Verfahren zur Herstellung von Dibenzocyclopentenen
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von neuen 5H-Dibenzo[a,d]cyclo- heptenen, die im 5-Kohlenstoffatom mit einem Alkylamino-propyliden-Radikal substituiert sind.
Die neuen, erfindungsgemäss erzeugten Verbindungen können durch die vorliegende, allgemeine Formel veranschaulicht werden:

Mehr als 1 dieser Substituenten kann an jedem Benzenoidring anwesend sein. Die erfindungsgemäss erzeugten Verbindungen können Substituenten an der Propyliden Kette, wie Niederalkylradikale, vorteilhafterweise von 1 bis 4 Kohlenstoffatomen, aufweisen.
Die durch die oben angeführte Formel veranschaulichten Verbindungen sind bei der Behandlung von Geistesstörungen nützlich, da sie antidepressiv wirken und zur Verbesserung der Gemütsverfassung oder zur psychischen Kräftigung dienen. Für diesen Zweck bewegt sich die tägliche Dosis innerhalb des Bereiches von 5 bis 250 mg, vorteilhafterweise in geteilten Mengen innerhalb des ganzen Tages eingenommen. Die erfindungsgemäss erzeugten Verbindungen werden vorzugsweise in Form ihrer sauren Additionssalze verabreicht.
Das erfindungsgemässe Verfahren zur Herstellung obiger Verbindungen ist dadurch gekennzeichnet, dass man eine Verbindung der Formel:

Dieses Verfahren geht vom bekannten Keton aus, welches gemäss dem von A. C. Cope und Mitarb. in ihrer Veröffentlichung Cyclic Polyolefins, XV, l-methylene-2,3,6,7-dibenzocycloheptatriene in J. A. C. 5.
73, 1673-1678 (1951) beschriebenen Verfahren hergestellt werden kann. Die Ausgangsverbindungen aber und insbesondere jene mit Substituenten an den Benzol Ringen können hergestellt werden gemäss Angaben von T. W. Campbell und Mitarb. in Synthesis of 2'-acetamido-2,3: 6,7-dibenzotropilidene and 2-acetamido-9,9dimethyl-fluorono in Helv. Chim. Acta 36, 1489 bis 1499 (1953). Die Herstellung der Ausgangsverbindungen mit einem Brom-Substituenten in entweder der 10oder ll-Stellung kann, wie von W. Treibs und H. J.
Klinkhammer, Chemische Berichte 84, 671-679 (1951) beschrieben, erfolgen.
Das Verfahren beruht auf der Bindung einer Diniederalkyl-propyliden-Kette am 5-Kohlenstoff des Kernes. Zu diesem Zwecke wird mit Erfolg ein Grignard Reagenz auf übliche Weise eingesetzt, und zwar mit Vorteil in einem inerten Lösungsmittel, wie Äther, wobei die Reaktion gewöhnlich bei Rückflusstemperatur beendet wird. Das sich gebildete Grignard-Additionsprodukt wird dann gewöhnlich auf übliche Art hydrolysiert, wobei aber bevorzugt wird, dass die Hydrolyse unter fast neutralen Bedingungen, wie sie durch Zugabe von Ammoniumchlorid-Lösung erhalten werden, durchgeführt wird. Die anschliessende Dehydratationsstufe erfolgt vorzugsweise mit Hilfe eines chemischen Dehydratisierungsmittels, wie Acetylchlorid, Essigsäure-anhydrid oder Trifluoressigsäureanhydrid, welches Mittel dann nachher durch Abdampfen entfernt werden kann.
In der nächsten Stufe wird die Verbindung mit Cyanhalogenid behandelt und die (N-Cyano-N-niederalkyl)aminopropylidenverbindung hydrolysiert.
Das oben angeführte Verfahren wird im nachfolgenden durch ein Beispiel näher erläutert.
Beispiel 10-Brom-5-(3 -methylaminopropyliden)-
5H-dibenzo a,d] cyclohepten A. 10-Brom-5-(3-dimethylaminopropyl)-5-oxy- 5 H-dibenzo[a,d]cyclohepten
Es wird 3 -Dimethylaminopropylmagnesium-chlorid aus 3,4 g, das sind 0,14 g Atome, Magnesium und 17,0 g, das sind 0,14 Mol, von 3-Dimethylaminopropylchlorid in 50 ml trockenem Tetrahydrofuran auf folgende Weise hergestellt. Die Magnesiumspäne werden in einen 200 ml, mit einem Rührer, einem Tropftrichter und einem Rückflusskondensator mit einem Trockenrohr ausgestatteten 3-Halskolben gegeben. Während der Reaktion wurde eine Atmosphäre von trockenem Stickstoff im Apparat aufrechterhalten. Dann wurde ein Jodkristall und 10 ml trockenes Tetrahydrofuran zugegeben.
Hernach fügte man 3 ml der 3-Dimethylaminopropylmagnesiumchlorid-Lösung aus einem vorherigen Versuch zu und erhitzte das Gemisch auf Rückfluss auf dem Dampfbad. Dann versetzte man das Gemisch durch tropfenweise Zugabe mit einer Lösung von 17,0 g, das sind 0,14 Mol, 3-Dimethylaminopropyl-chlorid in 35 mi trockenem Tetrahydrofuran, wobei der Rückfluss, durch Wärmezufuhr falls notwendig, aufrechterhalten wurde. Nach Beendigung dieser Zugabe wurde das Gemisch während 2 Stunden erhitzt, sobald fast sämtliches Magnesium gelöst wurde.
Statt der Verwendung von 3-Dimethylaminopropylmagnesiumchlorid zur Initierung der Reaktion, kann Äthylbromid in einer Menge von 0,05 Mol pro Mol Magnesium verwendet werden.
Eine Lösung von 20,0 g, das sind 0,07 Mol, 1 0-Brom-5H-dibenzo[a,d]cyclohepten-5-on in 40 ml Tetrahydrofuran wird zur Grignard-Lösung, unter Kühlung in einem Eisbad und unter Rühren innerhalb von 30 Minuten, zutropfen gelassen. Danach wird das Gemisch auf Zimmertemperatur unter Rühren während 2 Stunden erwärmen gelassen. Die Hauptmasse an Lösungsmittel wird dann unterhalb 500 C unter vermindertem Druck abdestilliert, der Rückstand wird in 50 ml Benzol gelöst und das Grignard-Additionsprodukt durch Zugabe von 25 ml Wasser zur Lösung unter Rühren und Kühlen hydrolysiert. Die Benzollösung wird dann vom Magnesiumbrei, durch Dekantieren abgeschieden, und der Brei wird mit weiteren Portionen Benzol extrahiert. Nach Waschen mit Wasser wird das Benzol aus den vereinigten Extrakten destilliert und ergibt 25,67 g eines gelben öligen Rückstandes.
Der Rückstand wird durch Erwärmen in 350 ml einer 0,5n Zitronensäure-Lösung gelöst und die Lösung mit Benzol rückextrahiert. Der Säureextrakt wird dann mit Natriumhydroxyd alkalisiert und die Base in Methylenchlorid extrahiert. Nach Waschen mit Wasser wird das Lösungsmittel abgedampft und der Rückstand aus einem Gemisch von Cyclohexan und Hexan kristallisiert. Der Ertrag an weissem kristallinem Produkt mit F. = 118 bis 119,50 C beträgt 20 g. Durch Umkristallisieren aus Hexan steigt der Schmelzpunkt auf 118,5 bis 1200 C.

14,75 g, das sind 0,0396 Mol,
10-Brom-5-(3 -dimethylaminopropyl) -5-oxy
5H-dibenzo [a,d]cyclohepten werden in 100 ml Trifluoressigsäure gelöst. Dann werden 50 ml Trifluor-essigsäureanhydrid zugesetzt, und die Lösung wird bei Rückfluss unter Rühren während 11/2 Stunden erhitzt. Der Überschuss an Trifluor-essigsäureanhydrid und an Trifluoressigsäure wird abdestilliert, der Rückstand in Wasser suspendiert und das Gemisch mit Natriumhydroxydlösung alkalisiert.
Die Base wird in Äther extrahiert, und nach Waschen des Extraktes mit Wasser wird der Äther abdestilliert, und es hinterbleibt als Rückstand das ölige Produkt in einem Ertrag von 13,75 g, das ist 98.
C. 1 0-Brom-5-[3 -(N-cyano-N-methyl)-amino- propyliden] -5 H-dibenzo [ a, d] cyclohepten
2,83 g, das sind 0,008 Mol, 1 0-Brom-5-(3-dimethylaminopropyliden)-
5H-dibenzo[a,d]cyclohepten in 2 ml trockenem Benzol werden zu einer Lösung von 1,02 g, das sind 0,0096 Mol, Cyan-bromid in 4 ml Benzol unter Rühren und unter Aufrechterhaltung der Temperatur von 25-300C zutropfen gelassen. Während der Zugabe wird ein langsamer Stickstoffstrom durch die Apparatur geführt. Nach Beendigung der Zugabe wird das Gemisch während 4 Stunden gerührt und dann während 15 Stunden bei Zimmertemperatur stehengelassen. Danach wird das Lösungsmittel unter reduziertem Druck abdestilliert. Der Rückstand wird in Methylenchlorid aufgenommen und zur Trockene eingeengt.
Danach wird der Rückstand nochmals in Methylenchlorid aufgelöst und die Lösung mit ln Chlorwasserstoffsäure extrahiert. Nach Waschen mit Wasser wird die Methylenchloridlösung über Natriumsulfat getrocknet und ergibt nach Abdampfen des Lösungsmittels 2,15 g des Produktes in Form eines öligen Rückstandes.
D. 10-Brom-5-(3-methylaminopropyliden)- 5H-dibenzoà,dcydohepten
2,0 g, das sind 0,00548 Mol, lO-Brom-5-[3-(N-cyano-N-methyl)-amino- propyliden]-5H-dibenzo[a,d]cyclohepten werden in 40 ml Eisessigsäure gelöst. Danach werden 27 ml Wasser und 4 ml konzentrierte Chlorwasserstoffsäure zugesetzt, und das Gemisch wird bei Rückfluss unter Rühren während 17 Stunden erhitzt. Das Lösungsmittel wird unter vermindertem Druck abdestilliert und der Rückstand in 65 ml Wasser gelöst und die Lösung mit Benzol extrahiert. Die wässrige Schicht wird mit Natriumhydroxyd alkalisch gemacht und das Produkt in Benzol extrahiert.
Nach Waschen mit Wasser und nach Abdestillieren des Benzols bleiben die geometrischen Isomere von
1 0-Brom5-(3 -methylaminopropyliden)-
5 H-dibenzo [a,d] cyclohepten in einem Ertrag von 1,86 g und in Form eines gelben öligen Rückstandes.
E. 1 0-Brom-5-(3-methylaminopropyliden)-5H- dibenzo[a,d]cyclohepten-hydrogenmaleat (a-Isomer)
1,65 g, das sind 0,00485 Mol, des gemäss Stufe D erhaltenen Produktes werden in 15 ml absolutem Alkohol gelöst. Dann wird eine Lösung von 0,620 g, das sind 0,00533 Mol, Maleinsäure in 4 ml absolutem Alkohol zugegeben und die Lösung mit absolutem Ather zwecks Einleitung der Trübung verdünnt. Es wurde ein erster Ertrag an Kristallen vom Gewicht von 0,55 g gesammelt. Dieses Material sinterte bei 153,50 C und schmolz bei 1590 C.
Ein zweiter Ertrag des Produktes, erhalten aus der Mutterlauge, hatte ein Gewicht von 0,25 g mit F. = 151,5-155,50 C. Die vereinigten Produkte wurden dreimal aus Methanol-Äther-Gemischen umkristallisiert und ergaben ein Material mit einem konstanten F. = 166-167,50 C.

Die Mutterlaugen aus einem anderen Experiment, wie z. B. E, wurden eingeengt, und das amorphe Hydrogenmaleat des ,B-Isomers wurde in die Base umgewandelt. 10,8 g dieser Base wurden in 20 ml Benzol gelöst, und es wurden jedesmal 10 ml dieser Lösung auf zwei Chromatographie-Säulen, erzeugt aus Polyäthylen-Röhren vom Durchmesser von 2 cm und gefüllt mit 60 g Tonerde von der Qualität Merck-Reagenz, und aktiviert in situ mittels Aceton, durchgesetzt. Jede Säure wurde dann durch Durchleiten von 100 ml Chloroform entwickelt.
Die Kolonnen wurden dann in der Länge aufgeschlitzt. Es ist eine Zone ungefähr 1/3 der Länge vom oberen Ende des Adsorbents sichtbar. Die Sektion der Säule unterhalb dieser Zone wird entfernt und mit siedendem Methanol eluiert. Nach Separieren des Feststoffes wird das Methanol abdestilliert und hinterlässt 1,66 g eines gelben öligen Rückstandes. 0,49 g dieser Base wird in 10 ml absolutem Äthanol gelöst und mit einer Lösung von 0,142 g, das sind 0,00158 Mol, Oxalsäure in 5 ml absolutem Methanol behandelt. Das Hydrogenoxalatsalz des Produktes scheidet sich als hellgelber Feststoff ab. Drei Umkristallisierungen aus absolutem Äthanol ergeben das Produkt mit F. = 219 bis 2200 C.

The present invention relates to a process for the preparation of new 5H-dibenzo [a, d] cycloheptens which are substituted in the 5-carbon atom with an alkylamino-propylidene radical.

More than 1 of these substituents can be present on each benzene ring. The compounds produced according to the invention can have substituents on the propylidene chain, such as lower alkyl radicals, advantageously of 1 to 4 carbon atoms.
The compounds represented by the above formula are useful in the treatment of mental disorders because they have antidepressant effects and are used for mood enhancement or psychological strengthening. For this purpose the daily dose is within the range of 5 to 250 mg, advantageously taken in divided amounts throughout the day. The compounds produced according to the invention are preferably administered in the form of their acidic addition salts.
The process according to the invention for the preparation of the above compounds is characterized in that a compound of the formula:

This process is based on the known ketone, which according to the method of A. C. Cope and colleagues. in their publication Cyclic Polyolefins, XV, l-methylene-2,3,6,7-dibenzocycloheptatriene in J.A.C. 5.
73, 1673-1678 (1951) can be prepared. The starting compounds, however, and in particular those with substituents on the benzene rings, can be prepared according to the information provided by T. W. Campbell and colleagues. in Synthesis of 2'-acetamido-2,3: 6,7-dibenzotropilidene and 2-acetamido-9,9dimethyl-fluorono in Helv. Chim. Acta 36, 1489-1499 (1953). The preparation of the starting compounds with a bromine substituent in either the 10 or II position can be carried out as described by W. Treibs and H. J.
Klinkhammer, Chemischeberichte 84, 671-679 (1951).
The process is based on the attachment of a di-lower alkyl propylidene chain to the 5-carbon of the core. For this purpose, a Grignard reagent has been successfully used in the usual manner, advantageously in an inert solvent such as ether, the reaction usually being terminated at reflux temperature. The Grignard addition product formed is then usually hydrolyzed in the usual way, but it is preferred that the hydrolysis is carried out under almost neutral conditions, such as are obtained by adding ammonium chloride solution. The subsequent dehydration stage is preferably carried out with the aid of a chemical dehydrating agent such as acetyl chloride, acetic anhydride or trifluoroacetic anhydride, which agent can then be removed afterwards by evaporation.
In the next step, the compound is treated with cyanide halide and the (N-cyano-N-lower alkyl) aminopropylidene compound is hydrolyzed.
The above-mentioned method is explained in more detail below by means of an example.
Example 10-Bromo-5- (3-methylaminopropylidene) -
5H-dibenzo a, d] cycloheptene A. 10-Bromo-5- (3-dimethylaminopropyl) -5-oxy-5 H-dibenzo [a, d] cycloheptene
3-Dimethylaminopropylmagnesium chloride is prepared from 3.4 g, that is 0.14 g of atoms, magnesium and 17.0 g, that is 0.14 mol, of 3-dimethylaminopropyl chloride in 50 ml of dry tetrahydrofuran in the following manner. The magnesium turnings are placed in a 200 ml 3-neck flask equipped with a stirrer, a dropping funnel and a reflux condenser with a drying tube. An atmosphere of dry nitrogen was maintained in the apparatus during the reaction. Then a crystal of iodine and 10 ml of dry tetrahydrofuran were added.
Then 3 ml of the 3-dimethylaminopropyl magnesium chloride solution from a previous experiment were added and the mixture was heated to reflux on the steam bath. A solution of 17.0 g, that is 0.14 mol, of 3-dimethylaminopropyl chloride in 35 ml of dry tetrahydrofuran was then added dropwise to the mixture, the reflux being maintained by supplying heat if necessary. After this addition was complete, the mixture was heated for 2 hours when almost all of the magnesium was dissolved.
Instead of using 3-dimethylaminopropyl magnesium chloride to initiate the reaction, ethyl bromide can be used in an amount of 0.05 mole per mole of magnesium.
A solution of 20.0 g, that is 0.07 mol, 1 0-bromo-5H-dibenzo [a, d] cyclohepten-5-one in 40 ml of tetrahydrofuran becomes the Grignard solution, with cooling in an ice bath and under Stir within 30 minutes, added dropwise. Thereafter, the mixture is allowed to warm to room temperature with stirring for 2 hours. The bulk of the solvent is then distilled off at below 500 ° C. under reduced pressure, the residue is dissolved in 50 ml of benzene and the Grignard addition product is hydrolyzed by adding 25 ml of water to the solution with stirring and cooling. The benzene solution is then separated from the magnesium slurry by decanting, and the slurry is extracted with additional portions of benzene. After washing with water, the benzene is distilled from the combined extracts and gives 25.67 g of a yellow oily residue.
The residue is dissolved by heating in 350 ml of a 0.5N citric acid solution and the solution is back-extracted with benzene. The acid extract is then made alkaline with sodium hydroxide and the base is extracted into methylene chloride. After washing with water, the solvent is evaporated and the residue is crystallized from a mixture of cyclohexane and hexane. The yield of white crystalline product with F. = 118 to 119.50 C is 20 g. Recrystallization from hexane increases the melting point to 118.5 to 1200 C.

14.75 g, that is 0.0396 mol,
10-bromo-5- (3-dimethylaminopropyl) -5-oxy
5H-dibenzo [a, d] cycloheptene are dissolved in 100 ml of trifluoroacetic acid. Then 50 ml of trifluoroacetic anhydride are added and the solution is heated at reflux with stirring for 11/2 hours. The excess of trifluoroacetic anhydride and trifluoroacetic acid is distilled off, the residue is suspended in water and the mixture is made alkaline with sodium hydroxide solution.
The base is extracted into ether, and after washing the extract with water, the ether is distilled off, and the oily product remains as a residue in a yield of 13.75 g, that is 98.
C. 10-Bromo-5- [3 - (N-cyano-N-methyl) -amino-propylidene] -5 H-dibenzo [a, d] cycloheptene
2.83 g, that is 0.008 mol, 1 0-bromo-5- (3-dimethylaminopropylidene) -
5H-dibenzo [a, d] cycloheptene in 2 ml of dry benzene are added dropwise to a solution of 1.02 g, that is 0.0096 mol, of cyanobromide in 4 ml of benzene with stirring and while maintaining the temperature of 25-300C calmly. During the addition, a slow stream of nitrogen is passed through the apparatus. After the addition is complete, the mixture is stirred for 4 hours and then left to stand for 15 hours at room temperature. The solvent is then distilled off under reduced pressure. The residue is taken up in methylene chloride and concentrated to dryness.
The residue is then redissolved in methylene chloride and the solution is extracted with 1N hydrochloric acid. After washing with water, the methylene chloride solution is dried over sodium sulfate and, after evaporation of the solvent, gives 2.15 g of the product in the form of an oily residue.
D. 10-Bromo-5- (3-methylaminopropylidene) -5H-dibenzoà, dcydohepten
2.0 g, that is 0.00548 mol, 10-bromo-5- [3- (N-cyano-N-methyl) -amino-propylidene] -5H-dibenzo [a, d] cycloheptene are in 40 ml of glacial acetic acid solved. Then 27 ml of water and 4 ml of concentrated hydrochloric acid are added and the mixture is heated at reflux with stirring for 17 hours. The solvent is distilled off under reduced pressure and the residue is dissolved in 65 ml of water and the solution is extracted with benzene. The aqueous layer is made alkaline with sodium hydroxide and the product extracted into benzene.
After washing with water and after distilling off the benzene, the geometric isomers of
1 0-bromo5- (3-methylaminopropylidene) -
5 H-dibenzo [a, d] cycloheptene in a yield of 1.86 g and in the form of a yellow oily residue.
E. 1 0-Bromo-5- (3-methylaminopropylidene) -5H-dibenzo [a, d] cycloheptene hydrogen maleate (a-isomer)
1.65 g, that is 0.00485 mol, of the product obtained in step D are dissolved in 15 ml of absolute alcohol. Then a solution of 0.620 g, that is 0.00533 mol, of maleic acid in 4 ml of absolute alcohol is added and the solution is diluted with absolute ether to induce turbidity. An initial crop of crystals weighing 0.55 g was collected. This material sintered at 153.50 C and melted at 1590 C.
A second crop of the product, obtained from the mother liquor, had a weight of 0.25 g with F. = 151.5-155.50 C. The combined products were recrystallized three times from methanol-ether mixtures to give a material with a constant F. = 166-167.50 C.

The mother liquors from another experiment, e.g. B. E, were concentrated and the amorphous hydrogen maleate of the, B isomer was converted to base. 10.8 g of this base were dissolved in 20 ml of benzene, and each time 10 ml of this solution were applied to two chromatography columns made from polyethylene tubes with a diameter of 2 cm and filled with 60 g of Merck reagent quality clay. and activated in situ using acetone. Each acid was then evolved by bubbling 100 ml of chloroform through it.
The columns were then slit lengthways. A zone about 1/3 the length from the top of the adsorbent is visible. The section of the column below this zone is removed and eluted with boiling methanol. After separating the solid, the methanol is distilled off and leaves 1.66 g of a yellow oily residue. 0.49 g of this base is dissolved in 10 ml of absolute ethanol and treated with a solution of 0.142 g, that is 0.00158 mol, of oxalic acid in 5 ml of absolute methanol. The hydrogen oxalate salt of the product separates out as a light yellow solid. Three recrystallizations from absolute ethanol give the product with F. = 219 to 2200 C.
