Verfahren zur Herstellung 11-basisch substituierter Dibenzolb, fl 11, 41thiazepine
Gegenstand der Erfindung ist ein Verfahren zur Herstehung von Amidinen der Dibenzo [bf] [1, 4] thiazopin-Reihe der Formel :

von Additionssalzen oder von quaternären Ammo nivmderivaten davon. In der Formel I bedeutet Z ein Schwefelatom oder eine Sulfinylgruppe (-SO-).
Rl und R2 sind gleich oder verschieden und bedeuten Wasserstoff, unsubstituierte oder im Aryles-t Substituenten von gleicher Art wie R3 enthaltende Aryloder Aralkylgruppen, Alkenyl-oder Alkylreste mit 1 bis 5 C-Atomen, die gegebenenfalls gemeinsam mit dem Stickstoffatom einen Ring bilden, welcher als weitere Heteroatome O, S oder N enthalten kann, wobei das N seinerseits Wasserstoff oder eine Alkyl-, Hydroxyalkyl-oder Alkoxyalkylgruppe trägt, oder schliesslich unsubstituierte oder am Stickstoffatom alkylierte Amino- oder Aminoalkylgruppen.
R3 und R4 sind gl'eich oder verschiedlen und bedeuten Wasserstoff, Halogenatome, Hydroxygruppen, 1 bis 3 C Atome enthaltende Alkyl-, Aqkoxy-oder Alkylmer- captogruppen odeur Trifluormethylgruppen.
Verbindungen der Formel I werden erfindungs- gemäss erhalten, wenn man ein Thiazepinderivat der Formel :

worin Z, R3 und R4 die oben genaninte Bedeutung besitzen und worin A ein Halogenatom, vorzugsweise ein Chlaratom, oder eine höchstens 3 C-Atome aufweisende Alkoxy-oder Alkylthiogruppe darstellt, mit einer Verbindung der Formel HNRtR2, beispiels- weise Diäthylamin, Piperidin, Morpholin, N-Methyl- piperazin, geradkettigen oder verzweigtketti'gen, gegebenenfalls alkylierten Alkylendiaminen und dergleichen, umsetzt.
Die Umsetzung des Imidoäthers, Imidothioäthers oder des Imid'halogenids mit dem Amin erfolgt zweck mässig, indem man d'ie Komponenten, je nach ihren physikalischen Eigenschaften eventuell unter Verwendung eines inerten Verdünnungsmittels, wie Di- oxan, Xylol, Mesitylen, Decali'n und dergleichen wäh rend einigen Stunden bis zu 3 Tagen auf eine Temperatur von vorzugsweise über 150 C erhitzt, seii es durch Rückflusserhitzung oder unter Verwendung des Einschlussrohres. Das Amin wird dabei vorzugsweise in mindestens dreifachem molarem tXberschuss verwendet.
Die Reaktion wird oftmals durch Säure kata lysiext, wofür dem Reaktionsgemisch einige Tropfen Eisessig (z. B. 5 Tropfen auf 10 g der Thiazepinverbindung) zugesetzt werden können. Nach Einengen des Reaktionsgemisches verteilt man den Rückstand zweckmässig zwischen Ather und Wasser und entzieht die gebildete Base z. B. durch Extraktion mit verdünnter Salzsäure oder Essigsäure.
Aus der nötigen falls mit Kohle geklärten Hydrochlorid- oder Acetatlösung kann man die Base mit Ammoniak ausfällen und, falls sie gut kristallisiert und in Ather schwer löslich ist, direkt durch Abfitrieren isolieren, andernfalls in Äther aufnehmen und die ätherische Lösung in üblicher Weise durch Auswaschen mit Wasser und Trocknen mit Natriumsulfat aufarbeiten. Die weitere Reinigung erfolgt z. B. durch Umkristallisieren oder Hochvakuumdestillation.
Es vaste'hot sich, dass man die basische Seitenkette, soweit Rz und Ruz nicht Oeichzeitig Wdsserstoff bedeuten, auch schrititweise einführen kann, indem man die Thiazepinverbindung der Formel II zunächst mit Ammoniak oder einem primären Amin umsettzt und Rt und/oder R2 nachtrdglich eibfiihrt.
Weitere N-Atome der basischen Seitenkette kaon- nen gegebenenfalls ebenfalls nacnträgllch alkyliert werden.
Diejenigen Produite, in welchen Z eine Sulfinylgruppe bedeutet, können auch dadurch erhalten werden, dal3 man ein entsprechendes in 5-Stellung sauer- stofffreies Thiazepinderivat in an sich bekannter Weise oxydiert.
Die nach diesem Verfahren erhalienen Basen sind gelb, in vielen Fällen kristallisierbar, sonst im Hochvakuum unzersetzt destillierbar, und besitzen schon auf Grund der Amidingruppierung, abgesehen von allfälligen weiteren basischen Stickstoff- atomen, genügende Basenstärke, um mit anorgani suc'hein oder organischen Säuren, beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Salpetersäure, Phosphorsäure, Essigsäure, Oxalsäure, Malonsäure, Bernsteinsäure, Maleinsäure, Äpfelsäure, Weinsäure, Toluolsulfonsäure und dergleien} in Wa'sser beständige Additionssalze zu bilden, in welcher Form die Produkte ebenfalls verwendet werden können.
Um die quaternären Ammoniumderivate der Ver- bindungen gemäss Formel I zu erhälten, kann man entweder Ausgangsstoffe verwenden, die bereits quaternäre Stickstoffatome aufweisen, oder man kann die der Quaternisierung zugänglichen Stickstoffatome nach erfolgter Bildung der Basen (I) nachträglich in an sich bekannter Weise quaternisieren, beispielsweise durci Behandlung mit einem Dialkylsulfat, Alkylhalogenid oder SulfousdurealkylQster.
Die als Ausgangsstoffe verwendeten Verbindungen der Formel II erhät man beispielsweise dlarch thermische Cyclisierung entsprechend substituierter o-Amino-o'-carboxy-diphenylsulfide zum Lactam, dessen tautomere Form der Formel II entspricht, worin A eine Hydroxylgruppe ist. Durch Behandeln des Lactams einerseits mit Phosphorpentasulfid in siedendem Pyridin kann man das in Alkalilauge isliche, gelbe Thiolactam (Formel II ; A = SH ; tautomere For), und daraus durch Alkylierung mit Alkali und Oìalkylsulfat den Imidothioäther (Formel II ; A = S Alkyl), beide Stufen in guter Ausbeute erhalten.
Anderseits kann man aus dem Lactam durch Behandeln mit einem Gemisch aus Phosphoroxychlorid und Phosphorpentachlorid das Imidchlorid, und in entsprechender Weise die anderen Imidhalogenidze erhal- ten.
Die iu erfindungsgemässer Weise erhaltenen Basen, quaternären Ammoniumderivate und Silure- Additionssalze sind neue Verbindungen, dzie als Wirk staffe in Arzneimitteln Verwendung finden können, insbesondere als Analgetika, Chemotherapeutika, Antihistaminika, Antiallergika, Sedativa, Adrenolytika und Neuroplegika. Eìnzelne davon eignen sich zur BchandDung psychotischer Zustände.
Beispielsweise zeigt das gemäss Beispiel 23 erhal- tene 2-Chlor-11-(N-methyl)piperazino-dibenzo [b, f] [1, A.] thiaepin im Tierversuch die Eigenschaften eines Heurol'eptikums mit stark motilitätsdämpfender Wirkung sowie kataleptischen und apomorphinantagonistischen Eigenschaften. Die motilitätsdämpfende Wirkung wurde einerseits durch Messung der Laufaktivität bei Mäusen nach der Methode von Caviezel und Baillod (Pharm. Acta Helv. 33, 469 (1958), anderseits im Open-field -Test an Ratten nach der Methodç von Janssen et al. (Psychopharmacologia 1, 389 (1960) an je 10 Tieren bestimmt.
Der ermittelte Durohschnittswert wird in der folgenden Tabelle I mit den entsprechenden Werten finir bekannte Neurolep- tika verglichen. In dieser sind ferner Vergleichrwerte fiiez die akute Toxizität an der Maus sowie finir die kataleptische Wirkung enthalten. Die letztre wurde an Ratten geprüfk, die man'in versclhiedenen Zeit- abstäadea nach s. c. Injektion verschiedener Substanzmengen mit beiden Vorderpfoten und eine 7 çm hohe Säule legte, wobei die Verharrungsdauer gemessen wurde.
Die in Tabelle I angegebenen Zahlenwerte entsprechen den graphisch ermittelten Substanzmengen, die 180 Minuten nach Injektion im Mittel von 10 Tieren eine Verharrungsdauer von 30 Sekunden bewirkten.

Beispiel 1
3, 5 g 11-Chlor-dibenzo [b, f] [1, 4] thiazepin werden ç in 40 ml absolutem Xylol gelöst, mit 4, 3 g Piperidin versetzt und 5 Stunden auf Rückfluss erhitzt. Das Reaktionsgemisch wird mit Wasser versetzt und mit konzeretrierter Natronlauge alkalisch gemacht. Die XylolsChicht wird a'bgetrennt, mit Wasser neural ge- waschen und mit Salzsäure extrahiert.
Die sauren Auszüge werden mit konzentrierter Ammoniaklösung alkalisch gemacht und das ausgefallene O1 wird ausgeäthert. Die ätherische Lösung wird mit Wasser ge- waschen und eingedampft. Der Rückstand wird aus ither/Petrol'äther kristallisiert. Man erhält 3, 4 g 11 Piperidino-dibenzo [b, f] [1, 4]thiazepin vom Schmelzpunkt 133-134 C.
In analoger Weise wie im oben beschriebenen Beispiel erhält man aus den entsprechenden Ausgangsstoffen die in der nachfolgenden Tabelle II genannten Produkte. Darin bedeutet A den entsprechenden Rest der Ausgangsverbindung der Fo'rmel II.

Process for the preparation of 11-basic substituted dibenzene, fl 11, 41thiazepine
The invention relates to a process for the preparation of amidines of the dibenzo [bf] [1, 4] thiazopine series of the formula:

of addition salts or of quaternary ammo nivmderivaten thereof. In formula I, Z denotes a sulfur atom or a sulfinyl group (-SO-).
Rl and R2 are identical or different and denote hydrogen, unsubstituted or aryl or aralkyl groups, alkenyl or alkyl radicals with 1 to 5 carbon atoms, which optionally together with the nitrogen atom form a ring together with the arylene-t substituents of the same type as R3 may contain O, S or N as further heteroatoms, the N in turn bearing hydrogen or an alkyl, hydroxyalkyl or alkoxyalkyl group, or finally unsubstituted or amino or aminoalkyl groups alkylated on the nitrogen atom.
R3 and R4 are identical or different and denote hydrogen, halogen atoms, hydroxyl groups, alkyl, alkoxy or alkyl mercapto groups containing 1 to 3 C atoms or trifluoromethyl groups.

in which Z, R3 and R4 have the meaning given above and in which A is a halogen atom, preferably a chlorine atom, or an alkoxy or alkylthio group having at most 3 carbon atoms, with a compound of the formula HNRtR2, for example diethylamine, piperidine, morpholine , N-methylpiperazine, straight-chain or branched-chain, optionally alkylated alkylenediamines and the like.
The reaction of the imidoether, imidothioether or the imide halide with the amine is expediently carried out by adding the components, depending on their physical properties, possibly using an inert diluent such as dioxane, xylene, mesitylene, decali and the like heated for a few hours up to 3 days to a temperature of preferably over 150 C, either by reflux heating or using the containment tube. The amine is preferably used in at least a three-fold molar excess.
The reaction is often catalyzed by acid, for which a few drops of glacial acetic acid (e.g. 5 drops per 10 g of the thiazepine compound) can be added to the reaction mixture. After concentrating the reaction mixture, the residue is expediently partitioned between ether and water and the base formed is removed, e.g. B. by extraction with dilute hydrochloric acid or acetic acid.
The base can be precipitated with ammonia from the hydrochloride or acetate solution, if it has been clarified with charcoal, and, if it is well crystallized and sparingly soluble in ether, it can be isolated directly by filtration, otherwise it can be taken up in ether and the ethereal solution washed out with it in the usual way Work up water and dry with sodium sulfate. Further purification takes place e.g. B. by recrystallization or high vacuum distillation.
It is evident that the basic side chain, if Rz and Ruz do not simultaneously mean hydrogen, can also be introduced step by step by first reacting the thiazepine compound of the formula II with ammonia or a primary amine and subsequently adding Rt and / or R2.
Further N atoms of the basic side chain can, if appropriate, also be alkylated subsequently.
Those products in which Z denotes a sulfinyl group can also be obtained by oxidizing a corresponding thiazepine derivative which is oxygen-free in the 5-position in a manner known per se.
The bases obtained by this process are yellow, in many cases crystallizable, otherwise they can be distilled without decomposition in a high vacuum, and due to the amidine grouping, apart from any other basic nitrogen atoms, they have sufficient base strength to react with inorganic suc'hein or organic acids, For example, hydrochloric acid, hydrobromic acid, sulfuric acid, nitric acid, phosphoric acid, acetic acid, oxalic acid, malonic acid, succinic acid, maleic acid, malic acid, tartaric acid, toluenesulfonic acid and the like} to form stable addition salts in water, in which form the products can also be used.
In order to obtain the quaternary ammonium derivatives of the compounds according to formula I, either starting materials can be used which already have quaternary nitrogen atoms, or the nitrogen atoms accessible to quaternization can be quaternized subsequently in a manner known per se after the bases (I) have been formed, for example by treatment with a dialkyl sulfate, alkyl halide or sulfousdurealkyl ester.
The compounds of formula II used as starting materials are obtained, for example, by thermal cyclization of appropriately substituted o-amino-o'-carboxy-diphenyl sulfides to the lactam, the tautomeric form of which corresponds to formula II, in which A is a hydroxyl group. By treating the lactam on the one hand with phosphorus pentasulfide in boiling pyridine, the yellow thiolactam (formula II; A = SH; tautomeric form) which is in alkali solution can be obtained, and from it the imidothioether (formula II; A = S alkyl) by alkylation with alkali and olalkyl sulfate. obtained both stages in good yield.
On the other hand, the imide chloride can be obtained from the lactam by treatment with a mixture of phosphorus oxychloride and phosphorus pentachloride, and the other imide halide salts in a corresponding manner.
The bases, quaternary ammonium derivatives and silicic addition salts obtained according to the invention are new compounds that can be used as active substances in drugs, in particular as analgesics, chemotherapeutics, antihistamines, antiallergics, sedatives, adrenolytics and neuroplegics. Some of them are suitable for the formation of psychotic states.
For example, the 2-chloro-11- (N-methyl) piperazino-dibenzo [b, f] [1, A.] thiaepin obtained according to Example 23 shows the properties of a heuroleptic with a strong motility-suppressing effect as well as cataleptic and apomorphine antagonistic properties. The motility-suppressing effect was on the one hand by measuring the running activity in mice according to the method of Caviezel and Baillod (Pharm. Acta Helv. 33, 469 (1958), on the other hand in the open-field test on rats according to the method of Janssen et al. (Psychopharmacologia 1, 389 (1960) on 10 animals each.
The average value determined is compared in Table I below with the corresponding values for known neuroleptics. This also contains comparative values for the acute toxicity in mice and for the cataleptic effect. The latter was tested on rats which were given in different periods of time according to s. c. Injection of various amounts of substance with both front paws and a 7 çm high column was placed, the duration of which was measured.
The numerical values given in Table I correspond to the graphically determined amounts of substance which, 180 minutes after injection, caused an average of 10 animals to persist for 30 seconds.

3.5 g of 11-chloro-dibenzo [b, f] [1, 4] thiazepine are dissolved in 40 ml of absolute xylene, mixed with 4.3 g of piperidine and heated to reflux for 5 hours. The reaction mixture is mixed with water and made alkaline with concentrated sodium hydroxide solution. The xylene layer is separated, neural washed with water and extracted with hydrochloric acid.
The acidic extracts are made alkaline with concentrated ammonia solution and the precipitated O1 is extracted with ether. The ethereal solution is washed with water and evaporated. The residue is crystallized from ither / petroleum ether. 3.4 g of 11-piperidino-dibenzo [b, f] [1,4] thiazepine with a melting point of 133-134 ° C. are obtained.
