Verfahren zur Herstellung von oral wirksamen Antidiabetika
Der menschliche Diabetes wird heute üblicher weise mit strengen Diätvorschriften und Insulin Injektionen behandelt. Es hat aber in den letzten Jahrzehnten nicht an Versuchen gefehlt, das Insulin durch ein per-oral wirksames Antidiabetikum zu ersetzen. Die bisherigen Präparate, z. B. die Bisguanidine mit einem höhermolekularen Alkylenrest, haben wegen ihrer Toxizität auf die Dauer nicht befriedigt. Andere derartige Substanzen (Glucokinine) erwiesen sich als unzuverlässig in der Wirkung.
Weitere Verbindungen sind zwar im Tierexperiment als wirksam befunden worden, sind aber wegen ihrer Nebenwirkungen, z. B. ihrer Toxizität, gar nicht bis in den klinischen Versuch gelant. Zu der letzten Gruppe gehören heterocyclische Derivate des Sulfanilamids, nämlich Sulfanilylthiodiazole mit einem Athyl-, Isopropyl-oder Butylrest.
Man wird also von einem Antidiabetikum geringe Toxizität, insbesondere an der Leber, den Nebennieren und dem Zentral-Nervensystem, und zuverlässige Wirkung verlangen müssen. Ausserdem ist es notwendig, dass die betreffende Substanz nicht stossartig, sondern kontinuierlich wirkt, damit bedrohliche Hypoglykamien vermieden werden.
Gegenstand des vorliegenden Patentes ist die Herstellung einer neuen Gruppe von Sulfonamiden, die diesen Ansprüchen weitgehend entsprechen. Während die Sulfanilylthiodiazole nur tierexperimentell eingesetzt werden konnten und unter den zahlreichen übrigen Sulfanilamiden keine blutzuckersenkenden Wirkungen bekanntgeworden sind, haben die neuen Sulfonamide eine zuverlässige und gleichmässige Wirkung auf den Kohlehydrathaushalt des Menschen ohne wesentliche toxische Nebenerscheinungen.

An sich ist die blutzuckersenkende Wirkung der N2-substituierten Sulfonylharnstoffe nicht auf die den Gegenstand des vorliegenden Patentes bildenden Verbindungen mit den oben definierten Resten beschränkt, wie aus eingehenden Untersuchungen der pharmakologischen Eigenschaften dieser Gruppe der Sulfonamide hervorgeht. Indessen scheinen Ausmass und Charakter der Nebenwirkungen weitgehend an die Art der N2-Substituenten gebunden zu sein.
Während nämlich die höheren Alkylderivate verhältnismässig toxisch sind, bewirkt die Anwesenheit eines Heteroatoms im N2-Substituenten eine überraschend starke Herabsetzung der Toxizität ohne entsprechende Beeinträchtigung der antidiabetischen Wirkung. Die Erkenntnis, dass durch die Anwesenheit des Heteroatoms O eine starke Herabsetzung der Toxizität bewirkt wird, findet durch den Vergleich auch der übrigen Sulfonylharnstoffe ihre Be stätigung und scheint allgemeine Gültigkeit zu besitzen.
Die Verbindungen der vorliegenden Erfindung besitzen keine den Sulfanilamiden vergleichbare bakteriostatische Wirkung, was für gewisse Fälle bei ihrer Anwendung als blutzuckersenkende Substanzen ein Vorteil sein kann. So findet z. B. eine Beein- flussung der Darmflora nicht statt, und ebenso bleibt eine durch Dauergebrauch eventuell zu befürchtende Gewöhnung pathogener Keime an Sulfanilamide ausser Betracht.
Die Herstellung der Sulfonylcarbamide gemäss obiger Formel kann nach den für den Aufbau von substituierten Harnstoffen üblichen Methoden erfolgen. Allerdings versagt im allgemeinen die einfachste Darstellung, nämlich die Umsetzung eines substituierten Benzolsulfonylchlorids mit einem substituierten Harnstoff, indem dabei relativ kleine Ausbeuten neben andern Stoffen erhalten werden. Dies hat seine Begründung darin, dal3 der Sulfonylrest nicht nur an das Stickstoffatom tritt, sondern sich bevorzugt mit dem Sauerstoff der Isoform verbindet, um dann Sulfonsäure abzuspalten, unter Ausbildung eines substituierten Cyanamids.
Erfindungsgemäss werden die neuen Verbindungen hergestellt, indem man ein Sulfonamid, zweckmässigerweise in Form des Natrium-oder Kaliumsalzes, mit einem Isocyansäureester umsetzt :

Anstelle des Isocyanates kann man auch Verbindungen einsetzen, welche unter den Reaktionsbedingungen in einen Isocyansäureester übergehen.
Als derartige Isocyanat-Bildnerp sind z. B. geeignet substituierte Carbaminsäurehalogenide, Urethane, N2-substituierte Harnstoffe, sowie deren N,-Acyl- derivate ; unter den letztgenannten Verbindungen sind insbesondere solche N2-substituierten Harnstoffe geeignet, die am N-Atom einsn niederen aliphatischen Acylrest, wie Acetyl, Propionyl und Butyryl, bzw. einen Benzoylrest tragen.
Man kann auch in umgekehrter Weise ein Sulfonylisocyanat mit einem entsprechenden Amin reagieren lassen gemäss der Formel :

Auch bei dieser Reaktion können anstelle des Sulfonylisocyanates Verbindungen eingesetzt werden, welche unter den Reaktionsbedingungen ein solches Sulfonylisocyanat bilden, z. B. Sulfonylurthane bzw.
-thiourethane, Sulfonylcarbaminsäurehalogenide, Sulfonylharnstoffe bzw. deren N2-Acylderivate ; unter den letztgenannten Verbindungen sind insbesondere Di-Sulfonylharnstoffe sowie solche N-Sulfonyl- harnstoffe geeignet, die am N2-Atom einen niederen aliphatischen Acylrest, wie Acetyl, Propionyl und Butyryl bzw. einen Benzoylrest, tragen.
Beispiel 1 Nl- (p-Chlor-benzolsulfonyl)-N.,- (3-methoxy- propyl)-harnstoff a) 79 g Di- (p-Chlor-benzolsulfonyl)-harnstoff (hergestellt aus p-Chlor-benzolsulfonamid und Phosgen in alkalischer Lösung) werden mit 79 cm3 Wasser verrührt und mit 19 g 3-Methoxypropylamin versetzt. Nach einigen Stunden kristallisiert das 3-Methoxypropylaminsalz des Di- (p-Chlor-benzol- sulfonyl)-harnstoffes aus ; Ausbeute : 88 g, Smp. 132 bis 137 C (Zers.). b) 88 g 3-Methoxy-propylaminsalz des Di- (p- Chlor-benzolsulfonyl)-harnstoffes werden 82 Minuten im Ölbad auf 116 C erhitzt ; es entsteht eine klare Schmelze.
Diese wird nach dem Abkühlen mit 200 cm3 Wasser und 10 cm3 konz. Ammoniak verrührt. Das bei der thermischen Zersetzung als Nebenprodukt entstandene Chlorbenzolsulfonamid wird abgesaugt und das Filtrat auf 40 C erhitzt, auf pH 6,0 gebracht und mit 5 g Kohle ausgerührt.
Nach dem Absaugen wird das Filtrat angesäuert, es fallen 39 g Nl-(p-Chlor-benzolsulfonyl)-N < -(3-meth- oxy-propyl)-harnstoff vom Smp. 109-111 C aus.
Nach derselben Methode wird der Nl-(p-Chlor- benzolsulfonyl)-N2- (2-äthoxy-äthyl)-harnstoff vom Smp. 108-110 C hergestellt.
Beispiel 2
N1- (p-Chlor-benzolsulfonyl)-N2-(3-methoxy propyl)-harnstoff
50 g N- (p-Chlor-benzolsulfonyl)-carbamidsäure- äthylester (dargestellt aus p-Chlor-benzolsulfamid und Chlorameisensäureäthylester in Aceton in Gegenwart von gemahlenem Kaliumcarbonat ; Smp.
92-93ç} C) und 21 g 3-Methoxy-propylamin werden in 150 cm3 Glykolmonomethyläther 8 Stunden am Rückfluss gekocht. Dann wird das Lösungsmittel unter vermindertem Druck abdestilliert und der Rückstand unter Zusatz von Natronlauge in 500 cm3 Wasser gelöst. Man klärt die Lösung mit Kohle und säuert sie mit Salzsäure vorsichtig an. Der zunächst etwas teigige, nach kurzem Digerieren in der Kälte kristallisierende Niederschlag wird abgesaugt, gut mit Wasser gewaschen und in 350 cm3 verdünntem Ammoniak gelöst. Die Lösung wird mit Kohle ge klärt und mit Salzsäure angesäuert. Der in guter Ausbeute ausfallende Nl-(p-Chlor-benzolsulfonyl)- N. schmilzt nach dem Umkristallisieren aus Acetonitril bei 111-112 C.
Beispiel 3 Nl- (m-Chlor-p-methyl-benzolsulfonyl)-N2- (3-methoxy-propyl)-harnstoff
70 g N- (m-Chlor-p-methyl-benzolsulfonyl)carbamidsäureäthylester vom Schmelzpunkt 81 bis 839 C (dargestellt aus 2-Chlor-toluol-4-sulfamid und Chlorameisensäure-äthylester in Aceton in Gegenwart von gemahlenem Kaliumcarbonat) und 24 g 3-Methoxypropylamin werden in 150 cm3 Glykolmonomethyläther acht Stunden am Rückfluss gekocht.
Man destilliert das Lösungsmittel unter vermindertem Druck ab und löst den Rückstand in verdünnter Natronlauge. Die Lösung wird mit Kohle geklärt und mit Salzsäure vorsichtig wieder angesäuert. Der zunächst etwas teigig ausfallende Niederschlag kristallisiert beim Stehen in der Kälte. Er wird einige Zeit in 1 Liter verdünntem Ammoniak digeriert.
Man saugt ab, klärt die Lösung mit Kohle und säuert mit Salzsäure an. Der ausgefallene Nl- (m-Chlor-p- methyl-benzolsulfonyl)-N2- (3-methoxy-propyl)-harn- stoff wird abgesaugt, gut mit Wasser gewaschen und aus Acetonitril umkristallisiert. Smp. 106-107 C.
Beispiel 4 Nl (m-Chlor-p-methoxy-benzolsulfonyl)-N2- (3-methoxy-propyl)-harnstoff
19,6 g N- (m-Chlor-p-methoxy-benzolsulfonyl)- carbamidsäuremethylester vom Schmelzpunkt 142 bis 144 C (hergestellt durch Umsetzung von 4-Meth oxy-3-chlor-benzolsulfamid mit Chlorameisensäure- methylester in Gegenwart von Kaliumcarbonat und Aceton) und 6,3 g 3-Methoxy-propylamin werden miteinander vermischt und 35 Minuten in einem öl- bad auf etwa 130 C erhitzt. Unter Abspaltung von Methanol bildet sich eine klare Schmelze, die nach dem Abkühlen und Anreiben mit Essigsäureäthyl- ester kristallisiert.
Nach dem Absaugen und Umkristallisieren aus verdünntem Athanol erhält man den N1- (m-Chlor-p-methoxy-benzolsulfonyl)-N2-(3- methoxy-propyl)-harnstoff in guter Ausbeute vom Schmelzpunkt 120-122 C.
In analoger Weise wird unter Verwendung von 3-Athoxypropylamin der Nl- (m-Chlor-p-methoxy benzolsulfonyl)-N2- (3-äthoxypropyl)-harnstoff hergestellt ; Schmelzpunkt 135-137 C (nach Umkristallisieren aus wässrigem Athanol).
Beispiel 5 N,- (3-Chlor-p-toluol-sulfonyl)-N2- (3-methoxy- butyl)-harnstoff
83 g 3-Chlor-p-toluol-sulfonyl-äthylurethan, 60 cm3 Dimethylformamid und 30 g 3-Methoxybutylamin werden 3 Stunden auf 105-110 C erhitzt. Man giesst die abgekühlte Lösung in 1 Liter Wasser, wobei sich das Reaktionsgemisch zunächst ölig abscheidet, aber schon nach kurzer Zeit kristal- lin erstarrt. Man saugt ab, wäscht gut mit Wasser nach und trocknet im Vakuum. Ausbeute 700/o der Theorie ; F. P. 126-127 C (Propanol).
Beispiel 6 Nj- (4-Brom-benzolsulfonyl)-N2-(3-methoxy- propyl)-harnstoff
29,4 g N- (4-Brom-benzolsulfonyl)-carbaminsäuremethylester und 8,9 g 3-Methoxypropylamin werden vermischt und die Mischung 35 Minuten auf etwa 120 C erhitzt. Die Schmelze wird anschlie- ssend aus 50 /oigem Methanol umkristallisiert. Der in guter Ausbeute kristallisierende N-(4-Brom- benzolsulfonyl)-N2- (3-methoxy-propyl)-harnstoff schmilzt zwischen 110 und 112 C.
Beispiel 7
N- (2-Methyl-6-chlor-benzolsulfonyl)-N2 (3-methoxy-propyl)-harnstoff
10,3 g 2-Methyl-6-chlor-benzolsulfonamid und 8,7 g N,-Acetyl-N2- (3-methoxy-propyl)-harnstoff und 6,9 g fein gepulvertes Kaliumcarbonat werden gut gemischt. Man erhitzt das Gemisch 11/2 Stunden im Olbad auf 150 C und behandelt nach dem Erkalten das Reaktionsgut mit 1 /oigem Ammoniak.
Man filtriert vom Ungelösten ab und säuert das Filtrat mit verdünnter Salzsäure an. Man erhält eine kristalline Fällung von Nl- (2-Methyl-6-chlor- benzolsulfonyl)-N2- (3-methoxy-propyl)-harnstoff, die man absaugt und aus verdünntem Athanol umkristallisiert. Die Substanz schmilzt zwischen 128 und 130 C.
Dieselbe Substanz kann beispielsweise auch erhalten werden durch Zusammenschmelzen von 13,2 g 2-Methyl-6-chlor-benzolsulfonyl-carbamin- säuremethylester (erhalten aus 2-Methyl-6-chlorbenzolsulfamid und Chlorameisensäuremethylester durch Umsetzung in Aceton in Gegenwart von Kaliumcarbonat ; Schmelzpunkt 180-181 C) mit 4,5 g 3-Methoxy-propylamin auf 125 bis 130 C während einer Stunde. Die Aufarbeitung geschieht analog der vorstehend angegebenen Vorschrift durch Behandeln mit Ammoniak, Ansäuern mit Salzsäure und Umkristallisieren aus Äthanol.
Beispiel 8 Ni- (4-Chlor-benzolsulfonyl)-N2-(3-methoxy- propyl)-harnstoff
20,2 g 4-Chlor-benzolsulfonylisocyanat (hergestellt aus 4-Chlor-benzolsulfamid und Phosgen, Kpojl8 = 110-111 C) werden in 15 cm3 absolutem wasserfreiem Dioxan gelöst und bei Zimmertemperatur unter Rühren tropfenweise mit 9 g 3-Methoxypropylamin, gelöst in 15 cm3 Dioxan, versetzt. Nach beendigtem Eintropfen rührt man noch eine Stunde bei 80 C nach, engt im Vakuum ein und fällt den N1- (4-Chlor-benzolsulfonyl)-N2-(3-methoxy-propyl)- harnstoff durch Zugabe von Wasser aus.
Durch Lösen in etwa l /oigem Ammoniak, Behandeln mit Tierkohle und Fällen mit verdünnter Salzsäure erhält man in guter Ausbeute den Ni-(4-Chlor-benzol- sulfonyl)-N2- (3-methoxy-propyl)-harnstoff, der nach dem Umkristallisieren aus Acetonitril bei 111 bis 112 C schmilzt.
Beispiel 9 N1- (3, 4-Dichlor-benzolsulfonyl)-N-(3-methoxy- propyl)-harnstoff
3,4 g N,-(3, 4-Dichlor-benzolsulfonyl)-N2-butyryl- harnstoff vom Schmelzpunkt 183-184 C werden in Essigester mit 1 g 3-Methoxy-propylamin zum Salz umgesetzt. Man saugt ab und erhitzt das Produkt, das bei raschem Erhitzen bei etwa 173 C schmilzt, im Ölbad auf 150 C. Nachdem das Reaktionsgemisch nach etwa 5 bis 10 Minuten geschmolzen ist, lässt man die Temperatur auf 125 bis 130 C absinken und erhitzt insgesamt 60 Minuten. Man lässt erkalten, löst das Reaktionsprodukt in etwa l böigem Ammoniak und säuert die Lösung mit Essigsäure an. Die ausfallende Schmiere kristallisiert langsam durch.
Man saugt ab und kristallisiert den erhaltenen rohen Nl- (3, 4-Dichlor-benzolsulfonyl)-N2- (3-methoxy- propyl)-harnstoff aus verdünntem Athanol um. Der Schmelzpunkt beträgt 111 bis 112 C.
Beispiel 10 Nl- (p-Chlor-benzolsulfonyl)-N2-(3-methoxy- propyl)-harnstoff
24 g p-Chlorbenzolsulfonylharnstoff (hergestellt aus p-Chlor-benzolsulfonamid und Kaliumcyanat, Smp. 180-182 C) und 12,5 g 3-Methoxy-propylamin-Hydrochlorid werden gut vermischt und 60 Minuten im Ölbad von 150 C erhitzt. Dann löst man die Reaktionsmischung in verdünntem Ammoniak, filtriert vom Unlöslichen ab und säuert das Filtrat mit Essigsäure an. Man erhält auf diese Weise 18 g Nl-(p-Chlor-benzolsulfonyl)-N9-(3-meth- oxy-propyl)-harnstoff vom Smp. 109-llloC.
Process for the production of orally active antidiabetic agents
Today, human diabetes is usually treated with strict diet regulations and insulin injections. However, in the last few decades there has been no lack of attempts to replace insulin with a per-orally effective antidiabetic agent. The previous preparations, z. B. the bisguanidines with a higher molecular weight alkylene group have not been satisfactory in the long run because of their toxicity. Other such substances (glucokinins) proved to be unreliable in their effect.
Other compounds have been found to be effective in animal experiments, but because of their side effects, e.g. B. their toxicity, did not even get into the clinical trial. The last group includes heterocyclic derivatives of sulfanilamide, namely sulfanilylthiodiazoles with an ethyl, isopropyl or butyl radical.
An antidiabetic agent will therefore have to be expected to be of low toxicity, especially in the liver, adrenal glands and central nervous system, and to be reliable. In addition, it is necessary that the substance concerned does not act suddenly, but rather continuously, so that threatening hypoglycemia is avoided.
The subject of the present patent is the preparation of a new group of sulfonamides which largely correspond to these claims. While the sulfanilylthiodiazoles could only be used in animal experiments and the numerous other sulfanilamides have no known blood-sugar-lowering effects, the new sulfonamides have a reliable and even effect on the carbohydrate balance of humans without significant toxic side effects.

As such, the blood sugar-lowering effect of the N2-substituted sulfonylureas is not limited to the compounds with the residues defined above, which are the subject of the present patent, as can be seen from detailed studies of the pharmacological properties of this group of sulfonamides. However, the extent and nature of the side effects appear to be largely linked to the nature of the N2 substituents.
While the higher alkyl derivatives are relatively toxic, the presence of a hetero atom in the N2 substituent causes a surprisingly strong reduction in toxicity without a corresponding impairment of the antidiabetic effect. The knowledge that the presence of the hetero atom O causes a strong reduction in toxicity is confirmed by a comparison of the other sulfonylureas and appears to be generally valid.
The compounds of the present invention do not have any bacteriostatic activity comparable to the sulfanilamides, which can be an advantage in certain cases when they are used as blood sugar-lowering substances. So z. For example, there is no influence on the intestinal flora, and any accustoming of pathogenic germs to sulfanilamides, which may be feared through long-term use, remains out of consideration.
The sulfonyl carbamides according to the above formula can be prepared by the methods customary for the synthesis of substituted ureas. However, the simplest representation generally fails, namely the reaction of a substituted benzenesulfonyl chloride with a substituted urea, in that relatively small yields are obtained in addition to other substances. This is due to the fact that the sulfonyl radical not only comes to the nitrogen atom, but preferentially combines with the oxygen of the isoform, in order then to split off sulfonic acid, with the formation of a substituted cyanamide.

As such isocyanate formers are z. B. suitably substituted carbamic acid halides, urethanes, N2-substituted ureas and their N, acyl derivatives; Among the last-mentioned compounds, those N2-substituted ureas are particularly suitable which have a lower aliphatic acyl radical, such as acetyl, propionyl and butyryl, or a benzoyl radical on the N atom.
