Verfahren zur Herstellung ll-basisch substituierter Dibenz[b,f]-1,40xazepine, Dibenzo[b,f]
1,4thiazepine und Dibenzo[h,e]-1,4,diazepine
Die Erfindung betrifft ein Verfahren zur Herstellung von ll-basisch substituierten Dibenz[b,f]-1,4-oxazepinen, Dibenzo- [b,f]-1,4-thiazepinen und Dibenzo[b,e] -1,4-diazepinen der allgemeinen Formel:

sowie von Säure-Additionssalzen davon. In Formel I stellt Z ein Sauerstoffatom, ein Schwefelatom, eine Sulfinylgruppe (-SO-) oder eine Iminogruppe (-NH-) dar, Rt ist ein Wasserstoffatom, eine Allylgruppe, eine höchstens 3 C-Atome enthaltende Alkyl- oder Hydroxyalkylgruppe oder eine höchstens 6 C-Atome enthaltende Alkoxyalkyl- oder Alkoylaloxyalkylgruppe.
Die Reste R2 und R3 sind gleich oder verschieden und bedeuten Wasserstoffatome oder Methylgruppen.
Die gewünschten Verbindungen der Formel I werden erhalten, wenn man Verbindungen der Formel:

worin Z und Rt die genannte Bedeutung haben, und X einen mit dem Wasserstoff von Ammoniak oder Aminen abspaltbaren Rest, insbesondere ein Halogenatom, bedeutet, mit Ammoniak bzw. einem Amin der Formel HNR2R3, worin R2 und R3 die genannte Bedeutung haben, umsetzt.
Mit einer Sulfochloridgruppe (-SO2 Cl) substituierte Ausgangsprodukte der Formel II erhält man z. B.
durch Diazotieren entsprechender Aminoverbindungen und anschliessende Meerwein-Reaktion.
In die nach dem beschriebenen Verfahren erhaltenen Verbindungen gemäss Formel I, in welchen Rl Wasserstoff bedeutet, können nicht Wasserstoff bedeutende Reste Rt nachträglich eingeführt werden, z.B.
durch Umsetzen mit reaktionsfähigen Estern von Alkoholen der Formel R-OH. Als reaktionsfähige Ester eignen sich solche anorganischer oder organischer Säuren, z. B. Halogenwasserstoffsäure- oder Toluolsulfonsäureester. Eine Alkylgruppe Rt kann auch nach der Methode der reduktiven Alkylierung eingeführt werden, d. h. durch Umsetzen mit entsprechenden Aldehy den entweder unter Verwendung von Wasserstoff in Gegenwart eines Katalysators oder unter Verwendung eines Reduktionsmittels wie Ameisensäure. Die Einführung einer Hydroxyalkylgruppe R, kann auch durch Umsetzen mit einem entsprechenden Alkylenoxyd erfolgen.
Bei erhaltenen Verbindungen gemäss Formel I, in welchen Rt eine Hydroxyalkylgruppe bedeutet, kann nachträglich eine Acylierung zur Bildung der mit einer Alkoyloxyalkylgruppe Rt substituierten Produkte vorgenommen werden. Als Acylierungsmittel kommen insbesondere Säurehalogenide und Säureanhydride in Betracht.
Die in der beschriebenen Weise erhaltenen Basen sind in den meisten Fällen kristallisierbar, sonst im Hochvakuum unzersetzt destillierbar, und bilden mit anorganischen und organischen Säuren, beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Salpetersäure, Phosphorsäure, Essigsäure, Oxalsäure, Maleinsäure, Bernsteinsäure, Weinsäure, Toluolsulfonsäure und dergleichen, in Wasser beständige Additionssalze, in welcher Form die Produkte ebenfalls verwendet werden können.
Die in der beschriebenen Weise erhaltenen Basen und ihre Säure-Additionssalze sind neue Verbindungen, die als Wirkstoffe in Arzneimittel oder als Zwischenprodukte zur Herstellung von solchen Verwendung finden. Sie üben eine günstige Wirkung auf das Zentralnervensystem aus und fallen unter anderem als Analgetika, Neuroleptika, Sedativa und insbesondere als neuroleptische Antidepressiva und als Antiemetika in Betracht.
Bei Verbindungen gemäss Formel I, in welchen R2 und R2 Methylgruppen bedeuten, steht die antiemetische Wirkung im Vordergrund. Diese zeigt sich pharmakologisch in einer starken apomorphinantagonistischen Wirkung bei Hunden und Ratten sowie einer im Vergleich zur apomorphinantagonistischen Wirkung geringen kataleptischen und motllitätsdämpfenden Wirkung. Antiemetisch besonders wirksam sind die gemäss Beispielen 1 und 2 erhaltenen Verbindungen, nämlich das 2-Dimethylaminosullonyl-1 1 - (4-methyl-1-piperazinyl) dibenz[b,f]-1,4-oxazepin und 2-Dimethylaminosulfonyl-1 1 (4-methyl-1-piperazinyl)- dibenzo[b,f] -1,4-thiazepin und deren Säure-Additionssalze.
Andere Verbindungen, insbesondere diejenigen mit 11-ständigem 1-Piperazinylrest, zeigen gleichzeitig das für Neuroleptika und Antidepressiva typische Wirkungsbild, wobei die antidepressive Wirkungskomponente pharmakologisch z. B. aus einem bei Ratten zu beobachtenden Antagonismus gegenüber Tetrabenazin ersichtlich ist.
Beispiel 1
Eine Lösung von 5,2 g des in der unten beschriebenen Weise erhaltenen rohen 2-Chlorsulfonyl-1 1-(4-methyl-1- piperazinyl)-dibenz[b,f] -1,4oxazepins in 80 ml Chloroform wird bei Zimmertemperatur gerührt und tropfweise mit 50 ml einer ca. 10 0/oigen Lösung von Dimethylamin in Toluol versetzt. Das Reaktionsgemisch wird hierauf während 2 Stunden bei Zimmertemperatur weitergerührt und anschliessend während 1 Stunde unter Rühren auf 400 C erwärmt.
Das Reaktionsgemisch wird anschliessend im Vakuum zur Trockne eingedampft, und der Rückstand wird in verdünnter Essigsäure aufgenommen. Die essigsauren Auszüge werden mit Aktivkohle geklärt und mit konzentrierter Ammoniaklösung alkalisch gestellt. Die ausgeschiedene Base wird in Benzol aufgenommen. Die Benzollösung wird dreimal mit Wasser gewaschen, mit Natriumsulfat getrocknet und eingedampft. Der Rückstand wird in Benzol gelöst und durch basisches Aluminiumoxid filtriert. Der nach Abdampfen des Lösungsmittels erhaltene Rückstand wird aus Aceton/ Petroläther kristallisiert, wobei man 3,2 g 2-Dimethylaminosulfonyl-11- (4-methyl-1-piperazinyl)- dibenz[b,f]-1,4-oxazepin vom Schmelzpunkt 148-150 C erhält.
Das als Ausgangsmaterial verwendete 2-Chlorsulfonyl-1 1-(4-methyl-1- piperazonyl)-dibenz[b,f] 1,4-oxazopin erhält man wie folgt:
15,4 g 2-Amino-1 1-(4-methyl-1-piperazinyl)- dibenz[b,f] -1,4-oxazepin (Smp. 153-156 C) werden in 50 ml Eisessig und 15 ml 38 0/obiger Salzsäure gelöst und bei 0 bis 5 C mit einer Lösung von 3,6 g Natriumnitrit in 6 ml Wasser in üblicher Weise diazotiert. Die erhaltene Diazoniumlösung wird bei 10 C unter Rühren und innert weniger Minuten zu 40 ml einer 300/oigen Lösung von Schwefeldioxid in Eisessig, welche 2 g Kupfer(II)chlorid enthält, gegeben.
Nach Abklingen der Stickstoffentwicklung bei Zimmertemperatur wird das Reaktionsgemisch während 15 Minuten auf 40 C erwärmt. Hierauf wird das Reaktionsgemisch mit Wasser auf ein Volumen von ca. 1 Liter verdünnt und anschliessend mit Aktivkohle geklärt. Unter Rühren und sorgfältiger Kühlung werden die basischen Anteile mit konzentrierter Natronlauge gefällt und in Chloroform aufgenommen.
Die Chioroformauszüge werden einmal mit verdünnter Natronlauge und einmal mit Wasser gewaschen, mit Natriumsulfat getrocknet und eingedampft. Als Rückstand erhält man rohes 2-Chlorsulfonyl-11-(4-methyl-1piperazinyl)-dibenz[b,f]-1,4-oxazepin.
Bei analogem Vorgehen wie in Beispiel 1 erhält man aus den entsprechenden Ausgangsprodukten weiterhin z. B. die in der nachfolgenden Tabelle erwähnten Verbindungen gemäss Formel I. In der Tabelle haben Z, Rt, R2 und R3 die früher genannte Bedeutung. In der Kolonne rechts bedeutet Ac Aceton, Ae Äther, CH Chloroform, Me Methanol und Pe Petrol äther.

Beispiel 11
5,4 g des gemäss Beispiel 10 erhaltenen 2-Dimethylaminosulfonyl 11-(1-piperazinyl)-dibenz[b,f]- 1,4-oxazepins werden in 50 ml Aceton gelöst, mit 1 g wasserfreiem Kaliumcarbonat und 2,24 g Athyljodid in 20 ml Aceton versetzt und während 3 Stunden unter Rühren auf Rückfluss erhitzt.
Anschliessend wird das Reaktionsgemisch im Vakuum eingedampft. Der Rückstand wird zwischen verdünnter Natronlauge und Äther verteilt, und die Ätherauszüge werden mit Wasser gewaschen und mit verdünnter Salzsäure erschöpfend ausgeschüttelt. Die sauren Auszüge werden mit konzentrierter Natronlauge alkalisch gestellt und mit Chloroform ausgeschüttelt. Die Chloroformauszüge werden mit Wasser gewaschen, mit Natriumsulfat getrocknet und im Vakuum zur Trockne eingedampft. Der Rückstand wird aus Aceton/Petroläther kristallisiert, wobei man 4,9 g 2-Dimethylaminosulfonyl-1 1- (4-äthyl-1-piperazinyl)dibenz[b,f]-1,4-oxazepin vom Schmelzpunkt 160-161 C erhält.
Beispiel 12
4,63 g desselben Ausgangsmaterials wie in Beispiel 11 werden in 80 ml Isopropanol gelöst und mit 1,6 g wasserfreiem Kaliumcarbonat versetzt, worauf unter Rühren und Erwärmen 3 g ss-Methoxyäthyl-p-toluolsulfonsäureester in 10 ml Isopropanol zugetropft werden. Nach beendeter Zugabe wird während 1 1/2 Stunden unter Rückfluss erhitzt. Anschliessend wird das Reaktionsgemisch im Vakuum eingedampft. Der Rückstand wird zwischen verdünnter Natronlauge und Äther verteilt, und die Ätherauszüge werden mit verdünnter Salzsäure erschöpfend ausgeschüttelt. Die sauren Ausu züge werden mit konzentrierter Natronlauge alkalisch gestellt und mit Ather ausgeschüttelt. Die mit Wasser gewaschenen Ätherauszüge werden mit Natriumsulfat getrocknet und im Vakuum eingeengt.
Der ölige Rückstand wird in der Wärme mit 1,2 g Maleinsäure in Aceton gelöst und auf Zusatz von Äther kristallisiert.
Man erhält 4,9 g 2-Dimethylaminosulfonyl-1 1- (4-ss-methoxyäthyl-1-piperazinyl)- dibenz[b,f] -1,4-oxazepin-Maleat vom Schmelzpunkt 124-140 C (Zers.).
Beispiel 13
4 g gemäss Beispiel 8 erhaltenen 2-Dimethylaminosulfonyl-1 1- (4-ss-hydroxyäthyl-1-piperazinyl) dibenz[b,f]-1,4-oxazepins werden mit 30 ml absolutem Pyridin und 15 ml Acetanhydrid vermischt und während 1 Stunde bei Zimmertemperatur stehengelassen und dann auf dem Dampfbad kurz erwärmt. Das Reaktionsgemisch wird hierauf im Vakuum eingedampft, und der Rückstand wird mit Wasser versetzt. Die basischen Anteile werden unter Kühlen mit konzentrierter Natronlauge gefällt und erschöpfend mit Äther extrahiert. Die Sither- phase wird mit Wasser gewaschen, mit Natriumsulfat getrocknet und eingedampft. Der Rückstand wird in Aceton gelöst und mit 1,8 g Maleinsäure versetzt.
Nach starker Konzentration und auf Zusatz von Äther fallen Kristalle an, welche aus Aceton/Äther umkristallisiert werden. Man erhält 3 g 2-Dimethylamlnosulfonyl-1 1-(4-ss- acetoxyäthyl-1 -pip erazinyl)- dibenz(b,f] -1,4-oxazepin-Maleat vom Schmelzpunkt 155-1580 C.

They have a beneficial effect on the central nervous system and can be used, among other things, as analgesics, neuroleptics, sedatives and, in particular, as neuroleptic antidepressants and as antiemetics.
In the case of compounds according to formula I, in which R2 and R2 are methyl groups, the antiemetic effect is the main focus. This is shown pharmacologically in a strong apomorphine-antagonistic effect in dogs and rats as well as a low cataleptic and motility-suppressing effect compared to the apomorphine-antagonistic effect. The compounds obtained according to Examples 1 and 2, namely 2-dimethylaminosulfonyl-1 1 - (4-methyl-1-piperazinyl) dibenz [b, f] -1,4-oxazepine and 2-dimethylaminosulfonyl-1 1, are particularly antiemetically effective (4-methyl-1-piperazinyl) - dibenzo [b, f] -1,4-thiazepine and their acid addition salts.
Other compounds, in particular those with the 11-position 1-piperazinyl radical, simultaneously show the pattern of action typical of neuroleptics and antidepressants, the antidepressant active component being pharmacologically z. B. can be seen from an antagonism to tetrabenazine observed in rats.
example 1
A solution of 5.2 g of the crude 2-chlorosulfonyl-1 1- (4-methyl-1-piperazinyl) -dibenz [b, f] -1,4oxazepine obtained in the manner described below in 80 ml of chloroform is stirred at room temperature and 50 ml of an approx. 10% solution of dimethylamine in toluene are added dropwise. The reaction mixture is then stirred for a further 2 hours at room temperature and then heated to 400 ° C. for 1 hour with stirring.
The reaction mixture is then evaporated to dryness in vacuo, and the residue is taken up in dilute acetic acid. The acetic acid extracts are clarified with activated charcoal and made alkaline with concentrated ammonia solution. The precipitated base is taken up in benzene. The benzene solution is washed three times with water, dried with sodium sulfate and evaporated. The residue is dissolved in benzene and filtered through basic aluminum oxide. The residue obtained after evaporation of the solvent is crystallized from acetone / petroleum ether, 3.2 g of 2-dimethylaminosulfonyl-11- (4-methyl-1-piperazinyl) -dibenz [b, f] -1,4-oxazepine having a melting point 148-150 C.
The 2-chlorosulfonyl-1 1- (4-methyl-1-piperazonyl) -dibenz [b, f] 1,4-oxazopine used as starting material is obtained as follows:
15.4 g of 2-amino-1 1- (4-methyl-1-piperazinyl) - dibenz [b, f] -1,4-oxazepine (melting point 153-156 C) are dissolved in 50 ml of glacial acetic acid and 15 ml of 38 Dissolved 0 / above hydrochloric acid and diazotized in the usual way at 0 to 5 C with a solution of 3.6 g of sodium nitrite in 6 ml of water. If the procedure is analogous to that in Example 1, the corresponding starting materials continue to give z. B. the compounds according to formula I mentioned in the table below. In the table, Z, Rt, R2 and R3 have the meaning given earlier. In the column on the right, acetone means acetone, Ae ether, CH chloroform, Me methanol and Pe petroleum ether.
