Verfahren zur Herstellung von Trisulfamylanilinen
Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer Trisulfamylaniline der Formel:

in der R Halogen, eine Trifluormethylgruppe, eine Alkyl- oder Alkoxygruppe, zweckmässig mit nicht mehr als 4 Kohlenstoffatomen, bedeutet oder der Salze von solchen. Diese Verbindungen oder ihre Salze können als Ausgangsstoffe zur Herstellung von substituierten Benzothiadiazinen der Formel':

verwendet werden.
Die erfindungsgemäss erhältlichen Verbindungen können als Arzneimittel verwendet werden, da gefunden wurde, dass die Trisulfamylaniline und ihre Salze wie auch die Benzothiadiazine eine ausgesprochene diuretische oder natriuretische oder diureflsche und auch natriuretische Wirkung haben.
Gemäss dem Verfahren der Erfindung wird unter wasserfreien Bedingungen ein substituiertes Anilin der Formel:

mit Chlorsulfonsäure oder einem Salz dieser Säure umgesetzt und das erhaltene Trisulfonylchlorid mit Ammoniak zur Reaktion gebracht. Die so erhaltene Trisulfamylverbindung kann dann als solche oder in Form eines Salzes isoliert werden.
Ein weiterer Gegenstand vorliegender Erfindung ist die Verwendung der nach obigem Verfahren erhaltenen Verbindungen zur Herstellung von Benzothiadiazinen, dadurch gekennzeichnet, dass man eine Verbindung der Formel I als solche oder in Form eines Salzes mit Oxalsäure zur entsprechenden N (4,6-Disulfamyl-phenyl)-oxaminsäure umsetzt, die zwecks Abspaltung von Wasser und Kohlendioxyd erhitzt wird und dabei das entsprechende 6-substituierte 7-Sulfamyl-1, ,2,4-benzotbiadiazin-l,l-dioxyd der Formel II liefert.

Die Herstellung des Benzothiadiazins unter Verwendung des so erhaltenen Trisulfamylanilins lässt sich wie folgt formulieren:

Von der zur Anwendung kommenden Ammoniakmenge ist es abhängig, ob man entweder die Trisulfamylverbindung oder das Ammoniumsalz erhält.
Aus dem letzteren kann die Trisulfamylverbindung durch Säurezusatz in Freiheit gesetzt werden.
Die Trisulfamylverbindungen sind in wässriger Lösung bei einem pH über g beständig. Beim Ansäuern bis zu einem pH von 5 tritt sogar bei Raumtemperatur Hydrolyse ein, wodurch der Sulfamyl bestandteil in der Aminogruppe abgespalten, wird und sich eine Disulfamylverbindung und Aminosulfon- säure bildet.
Als Ausgangsmaterial werden zweckmässig solche metasubstituierten Aniline verwendet, in denen der m-Substituent Chlor, Brom, eine Methylgruppe oder eine Methoxygruppe darstellt, da diese Verbindungen gewöhnliche, leicht zugänglich, e Handelsprodukte sind.
Es kommen aber z. B. auch Aniline mit Alkyl- oder Alkoxygruppen mit 2 bis 4 Kohlenstoffatomen oder einer Trifluormethylgruppe in der m-Stellung als Ausgangsmaterialien in Betracht.
Die Chiorsulfonierung wird zweckmässig durch tropfenweisen Zusatz einer Lösung des substituierten Anilins bei niedriger Temperatur, z. B. bei 0 bis 50 C, zur Chlorsulfonsäure oder einer Lösung derselben durchgeführt, wonach portionsweise Natriumchlorid zugesetzt, die Temperatur langsam auf 100 bis 1500 C erhöht und die Reaktionsmischung bei dieser Temperatur stehengelassen wird, bis die Ent- wicklung von Chlorwasserstoff beendet ist
Als Lösungsmittel wird zweckmässig ein inertes wasserfreies organisches Lösungsmittel mit einem Siedepunkt oberhalb derjenigen Temperatur verwendet, bei der die Entwicklung von Chlorwasserstoff stattfindet, z. B. Tetrachloräthan.
Als allfälliges Extraktionsmittel für das erhaltene Trisulfamylchlorid kann ebenfalls Tetrachloräthan verwendet werden oder andere Extraktionsmittel, wie z. B. Ather, Benzol, Toluol, Trichloräthylen oder Methylendichlorid.
Die Umwandlung des Trisulfonylchlorids in die Trisulfamylverbindung kann mit Hilfe von wässrigem oder alkoholischem Ammoniak oder mit flüssigem oder gasförmigem trockenem Ammoniak in einem organischen Lösungsmittel, z. B. einem solchen wie bereits erwähnt, durchgeführt werden. Im allgemeinen führt dies zum Ammoniumsalz, aus dem das Sulfamid selbst durch Zusatz einer Säure gewonnen werden kann. Das Sulfamid stellt eine Säure dar und kann leicht durch Behandlung mit einer Base in das Salz verwandelt werden. Im allgemeinen sind die Alkalimetallsalze und auch die Salze des Calciums und des Bariums leicht in Wasser löslich.
Gemäss einer zweckmässigen Ausführungsform des erfindungsgemässen Verfahrens wird die Behandlung mit Ammoniak in wässriger Lösung durchgeführt, wobei ein Ammoniumsalz gebildet wird, aus dem die Trisulfamylverbindung durch vorsichtigen Säurezusatz gefällt wird, da das Sulfamid selbst im wesentlichen in Wasser unlöslich ist.
Wird die so erhaltene Trisulfamylverbindung erfindungsgemäss als Ausgangsstoff zur Herstellung eines Benzothiazins verwendet, wird die erste Reaktionsstufe zweckmässig durch Mischen des Sulfamylanilins mit Oxalsäure durchgeführt, worauf die Mischung geschmolzen wird. Das Schmelzen wird zweckmässig auf einem ölbad bei einer Temperatur von 1500 C oder höher durchgeführt. Während des Schmelzens destilliert das Kristallwasser von der Oxalsäure ab, im allgemeinen in 30-60 Minuten, wonach sich die Schmelze wieder verfestigt. Ein möglicher Überschuss an Oxalsäure kann aus der erhaltenen Oxaminsäure durch Waschen mit Wasser entfernt werden, da die freie Säure selbst und auch ihr Na trium-und Ammoniumsalz in kaltem Wasser schwer löslich sind.
Eine weitere Reinigung kann, sofern erforderlich, durch Umkristallisieren aus Wasser durchgeführt werden.
Beim Erhitzen der so erhaltenen Oxaminsäure, in der Regel auf 225-250 C, schmilzt sie unter Schäumen, und ein Ringschluss findet unter Abspaltung von Wasser und Kohlensäure statt. Das erhaltene, in 6-Stellung substituierte 7-Sulfamyl-1,2,4- benzothiadiazin-l, l-dioxyd kann durch Umkristallisation aus einem wässrigen Alkohol, wie z. B. Sitha- nol, gereinigt werden.
Derselbe Prozess kann vorteilhafterweise in Lösung durch 2- bis 3stündiges Erhitzen der Oxaminsäure unter Rückfluss in einem geeigneten Lösungs mittel, wie z. B. Dimethylformamid, durchgeführt werden.
Gemäss einer anderen Ausführungsform des erfindungsgemässen Verfahrens werden als Ausgangsstoffe Salze der genannten Oxaminsäuren verwendet und diese einem Ringschluss in alkalischer Lösung unterworfen, so dass sich die Salze der 6-substituierten 7 Sulfamyl - 1,2,4 - benzothiadiazin - 1, 1-dioxyd-3-carboxylsäuren bilden, welche dann decarboxyliert werden. Die Abspaltung von Kohlensäure findet wie im Falle der Oxaminsäuren durch Erhitzen statt und führt zu den Salzen der entsprechenden 6-substituier- ten Sulfamyl-1,2,4-benzothiadiazin-1,1-dioxyde.
Beispiel 1
Herstellung von Trisulfamylverbindungen
32 g (0,25 Mol) m-Chloranilin werden in 100 cm3 symmetrischem Tetrachloräthan gelöst, und die Lösung wird tropfenweise bei einer Temperatur von 0-5 C einer Mischung von 225 cm3 Chlorsulfonsäure und 300 cm3 Tetrachloräthan zugesetzt.
Dann werden 240 g Natriumchlorid portionsweise der Reaktionsmischung zugesetzt, worauf die Temperatur langsam auf 1001100 C unter starker Chlorwasserstoffentwicklung erhöht wird.
Die Reaktionsmischung wird auf 1500 C gehalten, bis die Entwicklung von Chlorwasserstoff beendet ist (etwa 3 Stunden).
Nach dem Abkühlen wird der Überschuss von Natriumchlorsulfonat abfiltriert und auf dem Filter mit 300 cm3 Tetrachloräthan gewaschen. Aus der eingedampften Tetrachloräthanlösung kristallisiert nach Kühlung in einer Ausbeute von 85-90 % N-(4,6- Dichlorsulfonyl-3-chlorphenyl)-sulfamylchlorid aus.
Der Schmelzpunkt beträgt 125-126 C nach Umkri stallis ation aus Tetrachloräthan.
Berechnet für C6H3O6S3NCl4:

Das Produkt muss gegen Luftfeuchtigkeit geschützt werden, da es leicht zum 4,6-Dichlor-sulfonyl- 3-chloranilin hydrolysiert.
Das Sulfonylchlorid wird in Benzol gelöst, und trockenes Ammoniak wird zugesetzt. Dabei wird das N-(4,6-Disulfamyl-3-chlorphenyl)-sulfamid in das Ammoniumsalz übergeführt. Das Salz wird in einer kleinen Menge Wasser gelöst und ein Überschuss an gelöster Chlorwasserstoffsäure langsam zugesetzt, wo bei sich das Sulfamid, das in Wasser unlöslich ist, in einer Ausbeute von 85-90 % der Theorie niederschlägt. Die Verbindung, die bei 2850 C unter Zersetzung schmolz, stellt eine Säure mit einem pH von etwa 6,5 dar.
Berechnet für C6H9O6S3N4Cl:

In entsprechend'er Weise erhält man: a) Aus m-Bromanilin das N-(4,6-Dichlorsulfonyl- 3-bromphenyl)-sulfamylchlorid mit einem Schmelzpunkt von 145-146 C. Die Ausbeute beträgt 86 % der Theorie, und aus dieser Verbindung kann N-(4,6 Disulfamyl-3-bromphenyl)-sulfamid mit einem Schmelzpunkt von 257-258 C in einer Ausbeute von 88 % erhalten werden; b) Aus m-Toluidin das N-(4, 6-Dichlorsulfonyl-3- methylphenyl)-sulfamylchlorid mit einem Schmelzpunkt von 144-145 C.
Die Ausbeute beträgt 59 %, und aus dieser Verbindung kann N-(4,6-Disulfamyl- 3-methylphenyl)-sulfamid mit einem Schmelzpunkt von 259-261 C unter Zersetzung in einer Ausbeute von 76 % erhalten werden; c) Aus m-Anisidin das N-(4,6-Dichlorsulfonyl-3- methoxyphenyl)-sulfamylchlorid mit einem Schmelzpunkt von 125-126 C. Die Ausbeute beträgt 65 %, und aus dieser Verbindung kann N-(4,6-Disulfamyl- 3-methoxyphenyl)-sulfamid mit einem Schmelzpunkt von 248-249 C unter Zersetzung in einer Ausbeute von 89 % erhalten werden; d) Aus m-Trifluormethylanilin das N-(4,6-Di- chlorsulfonyl-3-trifluormethylphenyl)-sulfamylchlorid als nichtkristallisierendes Öl.
Aus dieser Verbindung wird N-(4,6-Disulfamyl-3-triflfuormethylphenyl)-sulfamid mit einem Schmelzpunkt von 288-290 C unter Zersetzung erhalten.
Beispiel 2
12,8 g N - (3 - Chlor - 4,ödisulfamylpbenyl)-sulf- amid (0,035 Mol) werden in einem Mörser mit 40 g Oxalsäuredihydrat gemischt, worauf die Mischung auf 1500 C auf einem Ölbad erhitzt und 30 Minuten lang auf dieser Temperatur gehalten wird. Die Mischung schmilzt, wenn das Hydratwasser der Oxalsäure abdestilliert, worauf sie sich wieder verfestigt.
Die Reaktionsmischung wird, um die zurückbleibende Oxalsäure zu entfernen, mit kaltem Wasser gewaschen und dann in 100 cm3 heissem Wasser gelöst.
Durch Stehenlassen beim Abkühlen kristallisieren 11,5 g N-(3-Chlor- 4,6-disulfamylphenyl)-oxaminsäure aus. Der Schmelzpunkt beträgt 234-235 C unter Zersetzung, und die Ausbeute liegt bei 91 % der Theorie.
20 g der Oxaminsäure (0,056 Mol) werden auf einem Nitritbad auf 2500 C erhitzt. Während des Schmelzens werden Kohlensäure und Wasser frei, worauf sich die Masse wieder verfestigt. Der feste Rückstand wird aus 75 % igem wässrigem Äthanol umkristallisiert, wobei man 11 g 6-Chlor-7-sulfamyl1,2,4-benzothiadiazin-1,1-dioxyd mit einem Schmelzpunkt von 342-343 C erhält. Die Ausbeute entspricht 67 % der Theorie.
Weitere 20 g der Oxaminsäure werden in 50 cm3 Dimethylformamid gelöst, und die Lösung wird 2 Stunden unter Rückfluss gehalten. Das Lösungsmittel wird im Vakuum abdestilliert, worauf der feste Rückstand aus 75 Sigem wässrigem Äthanol umkri, stallisiert wird. Die Ausbeute beträgt 13 g 6-Chlor-7 sulfamyl-1, 2, 4-benzothiadiazin-1, 1-dioxyd mit einem Schmelzpunkt von 342-343 C, entsprechend 79 %.
Eine Lösung von 15 g der Oxaminsäure in
100 cm3 10%igem Ammoniak wird eine Stunde ull- ter Rückfluss gehalten, worauf die Lösung im Vakuum zur Trockne eingedampft wird. Der Rückstand wird in 200 cm3 kochendem Wasser gelöst und 40 cm3 konzentrierte Chlorwasserstoffsäure werden zugesetzt. Beim Abkühlen kristallisiert oChlor-7- sulfamyl-1,2,4-benzothiadiazin-1,1-dioxyd-3-carbonsäure aus. Der Schmelzpunkt beträgt 2250 C (unter Zersetzung), die Ausbeute beträgt 12 g.
Eine Lösung von 10 g dieser Carbonsäure in 25 cm3 Dimethylformamid wird eine Stunde unter Rückfluss gehalten. Das Lösungsmittel wirdi im Vakuum abdestilliert, worauf der feste Rückstand aus 75 % igem wässrigem Äthanol umkristzallisiert wird.
Die Ausbeute beträgt 8 g 6-Chlor-7-sulfamyl-1, ,2,4- benzothiadiazin-1,1-dioxyd (92% mit einem Schmelzpunkt von 342-343 C.
Beispiel 3
Bei der Beispiel 2 analogen Arbeitsweise ergeben 14,3 g N-(3-Brom-4,6-disulfamyl)-sulfamid 11,3 g (84 %) N-(3-Brom-4,6-disulfamyl)-oxaminsäure mit einem Schmelzpunkt von 222-224 C unter Zersetzung. Unter Erhitzen auf 230-235 C erhält man aus 23 g (0,06 Mol) der genannten Oxaminsäure 16,5 g (81 S) 6-Brom-7-sulfamyl-1 ,2,4-benzothiadiazin-1 1- dioxyd mit einem Schmelzpunkt von 3473490 C (unter Zersetzung).
Beispiel 4
Gemäss der Verfahrensweise von Beispiel 2 ergeben 5,2 g (0,015 Mol) N-(3-Methyl-4,6-disulfamyl- phenyl)-sulfamid und 15 g Oxalsäuredihydrat 3,4 g (71 S) N- (3-Methyl-4, 6-disulfamylphenyl)-oxamin- säure mit einem Schmelzpunkt von 230-232 C (Zersetzung). Kocht man 3,2 g (0,01 Mol) dieser Oxaminsäure 2 Stunden unter Rückfluss in 10 cm3 Dimethylformamid, so werden 1,8 g (69 %) 6-Methyl7 - sulfamyl-1,2,4-benzothiadiazin-1,1-dioxyd mit einem Schmelzpunkt von 343-345 C (Zersetzung) erhalten.
Beispiel 5
Gemäss der in Beispiel 2 beschriebenen Arbeitsweise ergeben 10,9 g (0,03 Mol) N-(3-Methoxy-4,6- disulfamylphenyl)-sulfamid 8,1 g (80%) N-(3-Meth oxy -4,6- disulfamylphenyl) - oxaminsäure mit einem Schmelzpunkt von 232-235 C (Zersetzung). Nach zweistündigem Erhitzen dieser Säure am Rückfluss in 25 cm3 Dimethylformamid erhält man 5,0 g (86 %) 6 - Methoxy-7-sulfamyl-1,2,4-benzothiadiazin-1,1-dioxyd mit einem Schmelzpunkt von 309-310 C (Zersetzung).

It depends on the amount of ammonia used, whether either the trisulfamyl compound or the ammonium salt is obtained.
The trisulfamyl compound can be set free from the latter by adding acid.
The trisulfamyl compounds are stable in aqueous solution at a pH above g. When acidifying up to a pH of 5, hydrolysis occurs even at room temperature, whereby the sulfamyl component in the amino group is split off and a disulfamyl compound and aminosulfonic acid are formed.
Metasubstituted anilines in which the m-substituent represents chlorine, bromine, a methyl group or a methoxy group, since these compounds are common, easily accessible, commercial products are expediently used as the starting material.
But z. B. also anilines with alkyl or alkoxy groups with 2 to 4 carbon atoms or a trifluoromethyl group in the m-position as starting materials into consideration.
The chlorosulfonation is conveniently carried out by adding dropwise a solution of the substituted aniline at low temperature, e.g. B. at 0 to 50 C, carried out to chlorosulfonic acid or a solution thereof, after which sodium chloride was added in portions, the temperature slowly increased to 100 to 1500 C and the reaction mixture is allowed to stand at this temperature until the evolution of hydrogen chloride has ended
The solvent used is an inert anhydrous organic solvent with a boiling point above that temperature at which the evolution of hydrogen chloride takes place, z. B. tetrachloroethane.
Tetrachloroethane can also be used as any extraction agent for the trisulfamyl chloride obtained, or other extraction agents, such as. B. ether, benzene, toluene, trichlorethylene or methylene dichloride.
The conversion of the trisulfonyl chloride into the trisulfamyl compound can be carried out with the aid of aqueous or alcoholic ammonia or with liquid or gaseous dry ammonia in an organic solvent, e.g. B. such as already mentioned, be carried out. In general, this leads to the ammonium salt, from which the sulfamide itself can be obtained by adding an acid. Melting is expediently carried out on an oil bath at a temperature of 1500 ° C. or higher. During melting, the water of crystallization distills off from the oxalic acid, generally in 30-60 minutes, after which the melt solidifies again. A possible excess of oxalic acid can be removed from the oxamic acid obtained by washing with water, since the free acid itself and also its sodium and ammonium salt are sparingly soluble in cold water.
If necessary, further purification can be carried out by recrystallization from water.
When the oxamic acid obtained in this way is heated, usually to 225-250 ° C., it melts with foaming, and a ring closure takes place with elimination of water and carbonic acid. The product must be protected from atmospheric moisture as it easily hydrolyzes to 4,6-dichlorosulfonyl-3-chloroaniline.
The sulfonyl chloride is dissolved in benzene and dry ammonia is added. The N- (4,6-disulfamyl-3-chlorophenyl) sulfamide is converted into the ammonium salt. The salt is dissolved in a small amount of water and an excess of dissolved hydrochloric acid is slowly added, whereupon the sulfamide, which is insoluble in water, is precipitated in a yield of 85-90% of theory. The compound, which melted with decomposition at 2850 C, is an acid with a pH of about 6.5.
