Verfahren zur Darstellung von neuen Sulfanilamido-polyalkylpyrimidinen
Es wurden bisher schon eine grosse Reihe von Sulfanilamiden dargestellt. Von den am Amidstickstoff heterocyclisch substituierten Verbindungen ist eine ganze Reihe bekanntgeworden ; sie sind aber zum Teil wenig oder sogar unwirksam.
Auch einige Pyrimidinderivate sind bereits beschrieben worden. So erwies sich das 4-[p- Amino-benzolsulfamido]-pyrimidin (Roblin & Mit- arb., J. Am. Chem. Soc., 62, 2002, [1940]) als chemotherapeutisch inaktiv. Das 2-[p-Amino-

relativ gut wirksam. Daraus ist zu ersehen, dass auch unter den Pyrimidinderivaten verhältnismässig grosse Unterschiede in der Wirksamkeit bestehen.
Es wurde nun die überraschende Beobachtung gemacht, dass Sulfanilamido-polyalkylpyrimidine sich chemotherapeutisch als besonders aktiv erweisen und dabei sehr gut verträglich sind.
Gegenstand der vorliegenden Erfindung bildet ein
Verfahren zur Herstellung solcher Sulfanilamido- polyalkylpyrimidine, welches dadurch gekenn- zeichnet ist, dass man Benzolsulfonsäuren, deren
Kern durch eine Amino-oder eine in eine solche überführbare Gruppe in p-Stellung substituiert ist, bzw. reaktionsfähige Säureabkömmlinge, in an sich bekannter Weise in Polyalkylpyrimidin- benzolsulfonamidverbindungen überführt und gegebenenfalls in Aminogruppen überführbare
Gruppen in solche verwandelt. Dabei kann nach den bisher auf diesem Gebiet bekanntgewordenen Methoden gearbeitet werden.
Als Ausgangsstoffe kann man z. B. verwenden : p-Acylamino-benzolsulfbnsäuren, p-Nitrobenzol- sulfosäuren, p-Halogenbenzolsulfosäuren, bzw. deren Säurehalogenide,-amide,-anhydride,-ester oder andere reaktionsfähige Säurederivate. Zur Umsetzung eignen sich beliebige Amino-bzw. Halogen-polyalkylpyrimidine, wie z. B. 2-Amino- 4, 6-dimethyl-pyrimidin, 6-Amino-2, 4-dimethylpyrimidin, 6-Amino-5-halogen-2, 4-dimethylpyrimidin, 2-Amino-4, 5-dimethylpyrimidin, 6- Amino-2, 4-diäthylpyrimidin, 6-Amino-5-methyl- 2, 4-diäthyl-pyrimidin, 6-Amino-2, 4,5-triäthylpyrimidin, 6-Chlor-2, 4- bzw. 4, 5-dimethylpyrimidin, 2-Chlor-4, 6-dimethylpyrimidin u. dgl.
Geht man von Nitrobenzolsulfohalogeniden aus, so muss nach der Umsetzung die Nitrogruppe in eine Aminogruppe übergeführt werden. Ver- wendet man als Ausgangsstoffe Acylaminobenzol- sulfosäurederivate, so wird durch Verseifung die entsprechende Verbindung mit freier Aminogruppe dargestellt. Für die Umsetzung selbst kann man in Gegenwart oder Abwesenheit von Lösungsmitteln und/oder Kondensationsmitteln arbeiten. Als Lösungsmittel kann man Wasser oder auch organische Lösungsmittel, wie z. B.
Aceton, Essigester, Ketone, Dioxan, Benzol, Pyridin u. dgl. verwenden. Als Kondensationsmittel kommen insbesondere säurebindende Mittel, wie z. B. Pyridin, Dialkylaniline und andere tertiäre Basen, ferner Natriumacetat, sowie anorganische Basen wie z. B. Ätzalkalien, Soda u. dgl. in Frage. Die erhaltenen Sulfanilamide können in an sich bekannter Weise alkyliert werden.

Die neuen Verbindungen sollen therapeutische Verwendung finden oder als Zwischenprodukte zur Herstellung therapeutisch verwendbarer Verbindungen dienen.
Beispiel 1 : 123 Teile 6-Amino-2, 4-dimethylpyrimidin (Kyanmethin) werden in 200 Teilen trockenem Pyridin gelöst, zum Sieden erhitzt

und mit 225 Teilen p-Nitrobenzolsulfochlorid in Portionen versetzt. Nach dem Zufügen wird noch eine Stunde am Rückfluss erhitzt. Nach dem Erkalten wird die Lösung in Wasser gegossen.
Die bald erstarrende Fällung wird abgenutscht, in verdünnter Natronlauge gelöst, filtriert, mit Salzsäure gefällt, abgenutscht und aus verdünntem Alkohol umkristallisiert. Man erhält so das 6- [p-Nitrobenzolsulfamido]-2, 4-dimethyl- pyrimidin vom F. P. 188-189 o. Analyse : Ber.
N 18,18, Gef. N. 18, 03%.
20 Teile feines Eisenpulver werden mit 10 Teilen 2n-Salzsäure angeätzt und mit 400 Teilen 50% igem Alkohol versetzt. Zu dieser Reduktionsmischung fügt man 31 Teile 6-[p-Nitrobenzolsulfamido]- 2, 4-dimethylpyrimidin und kocht 4 Stunden am Rückfluss, destilliert den Alkohol ab, gibt 100 Teile 2n-Natronlauge zu, erwärmt kurz und filtriert.
Aus dem Filtrat wird mit Salzsäure das 6-[p-

Beispiel 2 : 123 Teile 2-Amino-4, 6-dimethylpyrimidin werden in 300 Teilen trockenem Pyridin gelöst, zum Sieden erhitzt und mit 225 Teilen p-Nitrobenzolsulfochlorid in Portionen versetzt. Nun wird noch eine Stunde am Rückfluss erwärmt und nach dem Erkalten in Wasser gegossen. Die Fällung erstarrt beim Ansäuern mit Salzsäure und wird abgenutscht. Zur Reinigung löst man in 2n-Sodalösung filtriert, fällt mit Säure und kristallisiert aus verdünntem Alkohol um.
Man erhält so das 2-[p-Nitrobenzolsulfamido]- 4, 6-dimethylpyrimidin vom F. P. 208 bis 210 .
