Verfahren zur Herstellung von Sulfonylurethanen
Es ist bekannt, dass man p-Toluolsulfonylurethane erhält, wenn man auf p- Toluolsulfonyl-isocyanat Alkohole einwirken lässt. Beispielsweise wird in der deutschen Patentschrift Nr. 845042 die Herstellung von N-(p-Toluolsulfonyl) - isopropyl -urethan durch Umsetzung von p-Toluolsulfonyl-isocyanat mit Isopropylalkohol beschrieben. In der gleichen Patentschrift wird auch das N-(p-Toluolsulfonyl)-methylurethan erwähnt. Ferner wird die Umesterung von N-(p-Toluolsulfonyl)-isopropylurethan bzw. von Toluolsulfonyl)-methylurethan mit Dodecylalkohol zu N-(p-Toluolsulfonyl)-dodecylurethan beschrieben.
Weiterhin sind beispielsweise aus Naturwissen- schaften 43 (1956), Seite 93, substituierte Benzolsulfonylharnstoffe bekannt, die eine starke und langandauernde Senkung des Blutzuckerwertes bewirken.
Zahlreiche weitere Abkömmlinge des Sulfonylharnstoffs wurden als orale Antidiabetika vorgeschlagen.
Es wurde nun gefunden, dass die blutzuckersenkende Wirksamkeit nicht, wie man auf Grund der bisherigen Forschungsergebnisse annehmen konnte, an die Sulfonylharnstoff-Gruppierung gebunden ist, sondern dass überraschenderweise auch Sulfonylurethane eine derartige Wirksamkeit besitzen.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung blutzuckersenkender Sulfonylurethane, das dadurch gekennzeichnet ist, dass man Sulfonylurethane der Formel R-SO2-NH-COOX worin R einen gegebenenfalls durch einen oder zwei gleiche oder verschiedene Alkyl- oder Alkoxygruppen mit höchstens sechs Kohlenstoffatomen substituierten Phenylrest oder einen Naphthyl-(2)-, einen 5,6,7, 8-Tetrahydronaphthyl-(2)-, einen 4-Phenoxyphenyl-, einen 4-Biphenylrest oder einen Alkyl- oder Cycloalkylrest mit 3-6 Kohlenstoffatomen oder den Cyclohexylmethylrest bedeutet und X für den Rest eines andern Alkohols als der nachfolgend genannten steht, mit aliphatischen Alkoholen mit 4 bis 6 Kohlenstoffatomen oder mit araliphatischen Alkoholen, deren Alkylengruppe 2-4 Kohlenstoffatome enthält, umestert.
Es war nicht zu erwarten, dass die vorliegenden Verfahrenserzeugnisse eine blutzuckersenkende Wirksamkeit besitzen würden, da Abwandlungen im Molekül der bisher durch eine derartige Wirksamkeit bekannten Sulfonylharnstoff-Abkömmlinge, beispielsweise ein Austausch der Carbonylgruppe in der Harnstoff-Gruppierung durch die Sulfonyl- oder Thiocarbonylgruppe oder ein Ersatz der Sulfonylgruppe durch eine weitere Carbonylgruppe, zu einem Verlust der spezifischen Wirksamkeit führen.
Als Ausgangsstoffe kommen neben der unsubstituierten Verbindung solche Benzolsulfonylurethane in Frage, die im Benzolkern ein oder zwei Alkylbzw. Alkoxygruppen mit höchstens 6 Kohlenstoffatomen enthalten, die sowohl geradkettig als auch verzweigt sein können. Beispielsweise seien genannt: o-, m- und p-Toluolsulfonylurethane; o-, und p-Äthyl-benzolsulfonylurethane, o-, m- und p-n-Propylbenzolsulfonylurethane; o-, m- und p-Isopropylbenzolsulfonylurethane ; die verschiedenen o-, m- und p-Butyl-, -Pentyl und -Hexyl-benzolsulfonyiurethane;
weiterhin entsprechende o-, m- und p-Alkoxy benzolsulfonylurethane, wie o-, m-und p-Methoxy-benzolsulfonylurethane;
Dialkylbenzol-sulfonylurethane,
Dialkoxy-benzolsulfonylurethane sowie Alkyl-alkoxybenzol-suifonylurethane.
Insbesondere sind solche Verbindungen geeignet, die in oder p-Stellung substituiert sind. Als Ausgangsstoffe sind auch geeignet geradkettige oder verzweigte Alkansulfonylurethane, wie n-Propan-, Isopropan-, n-Butan-, Isobutan-, sek.-Butan-, tert.-Butan-, 3- Methylbutan-, n - Hexansulfonylurethane, Cycloalkansulfonylurethane, wie Cyclopentan-, Cyclohexansulfonylurethane, sowie Cyclohexylmethansulfonyl- urethane, sowie Naphthyl-(2)- bzw. 5,6,7,8-Tetra- hydronaphthyl-(2)- oder 4-Phenoxy-benzol- bzw.
4-Biphenyl-sulfonylurethane. Als Alkoholkomponente für diese Sulfonylurethane kommen insbesondere die Methyl- oder Äthylgruppe in Betracht.
Zur Umsetzung mit den vorstehend genannten Verbindungen können beispielsweise herangezogen werden: Alkohole, wie Butanol-(l), Butanol-(2), Methylpropanol-(l), Methylpropanol-(2), Pentanol (1), Pentanol-(2), Pentanol-(3), 2-Methylbutanol-( 1), 2-Methylbutanol-(2), 2-Methylbutanol-(3), 2-Methylbutanol-(4), Dimethylpropanol, die verschiedenen Hexylalkohole sowie araliphatische Alkohole, wie Phenyläthanol Phenylpropanole, wie 3-Phenylpropanol-(l), und Phenylbutanole, wie 4-Phenyl butanol-(l).
Zur Durchführung des erfindungsgemässen Verfahrens arbeitet man zweckmässig in Gegenwart von katalytisch wirkenden tertiären organischen Basen, Alkalihydroxyden oder Alkalialkoholaten.
Die Verfahrenserzeugnisse bilden sehr gute kristallisierende, leicht zu reinigende Alkali-, insbesondere Natriumsalze, eine Eigenschaft, die für die Verwendung der betreffenden Produkte als orale Antidiabetika besonders wertvoll ist, um so mehr, als sich diese Salze durch grosse Stabilität und leichte Wasserlöslichkeit bei annähernd neutraler Reaktion auszeichnen.
Die Verfahrensprodukte, die, wie bereits erwähnt, auch in Form ihrer Salze, aber auch in Gegenwart von Stoffen, welche zu einer Salzbildung führen, verwendet werden können, stellen wertvolle Arzneimittel dar und zeichnen sich insbesondere durch eine starke blutzuckersenkende Wirkung aus. Im Tierversuch lässt sich die Wirkung auf den Blutzuckerspiegel beispielsweise von Kaninchen und Hunden nachweisen. Verabreicht man die Verfahrenserzeugnisse z. B. normal gefütterten Kaninchen in einer einmaligen Dosis von durchschnittlich 400 mg/kg, beispielsweise in Form ihrer Alkalisalze, so sieht man eine rasch einsetzende Senkung des Blutzuckerspiegels, die innerhalb von etwa 3 bis 4 Stunden 20 bis 35 0/0 des Ausgangswertes erreicht. Die Blutzuckerwerte können durch stündliche Analysen nach Hagedorn/Jensen erhalten werden.
Die Blutzuckersenkung wird durch Vergleichen mit den Blutzuckerwerten gleichartiger, nicht behandelter Kontrolltiere erhalten. Die Prüfung der Verbindungen am Hund bietet den Vorteil, dass die Resorptionsverhältnisse im Magen-Darm-Kanal denen beim Menschen ähnlich sind, und dass der Blutzuckerspiegel geringere individuelle Schwankungen als beim Kaninchen aufweist.
Man erhält beim Hund schon bei Applikation niedriger Dosen, auch unter Verwendung der freien Urethane, gut reproduzierbare Werte. Verabreicht man die in Frage stehenden Verbindungen an nüchterne Hunde in einer einmaligen Dosis von 100 mg/kg, beispielsweise in Form ihrer Natriumsalze, so beobachtet man nach 24 Stunden Senkungen des Blut zuckerwertes bis zu 40 0/G. Die genannten Verbindungen sind gut verträglich und zeigen nicht die von Sulfonamiden bekannten Nebenwirkungen wie die durch die Beeinflussung der Bakterienflora des Darmes bedingten Verdauungsstörungen. Sie sollen beispielsweise zur Herstellung von oral verabreichbaren Präparaten mit blutzuckersenkender Wirkung zur Behandlung der Zuckerharnruhr dienen.

N-Benzolsulfonyl-hexyl-urethan 400/o (+) in Form des Kaliumsalzes
Beispiel 1 N-(p-Toluolsultonyl)-n-butylureíhan
114,5 g N-(p-Toluolsulfonyl)-methylurethan werden in Gegenwart von 50,5 g Triäthylamin in 1 Liter Butanol etwa 16 Stunden lang unter Rückfluss zum Sieden erhitzt. Man zieht den überschüssigen n-Butylalkohol unter vermindertem Druck ab und verreibt den erhaltenen Rückstand mit verdünnter Salzsäure.
Nach Abdestillieren wird der erhaltene Sirup in verdünntem Ammoniak aufgenommen. Nach Zusetzen von konzentrierter Natronlauge im Überschuss erhält man spontan und in guter Ausbeute Kristalle von N-(p-Toluolsulfonyl)-n-butylurethan-natrium, die man absaugt, mit 2n-Natronlauge und Aceton wäscht, im Exsikkator trocknet und aus Butylalkohol umkristallisiert, wobei ein Überschuss von Alkali durch Zugabe einiger Tropfen Schwefelsäure neutralisiert wird. Man erhält N-(p-Toluolsulfonyl)-n-butylurethan-natrium in Kristallen vom Schmelzpunkt 200-203". Die Substanz löst sich leicht in Wasser mit annähernd neutraler Reaktion.
Beispiel 2
N-(p-Methoxy-benzolsulf onyl)-n-butylurethan
50 g N-(p-Methoxy-benzolsuffonyl)-methylurethan (hergestellt aus p-Methoxy-benzolsulfonylamid und Chlorameisensäuremethylester) werden mit 400 cm3 n-Butylalkohol und 20 g Triäthyl-amin 17 Stunden unter Rückfluss zum Sieden erhitzt. Man engt unter vermindertem Druck ein, verreibt mit Salzsäure, dekantiert und löst den erhaltenen Rückstand in Ammoniak auf. Durch Zusatz von konzentrierter Natronlauge im Überschuss erhält man einen Kristallbrei von N-(p-Methoxy- benzolsulfonyl)-n- butyl- urethan-natrium, den man absaugt und mit verdünnter Natronlauge und Aceton nachwäscht.
Man löst das rohe Natriumsalz in Aceton, das etwas Wasser enthält, neutralisiert die Lösung mit wenig konzentrierter Schwefelsäure, filtriert und fällt das Methoxy-benzolsulfonyl)-n-butyl-urethan-natrium mit Ather. Die Substanz, welche in kristalliner Form anfällt, schmilzt bei 186-187".
Beispiel 3 N-(3-Methyl-benzolsuUonyl)-butylurethan
Ein Gemisch von 57,3 g N-(3-Methyl-benzolsulfonyl)-methyl-urethan, 500 cm3 Butanol und 25,3 g Triäthylamin wird 17 Stunden unter Rückfluss zum Sieden erhitzt. Man zieht das überschüssige Butanol unter vermindertem Druck ab, verreibt den erhaltenen Rückstand mit verdünnter Salzsäure und dekantiert die wässrige Flüssigkeit ab. Der erhaltene sirupöse Rückstand wird in verdünntem Ammoniak gelöst; die Lösung wird filtriert und mit konzentrierter Natronlauge im Überschuss versetzt. Man erhält ein Kristallisat von N-(3-Methylbenzolsulfonyl)butylurethan-natrium, das man absaugt und mit etwas Aceton nachwäscht. Das Salz wird zur Reinigung aus Butylalkohol umkristallisiert. Es schmilzt bei 163 bis 164".
Beispiel 4
N-(3, 4-Dimethoxy-benzolsulf onyl)-carbaminsäure- (2-phenyl-äthyl)-ester
27,5 g N-(3 4-Dimethoxy-benzolsulfonyl)-carb- aminsäure-methylester, 10,1 g Triäthylamin und 200 cm3 2-Phenyl-äthanol-(1) werden 7 Stunden auf 150 erhitzt. Man dampft das überschüssige Phenyl äthanol unter vermindertem Druck möglichst vollständig ab, schüttelt den verbleibenden festen Rückstand mit verdünnter Salzsäure durch und äthert aus.
Die ätherische Lösung wird mit verdünnter Salzsäure und Wasser gewaschen und nach dem Trocknen über Natriumsulfat der Äther abdestilliert. Man löst den Rückstand in wenig Äthanol, neutralisiert durch Zugabe einer Natriumäthylatlösung und fällt durch langsamen Zusatz von trockenem Ather das Natriumsalz des N-(3 ,4-Dimethoxy-benzolsulfonyl)-carbaminsäure- (2-phenyl-äthyl)-esters in kristallisierter Form aus.
Dieses zeigt nach dem Umkristallisieren aus verdünntem Äthanol den Schmelzpunkt 2480.
Beispiel 5 N-(4-Athyl-benzolsulfonyl)-n-b utylure than
48,6 g N-(4-Sithyl-benzolsulfonyl)-carbaminsäure- methylester (hergestellt durch Umsetzung von 4-Äthyl- benzolsulfamid mit Chlorameisensäuremethylester in Gegenwart von wasserfreiem Kaliumcarbonat und Aceton), 20,5 g Triäthylamin und 600 cm3 n-Butanol werden 22 Stunden unter Rückfluss gekocht. Man dampft das überschüssige Butanol unter vermindertem Druck möglichst vollständig ab, nimmt den verbleibenden Rückstand in Essigester auf und schüttelt die Essigesterlösung nacheinander mit verdünnter Salzsäure und Wasser durch. Das gebildete Sulfonylurethan wird mit verdünntem Ammoniak (1 25) der Essigesterlösung entzogen.
Die ammoniakalischen Auszüge werden mit Kohle geklärt und das Filtrat langsam und unter Kühlung mit konzentrierter Natronlauge versetzt. Der ausgefallene kristallisierte Niederschlag wird abgesaugt, mit wenig Eiswasser gewaschen und aus 150 cm3 Wasser unter Zusatz von Tierkohle umkristallisiert. Man erhält auf diese Weise das Natriumsalz des N-(4-Äthyl-benzolsullonyl)-n- butylurethans in guter Ausbeute und vom Schmelzpunkt 210-212".
Beispiel 6 N-Cyclohexansulfonyl-butyl-(1)-urethan
45 g N-Cyclohexansulfonyl-methylurethan (dargestellt aus Cyciohexansulfonamid und Chlorameisensäuremethylester in Gegenwart von Kaliumcarbonat in Aceton) werden in 400 ml n-Butylalkohol gelöst und 17 Stunden unter Rückfluss zum Sieden erhitzt.
Nach dem Einengen im Vakuum wird das Produkt in 2 o/oigem wässrigen Ammoniak gelöst. Man klärt die Lösung mit Kohle und säuert sie mit Salzsäure an. Das ausgeschiedene N-Cyclohexansulfonyl-butyl (l)-urethan wird mit Ather gesammelt und nach dem Verdampfen des Ethers im Vakuum destilliert.
Kp0,00 175-180". Das Destillat kristallisiert beim Behandeln mit Diisopropyl-äther und schmilzt bei 63-65".
Beispiel 7 N-(Naphthalin-2 -sulfonyl)-n-b utylure than
44,2 g N-(Naphthalin-2-sulfonyl)-carbaminsäuremethylester, 16,8 g Triäthylamin und 400 cm3 n-Butanol werden 20 Stunden zum Sieden erhitzt und anschliessend wird das Butanol im Vakuum möglichst vollständig abdestilliert. Der Rückstand wird mit einem Überschuss von verdünnter Salzsäure versetzt und die organischen Anteile in Essigester aufgenommen. Die vereinigten Essigesterauszüge werden nacheinander mit 2n-Salzsäure und Wasser ausgeschüttelt.
Die Essigesterlösung wird anschliessend mit verdünntem Ammoniak (1: 25) durchgeschüttelt, wobei das gebildete Sulfonylurethan wieder in die wässerige Phase übergeführt wird. Die wässerige Lösung klärt man mit Kohle und versetzt das schwach gelbliche Filtrat unter Kühlung und unter Umschütteln mit konzentrierter Natronlauge. Der ausfallende weisse Niederschlag wird abgesaugt, mit kaltem Wasser gewaschen und aus 400 cm3 Wasser unter Zusatz von Tierkohle umkristallisiert. Man erhält auf diese Weise das Natriumsalz des N-Naphthalin-2-sulfonyl)-nbutylurethans in guter Ausbeute vom Schmelzpunkt 2212230.
In analoger Weise erhält man unter Verwendung von n-Hexanol das Natriumsalz des N-(Naphthalin2-sulfonyl)-n-hexylurethans in guter Ausbeute vom Schmelzpunkt 2121 60.
Beispiel 8
N-(5, 6, 7,8-Te trahydronap2ithalin-2 -suonyl)- n-butylurethan
20 g N-(5,6,7, 8-Tetrahydronaphthalin-2-sulfonyl- carbaminsäure-methylester, 7,5 g Triäthylamin und 160 cm3 n-Butanol werden 20 Stunden unter Rückfluss gekocht. Man engt die Reaktionsmischung im Vakuum möglichst weitgehend ein, versetzt nach dem Abkühlen mit 2n-Salzsäure und nimmt die organischen Anteile in Essigester auf. Die vereinigten Essigesterlösungen werden nacheinander mit verdünnter Salzsäure und Wasser durchgeschüttelt.
Durch Ausschütteln der Essigesterlösung mit ver dünntem Ammoniak (1 25) bringt man das gebildete Sulfonylurethan wieder in die wässerige Phase. Man klärt mit Kohle und versetzt das Filtrat unter Kühlung und Umschütteln mit konzentrierter Natronlauge.
Es scheidet sich langsam ein in Blättchen kristallisierender Niederschlag ab. Nach dem Abkühlen wird der Niederschlag abgesaugt, mehrmals mit wenig Eiswasser gewaschen und anschliessend aus 50 cm : 3 Wasser unter Zusatz von Tierkohle umkristallisiert.
Das in guter Ausbeute erhaltene Natriumsalz des N-(5,6,7, 8-Tetrahydronaphthalin-2-salfonyl)-n-butyl - urethans schmilzt bei 184 186 .
Beispiel 9 N-(B iphenyl-4-sulfonyl)-n-bu tylurethan
29 g N-(Biphenyl - 4 - sulfonyl) - carbaminsäure- methylester vom Schmelzpunkt 136-138" (hergestellt durch Umsetzung von Biphenyl-4-sulfamid mit Chlorameisensäuremethylester in Gegenwart von Kaliumcarbonat und Aceton), 11 g Triäthylamin und 250 cm3 n-Butanol werden 20 Stunden zum Sieden erhitzt und anschliessend wird das überschüssige Butanol möglichst vollständig abdestilliert. Der verbleibende Rückstand wird wie vorstehend beschrieben aufgearbeitet. Man erhält nach dem Umkristallisieren aus Wasser das Natriumsalz des N-(Biphenyl 4-sulfonyl)-carbaminsäure-n-butylesters vom Schmelzpunkt 268-270".
Process for the preparation of sulfonyl urethanes
It is known that p-toluenesulfonyl urethanes are obtained if alcohols are allowed to act on p-toluenesulfonyl isocyanate. For example, German Patent No. 845042 describes the preparation of N- (p-toluenesulfonyl) isopropyl urethane by reacting p-toluenesulfonyl isocyanate with isopropyl alcohol. The same patent also mentions N- (p-toluenesulfonyl) methyl urethane. The transesterification of N- (p-toluenesulfonyl) isopropyl urethane or of toluenesulfonyl) methyl urethane with dodecyl alcohol to give N- (p-toluenesulfonyl) dodecyl urethane is also described.
Furthermore, substituted benzenesulfonylureas are known, for example, from Naturwissenschaften 43 (1956), page 93, which bring about a strong and long-lasting lowering of the blood sugar value.
Numerous other derivatives of the sulfonylurea have been proposed as oral antidiabetic agents.
It has now been found that the blood sugar-lowering effectiveness is not, as could be assumed on the basis of previous research results, bound to the sulfonylurea grouping, but that, surprisingly, sulfonyl urethanes also have such an effectiveness.
The present invention relates to a process for producing blood sugar-lowering sulfonyl urethanes, which is characterized in that sulfonyl urethanes of the formula R-SO2-NH-COOX wherein R is a phenyl radical optionally substituted by one or two identical or different alkyl or alkoxy groups with at most six carbon atoms or a naphthyl (2), a 5,6,7, 8-tetrahydronaphthyl (2), a 4-phenoxyphenyl, a 4-biphenyl radical or an alkyl or cycloalkyl radical with 3-6 carbon atoms or the cyclohexylmethyl radical and X represents the radical of an alcohol other than the one mentioned below, transesterified with aliphatic alcohols having 4 to 6 carbon atoms or with araliphatic alcohols whose alkylene group contains 2-4 carbon atoms.
It was not to be expected that the present process products would have a blood sugar-lowering effect, since modifications in the molecule of the sulfonylurea derivatives known to date for such effectiveness, for example an exchange of the carbonyl group in the urea group by the sulfonyl or thiocarbonyl group or a replacement the sulfonyl group by a further carbonyl group, lead to a loss of the specific effectiveness.

Those compounds which are substituted in the or p-position are particularly suitable. Straight-chain or branched alkanesulfonyl urethanes, such as n-propane, isopropane, n-butane, isobutane, sec-butane, tert-butane, 3-methylbutane, n-hexanesulfonyl urethanes, cycloalkanesulfonyl urethanes, are also suitable as starting materials. such as cyclopentane, cyclohexane sulfonyl urethane, as well as cyclohexyl methane sulfonyl urethane, and naphthyl (2) or 5,6,7,8-tetrahydronaphthyl (2) or 4-phenoxy-benzene or
4-biphenyl-sulfonyl urethanes. Particularly suitable alcohol components for these sulfonyl urethanes are the methyl or ethyl groups.

The blood sugar reduction is obtained by comparing with the blood sugar values of similar, untreated control animals. Testing the compounds in dogs has the advantage that the absorption ratios in the gastrointestinal tract are similar to those in humans and that the blood sugar level shows less individual fluctuations than in rabbits.
In dogs, even when applying low doses, even when using the free urethanes, values that are readily reproducible are obtained. If the compounds in question are administered to fasting dogs in a single dose of 100 mg / kg, for example in the form of their sodium salts, decreases in the blood sugar level of up to 40% are observed after 24 hours. The compounds mentioned are well tolerated and do not show the side effects known from sulfonamides such as the digestive disorders caused by influencing the bacterial flora of the intestine. They should serve, for example, for the production of orally administrable preparations with blood sugar lowering effect for the treatment of sugar urinary dysfunction.

Example 4
N- (3, 4-Dimethoxy-benzenesulphonyl) -carbamic acid- (2-phenyl-ethyl) -ester
27.5 g of N- (3 4-dimethoxy-benzenesulfonyl) -carbamic acid methyl ester, 10.1 g of triethylamine and 200 cm3 of 2-phenyl-ethanol- (1) are heated to 150 for 7 hours. Example 6 N-Cyclohexanesulfonyl-butyl- (1) -urethane
45 g of N-cyclohexanesulfonyl-methyl urethane (prepared from cyclohexanesulfonamide and methyl chloroformate in the presence of potassium carbonate in acetone) are dissolved in 400 ml of n-butyl alcohol and refluxed for 17 hours.

44.2 g of methyl N- (naphthalene-2-sulfonyl) carbamate, 16.8 g of triethylamine and 400 cm3 of n-butanol are heated to boiling for 20 hours and the butanol is then distilled off as completely as possible in vacuo. The aqueous solution is clarified with charcoal and the pale yellowish filtrate is mixed with concentrated sodium hydroxide solution while cooling and while shaking. The resulting white precipitate is filtered off with suction, washed with cold water and recrystallized from 400 cm3 of water with the addition of animal charcoal. Example 8
N- (5, 6, 7,8-tetrahydronap2ithalin-2 -suonyl) - n-butyl urethane
20 g of methyl N- (5,6,7,8-tetrahydronaphthalene-2-sulfonylcarbamate, 7.5 g of triethylamine and 160 cm3 of n-butanol are refluxed for 20 hours.
