Die Erfindung betrifft ein Verfahren zur Herstellung von eine Nitrogruppe enthaltenden, 11-basisch substi tuierten Dibenz- [b, f]-1, 4-oxazepinen und Dibenzo- [b, f]1, 4-thiazepinen der Formel

In Formel 1 stellt Z ein Sauerstoff-oder Schwefelatom dar, Ri ist Wasserstoff, niedriges Alkyl, niedriges Hydroxyalkyl, welches acyliert sein kann, oder Alkoxyalkyl mit höch- stens 5 C-Atomen. Unter niedrigem Alkyl usw. wird solches mit 1 bis 3 C-Atomen verstanden.

worin Z die genannte Bedeutung hat, in die Thiolactame, gewünschtenfalls unter nachfolgender Alkylierung der letzteren, oder durch Umsetzen der Lactame (V) mit einem Halogenierungsmittel, wie Phosphoroxychlorid oder Phosphorpentachlorid, vorzugsweise in Gegenwart katalytischer Mengen von Dimethylanilin oder Dimethyl- formamid. Die Lactame (V) sind ihrerseits z. B. durch Ringschluss entsprechender o-Isocyanatodiphenyläther bzw. o-Isocyanatodiphenylsulfide mit Aluminiumchlorid erhältlich.
Je nach der chemischen Natur des Restes X sind die Verbindungen IV in den erhaltenen Reaktions- gemischen mehr oder weniger stark in die Nitriliumbzw. Imonium-Kationen dissoziiert, so dlass die Reaktionsgemische direkt für die Umsetzung mit dem Piperazin bzw. Piperazinderivat der Formel III verwendet werden können.
Zum Teil lassen sich die in dieser oder anderer Weise hergestellten Verbindungen der Formel IV in undissoziierter Form isolieren und liefern dann beim Auflösen in einem geeigneten, vorzugsweise polaren Lösungsmittel, gegebenenfalls unter Erwärmen und in Gegenwart von Piperazin bzw. des Piperazinderivats der Formel III, welches auch als Lösungsmittel dienen kann, die gewünschten Nitrilium-bzw. Imonium- Kationen (II). Kationen der Formeln II enthaltende Reaktionsgemische können ferner z. B. durch intramolekulare Ritter-Reaktion (Angriff einer Nitrilgruppe auf ein Phenyl-Kation) bei nitrierten o-Cyanodiphenyl- oxiden bzw.
o-Cyanodiphenylsulfiden, durch Beck- mannsche Umlagerung von nitriertem Xanthonoxim bzw. Thioxanthonoxim oder durch Schmidt-Reaktion von nitriertem Xanthon bzw. Thioxanthon mit Stickstoff- wasserstoffsäure erzeugt werden. Als anionoide Komponenten können in den genannten Reaktionsgemischen ausser denjenigen, welche sich vom Substituenten X der Formel IV herleiten, je nach der Bildungsweise der Kationen (II) zum Beispiel auch Anionen der Schwefel- säure, Toluolsulfonsäure, Phosphorsäure, Flusssäure, Borfluorwasserstoffsäure usw. auftreten.
Soweit nach diesem Verfahren Verbindungen gemäss Formel I erhalten wurden, in welchen Ri Wasserstoff ist, kann ein nicht Wasserstoff bedeutender Rest Rl nachträglich eingeführt werden, indem man das sekun- däre Amin mit einem reaktionsfähigen Ester eines Alkohols der Formel Rl-OH umsetzt, nötigenfalls nach vorausgehender oder unter gleichzeitiger Einwirkung eines basischen Katalysators oder Metallisierungsmittels, wie Natriumamid, Lithiumamid, Natriumhydrid, Butyl- lithium, Phenylnatrium, Natriumäthylat oder KaliumLt- butylat. Als Ester kommen solche anorganischer und organischer Säuren in Betracht, insbesondere Halogen wasserstoffsäureester.
Die in der beschriebenen Weise erhaltenen Basen sind in den meisten Fällen kristallisierbar, sonst im Hochvakuum unzersetzt destillierbar, und bilden mit anorganischen und organischen Säuren, beispielsweise
Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Sal petersäure, Phosphorsäure, Essigsäure, Oxalsäure, Wein säure, Toluolsulfonsäure und dergleichen, in Wasser be ständige Additionssalze, in welcher Form die Produkte ebenfalls verwendet werden können.
Die in der beschriebenen-Weise erhaltenen Basen und ihre Säureadditionssalze sind neue Verbindungen, die als Wirkstoffe in Arzneimitteln oder als Zwischenprodukte zur Herstellung von solchen Ve-rwendung finden. Sie üben eine günstige Wirkung auf das Zentral nervensystem aus und fallen insbesondere als Analgetika, Neuroleptika und Sedativa in Betracht. Einzelne Produkte besitzen eine wachstumshemmende Wirkung auf verschiedene Bakterien, Protozoen und Pilze und können daher als Chemotherapeutika Verwendung finden.
Das gemäss Beispiel, 1 erhaltene 2-Nitro-11- (4-methyl-1-piperazinyl)-dibenz- [b, f]-1, 4-oxazepin ; zeigt das für Neuroleptika typische Wirkungsbild, näm- lich Motilitätsdämpfung und kataleptische Wirkung. Die Motilitätsdämpfung wird durch Messung der Laufaktivität von Mäusen nach der Methode von Caviezel und Baillod [Pharm. Acta Helv. 33, 469 (1958)] erfasst.
Die kataleptische Wirkung wird dadurch ermittelt, dass man mit der Testsubstanz vorbehandelte Ratten mit den Vorderpfoten auf eine 7 cm hohe Säule legt und die Zeit bestimmt, während der die Tiere in dieser anomalen Stellung verharren. Das erwähnte 2-Nitro-11-(4-methyl-1-piperazinyl3-dibenz- [b, f]-1, 4-oxazepin vermag bei einer akuten Toxizität an der Maus von 52 mg/kg p. o. (LD 50) in einer oralen Dosis von 0, 04 mg/kg die Laufaktivität der Mäuse aus 50% der Kontrollen herabzusetzen und bewirkt in einer oralen Dosis von 0, 1 mg/kg, dass 50% der behandelten Ratten während 30 Sekunden in ihrer anomalen Stellung ver harren.
Beispiel 1
4, 9 g 2-Nitro-10, 11-dihydro-11-oxo-dibenz-[b, f]-1, 4oxazepin werden mit einer Mischung von 2 ml N, N Dimethylanilin und 60 ml Phosphoroxychlorid 4 Stunden unter Rückfluss erhitzt. Zur Entfernung des überschüssi- gen Phosphoroxychlorids wird das Reaktionsgemisch am Vakuum eingedampft. Der Rückstand wird mit Eis/ Wasser zersetzt und sofort mit Chloroform ausgeschüttelt. Die Chloroformauszüge werden mit verdünnter Salzsäure und Wasser gewaschen, mit Natriumsulfat getrocknet und im Vakuum zur Trockne eingedampft. Der kristallisierte Rückstand, bestehend aus rohem 2-Nitro-1l-chlor-dibenz- [b, f]-1, 4-oxazepin, wird mit 6 ml N-Methylpiperazin in 200 ml Xylol während 6 Stunden unter Rückfluss erhitzt.
Die organische Phase wird hierauf mit Wasser und verdünnter Salzsäure ausgeschüttelt. Die sauren Auszüge werden mit konzentrierter Natronlauge alkalisch gestellt, worauf die ausgeschiedene Base mit Chloroform extrahiert wird. Die Chloroformauszüge werden mit Wasser gewaschen, mit Natriumsulfat getrocknet und zur Trockne eingeengt.
Der Rückstand ergibt nach Kristallisation aus Chloro form/Äther 4, 7 g 2-Nitro-11- (4-methyl-1-piperazinyl)- dibenz- [b, f]-1, 4-oxazepin in Form von gelben Nadeln vom Schmelzpunkt 192 bis 193 C.
Beispiel 2
2, 0 g 2-Nitro-10, ll-dihydro-ll-oxo-dibenzo- [b, f]1, 4-thiazepin werden mit 15 ml Phosphoroxychlorid und 1 ml N, N-Dimlethylanilin während 5 Stunden auf Rück fluss erhitzt, worauf man, das Reaktionsgemisch im Vakuum zur Trockne einengt. Der Rückstand wird mit Xylol nachgetrocknet und anschliessend mit 15 ml N Methylpiperazin und 10 ml Dioxan während 16 Stunden auf Rückfluss erhitzt. Nach Einengen im Vakuum zur Trockne wird der Rückstand zwischen Äther und Ammoniakwasser verteilt. Die Ätherphase wird zweimal mit Wasser gewaschen und mit verdünnter Essigsäure ausgeschüttelt. Aus den essigsauren Extrakten wird die Base mit konzentrierter Ammoniaklösung freigesetzt und in Äther aufgenommen.
Die Ätherphase wird viermal mit Wasser gewaschen, mit Natriumsulfat getrocknet und eingeengt. Der erhaltene harzige Rückstand wird in Äther gelöst, und die Ätherlösung wird durch Alu miniumoxid filtriert und eingedampft. Der Rückstand wird aus Aceton/Petroläther kristallisiert, wobei man 1, 7 g 2-Nitro 11-(4-methylel-pipeázinyl)-dibenzo-[b, f] 1, 4-thiazepin in Form von gelben, verfilzten Nadeln vom Schmelzpunkt 141 bis 142 C erhält.
Bei analogem Vorgehen wie in den vorerwähnten Beispielen erhält man aus den entsprechenden Ausgangsprodukten weiterhin z. B. die nachfolgend angegebenen Verbindungen entsprechend Formel I : 2-Nitro-ll- (l-piperazinyl)-dibenz- [b, f]
1, 4-oxazepin vom Schmelzpunkt 190 bis 192 C (aus Aceton/Petroläther);
2-Nitro-11-(4-ss-hydroxyäthyl-1-piperazinyl) dibenz-[b,f]-1,4-oxazepin-Maleat vom Schmelz punkt 155 bis 156 C (aus Methanol/Aceton) ;

The chloroform extracts are washed with water, dried with sodium sulfate and evaporated to dryness.
After crystallization from chloroform / ether, the residue gives 4.7 g of 2-nitro-11- (4-methyl-1-piperazinyl) -dibenz- [b, f] -1, 4-oxazepine in the form of yellow needles with a melting point 192 to 193 C.
Example 2
2.0 g of 2-nitro-10, ll-dihydro-ll-oxo-dibenzo- [b, f] 1,4-thiazepine are refluxed with 15 ml of phosphorus oxychloride and 1 ml of N, N-dimethylaniline for 5 hours , whereupon the reaction mixture is concentrated to dryness in vacuo. The residue is dried with xylene and then refluxed for 16 hours with 15 ml of N methylpiperazine and 10 ml of dioxane. After concentrating to dryness in vacuo, the residue is partitioned between ether and ammonia water. The ether phase is washed twice with water and extracted with dilute acetic acid. The base is released from the acetic acid extracts with concentrated ammonia solution and taken up in ether.
The ether phase is washed four times with water, dried with sodium sulfate and concentrated. The resinous residue obtained is dissolved in ether, and the ether solution is filtered through aluminum oxide and evaporated. The residue is crystallized from acetone / petroleum ether, 1.7 g of 2-nitro 11- (4-methylel-pipeázinyl) -dibenzo- [b, f] 1,4-thiazepine in the form of yellow, matted needles with a melting point of 141 up to 142 C.
