Verfahren zur Herstellung basisch substituierter Heterocyclen Die Erfindung betrifft ein Verfahren zur Herstellung von eine Sulfonamidgruppe enthaltenden -11-basisch substituierten Dibenz[b,f]-1,4-,oxazepinen, Dibenzo[b,fl-1,4-thiazepinen und Dibenzo[b,e]-1,4-diazepiwn der Formel:

sowie von Säure-Additionssalzen davon. In Formel I stellt Z ein Sauerstoffatom, ein Schwefelatom oder eine Irriinogruppe (-NH-) dar.
R, ist Wasserstoff, niedriges Alkyl, niedriges Hy- droxyalkyl, welches acyliert sein kann, oder Alkoxy- alkyl mit höchstens<B>5</B> C-Atomen. Unter niedrigem Alkyl usw. wird solchzs mit<B>1</B> bis<B>3</B> C-Atomen ver standen.
Die gewünschten Produkte werden erhalten, wenn man eine Verbindung der Formel:

Als ein mit dem Wasserstoff von Aminen abspalt- barer Rest, welcher kovalent oder gegebenenfalls auch <B>C</B> ionisch an das Kohlenstoffatom gebunden sein kann, kommt vorzugsweise ein Halogenatom, die Sulfhydrit- Cyruppe, eine gegebenenfalls aktivierte Alkoxy- oder Alkylmercaptogruppe, beispielsweise die Methoxy-, Tliomethyl- oder p-Nitrobenzylthiogruppe, oder ein Tosylrest in Betracht.
Ausgangsstoffe der Formel<B>11</B> erhält man beispiels weise durch überführen von Lactamen der Formel:

Die in der beschriebenen Weise erhaltenen Basen sind in den meisten Fällen kristaDisierbar, sonst<B>im</B> Hochvakuum unzersetzt destillierbar, und bilden mit anorganischen und organischen Säuren, beispielsweise Salzsäure, Brorrrwasserstoffsäure. Schwefelsäure, Salpe tersäure, Phosphorsäure. Essigsäure, Oxalsäure, Wein säure, Toluolsulfonsäure und dergleichen, in Wasser beständige Additionssalze, in welcher Form die Pro dukte ebenfalls verwendet werden können.
Die in der beschriebenen Weise erhaltenen Basen und ihre Säure-Additionssalze sind neue Verbindungen, dieals Wirkstaffe in Arzneimitteln Verwendung finden. Sie üben eine günstige Wirkung auf das Zentralnerven system aus und fallen als Neuroleptika, Sedativa und insbesondere als Antiemetika in Betracht.
<I>Beispiel<B>1</B></I> oxo-dibenzo[b,f]-1,4-t#iazepin 4,5 g 2-Dimethvlaminosulfonyl-10.11-dih.ydro-11 (Smp. 283'-284'C) wer- <B>-</B> den mit 40 n-il Phosphoroxychlorid und<B>1,3</B> nil N,N- Dimethylanflin während 4.5 Stunden unter Rückfluss erhitzt.
Hierauf wird das überschüssige Phosphoroxy- chlorid im Vakuum vollständig abdestiffiert, und der Rückstand wird in Xylol gelöst. Die Xylollösung wird auf Eis/M'asser gegossen, zweimal mit verdünnter Salzr säure und einmal mit Wasser ausgeschüttelt und an schliessend mit Natriumsulfat getrocknet. Die Xylol- lösung wird hierauf im Vakuum auf etwa<B>100</B> ml eingeengt, mit<B>8</B> ml N-Methylpiperazin versetzt und während 4 Stunden unter Rückfluss erhitzt.
Das Reak tionsgemisch wird anschliessend mit verdünnter Natron lauge und Wasser versetzt. Die Xylolphase wird abge trennt und mit verdünnter Salzsäure ausgeschüttelt. Die sauren Auszüge werden mit konzentrierter Ammoniak- lösung alkalisch gestellt, und die abgeschiedene Base wird mit Chloroform extrahiert. Die mit Natriumsulfat getrockneten Chloroformauszüge werden im Vakuum eingedampft, und der Rückstand wird aus Aceton,! Petroläther kristallisiert.
Man erhält 4,9 g 2-Dimethyl- aminosulfonyl-1 1-(4-methvl-I-piperazinyl)-dib.--nzo[b,fl- 1,4-thiazepin in Form von schwach aelblichen Nadeln vom Schmelzpunkt 192-193' <B>C.</B>
Das als Ausean2smaterial verwendete 2-Dimethyl- anünosulfonyl-10,1 1-dihvdro-1 1-oxo-dibenzo[b,fl- 1,4- thiazepin erhielt man durch 30minutiges Erhitzen von 2-Amino-2'-carboxy-4'-dimethylaminosulfonyl-diphenyl- sulfid (Smp. 224-227' <B>C;
</B> Zers.) in Diphenyläther. Das 2-Anlino-2'-carboxy-4'-dimethyla-minosu'fonyl-diphenyl- sulfid wurde durch 6stündiges Erhitzen von 2-Brom-5- dimethylaminosulfonylb.e,nzoesäure und 2-Aminothio- phenol mit wasserfreicm Kaliumcarbonat in Isoamyl- alkehol erhalten.
<I>Beispiel 2</I> <B>1,8 g</B> 2-iDimethylamin < ),sulfonyl-10,11-dihydro-II- oxo-dibenz[b,f]-1,4-oxazepin (Smp. 243-2450C) wer den mit 20 m] Phosphoroxychlorid und<B>0.6</B> nil N,N-Di- methylanilin während 4 Stunden unter Rückfluss erhitzt. Hierauf wird das überschüssige Phosphoroxychlorid im Vakuum vollständig abdestilliert. und der Rückstand wird in Xylol gelöst.
Die Xylollösung wird auf Eis/ Wasser gegossen, zweimal mit verdünnter Salzsäure und einmal mit Wasser ausgeschüttelt und anschliessend mit Natriumsulfat getrocknet. Die Xylollösung wird hierauf im Vakuum auf etwa<B>50</B> ml eingeengt, nüt <B>3</B> ral N- Methylpiperazin versetzt und während 4 Stunden unter Rückfluss erhitzt.
Das Reaklionsgernisch wird anschlie ssend mit verdünnter Natronlauge und Wasser venetzt. Die Xylolphase wird abgetrenni und mit verdünnter Salz säure ausgeschütelt. Die sauren Auszüge werden mit kon zentrierter Ammoniaklösung alkalisch gestellt. und die abgeschiedene Base wird mit Chloroform extrahiert.
Die mit Natriumsulfat getrockneten Chloroformauszüge wer den im Vakuum eingedampft, und der Rückstand wird aus Äther/.Petroläther krisiallisiert. Man erhält 1.,8 <B>g</B> 2-Dimethylanlinosulfonyl <B>-1</B>144<B>-</B> methyl <B>- 1 -</B> piperazinyl)- dibenz[b,f]-1,4-oxazepin vom Schmelzpunkt 149-150c <B>C.</B>
,Das als Ausgangsmaterial verwendete 2-Dimethyl aminosulfonyl-10,1,1-dihydro-II-oxo-di-benz[b.f]-1.4- oxazepin erhielt man durch 20stündiges Erhitzen von 2-Brom-5-dimethylaminosulfonylbenzoesäure-2'-hydroxv- benzamid (Smp. 168-17V <B>C)</B> mit<B>1</B> Äquivalent Natron lauge in Wasser.
Das 2-Brom-5-dirnethvlaminosulfonyl- benzoesäure-2'-hydroxy-benzamid wuräe durch Um setzen von 2-Brom-5-dimethylaminosulfonyl-benzoe- säurechlorid und 2-Aminophenol in Äther mit wässriger Natriumbicarbonatlösung erhalten.
Bei analogem Vorgehen wie in den vorerwähnten Beispielen erhält man weiterhin die in der nachfolgenden Tabelle erwähnten Produkte entsprechend Formel<B>1,</B> In der Tabelle haben Z nach R, die früher genannte Be deutung. In der Kolonne rechts bedeutet Ac Aceton, Ae Äther, Ch Chloroform und Pe Petroläther.

The bases obtained in the manner described are in most cases crystallizable, otherwise they can be distilled without decomposition in a high vacuum, and form hydrochloric acid with inorganic and organic acids, for example hydrochloric acid. Sulfuric acid, nitric acid, phosphoric acid. Acetic acid, oxalic acid, tartaric acid, toluenesulfonic acid and the like, addition salts stable in water, in which form the products can also be used.

The xylene solution is poured onto ice / water, extracted twice with dilute hydrochloric acid and once with water and then dried with sodium sulfate. The xylene solution is then concentrated in vacuo to about 100 ml, mixed with 8 ml of N-methylpiperazine and heated under reflux for 4 hours.
The reaction mixture is then mixed with dilute sodium hydroxide solution and water. The xylene phase is separated off and extracted with dilute hydrochloric acid. The acidic extracts are made alkaline with concentrated ammonia solution and the deposited base is extracted with chloroform. The chloroform extracts, dried with sodium sulfate, are evaporated in vacuo, and the residue is made from acetone! Petroleum ether crystallizes.
4.9 g of 2-dimethylaminosulfonyl-1 1- (4-methvl-I-piperazinyl) -dib .-- nzo [b, fl-1,4-thiazepine are obtained in the form of pale yellow needles with a melting point of 192- 193 '<B> C. </B>
The 2-dimethyl-anünosulfonyl-10,1 1-dihvdro-1 1-oxo-dibenzo [b, fl-1,4-thiazepine used as Ausean2smaterial was obtained by heating 2-amino-2'-carboxy-4 'for 30 minutes. dimethylaminosulfonyl diphenyl sulfide (m.p. 224-227 'C;
</B> decomp.) In diphenyl ether. The 2-anlino-2'-carboxy-4'-dimethyla-minosu'fonyl-diphenyl-sulfide was made by heating 2-bromo-5-dimethylaminosulfonylb.e, nzoic acid and 2-aminothiophenol with anhydrous potassium carbonate in isoamyl- get alkehol.

The 2-dimethyl aminosulfonyl-10,1,1-dihydro-II-oxo-di-benz [bf] -1.4-oxazepine used as starting material was obtained by heating 2-bromo-5-dimethylaminosulfonylbenzoic acid-2'-hydroxv for 20 hours - Benzamide (melting point 168-17V <B> C) </B> with <B> 1 </B> equivalent of sodium hydroxide solution in water.
