Verfahren zur Herstellung von Dibenzo-cyclohepten-I :} erivaten
Die vorliegende Erfindung betrifft ein neues Ver fahren zur Herstellung von 5H-Dibenzo [a, d]-10, 11-di- hydrocycloheptenen und 5H-DibenzoLa, d] cycloheptenen, welche am 5-Kohlenstoffatom mit einem sekundären Aminopropyl-oder sekundären Aminopropyliden
Radikal substituiert sind.
Die erfindungsgemäss erzeugten Endverbindungen sind bei der Behandlung von Geisteskrankheiten nützlich, nachdem sie als Antidepressants wirken und somit zur Verbesserung der Gemütsverfassung oder zur psychischen Kräftigung dienen. Die Verbindungen werden vorteilhafterweise in Form ihrer sauren Additionssalze verabreicht. Die vorliegende Erfindung umfasst somit auch die Herstellung dieser sauren Additionssalze.

Die gestrichelten Linien bedeuten, da¯ die Verbindung gesÏttigt oder ungesÏttigt, in den angef hrten St. llungen, sein kann, Z ist Sauerstoff oder Schwefel ; R, welches während des Verfahrens entfernt wird, ist ein Alkyl-, Aryl-, Aralkyl-oder Alkarylradikal und X" steht für ein Halogen.
Wie aus dem oben angeführten Fliessschema hervorgeht, wird in dr ersten Stufe des erfindungsgemässen Verfahrens ein tertiäres Aminopropyliden oder ein tertiäres Aminopropyl-Derivat eines 5H-Dibenzo [a, d]- cycloheptens mit einem Haloformiat oder einem Halo thioformiat kondensiert, wobei ein Urethan-oder ein Thiourethan-Zwischenprodukt entsteht. In einem typi- schen Vorgang wird 5-[3-N-Dimethyl-aminopropyliden]5H-dibenzo[a,d]cyclohepten mit Methylchlorformiat umgesetzt, wobei sich das Urethanprodukt, das 5- [3- (N Methyl-N-carbomethoxy-amino)-propyliden]-5H-di- benzo[a,d]cyclohepten und Methylchlorid bildet.
In Ïhnlicher Weise wird 5-[3-(N-Dimethylamino)-propyliden]-5H-dibenzo[a,d]cyclohepten mit Methylchlorothioformiat zur Reaktion gebracht, wobei das Thiourethanprodukt, das 5- [3- (N-Methyl-N-methyl'mercapto- formylamino)-propyliden]-5H-dibenzo [a, d] cyclohepten und Methylchlorid entsteht.
Obschon die Reaktion in Abwesenheit eines Lösungs- mittels durchgeführtwerden kann, ist es von Vorteil, ein Losungsmittelzu verwenden. Geeignete Lösungsmittel sind j-ne, welche inert sind, wie aromatische Kohlenwasserstoffe, wie Benzol und Toluol, aliphatische Kohlen- wasserstoffe, wie Hepten, Hexan und ähnliche, Halogenkohlenwasserstoffe, wie Chloroform und Kohlenstoff- tetrachlorid und Site, wie tetrahydrofuran. Die Reaktion, kann bei Zimmertemperatur durchgeführt werden, obschon es zweckmässig ist, dieselbe unter R ckfluss und während einer geeigneten Zeitspanne, vorteil hafterweise bis zu mehreren Stunden, erfolgen zu lassen.
Nach Beendigung dieser Reaktion wird die Lösung in der Regel mit verdünnter Säure extrahiert und zur Trockne abgedampft, wobei das gewünschte Urethan erhalten wird. In einer bevorzugten Reaktion wird 5-f3- N-Methylamino]-propylF5H-dibenzota, d] cyclohepten aus der entsprechenden N-Dimethyl-Ausgangsverbindung hergestellt.
Das so erzeugte Urethan-Zwischenprodukt wird einer Hydrolyse unterworfen, um die substituierte Carbamylgruppe des Urethans oder die Thiocarbamylgruppe des Thiourethans in ein Wasserstoffatom überzuführen. Die Hydrolyse wird vorteilhafterweise unter basischen oder sauren Bedingungen durchgeführt.
In geeigneter Weiss erfolgt eine solche Hydrolyse in einer basischen Alkohol- l¯sung und unter Rückfluss. Zweckmäl3igerweise aber wird die Hydrolyse des Thiourethan-Zwischenproduktes so durchgeführt, dass man zunächst das Thiourethan mit einem Oxydationsmittel oxydiert, wobei vorteilhafter- weise Wasserstoffperoxyd in Essigsäure verwendet wird, obschon auch andere Oxydationsmittel, wie Perbenzoesaure, PeressigsÏure, ChromsÏure oder Kaliumpermanganat, ebensogut in Betracht kommen. Dann wird das oxydierte Zwischenprodukt mit einer basischen Lö sung, wie einer Natriumhydroxydlosung, in Berührung gebracht.
Das Urefhan-Zwischenprodukt wird vorzugsweise unter Stickstoff in einer r L¯sung von Kaliumhydroxyd in n-Butanol bei Rückfluss erhitzt. Nach Beeendigung der Hydrolyse wird das Lösungsmittel im Vakuum abgedampft und der Rückstand mit geeigneten Lösungs- mitteln, um das hydrolysierte Produkt zu isolieren, extrahiert. Das Hydrolyseprodukt ist das gewünschte sekundäre Aminopropyliden oder sekundäre Amino propyl-Derivat eines 5H-Dibenzo [a, d] cycloheptens oder 5H-Dibenzo [a, d]-10, 11-dihydrocycloheptens.
Beispiel 1 5- [3- (N-Methyl-N-carbäthoxyamino)-propyliden]-
5H-dibenzo [a, d] cyclohepten
Eine Lösung, bestehend aus 0, 1 Molen 5- [3- (N-Di- methyl amino)-propyliden]-5H-dibenzo [ad] cyclohepten in 100 ml Benzol, wurde innerhalb einer Stunde zu einer Lösung von 32, 4 g, das sind 0, 3 Mole, Athyl chloroformiat in 100 ml Benzol bei Zimmertemperatur zugesetzt. Diese Lösung wurde dann unter Rühren 2 Stunden bei Rückfluss erhitzt, dann auf Zimmertemperatur gekühlt und mit 3 X 100 ml von 2, 5n HCI extrahiert. Nach Waschen mit 2 X 50 ml Wasser, Trocknen über Magn siumsulfat, wird die Lösung zur Trockne abgedampft.
Es wird ein dickes 01 erhalten, das das reine Urethan darstellt. Der Ertrag besteht aus 29, 5 g oder 88 % der theoretischen Ausbeute.
29, 5 g dieses so erhaltenen 5- [3- (N-Methyl-N-carb- äthoxyamino)-propylidenl-5H-dibenzo [a, d] cycloheptens werden während 9 Stunden unter Stickstoff in einer Lösung von 24, 2 g Kaliumhydroxyd in 255 ml n-Butanol bei Rückfluss erhitzt. Nach Kühlen auf Zimmertemperatur wird das Lösungsmittel im Vakuum abgedampft, und der Rückstand wird mit 200 ml Wasser und 300 ml n-Hexan verrührt ; die Schichten werden separiert, und die Wasserschicht wird nochmals mit 100 ml n-Hexan extrahiert. Die vereinigten Schichten werden dann mit 2 X 100 ml Wasser und dann mit 100 ml, hernach mit 80 ml und schliesslich mit 100 ml 0, 5n Schwefelsäure gewaschen.
Die saure Lösung wird dann alkalisch gemacht und mit 2 X 150 ml und 1 X 100 ml waither extrahiert, über Magnesiumsulfat getrocknet und die Lösung zur Trockene abgedampft. Man erhielt 19, 7 g eines leicht gelben, klaren dicken Ols in 85 % iger theoretischer Ausbeute, welches O1 das im wesentlichen reine Mono methyl-Baseprodukt, das 5- [3- (N-Methylamino)-propyl- iden]-5H-dibenzo [a, d] cyclohepten, mit F. = 54-55 C darstellt.
Beispiel 2
27,7 g, das sind 0,1 Mole, von 5-[3-(N-Dimethylamino)-propyl]-5H-dibenzo[a,d]cyclohepten, gel¯st in 100 ml Benzol, werden innerhalb 1 Stunde zu einer Lösung von 32, 4 g, das sind 0, 3 Mole, Athylchloroformiat in 100 ml Benzol bei Zimmertemperatur zugesetzt. Die Lösung wird dann unter Rühren während 20 Stunden bei Rückfluss erhitzt, hernach auf Zimmertemperatur gek hlt und mit drei 100-ml-Portionen von 2,5n HCl extrahiert. Danach wird die L¯sung mit zwei 50-ml-Portionen Wasser gewaschen, über Magnesiumsulfat getrocknet und zur Trockne abgedampft. Man erhält ein dickes 01, das das reine Urethan darstellt, vom Gewicht von 29, 5 g oder in 88 % iger theoretischer Ausbeute.
29,5 g des erhaltenen 5-[3-(N-Methyl-N-carbÏthoxyamino)-propyl]-5H-dibenzo[a,d]cycloheptens werden während 24 Stunden unter Stickstoff in einer Lösung von 36, 3 g Kaliumhydroxyd in 378 ml n-Butanol bei Rückfluss erhitzt. Nach Kühlen auf Zimmertemperatur wird das Lösungsmittel im Vakuum abgedampft, der Rückstand mit 200 ml Wasser und mit 300 ml n-Hexan gerührt. Die Schichten werden separiert, worauf die Wasserschicht mit 100 ml n-Hexan extrahiert wird. Die vereinigten Hexanschichten werden dann mit 2 X 100 ml Wasser und dann mit 100 mil, dann mit 80 ml und schliesslich mit 80 ml 0, 5n Schwefelsäure gewaschen.
Die saure Lösung wird dann alkalisch gemacht und mit 2 X 150 ml und 1 X 100 ml Äther extrahiert, ber Magnesiumsulfat getrocknet und die Lösung dann zur Trockene abgedampft. Das Produkt besteht im wesentlichen aus dem reinen Monomethyl-Baseprodukt, welches dann im weiteren durch Uberführung in das Oxalat, mit F. = 181 C, Befreiung der freien Base und Konversion in das Hydrochlorid, mit F. = 169-170 C, gereinigt wird.
Beispiel 3
Gemäss dem in Beispiel 1 beschriebenen Verfahren und unter Verwendung gleichwertiger Mengen von
5- [3- (N-Dimethylamino)-propyliden]-5H dibenzo [a, d] 10, hydrocyclohepten gelangt man zum entsprechenden 5- [3- (N-Methyl-N-carboäthoxyamino)-propyliden]-
5H-dibenzo [a, d] 10, 11-dihydrocytlohepten.
28, 2 g 5- [3- (N-Methyl-N-carbäthoxyamino)- propyliden]-5H-dibenzo [a, d] 10, 11-dihydro- cyclohepten werden in einer Lösung von 23, 9 g Kaliumhydroxyd in 250 ml n-Butanol während 6 Stunden in einem Stickstoffstrom und unter Rühren bei Rückfluss erhitzt. Danach wird das Lösungsmittel im Vakuum entfernt und der Rückstand mit einem Gemisch von 260 ml Wasser und 310 ml 2n Schwefelsäure versetzt und, zur Bildung einer Lösung, erhitzt. Diese Lösung wird dann auf Zimmertemperatur gekühlt und mit 250 ml n-Hexan extrahiert.
Es bildeten sich drei Schichten, wobei die zwei Schichten beiseite gelegt werden und die untere Hexanschicht wird mit 100 mt n-Schwefelsäure extrahiert. Die ver einigten sauren Schichten werden dann mit 50 mi von 11, 7n Natriumhydroxydlösung alkalisch gemacht und dann mit 3 X 170-ml-Portionen ¯ther extrahiert, nochmals mit Wasser gewaschen, über Magnesiumsulfat getrocknet und zur Trockne abgedampft. Man erhielt 21, 5 g, das ist in 97% iger Ausbeute, an gewünschtem Monomethyl-Produkt, dem 5- [3- (N-Methylamino)-propyliden]-5H-dibenzo- [a, d] 10, 11-dihydrocyclohepten.
Beispiel 4 52, 4 g, das sind 0, 188 Mole, 5- [3- (N-Dimethylamino)-propyl]-5H-dibenzo- [a, d] 10, 11-dihydrocyclohepten werden in 216 ml Benzol gelöst und mit einer Lösung von 65 g Methylchloroformiat in 580 ml Benzol inner- halb etwa 15 Minuten unter Rühren und bei Zimmertemperatur versetzt. Die erhaltene Lösung wird dann bei Rückfluss unter Rühren während 2 Stunden erhitzt, danach auf Zimmertemperatur gek hlt und filtriert ; das Filtrat wird dann mit drei 200-ml-Portionen 2, 5n HCl extrahiert, mit Wasser gewaschen, über MgS04 getrock- net und im Vakuum abgedampft. Man erhält 56 g eines dicken, farblosen vols, das das Urethan darstellt.
Unter Befolgung des oben im einzelnen beschriebe- nen Verfahrens und unter Verwendung äquivalenter Mengen des erhaltenen 5- [3- (N-Methyl-N-carbomethoxyamino)-propyl]- 5Hqdibenzo [a, d] 10, 11-dihydrocycloheptens erhält man das entsprechende 5- [3- (N-Methylamino)-propyl]-5H-dibenzo- [a, d] 10, 11-dihydrocyclohepten.
Beispiel 5
6, 9 g, das sind 0, 025 Mole, 5- [3- (N-Dimethylamino)- propyliden]-5H-d'ibenzo [a, d] cyclohepten werden in 20 ml Benzol gelöst, und es wird eine Lösung von 2, 76 g, das sind 0, 025 Mole, Methylchlorothioformiat in 10 ml Benzol innerhalb von etwa 15 Min., währenddem das Gemisch bei Zimmertemperatur gerührt wird, zugesetzt. Die Lösung wird dann unter Rühren wÅahrend 11/2 Stunden bei Rückfluss erhitzt, auf Zimmertempera- tur gekühlt, filtriert, das Filtrat wird dann mit 3 X 25ml-Portionen 2, 5n HCl extrahiert, dann mit Wasser gewaschen, über Magnesiumsulfat getrocknet und im Vakuum abgedampft.
Man erhält 5, 8 g, das ist in 69 % iger theoretischer Ausbeute, eines dicken, farblosen Ols, das schnell kristallisiert. Für analytische Zwecke wird das Produkt der Formel
5- [3- (N-Methyl-N-methylmercaptoformylamino) propyliden]-5H-dibenzo [a, d] cyclohepten aus 15 ml Äthanol umkristallisiert und liefert 5, 09 g glitzernder Nadeln mit F. = 106-108 C.
Analyse : Berechnet für CzlH2INOS, Mol.-Gew. 335, 25 :

1, 68 g, das sind 0, 005 Mole, von 5- [3- (N-Methyl-N-methylmercaptoformylamino)- propyliden]-5H-dibenzo[a,d]cyclohepten werden in 13 ml EisessigsÏure unter mildem Erhitzen gelöst. Nach Kühlung auf Zimmertemperatur wird die Lösung mit 2 ml 30% iger H20s und ansc'hlie- ssend mit 7 ml Eisessigsäure versetzt.
Nach Stehen- lassen bei Zimmertemperatur während 21 Stunden unter Rühren wird eine lare, farblose Lösung er- halten, welche zu 40 ml einer 11, 7n Natriumhydroxydlösung, unter äusserer Kühlung, zugesetzt und mit 60 ml Wasser verdünnt wird. Die Lösung wird dann mit 40 ml und 2 X zu je 20 ml Benzol extrahiert, die vereinigten Benzolschichten werden mit 3 X 10 ml einer 2, 5n HCI gewaschen und die vereinigten HCMxtrakte mit 20 ml 11, 7n NaOH alkalisch gemacht ; die Lösung wird dann mit 2 X 25 ml Äther extrahiert, mit Magnesiumsulfat getrocknet und zur Trockne abgedampft.
Man erhält 0, 60 g des Produktes der Formel 5- [3- (N-Methylamino)-propyliden]-5H-dibenzo- [a, d] cyclohepten in Form eines dicken Öls, welches nach Impfung kri stallisiert. Das öl ist dadurch charakterisiert, dal3 es sein Salz mit Oxalsäure in ¯thanol bildet. Der Schmelz- punkt des Hemioxalats beträgt 202-203 C (Zers.).

As can be seen from the flow diagram above, a tertiary aminopropylidene or a tertiary aminopropyl derivative of a 5H-dibenzo [a, d] - cycloheptens with a haloformate or a halo thioformate is condensed in the first stage of the process according to the invention, with a urethane or a thiourethane intermediate is formed. In a typical process, 5- [3-N-dimethyl-aminopropylidene] 5H-dibenzo [a, d] cycloheptene is reacted with methyl chloroformate, the urethane product, the 5- [3- (N methyl-N-carbomethoxy- amino) propylidene] -5H-dibenzo [a, d] cycloheptene and methyl chloride.
In a similar way, 5- [3- (N-dimethylamino) propylidene] -5H-dibenzo [a, d] cycloheptene is reacted with methyl chlorothioformate, the thiourethane product, the 5- [3- (N-methyl-N- methyl'mercaptoformylamino) propylidene] -5H-dibenzo [a, d] cycloheptene and methyl chloride are formed.
Although the reaction can be carried out in the absence of a solvent, it is advantageous to use a solvent. Suitable solvents are those which are inert, such as aromatic hydrocarbons such as benzene and toluene, aliphatic hydrocarbons such as heptene, hexane and the like, halogenated hydrocarbons such as chloroform and carbon tetrachloride and sites such as tetrahydrofuran. The reaction can be carried out at room temperature, although it is expedient to allow it to take place under reflux and for a suitable period of time, advantageously up to several hours.
When this reaction is complete, the solution is usually extracted with dilute acid and evaporated to dryness to give the desired urethane. In a preferred reaction, 5-f3-N-methylamino] propylF5H-dibenzota, d] cycloheptene is prepared from the corresponding N-dimethyl starting compound.
The urethane intermediate thus produced is subjected to hydrolysis to convert the substituted carbamyl group of urethane or the thiocarbamyl group of thiourethane into a hydrogen atom. The hydrolysis is advantageously carried out under basic or acidic conditions.
In a suitable way, such a hydrolysis takes place in a basic alcohol solution and under reflux. However, the hydrolysis of the thiourethane intermediate is expediently carried out in such a way that the thiourethane is first oxidized with an oxidizing agent, hydrogen peroxide in acetic acid being advantageously used, although other oxidizing agents, such as perbenzoic acid, peracetic acid, chromic acid or potassium permanganate, are also possible . The oxidized intermediate is then brought into contact with a basic solution, such as a sodium hydroxide solution.
The urefhan intermediate is preferably refluxed under nitrogen in a solution of potassium hydroxide in n-butanol. After the hydrolysis has ended, the solvent is evaporated off in vacuo and the residue is extracted with suitable solvents in order to isolate the hydrolyzed product. The hydrolysis product is the desired secondary aminopropylidene or secondary amino propyl derivative of a 5H-dibenzo [a, d] cycloheptene or 5H-dibenzo [a, d] -10,11-dihydrocycloheptene.
Example 1 5- [3- (N-methyl-N-carbäthoxyamino) propylidene] -
5H-dibenzo [a, d] cycloheptene
A solution consisting of 0.1 mol of 5- [3- (N-dimethylamino) propylidene] -5H-dibenzo [ad] cycloheptene in 100 ml of benzene became a solution of 32.4 g within one hour , that is 0.3 moles of ethyl chloroformate in 100 ml of benzene at room temperature. 29.5 g of the 5- [3- (N-methyl-N-carbÏthoxyamino) propyl] -5H-dibenzo [a, d] cycloheptens obtained are in a solution of 36.3 g of potassium hydroxide in 378 for 24 hours under nitrogen ml of n-butanol heated to reflux. 5H-dibenzo [a, d] 10,11-dihydrocytloheptene.
28.2 g of 5- [3- (N-methyl-N-carbäthoxyamino) propylidene] -5H-dibenzo [a, d] 10, 11-dihydro-cycloheptene are added to a solution of 23.9 g of potassium hydroxide in 250 ml n-Butanol heated at reflux for 6 hours in a stream of nitrogen and with stirring. The solvent is then removed in vacuo and the residue is treated with a mixture of 260 ml of water and 310 ml of 2N sulfuric acid and heated to form a solution. This solution is then cooled to room temperature and extracted with 250 ml of n-hexane.
Three layers formed, the two layers being set aside and the lower hexane layer extracted with 100 ml of n-sulfuric acid. The combined acidic layers are then made alkaline with 50 ml of 11.7N sodium hydroxide solution and then extracted with 3 x 170 ml portions of ether, washed again with water, dried over magnesium sulfate and evaporated to dryness. 21.5 g, that is in 97% yield, of the desired monomethyl product, 5- [3- (N-methylamino) propylidene] -5H-dibenzo- [a, d] 10,11-dihydrocycloheptene, were obtained .
Example 4 52.4 g, that is 0.188 moles, 5- [3- (N-dimethylamino) propyl] -5H-dibenzo- [a, d] 10, 11-dihydrocycloheptene are dissolved in 216 ml benzene and mixed with a solution of 65 g of methyl chloroformate in 580 ml of benzene is added within about 15 minutes with stirring and at room temperature. Example 5
6, 9 g, that is 0, 025 moles, 5- [3- (N-dimethylamino) - propylidene] -5H-d'ibenzo [a, d] cycloheptene are dissolved in 20 ml of benzene, and a solution of 2.76 g, that is, 0.025 moles, of methyl chlorothioformate in 10 ml of benzene were added over the course of about 15 minutes while the mixture was stirred at room temperature. The solution is then refluxed for 11/2 hours with stirring, cooled to room temperature, filtered, the filtrate is then extracted with 3 × 25 ml portions of 2.5N HCl, then washed with water, dried over magnesium sulfate and in vacuo evaporated.
5.8 g are obtained, that is in 69% theoretical yield, of a thick, colorless oil which quickly crystallizes. For analytical purposes, the product is the formula
5- [3- (N-Methyl-N-methylmercaptoformylamino) propylidene] -5H-dibenzo [a, d] cycloheptene recrystallized from 15 ml of ethanol and yields 5.09 g of glittering needles with a F. = 106-108 C.

The solution is then extracted with 40 ml and 2 × to 20 ml each of benzene, the combined benzene layers are washed with 3 × 10 ml of 2.5N HCl and the combined HCl extracts are made alkaline with 20 ml of 11.7N NaOH; the solution is then extracted with 2 × 25 ml of ether, dried with magnesium sulfate and evaporated to dryness.
0.60 g of the product of the formula 5- [3- (N-methylamino) propylidene] -5H-dibenzo- [a, d] cycloheptene are obtained in the form of a thick oil which crystallizes after inoculation. The oil is characterized by the fact that it forms its salt with oxalic acid in ethanol. The melting point of the hemioxalate is 202-203 C (dec.).
