Verfahren zur Herstellung von oral wirksamen Antidiabetika
Der menschliche Diabetes wird heute üblicherweise mit strengen Diätvorschriften und Insulin Injektionen behandelt. Es hat aber in den letzten Jahrzehnten nicht an Versuchen gefehlt, das Insulin durch ein per-oral wirksames Antidiabetikum zu ersetzen. Die bisherigen Präparate, z. B. die Bisguanidine mit einem höhermolekularen Alkylenrest, haben wegen ihrer Toxizität auf die Dauer nicht befriedigt.
Andere derartige Substanzen (Glucokinine) erwiesen sich als unzuverlässig in der Wirkung. Weitere Verbindungen sind zwar im Tierexperiment als wirksam befunden worden, sind aber wegen ihrer Nebenerscheinungen, z. B. ihrer Toxizität, gar nicht bis in den klinischen Versuch gelangt. Zu der letzten Gruppe gehören heterocyclische Derivate des Sulfanilamids, nämlich Sulfanilyl-thiodiazole mit einem Athyl-, Isopropyl-oder Butylrest.
Man wird also von einem Antidiabetikum geringe Toxizität, insbesondere an der Leber, den Nebennieren und dem Zentralnervensystem, und zuverlässige Wirkung verlangen müssen. Ausserdem ist es notwendig, dass die betreffende Substanz nicht stossartig, sondern kontinuierlich wirkt, damit bedrohliche Hypoglykämien vermieden werden.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung einer neuen Gruppe von Sulfonamiden, die diesen Ansprüchen weitgehend entsprechen. Während die Sulfanilylthiodiazole nur tierexperimentell eingesetzt werden konnten und unter den zahlreichen übrigen Sulfanilamiden keine blutzuckersenkenden Wirkungen bekanntgeworden sind, haben die neuen Sulfonamide eine zuverlässige und gleichmässige Wirkung auf den Kohlehydrathaushalt des Menschen ohne wesentliche toxische Nebenerscheinungen.
Es handelt sich bei den neuen Verbindungen um Sulfonylcarbamide der Formel

An sich ist die blutzuckersenkende Wirkung der N2-substituierten Sulfonylharnstoffe nicht auf die erfindungsgemäss hergestellten Verbindungen mit den oben definierten Resten beschränkt, wie aus eingehenden Untersuchungen der pharmakologischen Eigenschaften dieser Gruppe der Sulfonamide hervorgeht. Indessen scheinen Ausmass und Charakter der Nebenwirkungen weitgehend an die Art der N2-Substituenten gebunden zu sein. Während nämlich die höheren Alkylderivate verhältnismässig toxisch sind, bewirkt die Anwesenheit von Heteroatomen in den N2-Substituenten eine überraschend starke Herabsetzung der Toxizität ohne entsprechende Beein trächtigung der antidiabetischen Wirkung ; so ist die Dosis letalis z.
B. beim N1-Toluolsulfonyl-N2-me- thoxypropyl-harnstoff rund 5-bis 6mal höher als beim N-Toluolsulfonyl-N2-n-butyl-harnstoff. Die die Grundlage der vorliegenden Erfindung bildende Erkenntnis, dass durch die Anwesenheit der Heteroatome S, O oder bzw. und N eine starke Herabsetzung der Toxizität bewirkt wird, findet durch den Vergleich auch der übrigen Sulfonylharnstoffe ihre Bestätigung und scheint allgemeine Gültigkeit zu besitzen.
Die Erzeugnisse des vorliegenden Verfahrens besitzen keine den Sulfanilamiden vergleichbare bakteriostatische Wirkung, was für gewisse Fälle bei ihrer Anwendung als blutzuckersenkende Substanzen ein Vorteil sein kann. So findet z. B. eine Beeinflussung der Darmflora nicht statt, und ebenso bleibt eine durch Dauergebrauch eventuell zu befürchtende Gewöhnung pathogener Keime an Sulfanilamide ausser Betracht.
Das erfindungsgemässe Verfahren zur Herstellung dieser Verbindungen ist dadurch gekennzeichnet, dal3 man Benzolsulfonylverbindungen der Formel

mit Verbindungen der Formel B-R, wobei einer der beiden Reste A und B eine Aminogruppe bedeutet, während der andere eine Gruppe, welche mit der Aminogruppe durch Anlagerung oder Kondensation die Harnstoffbrücke ergibt, bedeutet, umsetzt. Man setzt also z. B. ein Sulfonamid (zweckmässigerweise in Form des Natrium-oder Kaliumsalzes) mit einem Alkoxyalkyl-isocyanat um oder man kondensiert ein Sulfonylisocyanat mit einem Alkoxyalkylamin. Anstelle der Isocyanate können hierbei jeweils Verbindungen eingesetzt werden, welche unter den Reaktionsbedingungen in einen Isocyansäureester überzugehen vermögen. Als derartige Iso cyanat-Bildner sind z.
B. geeignet Carbaminsäurehalogenide, Urethane bzw. Thiourethane, Harnstoffe sowie deren Acylderivate.
Beispiel 1 N- (p-Toluolsulfonyl)-N2-(3-methoxypropyl)- harnstoff aus Di- (p-Toluolsulfonyl)-harnstoff und
Methoxypropylamin a) 186 g Di- (p-Toluolsulfonyl)-harnstoff (hergestellt aus p-Toluolsulfonamidnatrium und Phosgen in wässriger Lösung ; Smp. 108-109 C) werden in 400 cm3 Wasser von 50 C suspendiert und mit 37 g Methoxypropylamin versetzt. Es entsteht eine klare Lösung, die nach zweitägigem Stehen bei Zimmertemperatur durchkristallisiert (wenn Impfkristalle vorhanden sind, kann die Kristallisation durch Impfen, Abkühlen unter Zimmertemperatur und Rühren stark beschleunigt werden). Die Kristalle werden abgesaugt und mit 75 cm3 eiskaltem Wasser ausgewaschen.
Ausbeute 150 g 3-Methoxypropylaminsalz des Di- (p-Toluolsulfonyl)-harnstoffs vom Smp. 135 bis 138 C.
Die fehlenden Mengen sind noch in der Mutterlauge enthalten und können entweder durch mehrmalige Benutzung der Mutterlauge für die Salzbildung oder durch Eindampfen gewonnen werden. Man kann auch den gelösten Di- (p-Toluolsulfonyl)-harn- stoff durch Ansäuern auf PH 4-5 gewinnen. b) 100 g Methoxypropylaminsalz des Di- (p-To luolsulfonyl)-harnstoffs werden in einem Weithalskolben im Ölbad von 110 C erhitzt. Nach 15minütigem Erhitzen tritt Sinterung des Salzes ein, das bei weiterem Erwärmen zu einer klaren Flüssigkeit schmilzt. Nach 70 Minuten kühlt man ab und verrührt die dann erstarrte Masse mit 400 cm3 2,5 /oiger Natriumbicarbonatlösung.
Das bei der thermischen Zersetzung als Nebenprodukt gebildete Toluolsulfonamid bleibt dabei ungelöst und wird abgesaugt. Ausbeute 98-100''/o d. Th. In der Bicarbonatlösung ist der gebildete N-(p-Toluolsulfonyl)-N2-(3-methoxy- propyl)-harnstoff enthalten. Man säuert die Lösung bis zur Kongoreaktion an, wobei das Harnstoff-Derivat auskristallisiert. Nach dem Absaugen und Auswaschen mit Wasser erhält man 58 g Reinprodukt vom Smp. 98-101 C ; das sind 9211/o d. Th.
In der gleichen Weise kann auch der N-Toluol- sulfonyl-N2- (3-äthoxypropyl)-harnstoff vom Smp. 114 bis 119 C erhalten werden, wenn man anstelle von Methoxypropylamin Athoxypropylamin verwendet.
Wenn man anstelle von Di- (p-Toluolsulfonyl)- harnstoff Di-Benzolsulfonylharnstoff mit Methoxypropylamin bzw. mit Athoxypropylamin zum Salz umsetzt und bei 100-110 C thermisch zersetzt, erhält man den N,-Benzolsulfonyl-N2-methoxypropyl- harnstoff vom Smp. 102-105 C und den N,-Ben- zolsulfonyl-N2-äthoxypropyl-harnstoff vom Smp. 74 bis 76 C.
Beispiel 2 N- (p-Toluolsulfonyl)-N2-(3-methoxypropyl)- harnstoff aus Toluolsulfonamid und Methoxypropyl isocyansäureester
24,2 g trockenes p-Toluolsulfonamid-natrium werden in 50 cm3 absolut wasserfreiem Dimethylformamid suspendiert bzw. gelöst und mit 18 g 3-Methoxypropyl-isocyansäureester versetzt. Man erwärmt 8 Stunden auf 45-55 C. Sollte der Isocyansäure- ester noch nicht völlig verbraucht sein, so kann man noch einige Zeit nacherwärmen. Hierauf verdünnt man mit 200 cm3 Wasser und säuert auf einen pll-Wert von 8,5 an. Nach einigem Stehen wird von eventuell ausgefallenem p-Toluolsulfonamid abgesaugt. Die Mutterlauge wird beim pg 8,5 noch mehrfach ausgeäthert und daraufhin stark kongosauer gemacht.
Der gebildete Ni-Toluolsulfonyl-N2- methoxypropylharnstoff fällt dabei aus. Man erhält 28,7 g vom Smp. 98-101 C = 80 /o d. Th.
Setzt man anstelle von p-Toluolsulfonamid-natrium trockenes p-Methoxybenzolsulfonamid-kalium mit Methoxypropylisocyanat um, so erhält man in guter Ausbeute den N-(p-Methoxybenzolsulfonyl)- N2- (3-methoxy-propyl)-harnstoff vom Schmelzpunkt 135-138 C.
Beispiel 3
Nl- (p-Toluolsulfonyl)-N2- (3-methoxypropyl) harnstoff aus Toluolsulfonyl-thiourethan und
Methoxypropylamin
22,7 g p-Toluolsulfonyl-äthylthiourethan (dargestellt aus p-Toluolsulfonamid-Natrium und Dithio kohlensäure-di-S-äthylester unter Abspaltung von Athylmerkaptan) werden in 100 cm3 Dichlorbenzol suspendiert und mit 9 g Methoxypropylamin versetzt.
Man erwärmt die Reaktionsmischung auf 100 C, bis die Athylmerkaptan-Abspaltung beendet ist, was nach etwa 6-8 Stunden der Fall ist. Man destilliert einen Teil des Dichlorbenzols zusammen mit dem gebildeten Athylmerkaptan ab und schüttelt den Rückstand in der Kälte mit Sodalösung aus. Danach säuert man bis zum pH 8,5 mit Salzsäure an und trennt gegebenenfalls von etwas ausgefallenem p-Toluolsulfonamid ab. Die ein-bis zweimal ausgeätherte wässrige Lösung wird nun weiterhin bis kongosauer angesäuert, wobei der Toluolsulfonyl-methoxypropylharnstoff kristallin ausfällt. Man erhält etwa 25 g = 87 O/o d. Th. vom Smp. 98-101"C.
In gleicher Weise kann durch Verwendung von Tetrahydrofurfuryl-a-methylamin anstelle von Me thoxypropylamin der Nl-Toluolsulfonyl-N2-(a-tetra- hydrofuryl-methyl)-harnstoff vom Smp. 133-138 C gewonnen werden.
Beispiel 4 N- (p-Toluolsulfonyl)-N2-(2-isopropoxyäthyl)- harnstoff a) 120 g Di- (p-Toluolsulfonyl)-harnstoff werden mit 1 Liter Wasser und 35 g 2-Isopropoxy-äthyl- amin verrührt ; es entsteht ein dicker Kristallbrei.
Durch Absaugen erhält man 145 g Di- (p-Toluol- sulfonyl)-harnstoff-2-isopropaxyäthylamin-Salz vom Schmelzpunkt 133-135 C. b) 145 g 2-Isopropoxyäthylamin-Salz des Di- (p Toluolsulfonyl)-harnstoffs werden 75 Minuten im 01- bad von 110 C erhitzt ; es entsteht eine klare Lösung, die beim Erkalten erstarrt. Diese Reaktionsmischung wird mit 350 cm3 Wasser und 35 cm3 konz. Ammoniaklösung verrührt. Das bei der thermischen Zersetzung als Nebenprodukt entstandene p Toluolsulfonamid (Schmelzpunkt 134-136 C) kristallisiert aus und wird abgesaugt.
Durch Ansäuern des Filtrates fällt der N1-(p-Toluolsulfonyl)-N2-(2-iso- propoxyäthyl)-harnstoff in kristalliner Form aus ; Schmelzpunkt 114-116 C. Ausbeute 83 g = 901/o d. Th.
In gleicher Weise erhält man unter Verwendung von 3-Methoxybutylamin bzw. 3-Methylmercapto- propylamin die folgenden Sulfonylharnstoffe : N1- (p-Toluolsulfonyl)-N2-(3-methoxybutyl)- harnstoff, Smp. 127-131 C, N1- (p-Toluolsulfonyl)-N2-3-(methylmercapto- propyl)-harnstoff, Smp. 125-128 C.
Beispiel 5
Ni (p-Toluolsulfonyl)-N2-(3-butoxypropyl)-harnstoff
Man löst 39,4 g p-Toluolsulfonyl-isocyanat in 400 cm3 Benzol und tropft unter Rühren 26,2 g 3-Butoxy-propylamin langsam zu, wobei ein Temperaturanstieg auf etwa 45 C eintritt. eintritt. Lösung wird nach beendetem Zutropfen 11/2 Stunden unter Rückfluss zum Sieden erhitzt. Anschliessend zieht man das Benzol im Vakuum ab und erhitzt den Rückstand mit 1 Liter verd. Ammoniak 1 Stunde auf dem Dampfbad, filtriert und säuert das Filtrat mit verd.
Salzsäure an. Der zunächst ölig anfallende Ni (p- Toluolsulfonyl)-N2- (3-butoxypropyl)-harnstoff kristallisiert nach einiger Zeit und wird abgesaugt. Nach Trocknen im Exsikkator und Umkristallisieren aus Essigester schmilzt das Produkt bei 69-71 C.
Beispiel 6 Nl- (p-Toluolsulfonyl)-N2-(pyridyl-2)-harnstoff
In eine Lösung von 28,2 g 2-Aminopyridin und 600 cm3 abs. Benzol tropft man unter Rühren bei Zimmertemperatur 75 g p-Toluolsulfonyl-isocyanat ein. Man erhitzt 2 Stunden unter Rückfluss zum Sieden, kühlt ab und saugt den ausgefallenen Kristallbrei von N-(p-Toluolsulfonyl)-N2-(pyridyl-2)-harn- stoff ab. Die Substanz wird aus Acetonitril und anschliessend aus Essigester umkristallisiert. Schmelzpunkt 146-147 C.
In analoger Weise erhält man unter Verwendung der entsprechend substituierten Aminopyridine die folgenden Sulfonylharnstoffe : Nl- (p-Toluolsulfonyl)-N2-(6-methyl-pyridyl-2)- harnstoff, Schmelzpunkt 190 C unter Zersetzung, N- (p-Toluolsulfonyl)-N2-(4-methyl-pyridyl-2)-harn- stoff, Schmelzpunkt 169-171o C.
Beispiel 7
Na- (p-Toluolsulfonyl)-N2-(2-methoxyäthyl)-harnstoff
97 g N-(p-Toluolsulfonyl)-carbamidsäure-äthyl- ester (Schmelzpunkt 84-85 C, hergestellt aus p Toluolsulfonamid und Chlorameisensäure-äthylester in Gegenwart von Aceton und trockenem Kaliumcarbonat) werden zusammen mit 30 g 2-Methoxy äthylamin und 130 g Glykolmonomethyläther 19
Stunden auf 110-120 C erhitzt. Dann wird das Methylglykol im Vakuum möglichst vollständig abdestilliert und der verbleibende Rückstand unter Rüh- ren in 1 Liter Wasser eingegossen. Der ausfallende Niederschlag wird abgesaugt, mit Wasser gewaschen und in verd. Ammoniak (1 : 20) aufgenommen.
Die entstehende klare Lösung wird durch eine Kohleschicht gesaugt und mit 2n Salzsäure langsam angesäuert. Dann saugt man den kristallinen Niederschlag ab, wäscht ihn mit Wasser und kristallisiert aus 20fl/oigem Athanol um. Der so in sehr reiner Form und in guter Ausbeute erhaltene N,- (p-Toluol- sulfonyl)-N2- (2-methoxyäthyl)-harnstoff schmilzt bei 120-121, 5 C.
Beispiel 8 N- (p-Toluolsulfonyl)-N2-(3-methoxypropyl)-harnstoff aus Toluolsulfonamid und Methoxypropyl-carbamin säurechlorid
Eine Lösung von 15 g 3-Methoxypropyl-carbaminsäurechlorid in Pyridin (erhalten aus 3-Methoxypropyl-isocyanat und Pyridinhydrochlorid) wird tropfenweise zu einer Mischung von 25 cm3 Pyridin und
17,1 g p-Toluolsulfonamid gegeben. Die Mischung wird etwa 12 Stunden auf 80 C erhitzt und danach im Vakuum vom überschüssigen Pyridin befreit. Der Rückstand wird mit 200 cm3 Wasser versetzt und schwach ammoniakalisch gemacht. Das nicht umgesetzte Toluolsulfonamid bleibt ungelöst und wird abgesaugt.
Aus dem Filtrat kann der Nl- (p-Toluolsulfo- nyl)-N2- (3-methoxypropyl)-hamstoff durch Ansäuern ausgefällt werden ; Schmelzpunkt 100-104 C.
Beispiel 9 N1,-Benzolsulfonyl-N2-isopropoxyäthyl-harnstoff a) 170 g Di-Benzolsulfonyl-harnstoff (hergestellt aus Benzolsulfonamidnatrium und Phosgen in wässri ger Lösung) werden in 200 cm3 Wasser von 50 C suspendiert und mit 52 g Isopropoxyäthylamin ver setzt. Aus der erhaltenen Lösung kristallisiert nach längerem Stehen bei Zimmertemperatur das Isopro poxyäthylaminsalz des Di-Benzolsulfonyl-harnstoffs vom Schmelzpunkt 112-115 C aus. b) 100 g dieses Salzes werden eine Stunde im bl- bad von 106 C erhitzt, wobei das Salz sintert und schliesslich zu einer klaren Flüssigkeit schmilzt.
Man lässt eine Stunde bei Zimmertemperatur stehen, wor auf die erstarrte Schmelze mit 300 cm3 gesättigter
Ammoniaklösung verrührt wird. Man trennt vom zu rückgebliebenen Benzolsulfonamid ab, verrührt das
Filtrat mit Aktivkohle und säuert nach nochmaligem
Filtrieren bis zur Kongo-Reaktion an. Der hierdurch ausgefällte NX-Benzolsulfonyl-N2-isopropoxyäthyl- harnstoff schmilzt bei 100-103 C ; Ausbeute nach
Reinigung etwa 80non d. Th.
Beispiel 10
Nt- (p-Isopropyl-benzolsulfonyl)-N2-(3-äthoxypropyl)- harnstoff
28 g N- (4-Isopropyl-benzolsulfonyl)-carbamid- säure-methylester (Schmelzpunkt 99-101 C, herge stellt aus Isopropylbenzolsulfamid und Chlorkohlensäuremethylester in acetonischer Lösung in Gegenwart von Kaliumcarbonat), 30 g Xylol und 11,2 g 3-Athoxypropylamin werden sechs Stunden zum Sieden erhitzt. Man lässt erkalten und zieht die Xylollösung zweimal mit je 150 cm3 verdünntem Ammoniak (1 Vol. konzentriertes Ammoniak und 20 Teile Wasser) aus. Die ammoniakalischen Auszüge werden vereinigt, mit Kohle geklärt und mit verdünnter Salzsäure angesäuert. Man erhält eine kristalline Fällung von N- (4-Isopropyl-benzolsulfonyl)-N2- (3-äthoxypro- pyl)-harnstoff.
Das in guter Ausbeute erhaltene Produkt schmilzt nach dem Umkristallisieren aus Essigester bei 96-98 C.
Beispiel 11
Ni (p-Isopropyl)-benzolsulfonyl)-N2-(3-methoxy- propyl)-harnstoff
25,7 g N- (4-Isopropyl-benzolsulfonyl)-carbamid- säure-methylester werden in einem Rundkolben mit 8,9 g 3-Methoxypropylamin übergossen. Die Mischung wird auf dem Dampfbad kurz erwärmt, wobei man das Methoxypropylaminsalz des Sulfonylurethans in Gestalt einer klaren Schmelze erhält. Man erhitzt die Schmelze im Olbad unter Vakuum etwa eine Stunde auf 120-130 C und lässt erkalten. Man erhält in guter Ausbeute einen Kristallkuchen von Ni (4-Isopropyl-benzolsulfonyl)-N2- (3-methoxypropyl)harnstoff. Die Substanz schmilzt nach dem Umkri- stallisieren aus Essigester bei 104-106 C.
Beispiel 12
Nl- (4-Athyl-benzolsulfonyl)-N2-(3-methoxypropyl)- harnstoff
25 g N- (4-Athyl-benzolsulfonyl)-carbamidsäure- äthylester (hergestellt durch Umsetzung von 4-Athyl- benzolsulfamid mit Chlorameisensäureäthylester in Gegenwart von trockenem Kaliumcarbonat ; hellgelbes, klares, zähes Harz), 9 g 3-Methoxy-propylamin und 40 g 1,2-Dichlorbenzol werden acht Stunden auf 120 C erhitzt. Man kühlt ab, verdünnt mit Ather und schüttelt die so erhaltene Lösung mehrmals mit je etwa 25 cms 0, 5n-Natronlauge aus. Die vereinigten alkalischen Auszüge schüttelt man, um Spuren von
1,2-Dichlorbenzol zu entfernen, zweimal mit wenig Ather durch, behandelt zwecks Entfärbung mit Tierkohle und säuert mit 2n-Salzsäure langsam an.
Der weisse kristalline Niederschlag wird abgesaugt, mit Wasser gut gewaschen und aus 100 cms 50 /eigem Athanol umkristallisiert. Man erhält so den Ni (4- Athyl-benzolsulfonyl)-N2- (3-methoxypropyl)-hamstoff in guter Ausbeute in Form weisser Kristalle vom Schmelzpunkt 108-110 C.
Beispiel 13
N (3, 4-Dimethoxy-benzolsulfonyl)-N2-(3-methoxy- propyl)-harnstoff 18 g N- (3, 4-Dimethoxy-benzolsulfonyl)-carbamidsäure-methylester (Schmelzpunkt 113-115 C ; hergestellt aus 3,4-Dimethoxy-benzolsulfamid und Chlorkohlensäuremethylester in acetonischer Lösung in Gegenwart von Kaliumcarbonat) werden zusammen mit 30 g Xylol und 5,8 g 3-Methoxypropylamin sechs Stunden unter Rückfluss zum Sieden erhitzt. Man lässt erkalten und erhält einen Kristallbrei von Ni (3, 4-Di methoxy-benzolsulfonyl)-N2- (3-methoxypropyl)-harn- stoff, der mit verdünntem Ammoniak (1 Teil konzentriertes Ammoniak und 20 Teile Wasser) behandelt wird.

In analoger Weise erhält man aus 18 g N- (3, 4 Dimethoxy-benzolsulfonyl)-carbamidsäure-methylester und 6,7 g 3-Athoxypropylamin den Nl-(3, 4-Di methoxy-benzolsulfonyl)-N2- (3-äthoxypropyl)-harnstoff vom Schmelzpunkt 154-156 C.
Beispiel 14 Ni- (3-Methoxy-4-methyl-benzolsulfonyl)-N2- (3-methoxypropyl)-harnstoff
60 g N- (3-Methoxy-4-methyl-benzolsulfonyl) carbamid-säureäthylester (dargestellt aus 3-Methoxy4-methyl-benzolsulfamid und Chlorameisensäureäthylester in Aceton in Gegenwart von gemahlenem Kaliumcarbonat) und 24 g 3-Methoxy-propylamin werden in 150 cmS Glykolmonomethyläther acht Stunden am Rückfluss gekocht. Dann wird das Lösungsmittel unter vermindertem Druck abdestilliert und der Rück- stand unter Zusatz von Natronlauge in 500 cm3 Wasser gelöst. Man klärt die Lösung mit Kohle und säuert sie mit Salzsäure vorsichtig an.
Der zunächst etwas teigige, nach kurzem Digerieren in der Kälte kristallisierende Niederschlag wird abgesaugt, gut mit Wasser gewaschen und in 350 cm3 verdünntem Ammoniak gelöst. Die Lösung wird mit Kohle geklärt und mit Salzsäure angesäuert. Der in guter Ausbeute ausfallende N-(3-Methoxy-4-methyl-benzolsulfonyl)- N2- (3-methoxypropyl)-harnstoff schmilzt nach dem Umkristallisieren aus Acetonitril bei 128-129 C.
In analoger Weise erhält man aus 57 g N- (3- Methoxy-4-methyl-benzolsulfonyl)-carbamidsäure- äthylester und 22 g Tetrahydrofurfurylamin den Ni (3-Methoxy-4-methyl-benzolsulfonyl)-N2-tetrahydro- furfuryl-harnstoff vom Schmelzpunkt 133 C (aus Acetonitril).
Beispiel 15 N- (3, 4-Dimethyl-benzolsulfonyl)-N2-(3-methoxy- propyl)-harnstoff
24,3 g N- (3, 4-Dimethyl-benzolsulfonyl)-carbamid- säure-methylester vom Schmelzpunkt 90-92 C (hergestellt durch Umsetzung von 3,4-Dimethyl-benzolsulfonsäureamid mit Chlorameiseimauremethylester in Gegenwart von trockenem Kaliumcarbonat in Aceton), 9 g 3-Methoxy-propylamin und 40 g 1,2-Di chlorbenzol werden 11 Stunden auf 120 C erhitzt.
Entsprechend der im Beispiel 14 angegebenen Vorschrift erhält man nach Umkristallisieren aus 150 cm3 50 /oigem Athanol den Ni (3, 4-Dimethyl-benzolsul fonyl)-N2- (3-methoxypropyl)-harnstoff in guter Ausbeute vom Schmelzpunkt 116-117, 5 C.
Beispiel 16 Nl- (3, 4-Dimethyl-benzolsulfonyl)-N2-(3-äthoxy- propyl)-harnstoff
Durch Umsetzung von 24,3 g N- (3, 4-Dimethyl- benzolsulfonyl)-carbamidsäure-methylester mit 10,3 g 3-Athoxypropylamin in Gegenwart von 40 g 1,2-Dichlorbenzol erhält man analog der im Beispiel 17 angegebenen Arbeitsweise und nach dem Umkristalli- sieren aus 160 cm3 50 böigem Äthanol den Nl- (3, 4 Dimethyl-benzolsulfonyl)-N2- (3-äthoxy-propyl)-harn- stoff in sehr guter Ausbeute vom Schmelzpunkt 107 bis 108,5 C.
Process for the production of orally active antidiabetic agents
Today, human diabetes is usually treated with strict diet regulations and insulin injections. However, in the last few decades there has been no lack of attempts to replace insulin with a per-orally effective antidiabetic agent. The previous preparations, z. B. the bisguanidines with a higher molecular weight alkylene group have not been satisfactory in the long run because of their toxicity.
Other such substances (glucokinins) proved to be unreliable in their effect. Other compounds have been found to be effective in animal experiments, but because of their side effects, e.g. B. their toxicity, does not even get into the clinical trial. The last group includes heterocyclic derivatives of sulfanilamide, namely sulfanilyl-thiodiazoles with an ethyl, isopropyl or butyl radical.
An antidiabetic agent will therefore have to be expected to be of low toxicity, especially in the liver, adrenal glands and central nervous system, and to be reliable. In addition, it is necessary that the substance concerned does not act suddenly, but continuously, so that threatening hypoglycaemia is avoided.
The present invention relates to a process for the preparation of a new group of sulfonamides which largely correspond to these claims. While the sulfanilylthiodiazoles could only be used in animal experiments and the numerous other sulfanilamides have no known blood-sugar-lowering effects, the new sulfonamides have a reliable and even effect on the carbohydrate balance of humans without significant toxic side effects.
