Verfahren zur Herstellung basisch substituierter Heterocyclen Die Erfindung betrifft ein Verfahren zur Herstellung von eine Sulfonamidgruppe enthaltenden 11-basisch substituierten Dibenz[b,f]-1,4-oxazepinen, Dibenzo[b,f]-1,4-thiazepinen und Dibenzo[b,e]-1,4-diazepinen der Formel:

sowie von Säure-Additionssalzen davon.
In Formel I stellt Z ein Sauerstoffatom, ein Schwefelatom oder eine Iminogruppe (-NH-) dar.
R, ist Wasserstoff, niedriges Alkyl, niedriges Hy- droxyalkyl, welches acyliert sein kann, oder Alkoxy- alkyl mit höchstens 5 C-Atomen. Unter niedrigem Alkyl usw. wird solches mit 1 bis 3 C-Atomen ver standen.
Die gewünschten Produkte werden erhalten, wenn man Säureamide oder Thioamide der Formel:

worin Z und R, die obengenannte Bedeutung haben und Y ein Sauerstoff- oder Schwefelatom darstellt, durch intramolekulare Kondensation zum Ringschluss bringt.
Eine rein thermische Kondensation gelingt in der Regel bei den Säureamiden nicht, eher dagegen bei den Thioamiden, die man zum Beispiel durch Behandeln der Säureamide mit Phosphorpentasulfid erhält und vor der nachfolgenden Kondensation nicht zu 'isolieren braucht.
Insbesondere bei den Säureamiden ist es zweck- mässig, in Gegenwart. von Kondensationsmitteln, wie Phosphorpentachlorid, Phosphoroxychlorid, Phosgen, Polyphosphorsäure und dergleichen, zu arbeiten.
Es ist anzunehmen, dass der Ringschluss dabei zum Teil über Zwischenstufen, wie Imidchloride. Amidchloride, Imido- phosphate, Amidophosphate oder salzartige Derivate davon, die in der Regel nicht fassbar sind, verläuft. Die Kondensation der Thioamide kann durch Gegenwart von Quecksilbersalzen oder durch intermediäre Bildung von gegebenenfalls aktivierten Imidothioäthern begün stigt werden.
Erwärmen und gegebenenfalls Benützung eines inerten Verdünnungsmittels sind angezeigt, beim Arbeiten mit Phosphoroxychlorid und Phosphorpenta- cWorid auch Zusatz katalytischer Mengen von Dimethyl- formamid oder Dimethylanilin.
,Die in der beschriebenen Weise erhaltenen Basen sind in den meisten Fällen kristallisierbar, sonst im Hochvakuum uniersetzt destillierbar, und -bilden mit anorganischen und organischen Säuren, beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Salpe tersäure, Phosphorsäure, Essigsäure, Oxalsäure, Wein- säure, Toluolsulfonsäure und dergleichen, in Wasser be ständige Additionssalze,
in welcher Form die Produkte ebenfalls verwendet werden können.
Die in der beschriebenen Weise erhaltenen Basen und ihre Säure-Additionssalze sind neue Verbindungen, die als Wirkstoffe in Arzneimitteln Verwendung finden. Sie üben eine günstige Wirkung auf das Zentralnerven system aus und fallen als Neuroleptika, Sedativa und insbesondre als Antiemetika in Betracht.
<I>Beispiel 1</I> 4,34 g 2-Amino-2'-(4"-methyl-I"-piperazinyl-car- bonyl)-4'-dimethylaminosulfonyl-diphenylsulfid werden mit 20 ml Phosphoroxychlorid und 1 ml N,N-Dimethyl- anilin während 3 Stunden unter Rückfluss gekocht, worauf das Reaktionsgemisch im Vakuum zur Trockne eingedampft wird. Der Rückstand wird mit Xylol nach getrocknet und dann zwischen Benzol und verdünnter Salzsäure verteilt.
Aus den salzsauren Extrakten wird die Base mit konzentrierter Ammoniaklösung freige setzt und in Benzol aufgenommen. Die Benzollösung wird mit verdünnter Essigsäure erschöpfend extrahiert, und die essigsauren Extrakte werden mit Kohle geklärt. Die basischen Anteile werden: unter Eiskühlung mit konzentrierter Ammoniaklösung freigesetzt und in Chloroform aufgenommen. Die Chloroformauszüge wer den mit Wasser gewaschen, mit Natriumsulfat getrocknet und eingedampft.
Der Rückstand wird in Äther gelöst und durch Aluminiumoxid filtriert. Der nach Abdamp fen des Lösungsmittels erhaltene Rückstand wird syste matisch aus Aceton/ÄtherJ'Petroläther kristallisiert, wobei man zunächst 0,7 g an unumgesetztem Ausgangs- material erhält. Aus den leichter löslichen Anteilen erhält man 0,78 g 2-Dimethylaminosulfonyl-11-(4- methyl-l-piperazinyl)-dibenzo[b,f]-1,4-thiazepin vom Schmelzpunkt 192-193 C.
Bei analogem Vorgehen wie in dem vorerwähnten Beispiel erhält man weiterhin die in der nachfolgenden Tabelle erwähnten Produkte entsprechend Formel 1. In der Tabelle haben Z und R1 die früher genannte Be deutung.
In der Kolonne rechts bedeutet Ac Aceton, Ae Äther, Ch Chloroform und Pe Petroläther.
