Procédé de préparation d'un dérivé de la colchicine
La présente invention a pour objet un nouveau procédé de préparation d'un dérivé de la colchicine, la 1-hy droxyméthyl-colchicine de formule :

Le composé préparé par le procédé selon l'invention possède une activité biologique intéressante. Plus particulièrement, il exerce une action antimitotique remarquable et peut, d'autre part, être utilisé industriellement en agriculture pour les modifications de la mitose et la création de polyploides, soit par épandage de solutions aqueuses ou de suspensions des produits sur des sols cultivés, soit par traitement préalable des semences avec le produit pur ou dilué dans un solvant ou sur un support.
Le procédé selon la présente invention est caractérisé en ce que l'on fait réagir la 2-déméthylcolchicine avec le formaldéhyde, en présence d'une dialcoylamine inférieure, transforme la 1-dialcoylaminométhyl-2-déméthylcolchicine résultante par action d'un halogénure d'alcoyle inférieur en sel d'ammonium quaternaire correspondant, soumet ce dernier à l'hydrolyse alcaline, obtient la 1-hy droxyméthyl-2-déméthyl-colchicine que l'on soumet à l'action d'un agent de méthylation et isole la 1-hydroxy- méthyl-colchicine.
L'intérêt du présent procédé réside dans le fait que le produit utilisé comme produit de départ, la 2-déméthyl- colchicine, est un sous-produit industriel, ce qui permet d'obtenir le produit final dans des conditions avantageuses.
Le procédé de l'invention est de préférence exécuté dans les conditions suivantes : -on fait réagir la 2-déméthyl-colchicine avec le form
aldéhyde, en présence de diméthylamine, et obtient
la 1-diméthylaminométhyl-2-déméthyl-colchicine ; -on transforme la 1-diméthylaminométhyl-2-démé-
thyl-colchicine en son sel d'ammonium quaternaire
par action de l'iodure de méthyle dans l'acétone, et
obtient le métho-iodure correspondant ; --on effectue l'hydrolyse alcaline du métho-iodure de
la 1-diméthylaminométhyl-2-déméthyl-colchicine au
moyen d'une solution aqueuse de bicarbonate de so
dium, et obtient la 1-hydroxyméthyl-2-déméthyl-col
chicine ;
-on effectue la méthylation de la 1-hydroxyméthyl-2
déméthyl-colchicine dans le méthanol au moyen d'une
solution de diazométhane dans le chlorure de méthv
lène, et obtient la 1-hydroxyméthyl-colchicine.

Préparation de la l-hydroxyméthyl-colchicine
Stade A : l-diméthylaminométhyl-2-déméthyl-
colchicine
On dissout 5 g de 2-déméthyl-colchicine dans 60 cm3 de méthanol et ajoute lentement, entre 0 et 5o C, 16 cm3 de formaldéhyde et 16 cm3 de diméthylamine.
On porte le mélange réactionnel au reflux pendant neuf heures et l'abandonne à la température ambiante pendant une nuit. On concentre, ajoute 100cm3 d'eau, sature de chlorure de sodium, extrait au chloroforme, lave à l'eau, sèche sur sulfate de magnésium et évapore à sec sous vide.
On reprend le résidu par 50cm3 de benzène qu'on évapore ensuite, ajoute 100cm3 d'éther isopropylique, concentre, glace, essore, lave à l'éther isopropylique et sèche à 900 C.
On obtient ainsi 5, 5g de 1-diméthylaminométhyl-2 déméthyl-colchicine brute (F = environ 2000 C) rendement 96"/o, qu'on purifie par chromatographie sur alumine. On élue par le chloru. re de méthylène à 1 /o de méthanol et recueille la 1-diméthylaminométhyl-2-démé thyl-colchicine pure. F = 198 C, [a] 2D0 =-920 2 (c=0,65%, chloroforme) qui cristallise dans le mélange acétone-éther isopropylique.
Le produit se présente sous forme de cristaux jaunes, solubles dans l'eau, les acides et alcalis dilués, l'alcool et le chloroforme, peu solubles dans l'acétone et le benzène, et insolubles dans l'éther.

2-dÚmÚthyl-colchicine
On abandonne pendant quatre jours à la température ambiante le mélange suivant : 1-diméthylaminométhyl-2-déméthyl-colchicine. 0, 5 g acétone 10 cm3 iodure de méthyle 2, 5 cm3
On essore le produit cristallisé formé, lave à l'acétone, laisse sécher à l'air pendant trois heures, et obtient ainsi 475 mg de métho-iodure de 1-diméthylaminométhyl-2 déméthyl-colchicine, soit un rendement de 72 /o, F= environ 2600 C, qui se présente sous forme de produit jaune, soluble dans l'eau, les acides et alcalis dilués et l'alcool, peu soluble dans l'acétone,
le benzène et le chloroforme et insoluble dans l'Úther.

On abandonne pendant quarante-huit heures à la température ambiante le mélange suivant :

solution saturée de bicarbonate de sodium 6 cm3
On acidifie par addition d'acide chlorhydrique 2N, sature de chlorure de sodium, extrait au chloroforme, lave à l'eau salée, sèche sur sulfate de magnésium et Úvapore à sec sous vide.
On obtient 470 mg de 1-hydroxyméthyl-2-déméthylcolchicine brute.
Ce composé n'est pas décrit dans la littérature.
L'extraction des eaux mères par le chloroforme à 10 /o d'éthanol fournit 50 mg supplémentaires.
On dissout les deux fractions dans 5 cm3 de méthanol, ajoute 30 cm3 d'une solution à 0, 4 ouzo de diazométhane dans le cholrure de méthylène et abandonne le mé- lange pendant une nuit à 00 C.
On évapore à sec sous vide et chromatographie sur alumine, par élution au chlorure de méthylène à 1, 5 ouzo de méthanol, on obtient 90 mg de 1-hydroxyméthyl-col- chicine qui cristallise dans l'acétate d'éthyle. F = 2600 C.

The compound prepared by the process according to the invention has an interesting biological activity. More particularly, it exerts a remarkable antimitotic action and can, on the other hand, be used industrially in agriculture for the modifications of mitosis and the creation of polyploids, either by spreading aqueous solutions or suspensions of the products on cultivated soils, either by pretreating the seeds with the pure product or diluted in a solvent or on a support.
The process according to the present invention is characterized in that the 2-demethylcolchicine is reacted with formaldehyde, in the presence of a lower dialkylamine, converts the resulting 1-dialkoylaminomethyl-2-demethylcolchicine by the action of a halide of lower alkyl in the corresponding quaternary ammonium salt, subjects the latter to alkaline hydrolysis, obtains 1-hy droxymethyl-2-demethyl-colchicine which is subjected to the action of a methylating agent and isolates the 1 -hydroxy-methyl-colchicine.
The advantage of the present process lies in the fact that the product used as starting product, 2-demethyl-colchicine, is an industrial by-product, which makes it possible to obtain the final product under advantageous conditions.

Preparation of 1-hydroxymethyl-colchicine
Stage A: 1-dimethylaminomethyl-2-demethyl-
colchicine
5 g of 2-demethyl-colchicine are dissolved in 60 cm3 of methanol and slowly added, between 0 and 5o C, 16 cm3 of formaldehyde and 16 cm3 of dimethylamine.
The reaction mixture is refluxed for nine hours and left at room temperature overnight. Concentrated, added 100cm3 of water, saturated with sodium chloride, extracted with chloroform, washed with water, dried over magnesium sulfate and evaporated to dryness in vacuo.
The residue is taken up in 50cm3 of benzene which is then evaporated, 100cm3 of isopropyl ether is added, concentrated, ice, filtered, washed with isopropyl ether and dried at 900 C.
In this way 5.5 g of crude 1-dimethylaminomethyl-2-demethyl-colchicine (F = approximately 2000 C) are obtained, yield 96 "/ o, which is purified by chromatography on alumina. Elution is carried out with 1 / methylene chloro. o of methanol and collects pure 1-dimethylaminomethyl-2-demethyl-colchicine. F = 198 C, [a] 2D0 = -920 2 (c = 0.65%, chloroform) which crystallizes in an acetone-isopropyl ether mixture .
The product is in the form of yellow crystals, soluble in water, dilute acids and alkalis, alcohol and chloroform, sparingly soluble in acetone and benzene, and insoluble in ether.

Acidified by addition of 2N hydrochloric acid, saturated with sodium chloride, extracted with chloroform, washed with salt water, dried over magnesium sulfate and evaporated to dryness under vacuum.
470 mg of crude 1-hydroxymethyl-2-demethylcolchicine are obtained.
This compound is not described in the literature.
Extraction of the mother liquors with chloroform at 10% ethanol provides an additional 50 mg.
The two fractions are dissolved in 5 cm3 of methanol, added 30 cm3 of a 0.4 ouzo solution of diazomethane in methylene cholride and the mixture is left overnight at 00 ° C.
Evaporated to dryness under vacuum and chromatography on alumina, by elution with methylene chloride at 1.5 ouzo of methanol, 90 mg of 1-hydroxymethyl-col chicine are obtained, which crystallizes in ethyl acetate. M = 2600 C.
