Beispielsweise seien genannt: o-, m- und p-Toluolsulfonylharnstoff; o-, m- und p-Äthyl-benzolsulfonylharnstoff, o-, m- und p-n-Propylbenzolsulfonylharnstoff; o-, m- und p-Isopropyl-benzolsulfonylharnstoff; die verschiedenen o-, m- und p-Butyl-, -Pentyl- und -Hexyl-benzolsulfonylhamstoffe; weiterhin entsprechende o-, m- und p-Alkoxy-benzol sulfonylharnstoffe, wie o-, m- und p-Methoxy-benzolsulfonylharnstoff;
Dialkylbenzol-sulfonylharnstoffe,

Process for the preparation of sulfonyl urethanes
It is known that p-toluenesulfonyl urethanes are obtained if alcohols are allowed to act on p-toluenesulfonyl isocyanate. For example, German patent specification No. 845042 describes the preparation of N - (p - toluenesulfonyl) - isopropyl urethane by reacting p-toluenesulfonyl isocyanate with isopropyl alcohol. The same patent also mentions N- (p-toluenesulfonyl) methyl urethane. The transesterification of N- (p-toluenesulfonyl) 4sopropylurethane or of N- (p-toluenesulfonyl) -methylurethane with dodecyl alcohol to give N- (p-toluenesulfonyl) -dodecylurethane is also described.
Furthermore, substituted benzenesulfonylureas are known, for example, from Naturwissenschaften 43 (1956), page 93, which bring about a strong and long-lasting lowering of the blood sugar value.
Numerous other derivatives of the sulfonylurea have been proposed as oral antidiabetic agents.
It has now been found that the blood sugar-lowering effectiveness is not, as could be assumed on the basis of previous research results, bound to the sulfonylurea grouping, but that, surprisingly, sulfonyl urethanes also have such an effectiveness.
The present invention relates to a process for the preparation of blood sugar-lowering sulfonyl urethanes, which is characterized in that sulfonylureas with a disubstituted terminal amino group of the formula R-SO, -NH-CO-N <where R is optionally replaced by one or two identical or different alkyl or alkoxy groups with at most six carbon atoms substituted phenyl radical or a naphthyl (2) -, a 5,6,7, 8-tetrahydronaphthyl (2) -, a 4-phenoxyphenyl, a 4-biphenyl radical or an alkyl or cycloalkyl radical 3-6 carbon atoms or the cyclohexylmethyl radical means, with aliphatic alcohols with 4-6 carbon atoms or with araliphatic alcohols whose alkylene group contains 2 to 4 carbon atoms, to form the corresponding esters.
It was not to be expected that the present process products would have a blood sugar-lowering effectiveness, since modifications in the molecule of the sulfonylurea derivatives known to date for such effectiveness, for example an exchange of the carbonyl group in the urea group by the sulfonyl or thiocarbonyl group or a replacement of the Sulphonyl group through a further carbonyl group lead to a loss of the specific effectiveness.
Suitable starting materials are those benzenesulfonylureas which are disubstituted in the terminal amino group and which contain one or two alkyl or alkoxy groups with at most 6 carbon atoms in the benzene nucleus, which groups can be either straight-chain or branched.
Examples include: o-, m- and p-toluenesulfonylurea; o-, m- and p-ethylbenzenesulfonylurea, o-, m- and p-n-propylbenzenesulfonylurea; o-, m- and p-isopropyl-benzenesulfonylurea; the various o-, m- and p-butyl-, -pentyl- and -hexylbenzenesulfonylureas; also corresponding o-, m- and p-alkoxy-benzene sulfonylureas, such as o-, m- and p-methoxy-benzenesulfonylurea;
Dialkylbenzenesulfonylureas,
Dialkoxy-benzenesulfonylureas and alkyl-alkoxy-benzenesulfonylureas.
Those compounds which are substituted in the or p-position are particularly suitable. Straight-chain or branched alkanesulfonylureas, such as n-propane, isopropane, n-butane, isobutane, secondary butane, tert-butane, 3-methylbutane, n-hexane-sulfonylurea, cycloalkanesulfonylureas, are also suitable as starting materials. such as cyclopentane, cyclohexane-sulfonylureas, as well as cyclohexylmethanesulfonylurea, as well as naphthyl- (2) - or 5,6,7,8 tetrahydronaphthyl- (2) - or 4-phenoxy-benzene or
4-biphenylsulfonylureas. All of the above-mentioned ureas contain two substituents in the terminal amino group.
The following can be used for the reaction with the above-mentioned compounds: aliphatic alcohols such as butanol (1), butanol (2), methylpropanol (1), methylpropanol (2), pentanol (1), pentanol (2), Pentanol (3), 2-methylbutanol (1), 2-methylbutanol (2), 2-methylbutanol (3), 2-methylbutanol (4), dimethylpropanol, also hexyl alcohols and araliphatic alcohols such as phenylethanol, phenylpropanols such as 3-phenylpropanol- (l), and phenylbutanols such as 4-phenylbutanol- (l).
The products of the process form very good crystallizing, easy-to-clean alkali salts, especially sodium salts, a property which is particularly valuable for the use of the products in question as oral antidiabetic agents, all the more since these salts are almost neutral due to their great stability and easy water solubility Excellent response.
The products of the process, which, as already mentioned, can also be used in the form of their salts, but also in the presence of substances which lead to salt formation, are valuable medicaments and are characterized in particular by a strong blood sugar-lowering effect. The effect on blood sugar levels in rabbits and dogs, for example, can be demonstrated in animal experiments. If the process products are administered z. B. normal fed rabbits in a single dose of an average of 400 mg / kg, for example in the form of their alkali salts, one sees a rapid onset of lowering of the blood sugar level, which reaches 20 to 35% of the initial value within about 3 to 4 hours. The blood sugar values can be obtained by hourly analyzes according to Hagedorn / Jensen.
The blood sugar reduction is obtained by comparison with the blood sugar values of similar, untreated control animals. Testing the compounds in dogs has the advantage that the absorption ratios in the gastrointestinal tract are similar to those in humans and that the blood sugar level shows less individual fluctuations than in rabbits. In dogs, even when applying low doses, even when using the free urethanes, values that are easily reproducible are obtained.
If the compounds in question are administered to fasting dogs in a single dose of 100 mg / kg, for example in the form of their sodium salts, decreases in blood sugar levels of up to 40 / c are observed after 24 hours. The compounds mentioned are well tolerated and do not show the side effects known from sulfonamides such as the digestive disorders caused by influencing the bacterial flora of the intestine. They should serve, for example, for the production of orally administrable preparations with blood sugar lowering effect for the treatment of sugar urinary dysfunction.
The effectiveness of some of the process products can be seen in the table below.

19.7 g of p-toluenesulphonyl isocyanate and 16.9 g of diphenylamine are combined and mixed well in accordance with the instructions given in Example 3 of German Patent No. 845042, and solution occurs. The reaction mixture warms up and soon solidifies with crystallization of N- (4-methylbenzenesulfonyl) -N'-diphenylurea. 250 cm3 of n-butanol are added and the reaction mixture is refluxed for several hours. After the reaction has ended, the solution is concentrated in vacuo and the residue obtained is treated with 10% aqueous ammonia.
The undissolved material is filtered off with suction and the N- (4-methylbenzenesulfonyl) butyl urethane is precipitated by acidification with dilute hydrochloric acid. The precipitated grease is then taken up in ether, the ether solution is washed in water and dried over sodium sulfate. After concentrating the ether, the residue is dissolved in a little methanol, the solution is treated with sodium methylate up to a pH of 6.5 and the N- (4-methylbenzenesulfonyl) butyl urethane sodium salt is added by adding absolute ether Crystallization. After recrystallization from n-butanol, the substance has a melting point of 204 to 206.
In an analogous manner, N- (4-methylbenzenesulfonyl) -isobutyl-urethane, the sodium salt of which melts at 201-203 ", is obtained from the reaction product of p-toluenesulfonyl isocyanate and caprolactam, obtained according to Example 1 of German Patent No. 845042, and isobutyl alcohol.
