Verfahren zur Herstellung von 2,2 disubstituierten 3-Acyl-5 amino-thiazolidin-4-carbonsäuren und deren funktionellen Derivaten
Gegenstand der vorliegenden Erfindung ist ein Methodikverfahren zur Herstellung von 2,2-disubstituierten 3-Acyl-Sa-amino-thiazolidin-4-carbonsäuren und deren funktionellen Derivaten, das zur Darstellung von wertvollen Zwischenprodukten und insbesondere bei der erstmaligen synthetischen Herstellung der 7-Amino-cephalosporansäure und ihrer Derivate Anwendung fand und zu dieser eigenartigen Synthese besonders geeignet ist.

Derivate sind in erster Linie N-Acylverbindungen, worin Acylreste, insbesondere diejenigen von wirksamen N-Acylderivaten der 7-Amino-cephalosporansäure, wie der Thienylacetyl-, Cyanacetyl-, Chloräthylcarbamyl- oder Phenylacetylrest, oder leicht abspaltbare Acylreste, wie der Rest eines Halbesters der Kohlensäure, z. B. der tert.-Butyloxycarbonylrest, bedeuten.
Die Synthese dieser für die Herstellung wertvoller Arzneimittel wichtigen Verbindung und ihrer Derivate beruht auf der Idee, von einer 3,5-unsubstituierten 2,2disubstituierten Thiazolidin-4-carbonsäure, z. B. einer Verbindung der Formel I

Zu den oben erwähnten Verbindungen gelangt man, indem man in einer 2,2-disubstituierten 3-Acyl Sa-azido-thiazolidin-4-carbonsäure oder einem funktionellen Säurederivat davon die Azidogruppe zu einer Aminogruppe reduziert.
Wenn erwünscht, kann man in einer erhaltenen Verbindung einen Substituenten in einen anderen überführen, und/oder, wenn erwünscht, ein erhaltenes Isomerengemisch in die einzelnen Isomeren auftrennen.
Die Umwandlung der Azido- in die Aminogruppe kann reduktiv, z. B. mit Hilfe von katalytisch aktiviertem Wasserstoff in Gegenwart eines Katalysators, enthaltend ein Metall der Gruppe VIII des periodischen Systems, wie Palladium, unter Normaldruck oder erhöhtem Druck, oder mit chemischen Reduktionsmitteln, wie nascierendem Wasserstoff, erhalten, z.B.
durch die Einwirkung von Metallen, Metallegierungen oder Amalgamen auf wasserstoffabgebende Mittel, wie Zink, Zinklegierungen, wie Zinkkupfer, oder Zinkamalgam in Gegenwart von Säuren, wie organischen Carbonsäuren, z.B. Essigsäure, oder Alkoholen, wie Methanol oder Alkalimetallamalgamen, z.B. Natriumoder Kaliumamalgam, Erdalkalimetall-, z.B. Magnesiumamalgam, oder Aluminiumamalgam in Gegenwart von feuchten oder wasserstoffabgebenden Mitteln, wie feuchtem Äther, oder Niederalkanolen, wie Methanol, durchgeführt werden. Ferner kann man Metallhydride, wie Alkalimetall-aluminiumhydride, z. B. Lithium-aluminiumhydrid, Alkalimetall-trialkoxy-aluminiumhydride, z. B.
Lithium-tritert.-butyloxyaluminiumhydrid, oder Organo-zinn-hydride, wie Diisobutyl-zinnhydrid, sowie reduzierende Metallsalze, wie Chrom-II-verbindungen, wie Chrom-II-acetat oder Chrom-II-chlorid, verwenden.
Die obige Reaktion wird in An- oder Abwesenheit von Verdünnungsmitteln, besonders solchen, welche sich, sofern dies erwünscht ist, gegenüber den Reaktionsteilnehmern inert verhalten und/oder diese zu lösen vermögen, und/oder von katalytisch wirkenden Substanzen, unter Kühlen, bei Zimmertemperatur oder unter Erwärmen, wenn notwendig, in einem geschlossenen Gefäss, unter Druck und/oder in einer Inertgasatmosphäre, vorgenommen.
In erhaltenen Verbindungen können Substituenten nach an sich bekannten Methoden in andere übergeführt werden. Erhaltene Säurederivate, wie Ester, können in die freie Säure umgewandelt werden, ohne dass die Acylgruppe, besonders eine leicht abspaltbare Acylgruppe, wie die tert.-Butyloxycarbonylgruppe, in 3-Stellung entfernt wird. So lässt sich eine Carbodiphenylmethoxygruppe unter sauren Bedingungen, z.B. in Gegenwart von katalytischen Mengen einer Säure, wie Trifluoressigsäure, in eine freie Carboxylgruppe umwandeln. In eigenartiger Weise lässt sich sodann z. B.
eine mit einem 2,2,2-Trihalogenäthanol, besonders 2,2,2-Trichloräthanol, veresterte Carboxygruppe mittels reduzierender Mittel in die freie Carboxylgruppe überführen. Geeignete Reduktionsmittel sind chemische Reduktionsmittel, wie die oben erwähnten Metalle, Metallegierungen oder -amalgame in Gegenwart von wasserstoffabgebenden Mitteln, u. a. auch Alkalimetalle, z.B. Lithium, Natrium oder Kalium, oder Erdalkalimetalle, z. 13. Calcium, in flüssigem Ammoniak, gegebenenfalls unter Zugabe von Alkoholen, wie eines Niederalkanols. Ferner kann ein Ester mit einem 2,2,2-Trihalogenäthanol, insbesondere 2,2,2-Trichloräthanol auch durch Behandeln mit starkreduzierenden Metallsalzen, wie Chrom-II-verbindungen, z. B.
Chrom-II-chlorid oder Chrom-II-acetat, vorzugsweise in Gegenwart von wässrigen Medien enthaltend mit Wasser mischbare Lösungsmittel, wie Niederalkanole, Niederalkancarbonsäuren oder Äther, z.B.
Methanol, Äthanol, Essigsäure, Tetrahydrofuran, Dioxan, Äthylenglykol- dimethyläther oder Diäthylenglykoldimethyläther, in die freie Säure übergeführt werden.
Dabei kann die an sich eigenartige reduktive Spaltung eines 2,2,2-Trihalogenäthylesters gleichzeitig mit der reduktiven Überführung der Azidogruppe in die gewünschte Aminogruppe erfolgen.
In einer erhaltenen Verbindung mit einer freien Carboxylgruppe kann diese nach an sich bekannten Methoden in ihre funktionellen Derivate, wie ihre Ester, Amide, Hydrazide oder Azide, umgewandelt werden. So kann sie z.B. durch Behandeln mit einer Diazoverbindung, wie einem Diazoniederalkan, z.B.
Diazomethan oder Diazoäthan, oder mit einem Phenyldiazoalkan, z.B. Phenyldiazomethan oder Diphenyldiazomethan, oder durch Umsetzen mit einer zur Veresterung geeigneten Hydroxyverbindung, wie z.B.
einem Alkohol, sowie einer Phenolverbindung, oder einer N-Hydroxy-stickstoffverbindung, z. B. einer Hydroxamsäure, in Gegenwart eines Veresterungsmittels, wie eines Carbodiimids, z. B. Dicyclohexylcarbodiimid, sowie von Carbonyldiimidazol, oder nach irgendeinem anderen bekannten und geeigneten Veresterungsverfahren, wie Reaktion mit einem reaktionsfähigen Ester der Hydroxyverbindung, besonders eines Alkohols, und einer starken anorganischen Säure oder einer starken organischen Sulfonsäure, wenn erwünscht, in Gegenwart einer salzbildenden Base, verestert werden.
Eine funktionell abgewandelte Carboxygruppe in einer erhaltenen Verbindung kann nach an sich bekannten Methoden auch in eine andere funktionell abgewandelte Carboxygruppe übergeführt werden, z.B.
veresterte Gruppen durch Umesterung, wie Behandeln mit einer Hydroxyverbindung in Gegenwart eines Umesterungskatalysators. Ferner können Ester und insbesondere aktivierte Ester, wie z. B. Ester mit N-Hydroxystickstoffverbindungen, oder Anhydride, z.B. mit Chlorameisensäureester, wie -methylester, durch Umsetzen mit anderen Hydroxyverbindungen, wie Alkoholen oder Phenolen bzw. mit Ammoniak, mit höchstens sekundären Aminen oder Hydrazinen, in andere Ester bzw. in Amide oder Hydrazide übergeführt werden. In einer erhaltenen Amid- oder Hydrazidverbindung mit einem wasserstoffenthaltenden Stickstoffatom kann dieses nachträglich substituiert werden, z.
B. durch Behandeln mit einem Carbonsäure- oder Sulfonsäurederivat, wie mit einem Säurehalogenid, und/oder einem reaktionsfähigen veresterten Alkohol oder mittels anderen geeigneten Reagentien; ein N-unsubstituiertes Amid kann z. B. durch Dehydratisierung in ein Nitril umgewandelt werden.
Wenn notwendig, kann die freie Aminogruppe während diesen zusätzlichen Verfahrensmassnahmen vorübergehend, z.B. durch geeignete Acylierung, geschützt werden.
Erhaltene Gemische von Isomeren können nach an sich bekannten Methoden, z.B. durch fraktioniertes Kristallisleren oder Destillieren, durch Adsorptionschromatographie (Kolonnen- oder Dünnschichtchro- matographie) oder anderen Verfahren in die einzelnen Isomeren getrennt werden; erhaltene Racemate können durch Bilden eines Gemisches von diastereoisomeren Salzen mit optisch aktiven, salzbildenden Mitteln, Trennen des Gemisches in die diastereoisomeren Salze und Überführen der abgetrennten Salze in die freien Verbindungen oder in die Antipoden getrennt werden.
Das Verfahren umfasst auch diejenigen Ausführungsformen, wonach Ausgangsstoffe in Form von Derivaten, z. B. von Salzen, verwendet oder während der Reaktion gebildet werden.
Vorzugsweise werden solche Ausgangsstoffe verwendet und die Reaktionsbedingungen so gewählt, dass man zu den eingangs als besonders bevorzugt aufgeführten Verbindungen gelangt.
Die im obigen Verfahren verwendeten Ausgangsstoffe werden nach dem in Anmeldung Gesuch Nr.
497 448 beschriebenen Verfahren hergestellt.
Die erfindungsgemäss erhaltenen Verbindungen sind -insbesondere 2,2-disubstituierte 3-Acyl-Sa-aminothiazolidin-4-carbonsäuren, besonders deren funktionelle Derivate, wie die Verbindungen der Formel VIIIa

worin Ac für eine Acylgruppe, besonders eine der oben erwähnten Acylgruppen, steht, X das disubstituierte Kohlenstoffatom des Thiazolidinrings bedeutet und R1 für eine freie oder funktionell abgewandelte Carboxylgruppe steht.
Acylreste Ac sind in erster Linie solche, welche in pharmakologisch wirksamen N-Acylderivaten der 7-Aminocephalosporansäure vorkommen, wie der Thienylacetyl-, z. B. 2-Thienylacetyl-, Chloräthylcarbamyl- oder Phenylacetylrest, oder leicht abspaltbare Acylreste, wie der Rest eines Halbesters der Kohlensäure, z. B. der tert.-Butyloxycarbonylrest, oder irgendwelche andere geeignete Acylreste organischer Säuren.

Die Gruppe Rt steht für eine freie oder vorzugsweise funktionell abgewandelte Carboxylgruppe, insbesondere eine veresterte Carboxylgruppe. Letztere ist mit zur Veresterung von Carbonsäuren geeigneten Hydroxyverbindungen jeglicher Art, insbesondere mit aliphatischen Alkoholen, wie Alkanolen, insbesondere Niederalkanolen, z.B. Methanol, Methanol, n-Propanol oder tert.-Butanol, cycloaliphatischen Alkoholen, wie Cycloalkanolen, z. B.
Cyclohexanol, oder araliphatischen Alkoholen, wie Phenylalkanolen, z.B. Benzylalkohol oder Diphenylmethanol, sowie mit Phenolverbindungen verestert, wobei die obgenanuten Hydroxyverbindungen unsubstituiert sind oder Niederalkyl-, Niederalkoxy- oder Trifluormethylgruppen oder insbesondere Halogenatome sowie andere Gruppen als Substituenten enthalten können; besonders geeignet als die Carbonsäure veresternde substituierte Hydroxyverbindungen sind halogenierte Niederalkanole, wie 2,2,2-Trichloräthano1.
Andere funktionell abgewandelte Carboxylgruppen Rl sind z. B. stickstoffhaltige funktionell abgewandelte Carboxylgruppen, wie Carbamylgruppen, welche am Stickstoffatom unsubstituiert oder durch gegebenenfalls Niederalkyl-, freie, veresterte oder verätherte Hydroxy-, wie Niederalkoxy-, Aralkoxy-, Niederalkanoyloxy- oder Aroyloxygruppen oder Halogenatome, Nitro- oder Trifluormethylgruppen enthaltende aliphatische, alicyclische, aromatische oder araliphatische Kohlenwasserstoffreste oder heterocyclische Reste aromatischen Charakters, wie Niederalkyl-, Cycloalkyl-, Phenyl-, Phenyl-niederalkyl-, Phenyl-niederalkylidenoder Pyridylreste, sowie durch freie, verätherte oder veresterte Hydroxylgruppen, wie den oben erwähnten Gruppen dieser Art, durch phosphorhaltige Reste, oder durch Acylreste, wie Reste von Carbonsäuren, z.B.
Reste von Halbestern oder Halbamiden der Kohlensäure oder Niederalkanoylreste, oder von Sulfonsäuren, wie Arylsulfonsäuren, z.B. Die erfindungsgemäss erhaltenen Verbindungen können, wie im Formelschema gezeigt wird, in 7-Amino-cephalosporansäure und deren Derivate umgewandelt werden; diese Umwandlung kann z. B. nach dem in den Anmeldungen G.Nr. 497 456, G.Nr. 497 371, G.Nr. 497 369, G.Nr. 497 457, G.Nr. 497 460, G.Nr.
497 379, G.Nr. 497 461 beschriebenen Verfahren erfolgen.
In den folgenden Beispielen werden die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
Zu einem Gemisch von 1,4 g Lithiumaluminiumhydrid in 35 ml trockenem Tetrahydrofuran wird bei 0 und unter Rühren in einer Stickstoffatmosphäre tropfenweise eine Lösung von 9,1 g tert.-Butanol in 10 ml Tetrahydrofuran zugegeben. Nach einstündigem Rühren bei 0 wird eine Lösung von 3,45 g L-2,2-Dimethyl-3 -tert.-butyloxycarbonyl5a-azido-thiazolidin4-carbonsäuremthylester in 5 ml Tetrahydrofuran zugegeben; das Reaktionsgemisch wird während 1,5 Stunden bei Zimmertemperatur gehalten, dann in 200 mol Eiswasser ausgegossen und mit Methylenchlorid extrahiert. Die organische Phase wird verdampft und das erhaltene Öl in 80 ml Pentan gelöst.
Die Lösung wird durch 1 g Aktivkohle filtriert, letzteres mit 50 ml Pentan nachgewaschen und die vereinigten Filtrate eingedampft. Der viskose Rückstand wird in 40 ml Pentan gelöst; nach 3-stündigem Stehenlassen bei -15" kristallisiert der L-2,2-Dimethyl-3 -tert.-butyl oxycarbonyl-5a-amino-thiazolidin- 4-carbonsäuremethylester der Formel

Beispiel 2
Durch eine Lösung von 0,337 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl Sa-azido-thiazolidin-4-carbonsäure- methylester in 3 ml Äthanol wird während 4 Stunden Wasserstoff durchgeleitet, wobei während den ersten 3 Stunden 0,6 g eines Palladiumkatalysators (10 01o Palladium auf Calciumcarbonat) in 3 Portionen zugegeben wird.
Nach Filtration durch ein Filterhilfsmittel (Hyflo) wird das braune Filtrat verdampft. Durch Extraktion des viskosen Rückstandes mit Zitronensäure erhält man als Rohprodukt den L-2,2-Dimethyl-3 -tert.-butyloxycarbonyl- 5a-amino-thiazolidin-4-carbonsäure- methylester; dieser wird mit dem nach dem Verfahren des Beispiels 3 erhaltenen Produkt vereinigt und wie im Beispiel 3 beschrieben weiter aufgearbeitet.
Beispiel 3
Zu einer wassergekühlten Lösung von 0,321 g L-2,2-Dimethyl-3-tert.- butyloxyzarbonyl-5se-azido-thiazolidin- 4-carbonsäure-methylester in 8 ml 900/obiger Essigsäure werden 2 g Zinkpulver während 5 Minuten zugegeben. Nach weiteren 10 Minuten wird das Gemisch in 100 ml einer gesättigten wässrigen Natriumhydrogencarbonatlösung ausgegossen, festes Natriumhydrogencarbonat bis zu pH 8-9 zugegeben und mit Methylenchlorid extrahiert. Der organische Extrakt wird getrocknet und verdampft und der viskose Rückstand durch Extrahieren mit Zitronensäure in einen neutralen und einen basischen Anteil getrennt.
Die neutrale Phase des organischen Lösungsmittels ergibt den U2, 2-Dimethyl-3 -tert. -butyloxycarbonyl- Se-acetyloxy-thiazolidin-4-carbonsäure- methylester, während das Produkt des sauren wässrigen Extrakts d. h. der basische Anteil, mit dem nach dem Verfahren des Beispiels 2 erhaltenen Rohprodukt vereinigt und an 10 g Silicagel chromatographiert wird. Mit einem 3:1-Gemisch Benzol und Essigsäureäthylester erhält man den reinen L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5a-amino-thiazolidin-4-carbonsäuremethylester, der nach Umkristallisieren aus Pentan bei 62-65 schmilzt.
Beispiel 4
Nach kurzem Behandeln mit einer 10 %igen wässrigen Natriumhydroxydlösung werden 2 g fein granuliertes Aluminium mit Wasser gewaschen und mit einer 0,5 3/obigen wässrigen Quecksilber-II-chloridlösung behandelt. Zum mit Wasser gewaschenen Amalgam wird eine Lösung von 0,746 g L-2,2-Dimethyl-3-tert.butyloxycarbonyl-5a-azido-thiazolidin4-carbonsäuremethylester in 10 ml Ather zugegeben; nach 10 Minuten wird die Ätherlösung abgeschüttet, getrocknet und eingedampft.
Der viskose Rückstand wird an 25 g Silicagel chromatographiert und der erwünschte L-2,2-Dimethyl-3 -tert .-butyloxycarbonyl5α-amino-thiazolidin-4-carbonsäuremethyl- ester mit einem 3:1-Gemisch von Benzol und Essigsäure äthylester eluiert. Nach Kristallisieren aus Pentan schmilzt das Produkt bei 64-66 .
Beispiel 5
Zu einer Lösung von 3,16 g L-2,2-Dimethyl-3 -tert.-butyloxycarbonyl 5a-azido-thiazolidin-4-carbonsäure- methylester in 50 ml wasserfreiem Benzol werden tropfenweise in einer Stickstoffatmosphäre 2,2 g Tri-n-butyl-phosphin zugegeben. Nach 5-stündigem Stehenlassen bei Zimmertemperatur wird die gelbe Lösung, enthaltend den L-2,2-Dimethyl-3-tert.-butyloxycarbonyl5α-tri-n-butylphosphinimino-thiazolidin-4- carbonsäuremethylester in 50 ml Wasser ausgegossen und das Gemisch während 15 Stunden gerührt. Nach dem Eindampfen wird der Rückstand der Benzollösung mit Pentan trituriert, die Pentanlösung 3 mal mit je 100 ml Wasser gewaschen und eingedampft.
Der viskose Rückstand wird an 200g Silicagel chromatographiert und der erwünschte L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5a-amino-thiazolidin-4-carbonsäuremethylester mit einem 3:1-Gemisch Benzol und Essigsäureäthylester eluiert; er ist mit dem nach den Beispielen 1, 3 und 4 erhältlichen Produkt identisch.
Der L-2,2-Dimethyl-3-tert.-butyloxycarbonyl Sa-triphenyl-phosphiniminothiazolidin-4-carbonsäure-methylester, der in ähnlicher Weise verwendet werden kann, wird wie folgt erhalten:
Eine Lösung von 0,3164 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl5a-azido-thiazolidin-4-carbonsäuremethylester in 15 ml reinem Methylenchlorid wird mit 0,2623 g umkristallisiertem Triphenylphosphin versetzt und das Gemisch während 3¸ Stunden am Rückfluss gekocht.
Nach dem vorsichtigen Verdampfen erhält man ein weisses schaumartiges Material, welches aus Hexan, enthaltend eine kleine Menge Methylenchlorid, kristallisiert; der so erhaltene L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5a-triphenyl-phosphinimino-thiazolidin 4-carbonsäuremethylester der Formel

Zu einer Lösung von 0,485 g reinem L-2,2-Dimethyl-3-tert.-butyloxycarbonyl 5a-azido-thiazolidin-4-carbonsäure- methylester in 5 ml Hexan wird eine Lösung von 0,427 g Triphenylphospin in 5 ml Hexan gegeben und das Gemisch bei 70" erhitzt. Nach etwa 2 Minuten setzt Kristallisation ein, das Gefäss wird verschlossen und man lässt abkühlen. Nach 2 Stunden bei 0 wird die überstehende Lösung abgegossen und das kristalline Material mit 5 ml Pentan gewaschen und getrocknet; das erhaltene Produkt schmilzt bei 180-181 und ist mit dem nach dem obigen Verfahren erhaltenen identisch.
Beispiel 6
Ein Gemisch von 58,2 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl 5a-azido-thiazolidin-4-carbonsäure- methylester in 750 ml Methanol, 750 mal Cyclohexan und 350 mal Hexan wird unter Rühren auf -18" gekühlt und in einem mit einem Vibrierrührer ausgerüsteten Gefäss mit Alumininmamalgam (hergestellt durch Behandeln von 10 g Aluminiumspänen mit 2-n. Natronlauge, dann mit Quecksilber-II-chlorid und Waschen mit Wasser, dann mit trockenem Methanol) versetzt, wobei mit einer kleinen Menge Methanol nachgewaschen wird.
Nach 40 Stunden bei -15 bis -180 kann noch unreagiertes Ausgangsmaterial festgestellt werden; 5 g frisch zubereitetes Aluminiumamalgam wird zugegeben und es wird während 8 Stunden bei -18" weitergerührt. Nach dem Filtrieren und Waschen mit Methanol wird der anorganische Rückstand mit Methanol kurze Zeit ausgekocht und die Lösung mit dem Filtrat vereinigt. Die kombinierten organischen Lösungen werden eingedampft und der ölige Rückstand mit Pentan extrahiert. Die Pentanlösung wird auf ein Volumen von 250-300 ml eingedampft und bei -18" stehen gelassen und dann vom semikristallinen Produkt dekantiert.
Letzteres wird mit etwas Pentan gewaschen und aus Pentan umkristallisiert; der so erhaltene L-2,2-Dimethyl-3-tert.-butyloxycarbonyl 5a-amino-thiazolidin-4-carbonsäure- methylester wird wiederum in Pentan gelöst, durch wasserfreies Magnesiumsulfat filtriert und auf ein Volumen von 150-200 ml konzentriert und kristallisiert; nach weiterem Umkristallisieren aus Pentan schmilzt der erhaltene Ester bei 665O.
Beispiel 7
Eine Lösung von 0,141 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5a-azido-thiazolidin-4-carbonsäure- methylester in 15 ml Methanol wird auf -15" gekühlt und mit 0,3 g Chrom-II-acetat versetzt; nach 35 Minuten wird eine weitere Menge von 0,24 g des Reduktionsmittels zugegeben. Nach 4-stündigem Rühren bei -10 bis -180 wird das Reaktionsgemisch filtriert, der Rückstand mit Methanol und 50 ml Cyclohexan gewaschen und das Filtrat unter vermindertem Druck eingedampft. Der Rückstand wird mit Cyclohexan extrahiert und man erhält ein Gemisch des L-2,2-Dimethyl-3-tert.-butyloxycarbonyl 5a-amino-thiazolidin-4-carbonsäuremethylesters und der entsprechenden 5ss-Aminoverbindung, welches mittels Dünnschichtchromatogramm getrennt werden kann.
Beispiel 8
Eine Lösung von 0,32 g L-2,2-Dimethyl-3 -tert. butyloxycarbonyl- 5a-azido-thiazolidin-4-carbonsäuremethylester in einem Gemisch von 0,47 g Di-n-butyl-zinn-dihydrid und 1 mal Hexan wird unter Stickstoff während 18 Stunden bei Zimmertemperatur stehen gelassen. Nach Zugabe von 30 ml Pentan wird 3 mal mit je 20 ml 20 O/oiger wässriger Zitronensäurelösung extrahiert, die wässrigen Lösungen mit festem Natriumhydrogencarbonat gesättigt und mit Methylenchlorid extrahiert. Der Rückstand aus den organischen Extrakten wird in Pentan gelöst und durch 2,5 g Aktivkohle (Carboraffin) filtriert und mit 40 ml Methylenchlorid nachgewaschen.
Nach dem Eindampfen wird der Rückstand aus Pentan bei 200 kristallisiert; der so erhaltene reine L-2,2-Dimethyl-3-tert.-butyloxycarbonyl 5a-amino-thiazolidin-4-carbonsäure- methylester schmilzt bei 64,5-65,50.
Beispiel 9
Ein Gemisch von 0,3164 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5a-azido-thiazolidin-4-carbonsäure- methylester in 10ml Metanol und 1 mol Essigsäure wird mit 5g Natriumamalgam (5 0/o) versetzt, während 2 Minuten gerührt, filtriert und verdampft. Der Rückstand, der sommer noch Ausgangsmaterial enthält, wird in 10ml Dioxan und 0,6 ml Essigsäure gelöst, 5 g Natriumamalgam werden zugegeben und das Gemisch während 15 Minuten gerührt. Nach dem Aufarbeiten wie oben wird der Rückstand in 20 ml Methylenchlorid gelöst und 3 mal mit je 5 ml 20 0/obiger wässriger Zitronensäure extrahiert.
Die saure Lösung wird wie in den vorstehenden Beispielen beschrieben aufgearbeitet und man erhält ein Rohprodukt, welches zur Hauptsache aus L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5a-amino-thiazolidin-4-carbonsäure- besteht.
Beispiel 10
Eine Lösung von 0,973 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl 5a-azido-thiazolidin-4-carbonsäure- methylester in 25 ml Methanol und 2,40 ml Essigsäure wird auf -8" gekühlt und mit 0,5 g Magnesiumspänen versetzt und die Reaktion durch Zugabe von einigen Kristallen
Quecksilber-II-chlorid ausgelöst. Nach 90-minütigem
Rühren bei -5" bis -8" werden 0,25 g Magnesium, ge folgt won 1,2 ml Essigsäure und einer Spur Quecksil ber-II-chlorid zugegeben. Nach weiteren 2 Stunden
Rühren wird das Reaktionsgemisch filtriert und mit Methanol nachgewaschen; die organischen Lösungen werden vereinigt und unter vermindertem Druck zur Trockne eingedampft.
Der Rückstand wird in 130 ml Wasser aufgenommen (pHcr 8), mit 3 Portionen zu je 50 ml Methylenchlorid extrahiert und die getrockneten Extrakte eingedampft. Das erhaltene Öl kristallisiert aus Pentan (-10 ) und der L-2,2-Dimethyl-3-tert.-butyloxycarbonyl 5a-amino-thiazolidin-4-carbonsäure- methylester schmilzt nach Umkristallisieren aus dem gleichen Lösungsmittel bei 63-65 .
Beispiel 11
Eine Lösung von 9,96 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl 5a-azido-thiazolidin-4-carbonsäure- methylester in 300 ml Methanol wird auf -9" gekühlt, 9 g Alumi ninmamalgam werden zugegeben und das Gemisch während 6 Stunden bei -100 und 0 gerührt. Nach dem Abfiltrieren und Nachwaschen mit Methanol werden die vereinigten organischen Lösungen unter vermindertem Druck verdampft; der farblose sirupartige Rückstand wird mit 200 ml warmem Pentan extrahiert und die organischen Extrakte eingedampft. Der pentanunlösliche Rückstand wird mit 250 ml Methanol trituriert und die abgetrennte Methanollösung unter vermindertem Druck verdampft.
Die Pentan- und Methanol-löslichen Fraktionen werden vereinigt und aus etwa 30ml Pentan kristallisiert; nach Umkristallisieren aus Pentan schmilzt der L-2,2-Dimethyl-3-tert.-butyloxycarbonyl5a-amino-thiazolidin-4-carbonsäuremethylester bei 64-660.
Durch Verlängern der Reaktionszeit auf 11 Stunden bei -10 bis 150 kann die Ausbeute an L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5a-amino-thiazolidin-4-carbonsäuremethylester gesteigert werden; eine weitere Menge des erwünschten Materials kann aus der Kristallisationsmutterlauge erhalten werden.
Beispiel 12
Ein Gemisch von 0,316 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5a-azido-thiazolidn-4-carbonsäure- methylester in 3 ml Methanol und 2,5 ml Essigsäure wird unter Rühren bei Zimmertemperatur mit 0,05 g Aluminiumamalgam versetzt; nach einstündigem Rühren wird nach dem Abfiltrieren, Waschen und Eindampfen der Rückstand mit Pentan extrahiert; das nach dem Verdampfen erhaltene Produkt zeigt immer noch unreagiertes Ausgangsmaterial und 0,215 g des Rohproduktes werden während weiteren 230 Minuten in 3 ml Methanol und 21/2 ml Essigsäure mit 0,07 g Alumindum- amalgam gerührt.
Man erhält nach dem üblichen Aufarbeiten den L-2,2-Dimethyl-3 -tert .-butyloxycarbonyl 5a-amino-thiazolidin-4-carbonsäure- methylester, der nach Umkristallisieren aus Pentan bei 61-65 schmilzt.
Beispiel 13
Eine Lösung von 0,26 g L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5a-azido-thiazolidin-4-carbonsäure- 2,2,2-trichloräthylester in 8 ml 90 6/obiger wässriger Essigsäure wird portionenweise unter Rühren innert 5 Minuten mit 2g Zinkstaub versetzt. Nach 2112-stün < ligem Rühren bei Zimmertemperatur wird das Reaktionsgemisch filtriert, der Rückstand mit 2 ml Eisessig und 5 ml Methylenchlorid gewaschen und das Filtrat eingedampft. Der Rückstand wird mehrmals mit Methylenchlorid verrührt, die organische Lösung mit 10 ml Wasser gewaschen, getrocknet und eingedampft.

Die Lösung des Rohproduktes in Methylenchlorid wird 3 mal mit je 20 ml 20 0/obiger wässriger Zitronensäure extrahiert. Die sauren Extrakte werden mit Natriumcarbonat auf pH ca. 5 gestellt und die wässrige Lösung 3 mal mit 20 ml Methylenchlorid extrahiert.

Eine Lösung von 0,121 g L-2,2-Dimethyl-3 -tert.-butyloxycarbonyl5a-azido-thiazolidin-4-carbonsäure2,2,2-trichloräthylester in 10 ml Methanol wird auf -200 abgekühlt und mit 0,5 g Aluminiumamalgam behandelt. Das Reaktionsgemisch wird während 55 Stunden bei -20 , während 22 Stunden bei OC und während 18 Stunden bei +25 gerührt.
Das unlösliche Material wird abfiltriert und das Filtrat eingedampft; man erhält einen öligen Rückstand welcher aufgrund des Dünnschichtchromatogramms Ausgangsmaterial und eine etwa gleich grosse Menge des polareren L-2,2-Dimethyl-3-tert.-butyloxycarbonyl- 5a-amino-thiazolidin-4-carbonsäure2,2,2-trichloräthylesters der Formel
