Eis und 100 ml l-n. Salzsäure gegossen und noch 1/2 Stunde gerührt.
Das in weissen Kristallen ausfallende 6,7,8,9-Tetra hydro-dibenzofuran-2-ol wird abgenutscht und so lange mit kaltem Wasser gewaschen, bis das Waschwasser neutral ist. Nach Trocknen im Hochvakuum erhält man reines 6,7,8,9-Tetrahydro-dibenzofuran-2-ol als weisses Pulver, Smp. 106-1070.
Das als Ausgangsstoff verwendete 6,7,8,9-Tetrahydro -dibenzofuran-3-ol kann folgendermassen hergestellt werden: g) In einem Dreihalsrundkolben mit Thermometer, Rührer und Rückflusskühler werden 300,0 g (2,05 Mol) des Natriumsalzes des Resorcin-monomethyläthers portionsweise in eine Lösung von 382,0 g (2,16 Mol) 2-Brom -cyclohexanon in 825 ml absolutem Toluol eingetragen, wobei die Temperatur von 250 auf 600 steigt. Der so erhaltene gelbe Brei wird nun 2 Stunden unter Rückfluss erhitzt, wobei sich das Natriumsalz des Resorcinmonomethyläthers allmählich löst und gleichzeitig Natriumbromid ausfällt.
Nach dem Abkühlen verteilt man das Reaktionsgemisch zwischen Äther und Wasser, trennt die Ätherpha se ab und wäscht diese viermal mit insgesamt 2,0 115%- iger Kalilauge und Wasser, trocknet sie über Magnesiumsulfat und dampft sie im Vakuum ein.
Man erhält so das rohe 2-(3-Methoxy-phenoxy)-cyclohexanon als gelbes öl. Nach zweimaligein Umkristallisieren aus Äther-Hexan erhält man das reine 2-1(3-Meth- oxy.phenoxy)-cyclohexanon in Form blassgelber Kristalle vom Smp. 72,5-730. Die nicht kristallisierenden Mutterlaugen können ebenfalls nach h) weiterverarbeitet werden.
h) In einem Rundkolben mit Rührer werden 134,0 g (0,61 Mol) 2-(3-Methoxy-phenoxy)-cyclohexanon portionsweise in 1340 ml Phosphorsäure (d = 1,71) eingetragen, wobei eine grüne Lösung entsteht, die anschliessend 2 Stunden auf 1050 erhitzt wird. Nach dem Abkühlen giesst man das Reaktionsgemisch auf Eis und extrahiert mit Äther. Man wäscht die ätherische Lösung mit l-n. Natronlauge und Wasser, trocknet sie über Magnesilumsulfat und dampft sie im Vakuum ein. Man erhält ein Gemisch von 3- und 1-Methoxy-6,7,8,9-tetrahydro-dibenzo- furan in Form eines braunen öls, das bei 0,005 Torr zwischen 99-1080 destilliert wird.
Auf diese Weise erhält man ein farbloses öl, das ausser 3Methoxy.6,7,8,9-tetrahydro- -dibenzofuran laut NMR-Spektrum ca. 8% 1-Methoxy -6,7,8,9-tetrahydro-dibenzofuran enthält und ohne weitere Reinigung weiterverarbeitet wird.
i) In einem Rundkolben mit Rückflusskühler und Kaliumhydroxid-Trockenrohr werden 129,1 g (0,64 Mol) eines nach b) erhaltenen Gemisches aus 3- und 1-Meth oxy.6,7,8,9.tetrahydrodibenzofuran 23/; Stunden unter Rühren mit 401,1 g Pyridinhydrochlorid auf 1700 erhitzt.
Dann wird das noch heisse Reaktionsgemisch auf eine Mischung von 800 g Eis und 400 mi 1-n. Salzsäure gegossen und noch 1/2 Stunde gerührt. Das ausgefallene öl wird mit Äther extrahiert und die ätherische Lösung eingeengt, wobei unter Kühlung das rohe 6,7,8,9-Tetrahydrodibenzofuran-3-ol kristallisiert. Man saugt es ab und kristallisiert es noch zweimal aus Äther-Benzin. Man erhält so reines 6,7,8,9-Tetrahydro-dibenzofuran-3 -ol in Form hellgelber Kristalle vom Smp. 105-1060, während das 6.7, 8,9-Tetrahydro-dibenzofuran-1-ol in den Mutterlaugen verbleibt.
Das als Ausgangsmaterial verwendete 6,7,8,9-Tetra hydro-dibenzofuran-2-thiol kann wie folgt hergestellt werden: j) In einem Rundkolben mit Rückflusskühler, Rührer, Kaliumhydroxid-Trockenrohr, Thermometer und Gaseinleitungsrohr fügt man in kleinen Portionen 0,48 g (10,0 mMol) 5()%ige Natriumhydriddispersion zu einer Lösung von 1,88 g (10,0 ImMol) 6,7,8,9.Tetrahydro.dibenzofuran- -2-ol in 10 ml Dimethylformamid unter Stickstoff. Nach 1/2 Stunde ist die Wasserstoffentwicklung beendet. Die entstandene dunkelbraune Suspension wird nun auf 100 abgekühlt und dann 1,65 g (13,0 mMol) Dimethylthiocarbaminsäurechlorid auf einmal zugegeben. Dabei steigt die Temperatur sofort auf 180 an.
Anschliessend wird noch eine Stunde auf 800 unter Rühren erhitzt, wobei in der nun hellbraun gefärbten Lösung Natriumchlorid ausfällt. Nach dem Abkühlen wird im Vakuum eingedampft und das zurückbleibende braune öl zwischen Äther und Wasser verteilt. Man wäscht die Ätherphase mehrmals mit kalter verdünnter Natronlauge und Wasser, trocknet über Magnesiumsulfat und dampft sie ein, wobei der rohe Dimethylthiocarbaminsäure.O.(6,7,8,9.tetrahydro -diben zofuran-2-yl)-ester als gelbbraunes Öl zurückbleibt, das durch Säulenchromatographie [Kieselgel 0,05-0,2 mm, Merck, Lösungsmittel Benzol-Essigester (9:1)] gereinigt wird. Nach Eindampfen der reinen Fraktionen werden diese zweimal aus wässrigem Methanol unter Zusatz von Aktivkohle umkristallisiert.
Man erhält so den reinen Dimethylthiocarbaminsäure-0-(6,7,8,9-tetrahydro - dibenzofuran-2-yl)-ester vom Smp. 129-1310.
k) In einem mit Magnetrührer und Gaseinleitungsrohr versehenen Rundkolben werden 9,6 g (35 mMol) Dime thylthiocarbaminsäure - 0- (6,7,8,9-tetrahydro - dibenzofu ran-2.yl)-ester 31,; Stunden bei 280-2950 unter Stickstoff und Rühren erhitzt. Das so entstandene dunkelbraune öl kann direkt nach c) weiterverarbeitet werden. Gewünschtenfalls reinigt man es durch Säulenchromatographie [Kieselgel 0,05-0,2 mm, Merck, Lösungsmittel Benzol-Essigester (9:1)]. Die reinen Fraktionen werden vereinigt und eingedampft. Nach zweimaligem UmkristaDi- sieren aus wässrigem Methanol erhält man den reinen Dimethylthiocarbaminsäure-S-(6,7,8,9 - tetrahydro - dibenzofuran-2-yl)-ester vom Smp. 73-74 .
1) In einem Rundkolben mit Rückflusskühler, Rührer, Kaliumhydroxid-Trockenrohr und Gaseinleitungsrohr wird eine Lösung von 3,0 g (11,0 mMol) Dirnethylthio- carbaminsäure-S - (6,7,8,9- tetrahydro-dibenzofuran-2-yl) -ester (Rohprodukt) in 18,8 ml 0%ihrer Natronlauge und 60 ml Methanol 31/2 Stunden unter Stickstoff und Rückfluss gekocht. Nach dem Abkühlen wird das Methanol im Vakuum abgedampft, der Rückstand mit 2-n. Salzsäure angesäuert und mit Äther extrahiert. Nach Waschen der ätherischen Lösung mit Wasser bis pH = 7 und Trocknen über Magnesiumsulfat wird erneut eingedampft.
Man erhält das rohe 6,7,8,9-Tetrahydro-dibenzofuran-2-thiol als gelbes öl, das durch Säulenchromatographie [Kieselgel 0,05-0,2 mm, Merck, Lösungsmittel Benzol-Essigester (9:1)] gereinigt wird. Die reinen Fraktionen werden vereinigt und eingedampft. Nach Umkristallisation aus wässerigem Äthanol erhält man das 6,7,8,9-Tettrahydro-dibenzofuran-2-thiol in Form blassgelber Kristalle vom Smp. 44,5460.
Das als Ausgangsmaterial verwendete 6,7,8,9-Tetrahydro-dibenzofuran-3-thiol kann wie folgt hergestellt werden: m) Analog Beispiel 1j) erhält man aus 22,6 g (0,12 Mol) 6,7,8,9-Tetrahydro-dibenzofuran-3 -ol und 19,8 g (0,16 Mol) Dimethylthiocarbaminsäurechlorid den Dimethylthiocarbaminsäure-O-(6,7,8,9-tetra- hydro-dibenzofuran-3-yl)-ester, Smp. 158-1590 (aus Essigester).
n) Analog Beispiel lk) erhält man aus 19,0 g (69,0 mMol) Dimethylthiocarbaminsäure-O -(6,7,8,9-tetrahydro-dibenzofuran-3-yl)-ester den Dime thylthiocarbaminsäure-S-(6,7,8,9-tetrahydro-dibenzofuran- -3-yl)-ester, Smp. 102-1030 (aus Äthanol-Wasser).
o) Analog Beispiel 11) erhält man aus 11,75 g (42,0 mMol) Dimethylthiocarbaminsäure-S -(6,7,8,9-tetrahydro-dibenzofuran-3-yl)-ester das 6,7,8,9 -Tetrahydro-dibenzofuran-3-thiol, Smp. 73-74 (aus Methanol-Wasser).
Beispiel 2
Analog Beispiel 1) erhält man aus 0,80 g (ca. 2 mMol) roher 2-(6,7,8,9-Tetrahydro-di- benzothiophen-2-ylthio)-2-hexyl-malonsäure die 2-(6,7,8,9-Tetrahydro-dibenzothiophen-2-ylthio)-octansäure vom
Smp. 91-92 (aus Hexan); aus 1,20 g (ca. 3 mMol) roher 2-(6,7,8,9-Tetrahydro-dibenzothiophen-3-ylthio)-2-hexyl-malonsäure die 2-(6,7, o,- 9-Tetrahydro-dibenzothiophen-3 -ylthio)-octansäu re, nD : 1,5718 nach chromatographischer Reinigung an Kieselgel, Elution mit Benzol und Benzol-Eisessig (49:1).
Die Ausgangsstoffe werden analog Beispiel la), jedoch nur zweistündigem Kochen aus 0,924 g (2,0 mMol) 2-(6,7,8,9-Tetrahydro-dibenzothio phen-2-ylthio)-2-hexyl-malonsäure-diäthylester bzw.
1,386 g (3,0 mMol) 2-(6,7,8,9-Tetrahydro-dibenzothio phen.3 -ylthio)-2-hexyl-malonsäure-diäthylester erhalten.
Analog Beispiel 3a erhält man die folgenden Zwischenprodukte: aus 1,0 g (4,54 mMol) 6,7,8,9-Tetrahydro-dibenzothiophen-2-thiol und 1,47 g (4,54 mMol) 2-Brom-2-hexyl-ma- lonsäurediäthylester den 2-(6,7,8,9-Tetrahydro-dibenzo thiophen-2-ylthio)-2-hexyl-malonsäure-diäthylester, nD20 : 1,5598; aus 2,20 g (10,0 mMol) 6,7,8,9-Tetrahydro-dibenzothiophen-3-thiol und 3,23 g (10,0 mMol) 2-Brom-2-hexyl-malonsäure-diäthylester den 2-(6,7,8,9-Tetrahydro-dibenzo thiophen- 3 -ylthio)-2-hexyl-malonsäure-diäthylester, 20, 1,5601.
Das als Ausgangsmaterial verwendete 6,7,8,9-Tetra hydrodibenzothioplaen-2-thiol kann wie folgt hergestellt werden: a) Zu der auf ca. 5 gekühlten Lösung von 10,0 g (49,0 mMol) 6,7,8,9-Tetrahydro-dibenzothiophen-2-ol in 50 ml abs. Dimethylformamid gibt man unter Rühren und Einleiten von Stickstoff portionsweise 2.35 g (49,0 mMol) Sie Natriumhydriddispersion. Nach 1/2 Stunde Rühren bei Raumtemperatur und 5 Minuten bei 800 ist die Wasserstoffentwicklung beendet. Nun werden bei 5-10 8,07 g (65,4 mMol) Dimethylthiocarbamidsäurechlorid in
10 ml abs. Dimethylformamid innerhalb ca. 2 Minuten zugetropft und das Reaktionsgemisch noch 2 Stunden bei 800 gerührt.
Nach dem Abkühlen dampft man es im Vakuum ein, nimmt den Rückstand in Wasser auf und extrahiert ihn gründlich mit Äther und Chloroform. Die vereinigten organischen Phasen werden mit Wasser gewaschen, über Magnesiumsulfat getrocknet, eingedampft und der Rückstand durch Säulenchromatographie an Kieselgel 0,05-0,2 mm, Merck, Elution mit Benzol-Essigester [9:1] gereinigt.
Man erhält den Dimethylthiocarbaminsäure-0-(6,7,8, 9-tetrahydro-dibenzothiophen-2-yl)-ester vom Smp. 154
1550. (Methanol oder Essigester).
b) in einer Stickstoffatmosphäre werden 9,0 g (30,9 mMol) Dimethylthiocarbaminsäure-0-(6,7,8,9-tetrahydro- -dibenzothiophen-2-yl)ester bei 2500 zum Schmelzen gebracht und anschliessend 3 Minuten bei 3500 erhitzt. Nach dem Abkühlen (mittels Luftstrom) reinigt man den gelb gefärbten Rückstand durch Säulenchromatographie an Kieselgel Merck fElution mit Benzol-Essigester (19:1)].
Die das gewünschte Produkt enthaltenden Fraktionen werden zusammengefasst und aus Methanol umkristallisiert. Man erhält so den Dimethylthiocarbaminsäure-S -(6,7,8,9-tetrahydro-dibenzothiophen-2.yl)-ester vom Smp.
98-990 (aus Methanol).
c) Unter Rühren und Einleiten von Stickstoff werden 5,1 g (17,5 mMol) Dimethylthiocarbaminsäure-S-(6.7,8,9- -tetrahydro-dibenzothiophen-2-yl)-ester in 100 ml Methanol und 80 ml lttOiger Natronlauge 3 Stunden unter Rückfluss gekocht. Das organische Lösungsmittel wird dann im Vakuum abgedampft, der Rückstand mit 1-n.
Salzsäure angesäuert und mit Äther extrahiert. Die mit Wasser gewaschene und über Magnesiumsulfat getrocknete Ätherphase dampft man im Vakuum ein und chromatographiert den Rückstand an Kieselgel, Merck, Elution mit Benzol und Benzol-Essigester (19:1). Nach Umkristallisation aus Methylenchlorid-Hexan erhält man das 6,7,8,9-Tetrahydro-dibenzothiophen-2-thiol vom Smp. 64650.
Das als Ausgangsmaterial verwendete 6,7,8,9-Tetra hydfo-diben2othiophen-3-thiol kann analog zu der im Beispiel 2a), b) und c) beschriebenen Reaktionsfolge hergestellt werden: d) Analog Beispiel 2a) erhält man aus 16,0 g (78,3 mMol) 6,7,8,9-Tetrahydro-dibenzothiophen-3-ol und 12,95 g (104,9 mMol) Dimethylthiocarhaminsäurechlorid den Dimethylthiocarbaminsäure-0-(6,7,8,9-tetralaydro - dihen- zothiophen-3-yl)-ester vom Smp. 139,5-1400 (aus Methanol).
e) Analog Beispiel 2b), jedoch bei einer Reaktionstemperatur von 2600 und einer Reaktionsdauer von 5 Stunden, erhält man aus 12,10 g (41,5 mMol) Dimethylthio carbaminsäure-0-(6,7,8,9-tetrahydro-dibenzothiophen- 3 -yl)-ester den Dimethylthiocarbam'insäure-S- (6,7,8,9-tetra- hydro-diSbenzothiophen-3-yl)-ester vom Smp. 98-990 (aus Methanol).
f) Analog Beispiel 2c) erhält man aus 8,74 g (30,0 mMol) lDimethylthiocarbaminsäure-S-(6,7,8,9-tetrahydro- -dibenzothiophen-3-yl)-ester das 6,7,8,9-Tetrahydro-dibenzothiophen-3-thiol vom Smp. 36-36,50 (aus Hexan).
Beispiel 3
0,78 g (ca. 2 mMol) rohe 2-(6'7,8,9-Tetrahydro-diben- zothiophen-2-yloxy)-2-hexyl-malon säure werden unter Stickstoff eine halbe Stunde bei 1500 erhitzt. Das decarboxylierte Rohprodukt reinigt man durch Chromatographie an Kieselgel [Elution mit Benzol und Benzol-Eisessig (19:1)]. Nach Umkristallisation der reinen Fraktionen aus Hexan erhält man die 2-(6,7,8,9-Tetrahydro-dibenzothio- phen-2-yloxy)-octansäure, Smp. 90-910.
Aus 0,78 g (ca. 2 mMol) roher 2-(6,7,8,9-Tetrahydro -dibenzothiophen-3 -yloxy)-2-hexyl-malonsäure erhält man durch (Erhitzenjauf 1240-2500 während 5 Minuten und Aufarbeitung wie oben die 2-(6.7,8,9-Tetrahydro-dibenzo thiophen-3-yloxy)-octansäure vom Smp. 106-1070 (aus Hexan).
Die Ausgangsstoffe werden analog Beispiel la), jedoch nur zweistündigem Kochen aus 0,892 g (2,0 mMol) 2-(6,7,8,9-Tetrahydro-dibenzothio- phen-2-yloxy)-2-hexyl-malonsäure-diäthylester bzw.
0,892 g (2,0 mMol) 2-(6,7,8,9-Tetrahydro-dibenzothio- phen-3 -yloxy)-2-hexyl-malonsäure-diäthylester erhalten.
Der als Ausgangsstoff eingesetzte 2-(6,7,8,9-Tetrahy dro - diben,zothiophen - 2 - yloxy)-2-hexyl - malonsäure - di äthylester wird wie folgt hergestellt: a) Zu einer Lösung von 0,258 g (11,2 mMol) Natrium in 15 ml abs. Äthanol fügt man 2,29 g (11,2 mMol) 6,7,8,9 -Tetrahydro-dibenzothiophen-2-ol. Unter Rühren und Einleiten von Stickstoff versetzt man diese Lösung tropfenweise mit 3,62 g (11,2 mMol) 2-Brom-2-hexyl-malonsäure-diäthylester in 15 ml abs. Äthanol und kocht das Reaktionsgemisch 4 Stunden unter Rückfluss. Nach dem Abkühlen wird das Äthanol im Vakuum abgedampft und der Rückstand zwischen Wasser und Äther verteilt. Die abgetrennte und mit Wasser gewaschene ätherphase trocknet man über Magnesiumsulfat und dampft sie ein.
Die Reinigung des Rohproduktes erfolgt durch Säulenchromatographie an Kieselgel Elution mit Benzol-Hexan (2:1)]. Man erhält den 2-(6,7,8,9-Tetrahydro-dibenzothio- phen-2 - yloxy)-2 - hexyl-malonsäure - diäthylester, nD ¯ 1,5383.
Analog erhält man aus 2,04 g (10,0 mMol) 6,7,8,9-Tetrahydro-dibenzothio phen-3-ol und 3,23 g (10,0 mMol) 2-Brom-2-hexyl-malonsäure-diäthylester den 2-(6,7,8,9-Tetrahydro-dibenzothio- phen-3-yloxy) -2-hexyl-malonsäure-diäthylester, nD20t: 1,5361.
Das als Ausgangsmaterial verwendete 6,7,8.9-Tetra- hydro-dibenzothiophen-2-ol kann wie folgt hergestellt werden: b) 12 g (50,78 mMol) 2-(p-Methoxy-phenylthio)-cyclo- hexanon werden unter Rühren und Einleiten von Stickstoff zu 120 ml konz. Phosphorsäure (d = 1,71) gegeben.
Man erhitzt das Reaktionsgemisch auf 850 und rührt es bei dieser Temperatur während 14 Stunden. Nach dem Abkühlen auf Raumtemperatur wird es auf Eis gegossen und mit Äther extrahiert. Die vereinigten Ätherphasen werden mit 2-n. Natronlauge und Wasser gewaschen, über Natrium getrocknet und im Vakuum eingedampft. Die Reinigung des kristallin anfallenden Rohproduktes erfolgt durch Säulenchromatographie an Kieselgel 0,050,2 mm, Merck, Elufionsmittel Benzol-Hexan [3:2].
Man erhält so reines 2-Methoxy-6,7,8, 9-tetrahydro- di- benzothiophen, Smp. 850 (aus Hexan).
c) 3,5 g (16,03 mMol) 2-Methoxy-6,7,8,9-tetrahydro -dibenzothiophen werden mit 28 g (0,24 Mol) geschmolzenem Pyridinhydrochlorid versetzt. Das sich unter Stickstoff befindende Reaktionsgemisch rührt man 2 Stunden bei 160-1650 und verteilt es nach dem Abkühlen zwischen 2-n. Salzsäure und Äther. Die vereinigten Ätherextrakte werden neutral gewaschen, über Natriumsulfat getrocknet und im Vakuum eingedampft. Das kristallin anfallende Rohprodukt reinigt man durch Säulenchromatographie an Kieselgel 0,02-0,5 mm, Merck. [Elution mit Benzol und Benzol-Essigester (9:1)]. Die das gewünschte Produkt enthaltenden Fraktionen werden eingedampft und aus Methylenchlorid-Hexan umkristallisiert. Das erhaltene 6,7,8,9-Tetrahydro-dibenzothiophen-2-ol schmilzt bei 1131140.
Das als Ausgangsmaterial eingesetzte 6,7,8,9-Tetrahy dro-dibenzothiophen-3-ol wird wie folgt hergestellt: d) Zu einer Lösung von 23,0 g (1,0 Mol) Natrium in 700 ml abs. Äthanol gibt man unter Rühren und Einleiten von Stickstoff 140,2 g (1,0 Mol) m-Methoxy-thiophenol. Im Verlaufe von 15 Minuten werden dann 177,0 g (1,0 Mol) 2-Brom-cyclohexanon zugetropft, wobei sich das Reaktionsgemisch erwärmt. Man kocht es anschliessend noch ll/2 Stunden unter Rückfluss. Das Äthanol wird anschliessend im Vakuum abgedampft und der Rückstand zwischen Wasser und Äther verteilt. Den mit Wasser neutral gewaschen und über Natriumsulfat getrockneten Ätherextrakt dampft man ein.
Zur Reinigung wird das Rohprodukt im Hochvakuum fraktioniert (20 cm Vi greux-Kolonne). Man erhält das 2-(,m-Methoxy-phenyl- thio3-cyclo,hexanon vom Kp. 146-1470/0,15 Torr, in Form eines gelb gefärbten öls, nD: 1,5786.
e) 118,5 g (0,5 Mol) 2-[m-Methoxy-phenylthio3-cyclo- hexanon werden unter Rühren und Einleiten von Stick stoff zu 1200 ml konzentrierter Phosphorsäure (d = 1,71) gegeben. Man erhitzt das Reaktionsgemisch auf 1050 und rührt es 5 Stunden bei dieser Temperatur. Nach dem Abkühlen auf Raumtemperatur wird es auf Eis gegossen und mit Äther extrahiert. Die vereinigten Ätherphasen werden mit 2-n. Natronlauge und Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum eingedampft.
Die Reinigung des Rohproduktes erfolgt durch Chromatographie an Kieselgel [Merck, 0,05-0,2 mm, Elution mit Benzol-Hexan (1:3)]. Man erhält das 3-Methoxy-6,7,8,9 -tetrahydro-dibenzothiophen vom Smp. 46-46,50 (aus Methanol).
Als Nebenprodukt wird noch 1-Methoxy-6,7,8,9 1 -Methoxy-6,7,8'9-tetra- hydro-dibenzothiophen, Smp. 57-580 (aus Methanol) isoliert.
f) 54,57 g (0,25 Mol) 3-Methoxy-6,7,8,9:tetrahydro-di- benzothiophen werden unter Rühren und Einleiten von Stickstoff zu einer Schmelze von 150 g (0,77 Mol) frisch destilliertem Pyridin-hydrochlorid gegeben. Man erhitzt das Gemisch 1,25 Stunden auf 2200 und trägt dann die Schmelze in ein Gemisch von 400 ml 2-n. Salzsäure und 200 g Eis ein. Das nach Extraktion mit Äther-Methylenchlorid (3:1) und Waschen der organischen Phase mit Wasser, Trocknen über Natriumsulfat und Eindampfen im Vakuum resultierende kristalline Rohprodukt wird über Kieselgel tMerck, 0,05-0,2 mm, Elution mit Benzol Äthylacetat (9:1)] filtriert und aus Methylenchlorid-Hexan umkristallisiert. Man erhält das 6,7,8,9-Tetrahydro -dibenzothiophen-3-ol vom Smp. 117-1180 (aus Methanol).
Beispiel 4
1,1 g (3,5 mMol) 2-(6,7,8,9-Tetrahydroaibenzofuran- -2-yloxy)-heptansäure werden in 20 ml abs. Äthanol gelöst. Zu der erhaltenen Lösung fügt man eine Lösung von 69 mg (3,0 mMol) Natrium in 5 ml abs. Äthanol, dampft zur Trockne und verreibt den weissen, festen Rückstand mit ca. 20 ml Äther. Dann wird abgenutscht und mit Äther nachgewaschen. Man erhält das reine Natriumsalz als weisses Pulver, das zwischen 290-3080 unter Zersetzung schmilzt.
Beispiel 5
Analog Beispiel 49 erhält man aus 3,465 g (10,0 mMol) 2-(6,7,8,9-Tetrahydro-dibenzothiophen-3-yloxy)-octansäure das Natriumsalz der 2-(6,7,8,9 - Tetrahydro - dibenzothiophen-3- yloxy) - octansäure vom Smp. 320-3220.
Beispiel 6
110 mg (2,75 mMol) Calcium werden in 10 ml Wasser unter Stickstoff zersetzt. Zu der so erhaltenen Calciumhydroxid-Suspension werden 2,05 g (6,22 mMol) 2-(6,7, 8,9-Tetrahydro-dibenzofuran-2-yloxy)-octansäure in 50 ml Methanol zugegeben und das Gemisch 15 Minuten unter Rückfluss gekocht. Nach dem Abkühlen wird die so erhaltene, weisse Suspension auf ca. 20 ml eingeengt, das ausfallende, rohe Calciumsalz abgesaugt und mit wenig Äther gewaschen. Anschliessend extrahiert man es dreimal mit je 120 ml kochendem Methanol. Die Methanolextrakte werden vereinigt, filtriert und auf ca. 20 ml eingeengt, wobei das Calciumsalz auskristallisiert. Man verdünnt die Methanolphase noch mit ca. 30 ml Äther, saugt dann das Kristallisat ab und wäscht es mit Äther.
Nach Trocknen im Hochvakuum erhält man das reine Calciumsalz der 2-(6,7,8,9-Tetrahydro-dibenzofuran-2-yl- oxy)-octansäure, das zwischen 305-3150 unter Zersetzung schmilzt.
Beispiel 7
Zu einer durch Zersetzen von 137 mg (3,42 mMol) Calcium in 15 ml Wasser hergestellten Suspension von Calciumhydroxid gibt man unter Rühren und Einleiten von Stickstoff eine Lösung von 2,50 g (7,2 mMol) 2-(6,7, 8,9-Tetrahydro-dibenzothiophen-2 -yloxy) - octansäure in 80 ml Methanol. Das Reaktionsgemisch wird 15 Minuten unter Rückfluss gekocht und im Vakuum eingeengt. Das ausgefallene, rohe Calciumsalz wird abfiltriert, mit Äther gewaschen und zweimal aus abs. Methanol umknstalli- siert. Man erhält so das reine Calciumsalz der 2-(6,7,8,9 -Tetrahydro- dibenzothiophen- 2 - yloxy) - octansäure, das sich zwischen 290 und 3050 unter Braunfärbung zersetzt.

The 6,7,8,9-tetrahydro-dibenzofuran-2-thiol used as the starting material can be prepared as follows: j) In a round bottom flask equipped with a reflux condenser, stirrer, potassium hydroxide drying tube, thermometer and gas inlet tube, 0, 48 g (10.0 mmol) of 5% strength sodium hydride dispersion to a solution of 1.88 g (10.0 µmoles) of 6,7,8,9 tetrahydro.dibenzofuran- -2-ol in 10 ml of dimethylformamide under nitrogen . 0.78 g (approx. 2 mmol) of crude 2- (6,7,8,9-tetrahydro-dibenzothiophen-2-yloxy) -2-hexyl-malonic acid are heated at 1500 for half an hour under nitrogen. The decarboxylated crude product is purified by chromatography on silica gel [elution with benzene and benzene-glacial acetic acid (19: 1)]. After recrystallization of the pure fractions from hexane, 2- (6,7,8,9-tetrahydro-dibenzothiophene-2-yloxy) -octanoic acid, melting point 90-910, is obtained.

Similarly, from 2.04 g (10.0 mmol) of 6,7,8,9-tetrahydro-dibenzothiophen-3-ol and 3.23 g (10.0 mmol) of 2-bromo-2-hexyl-malonic acid diethyl ester 2- (6,7,8,9-tetrahydro-dibenzothiophene-3-yloxy) -2-hexyl-malonic acid diethyl ester, nD20t: 1.5361.

The crystalline crude product resulting after extraction with ether-methylene chloride (3: 1) and washing of the organic phase with water, drying over sodium sulfate and evaporation in vacuo is purified over silica gel tMerck, 0.05-0.2 mm, elution with benzene, ethyl acetate (9 : 1)] filtered and recrystallized from methylene chloride-hexane. The 6,7,8,9-tetrahydro-dibenzothiophen-3-ol with a melting point of 117-1180 (from methanol) is obtained.
Example 4
1.1 g (3.5 mmol) 2- (6,7,8,9-tetrahydroaibenzofuran- -2-yloxy) -heptanoic acid are dissolved in 20 ml of abs. Dissolved ethanol. A solution of 69 mg (3.0 mmol) of sodium in 5 ml of abs is added to the resulting solution. Ethanol, evaporates to dryness and rubs the white, solid residue with approx. 20 ml of ether. Then it is sucked off and washed with ether. The pure sodium salt is obtained as a white powder, which melts between 290-3080 with decomposition.
Example 5
Analogously to Example 49, the sodium salt of 2- (6,7,8,9-tetrahydro) is obtained from 3.465 g (10.0 mmol) of 2- (6,7,8,9-tetrahydro-dibenzothiophen-3-yloxy) -octanoic acid - dibenzothiophen-3-yloxy) octanoic acid of m.p. 320-3220.
Example 6
110 mg (2.75 mmol) calcium are decomposed in 10 ml water under nitrogen. To the calcium hydroxide suspension obtained in this way, 2.05 g (6.22 mmol) of 2- (6.7, 8,9-tetrahydro-dibenzofuran-2-yloxy) -octanoic acid in 50 ml of methanol are added and the mixture under Boiled under reflux. After cooling, the resulting white suspension is concentrated to approx. 20 ml, and the crude calcium salt which precipitates is filtered off with suction and washed with a little ether. It is then extracted three times with 120 ml of boiling methanol each time. The methanol extracts are combined, filtered and concentrated to approx. 20 ml, the calcium salt crystallizing out. The methanol phase is diluted with about 30 ml of ether, then the crystals are suctioned off and washed with ether.
After drying in a high vacuum, the pure calcium salt of 2- (6,7,8,9-tetrahydro-dibenzofuran-2-yloxy) octanoic acid is obtained, which melts between 305-3150 with decomposition.
Example 7
To a suspension of calcium hydroxide prepared by decomposing 137 mg (3.42 mmol) of calcium in 15 ml of water, a solution of 2.50 g (7.2 mmol) of 2- (6.7, 8,9-Tetrahydro-dibenzothiophene-2-yloxy) octanoic acid in 80 ml of methanol. The reaction mixture is refluxed for 15 minutes and concentrated in vacuo. The precipitated, crude calcium salt is filtered off, washed with ether and extracted twice from abs. Methanol umknstalli- siert. The pure calcium salt of 2- (6,7,8,9-tetrahydrodibenzothiophen-2-yloxy) -octanoic acid is obtained, which decomposes between 290 and 3050 while turning brown.
