Verfahren zur Herstellung von Sulfonylharnstoffen Es wurde gefunden, dass sich Verbindungen der Formel I - R-502 NH-CO-NH-R' .
I worin R einen aliphatischen oder cycloaliphatischen Kohlenwasserstoffrest mit<B>3-8-</B> Kohlenstoffatomen und R' einen gesättigten oder ungesättigten, offen kettigen oder ringförmigen, Sauerstoff oder Schwefel enthaltenden aliphatischen Kohlenwasserstoffrest, vorzugsweise mit bis zu 8 Kohlenstoffatomen, bedeuten, und die Salze didser Verbindungen,
bei geringer Toxizität durch eine starke blutzucker- senkende Wirksamkeit auszeichnen.
Das erfindungsgemässe Verfahren zur Herstel lung solcher Sulfonylharnstoffe ist dadurch gekenn zeichnet, dass man Verbindungen der Formeln R-502 A und R' B, worin A und B zwei durch Anlagerung oder Kon densation die Harnstoffgrappe bildende Reste be deuten, miteinander umsetzt.
Man kann beispielsweise Alkyl-, Cycloalkyl- oder Cycloalkylalkylsulfonylisocyanate mit Verbindungen der Formel R' NH2 umsetzen. Man kann aber auch in umgekehrter Reaktion die gewünschten Harnstoffe dadurch gewinnen, dass man Verbindun gen der Formel R' N=C=O mit z. B. Alkyl-, Cy- cloalkyl- oder Cycloalkylalkyfsulfonsäureamiden zur Reaktion bringt.
Dabei werden zur Umsetzung mit den Isocyanaten die entsprechenden aliphatischen Sulfonsäureamide zweckmässig in Form ihrer Salze, insbesondere der Natrium- bzw. Kaliumsalze, ver wendet. Eine weitere Möglichkeit, die neuen Ver bindungen herzustellen, besteht darin, dass man z. B.
Alkyl-, Cycloalkyl- oder Cycloalkylalkylsulfonyl- urethane mit Verbindungen der Formel R' NH2 zur Umsetzung bringt. In vielen Fällen führt das einfache Erhitzen der Reaktionsteilnehmer auf Temperaturen oberhalb 100 C zu besonders guten Ausbeuten. Weiter kann man Sulfonamide der For mel R-502 NH2 mit einfach aliphatisch bzw.
cycloaliphatisch substituierten Harnstoffen umsetzen, wobei es von besonderem Vorteil ist, die Sulfonamide in Form ihrer Alkalisalze .und die Harnstoffe in Form entsprechender acylierter oder nitrierter Ver bindungen einzusetzen und die Komponenten in Abwesenheit von Lösungsmitteln auf höhere Tempe raturen, vorzugsweise 130-160 C, zu erhitzen.
Die zur Herstellung der Verfahrensprodukte geeigneten Methoden können in ihren Reaktions bedingungen weitgehend variiert und den jeweiligen Verhältnissen angepasst werden. Beispielsweise kön nen, die Umsetzungen in vielen Fällen durch ein faches Erhitzen der Komponenten, aber auch unter Verwendung von Lösungsmitteln, wie Aceton, To luol, Xylol und Chlorbenzol, bei Zimmertempera tur oder bei erhöhter Temperatur durchgeführt werden.
Um die Verfahrensprodukte in möglichst reiner Form zu erhalten, nimmt man zweckmässig eine möglichst vollständige Abtrennung von den z. B. als Ausgangsstoffe eingesetzten oder im Ver lauf der Reaktion entstehenden Sulfonsäureamiden vor, die vorteilhaft dadurch erreicht werden kann, dass man die Verfahrensprodukte mit verdünntem Ammoniak behandelt, worin diese Sulfonylharnstoffe verhältnismässig leicht löslich sind,
und sie aus den Lösungen durch Ansäuern mit organischen oder anorganischen Säuren wieder ausfällt.
Als Ausgangsstoffe kommen für das Verfahren gemäss der Erfindung z. B. folgende Sulfonylverbin- dung in Betracht: Alkyl-, Cycloallcyl- oder Cyclo- alkylalkylsulfamide, die als Alkylreste einen Propyl-, Butyl (1)-, Butyl-(2)-, 2-Methyl-propyl-(1)-, Pentyl-(1)-, Pentyl-(2)-, .
Pentyl-(3)-, 3-Methyl-butyl (1)-, 2 Methyl-butyl-(1)-, Hexyl-(1)-, Hexyl-(2)-, 4-Methyl-pentyT-(2)-, 2 Äthyl-butyl-(1)-, Heptyl-(1)-, Heptyl-(2)-, Heptyl-(4)-, 2;
4 Dimethyl-pentyl-(1)-, 2,4-Dimethyl-pentyl-(3)-, Octyl-(1)- und _ Octyl-(2)-rest, als Cycloalkylrdste einen 'Cyclohexyl- und Cyclo- heptylrest und als Cycloalkylalkylreste einen; Cyclo- hexyhnethyl- und Cyclohexyläthylrest tragen können.
Weiterhin können herangezogen werden: Entspre chende Alkyl-,- Cycloalkyl- oder Cycloallcylalkyl- sulfonylcarbaminsäuren, die mit einem niedrigmole- kularen Alkanol, beispielsweise Methanol, Äthanol, oder einem Phenol verestert sind;
entsprechende Alkyl-, Cycloalkyl- oder Cycloalkylalkylsulfonyliso- cyanate, Sulfonylcarbaminsäurehalogenide und Sulfo- nylharnstoffe.
Anstelle der erwähnten Sulfonylisocyanate kön nen auch entsprechende Verbindungen verwendet werden, die im Verlauf der Reaktion, wie Isocyanate, reagieren.
Für die Umsetzung mit den vorstehend genann ten Sulfonylisocyanaten, Sulfonylurethanen oder Sulfonylharn'stoffen können z. B. folgende primäre Amine verwendet werden:
Methoxypropylamin; Äthoxypropylamin, Propoxypropylamin, Propoxyäthylamin, Äthoxyäthylamin, Äthoxyäthylenoxyäthylamin, Methoxyäthylenoxyäthylamin, a-Tetrahdrofurfurylmethylamin, 6-Propyl-4-aminodioxan-(1,3), ss,ss-Diäthoxy-äthylamin, Methylmercaptopropylamin und Athylmercaptopropylamin. Anstelle dieser Amine können zur Umsetzung mit oben erwähnten Sulfonylverbindungen,
die aus diesen Aminen ableitbaren Isocyanate, Urethane und Harn stoffe verwendet werden. Anstelle solcher Isocyanate können auch entsprechende Verbindungen verwen det werden, die im Verlauf der Reaktion wie Iso- cyanate reagieren. Die Verfahrenserzeugnisse stellen wertvolle Heil mittel dar, die sich insbesondere durch eine starke blutzuckersenkende Wirksamkeit und sehr geringe Toxizität auszeichnen.
Die letztgenannte Eigenschaft lässt sie zur Verwendung als perorale Antidiabetika als besonders geeignet erscheinen, da derartige zur Behandlung der Zuckerharnruhr 'geeignete Verbin dungen unter Umständen über lange Jahre gegeben werden müssen. - Die Verbindungen können als solche oder in Form ihrer Alkalisalze bzw. in Gegen wart von Stoffen, welche zu einer Salzbindung führen, Verwendung finden.
Zur Salzbildung können beispielsweise herangezogen werden: Alkali- oder Erdalkalihydroxyde, Alkalicarbonate- oder -bicarbo- nate, ferner physiologisch verträgliche organische Basen.
Im Tierversuch lässt sich die Wirkung auf den Blutzuckerspiegel beispielsweise von Kaninchen nachweisen. Verabreicht man normal gefütterten Kaninchen Verbindungen der Formel I in einer ein- maligen Dosis von<B>*</B> durchschnittlich 400 mglkg in beispielsweise bicarbonat-alkalischer Lösung, so sieht man eine rasch einsetzende Senkung des Bl'utzucker- \spiegels, die innerhalb von 3-4 Stunden ein Maxi mum von etwa 309/o des Ausgangswertes erreicht.
\Beispielsweise erniedrigt die angegebene Dosis von N- [3 -Methyl-butan-(1)4;sulfonyl] N'-[3' methoxy- propyl-(1')]-harnstoff den Blutzucker nach zwei Stunden um etwa 309/o; die Wirkung hält etwa sechs Stunden an.
Im Falle der Verwendung von N-[3-Me- thyl - butan - (1) - sulfonyl] N'-[3'-äthoxy-propyl-(1')]- 'harnstoff erhält man nach zwei Stunden eine Blut zuckersenkung um 209/o, die nach sechs Stunden noch 16% beträgt. Im Falle der Verwendung von N-Cyclohexansulfonyl-N^-[3' methoxy-propyl-(1')
]- harnstoff beträgt die Senkung des Blutzuckers nach einer Stunde etwa 309/o; diese Senkung hält über sechs Stunden hinaus an. Im Falle der Verwendung von N - Cyclohexylmethansulfonyl - N'- [3'-methoxy- propyl-(1')]-hamstoff beträgt die Blutzuckersenkung nach zwei Stunden 409/o und nach sechs Stunden noch 3 5 0/0. ' Die Blutzuckerwerte können durch stündliche Analysen nach Hagedornliensen ermittelt werden.
Die Blutzuckersenkung wird durch Vergleich mit den Blut zuckerwerten gleichartig gehaltener, nicht behandelter Kontrolltiere ermittelt.
Es ist bereit'ls bekannt, dass der N-(4-Amino- benzolsulfonyl)-N'-n-butyl-hamstoff blutzuckersen- kende Eigenschaften: aufweist. Weiterhin ist bekannt, dass diese Verbindung auf Grund ihres Sulfanilyl- charakters auch chemotherapeutisch wirksam ist.
Da für die Verwendung als orales Antidiabetikum die Dauergabe über einen langen Zeitraum hinaus erfor derlich ist, ist. es jedoch wünschenswert, dass die applizierte Verbindung keinen Sulfanilylcharakter hat und somit von andersartigen Wirkungen möglichst frei ist, um evtl. Schädigungen, beispielsweise dter Darmflora, sowie Allergien und die Resistenzbildung pathogener Keime gegenüber Sulfonanilylamiden aus zuschliessen.
<I>Beispiel 1</I> N-[3-Methyl-butan-(1)-sulfonyl] N'-[3'-methoxy- propyl-(1')]-harnstoff 31,5g N-[3 Methyl-butan-(1)-sulfonyl]-carbamin- säure-methylester (hergestellt aus 3-Methyl-butan- (1)-sulfonsäureamid und Chlorkohlensäure-methyl- ester in acetonischer Lösung in Gegenwart von Ka- liumearbonat);
45g Xylol und 13,4 g 3-Methoxy-n- propylamin werden unter Rückfluss sechs Stunden zum Sieden erhitzt. Nach dem Erkalten wird das Ge misch zweimal mit je 150 cms verdünntem Ammo niak (1 Volumen konzentriertes Ammoniak und 20 Volumen Wasser) ausgeschüttelt. Die vereinigten Auszüge werden mit Kohle behandelt und filtriert; das Filtrat wird mit Salzsäure angesäuert.
Man erhält in guter - Ausbeute eine Fällung von N-[3 Methyl- butarx-(1) sulfonyf]-Nr4[3 methoxy-propyl-{1')]-ham- stoff. Die Substanz wird getrocknet und aus Essig ester umkristallisiert. Der Schmelzpunkt beträgt 103 bis 105 C.
In analoger Weise erhält man aus 31,5g N-[3- Methyl-butan-(1)-sulfonyl]-carbaminsäure-methylester und 15,5 g 3-Äthoxy-n-propylamin den N-[3 Methyl- butan-(1)-sulfonyl]-N'-[3'-äthoxypropyl-(1')]-harnstoff vom Schmelzpünkt 74-76 C.
Beispiel <I>2</I> N-Cyclohexansulfonyl-N'-(2'-methoxy-äthyl)- harnstoff Zu einer Lösung von 38g Cyclohexansulfonyliso- cyanat (dargestellt aus Cyclohexansulfonsäureamid und Pho'sgen in 200 ml absolutem Benzol)
werden unter Rühren bei Zimmertemperatur 18 g 2-Methoxy- äthylamin zugetropft. Man kocht das Reaktions gemisch zwei Stunden am Rückfluss und destilliert dann das Lösungsmittel unter vermindertem Druck vollständig ab. Der Rückstand wird unter Zusatz von Ammoniak in 300 cm3 Wasser gelöst, die Lö sung mit Kohle geklärt und dann mit Salzsäure vorsichtig angesäuert.
Der in guter Ausbeute aus fallende N-Cyclohexansulfonyl-N'-(2'-methoxy-äthyl)- harnstoff wird abgesaugt, gut mit Wasser gewaschen und nach dem Trocknen aus einem Gemisch von Diisopropyläther und gssigsäure-methylester im Verhältnis 3 : 1 umkristallisiert.
Schmelzpunkt 108-109 C.
In analoger Weise erhält man aus 25g Cyclo- hexansulfonylisocyanat und 14 g 3-Methoxy-propyl- amin-(1) den N-Cyclohexansulfonyl-N'-[3'-methoxy- propyl-(1')]-harnstoff. Der Schmelzpunkt beträgt nach dem Umkristallisieren aus Acetonitril 129 bis <B>1300</B> C.
Aus 38 g Cyclohexansulfonylisocyanat und 24 g Tetrahydrofurfurylamin erhält man in analoger Weise den N - Cyclohexansulfonyl - N' - tetrahydrofurfuryl- harnstoff. Der Schmelzpunkt liegt nach dem Um- kristallisieren aus einem Gemisch von Diisopropyl- äther und Essigsäure-methylester im Verhältnis 4 : 1 bei 122-12311 C.
<I>Beispiel 3</I> N-Cyclohexylmethansulfonyl-N'-[3'-methoxy- propyl-(1')]-harnstoff 12 g Cyclohexylmethansulfonylcarbaminsäure- methylester und 4,5 g 3-Methoxy-n-propylamin wer den unter schwach vermindertem Druck eine Stunde im Ölbad auf 130 C erhitzt. Nach dem Erkalten wird das Reaktionsgut in etwa 1% igem Ammoniak aufgenommen.
Die erhaltene Lösung wird mit Tier kohle geklärt und nach Filtration mit Salzsäure congosauer gestellt. Man erhält eine Fällung von N - Cyclohexyhnethansulfonyl-N'[3'-methoxy - propyl- (1')]-harnstoff, die man absaugt und trocknet. Der erhaltene Harnstoff schmilzt nach dem Umkristalli- sieren aus Essigester bei 116-117 C.
<I>Beispiel 4</I> N-Cyclohexansulfonyl-N'-(2'-äthylthio-äthyl)- harnstoff 17,7 g N-Cyclohexansulfonylcarbaminsäure-me- thylester und 8,4 g 2-Athylthio-äthylamin-(1) werden miteinander vermischt und 45 Minuten auf 120 bis 140 C erhitzt. Den erkalteten Rückstand, der eine klare Schmelze darstellt, kristallisiert man aus einem Liter 50 /oigen Methanol um.
Man erhält in sehr guter Ausbeute den M-Cyclohexansulfonyl-N'-(2'- äthylthio-äthyl) harnstoff vom Schmelzpunkt 96 bis 98 C.
<I>Beispiel 5</I> N-Cyclohexansulfonyl-N'-[3'-methoxy-propyl-(1')]- harnstoff In 38g Cyclohexarisulfonylisocyanat vom Kpo,5 88-94 C (hergestellt durch Umsetzung von Cyclo- hexansulfonamid mit Phosgen in der Hitze) wird die berechnete Menge trockener Chlorwasserstoff ein geleitet und das so erhältliche N-Cyclohexansulfonyl- carbaminsäurechlorid unter Kühlung und unter gutem Rühren in 24 g 3-Methoxy-propylamin-(1) eingetragen.
Man rührt noch zwei Stunden bei 20 bis 30 C nach, verdünnt die Reaktionsmischung mit Essigester und schüttelt anschliessend mehrmals mit verdünntem Ammoniak durch.
Die vereinigten ammoniakalischenLösungen werdenmitKohle geklärt und mit verdünnter Salzsäure angesäuert. Der ausge- fallene Niederschlag wird) abgesaugt, mit Wasser ge- waschen und aus Acetonitril umkristallisiert. Man erhält so den N-Cyclohexansulfonyl-N'-[3'-methoxy- propyl-(1')
]-harnstoff vom- Schmelzpunkt 129 bis 130 C.
<I>Beispiel 6</I> N-Cyclohexansulfonyl-N'-[3' methoxy-propyl-(1')]- harnstoff 9,3g Cyclohexansulfonamid natrium und 8,7 g N - (3 - Methoxy - propyl) - N' - acetyl - harnstoff vom Schmelzpunkt 92-94 C (hergestellt aus N-[3- Methoxy-propyl-(1)]-harnstoff und Essigsäureanhy- drid durch Erhitzen auf dem Dampfbad)
werden gut vermischt und im Ölbad l1/2 Stunden auf 140 bis 150 C erhitzt. Nach dem Erkalten der Schmelze behandelt man das Reaktionsgemisch mit lo/oigem wässerigem Ammoniak, filtriert von geringen Mengen Ungelöstem ab und erhält durch Ansäuern des Fil trats mit verdünnter Salzsäure in guter Ausbeute eine kristalline Fällung von N-Cyclohexansulfonyl-N'-[3'- methoxy-propyl-(1@]-harnstoff,
der nach dem Um kristallisieren aus Äthanol bei 129-130 C schmilzt. <I>' Beispiel ?</I> N-Cyclohexansulfonyl-N'-[3' n-butoxy-propyl-(1')]- harnstoff 90 g Cyclohexansulfonamid-natrium werden in 1 1 Aceton suspendiert und unter Rühren bei einer Temperatur von etwa 55 C tropfenweise mit 80 g 3-n-Butoxy propyl-(1) isocyanat vom Kp" 76 bis 78 C versetzt.
Man rührt 12 Stunden bei 55 C nach und destilliert anschliessend das Aceton im Vakuum ab. Den verbleibenden Rückstand löst man in Wasser auf, klärt die Lösung mit Kohle und säuert mit Salzsäure an. Der ausfallende Sulfonyl- harnstoff wird in Essigester aufgenommen und die sem durch verdünnten Ammoniak (1 :20) wieder entzogen.
Die ammoniakalische Lösung erhitzt man kurze Zeit im -Vakuum, um den gelösten Essigester zu entfernen, kühlt sie ab und trägt sie langsam unter Kühlung und unter gutem Rühren in verdünnte Salzsäure ein.
Der kristallin ausfallende Niederschlag, der den in der Überschrift genannten Sulfonylharn- stoff darstellt, wird abgesaugt, mit Wasser gewaschen und getrocknet. Der Schmelzpunkt beträgt 63-65 C.
<I>Beispiel 8</I> N-Cyclohexansulfonyl-N'-[3' methoxy propyl-(1')]- harnstoff ' 9,3g Cyclohexansulfonamidnatrium werden mit 14 g gemahlenem Kaliumcarbonat gut verrieben und das Gemisch mit 14,7g N-[3-Methoxy propyl-.(1)]- methylurethan vom Kp15114 C [hergestellt aus 3-Methoxy-propylamin-(1)
und Chlorameisensäure- methylester] drei Stunden auf 130 C erhitzt. Nach dem Erkalten versetzt man den Ansatz mit Wasser, bis eine homogene Lösung entstanden ist, säuert mit Salzsäure an und kühlt zur besseren Kristallisation mit Eis.
Der Niederschlag wird abgesaugt, mit etwa 5 O/oiger Natriumcarbonatlösung behandelt, Ungelöstes abfiltriert, das Filtrat wiederum mit SaIz- säure versetzt und gekühlt. Der abgeschiedene N-Cy- clohexansulfonyl -N'- [3'-methoxy-propyl-(1')
]-harn- stoff schmilzt nach dem Umkristallisieren aus Äthanol/Wasser bei 128-130 C.
<I>Beispiel 9</I> N-Cyclohexansulfonyl-N'-[3'-methoxy-propyl-(1')]- harnstoff 20,6 g N-Cyclohexansulfonyl-harnstoff werden mit 9 g 3-Methoxy-propylamin-(1) versetzt. Man er hitzt das entstandene Salz für l1/2 Stunden auf 130 bis 140 C. Nach dem Erkalten wird die Schmelze mit etwa lp/oigem Ammoniak behandelt, wobei Lö sung eintritt. Man klärt mit Kohle und säuert das Filtrat mit Salzsäure an.
Der erhaltene Niederschlag wird abfiltriert, mit Wasser gewaschen und aus Äthanol umkristallisiert. Man erhält so den N-Cyclo- hexansulfony2l-N'@[3' methoxy-propyl-(1')]-harnstoff vom Schmelzpunkt 129-130 C.

The effect on blood sugar levels in rabbits, for example, can be demonstrated in animal experiments. If compounds of the formula I are administered to normally fed rabbits in a single dose of an average of 400 mg / kg in, for example, bicarbonate-alkaline solution, a rapid drop in the blood sugar level can be seen reached a maximum of about 309 / o of the initial value within 3-4 hours.

It is already known that N- (4-Amino-benzenesulfonyl) -N'-n-butyl-urea has blood-sugar-lowering properties. It is also known that this compound is also chemotherapeutically effective due to its sulfanilyl character.
Since long-term administration is required for use as an oral antidiabetic agent. However, it is desirable that the applied compound does not have a sulfanilyl character and is thus as free as possible from other types of effects in order to exclude possible damage, for example to the intestinal flora, as well as allergies and the formation of resistance of pathogenic germs to sulfonanilylamides.
Example 1 N- [3-methyl-butan-(1) -sulfonyl] N '- [3'-methoxypropyl- (1')] urea 31.5 g of N- [3 methyl -butane (1) sulfonyl] carbamic acid methyl ester (prepared from 3-methyl-butane (1) sulfonic acid amide and chlorocarbonic acid methyl ester in acetone solution in the presence of potassium carbonate);
45 g of xylene and 13.4 g of 3-methoxy-n-propylamine are heated to boiling under reflux for six hours. After cooling, the mixture is extracted twice with 150 cms of diluted ammonia each time (1 volume of concentrated ammonia and 20 volumes of water). The combined extracts are treated with charcoal and filtered; the filtrate is acidified with hydrochloric acid.
A precipitation of N- [3 methyl-butarx- (1) sulfonyf] -Nr4 [3 methoxy-propyl- {1 ')] - urea is obtained in good yield. The substance is dried and recrystallized from ethyl acetate. The melting point is 103 to 105 C.
In an analogous manner, from 31.5 g of N- [3-methyl-butane (1) -sulfonyl] -carbamic acid methyl ester and 15.5 g of 3-ethoxy-n-propylamine, N- [3 methyl-butane ( 1) -sulfonyl] -N '- [3'-ethoxypropyl- (1')] - urea from melting point 74-76 C.
Example <I> 2 </I> N-Cyclohexanesulfonyl-N '- (2'-methoxy-ethyl) - urea To a solution of 38g cyclohexanesulfonyl isocyanate (prepared from cyclohexanesulfonic acid amide and pho'sgene in 200 ml of absolute benzene)
18 g of 2-methoxyethylamine are added dropwise with stirring at room temperature. The reaction mixture is refluxed for two hours and the solvent is then completely distilled off under reduced pressure. The residue is dissolved in 300 cm3 of water with the addition of ammonia, the solution is clarified with charcoal and then carefully acidified with hydrochloric acid.
The N-cyclohexanesulfonyl-N '- (2'-methoxy-ethyl) - urea which precipitates in good yield is filtered off with suction, washed well with water and, after drying, recrystallized from a mixture of diisopropyl ether and methyl acetate in a ratio of 3: 1.
Melting point 108-109 C.
In an analogous manner, N-cyclohexanesulfonyl-N '- [3'-methoxypropyl- (1')] urea is obtained from 25 g of cyclohexanesulfonyl isocyanate and 14 g of 3-methoxypropylamine- (1). After recrystallization from acetonitrile, the melting point is 129 to 1300 C.
From 38 g of cyclohexanesulfonyl isocyanate and 24 g of tetrahydrofurfurylamine, N-cyclohexanesulfonyl-N'-tetrahydrofurfuryl urea is obtained in an analogous manner. After recrystallization from a mixture of diisopropyl ether and methyl acetate in a ratio of 4: 1, the melting point is 122-12311 C.
<I> Example 3 </I> N-Cyclohexylmethanesulfonyl-N '- [3'-methoxypropyl- (1')] - urea 12 g of methyl cyclohexylmethanesulfonylcarbamate and 4.5 g of 3-methoxy-n-propylamine are the heated to 130 ° C. under slightly reduced pressure in an oil bath for one hour. After cooling, the reaction mixture is taken up in about 1% ammonia.
The solution obtained is clarified with animal charcoal and, after filtration, made congo acidic with hydrochloric acid. <I> Example 4 </I> N-Cyclohexanesulfonyl-N '- (2'-ethylthio-ethyl) -urea 17.7 g of methyl N-cyclohexanesulfonylcarbamate and 8.4 g of 2-ethylthio-ethylamine- (1st ) are mixed together and heated to 120 to 140 C for 45 minutes. The cooled residue, which is a clear melt, is recrystallized from one liter of 50% methanol.
The M-Cyclohexanesulfonyl-N '- (2'-ethylthio-ethyl) urea with a melting point of 96 to 98 ° C. is obtained in very good yield.

<I> Example 6 </I> N-Cyclohexanesulfonyl-N '- [3' methoxy-propyl- (1 ')] - urea 9.3 g cyclohexanesulfonamide sodium and 8.7 g N - (3 - methoxy - propyl) - N '- acetyl urea with a melting point of 92-94 C (made from N- [3-methoxy-propyl- (1)] -urea and acetic anhydride by heating on the steam bath)
