Die genannten Amide haben sich als ausserordent lich gute Lokalanästhetika erwiesen, deren Effekt denjenigen, des Procains vielfach übertrifft.

erhalten wird, worin n 3 oder 4 ist, welche durch Be handlung mit Mineralsäuren unter Bildung von einem Piperidin- bzw,
Pyrrolydin-a-carbonsäureamid ring geschlossen und decarboxyliert wird, und dass das so erhaltene Produkt durch Alkylierung des Ringstick stoffes in ein N - Alkyl - piperidin- bzw. N-Alkyl- pyrrolidin-a-carbonsäureamid übergeführt wird. <I>Beispiel 1</I> 121 Gewichtsteile 2,6-Xylidin werden während 1 Stunde mit 400 Gewichtsteilen Diäthylmalonat bei 160 erhitzt, wobei der bei der Reaktion gebildete Alkohol abdestillieren soll.
Danach wird auf 80 ab- gekühlt, und 500 Teile Alkohol werden zugesetzt. Nach der Kühlung wird das Dixylidid abgesaugt und die Alkohollösung wird mit dem Malonestermonoxy- lidid in 2000 Teile Wasser gegossen. Das Monoxy- lidid fällt aus, wird abgenutscht und mit Wasser ge waschen sowie in verdünntem Alkohol umkristalli siert.
Die Nitrosierung geschieht dann durch Lösen des getrockneten Monoxylidids in Chloroform sowie Zuführung des Nitrosylchlorids bei 0 , bis die Nitrö- sierung fertig ist. Das Isonitrosomalonesterxylidid wird abgesaugt und getrocknet. Die Reduktion ge schieht dann mit Zinkpulver und Ameisensäure bei 90 bis 100 . Die Ameisensäure wird abdestilliert, der Rest in warmem Benzol gelöst und mit Bicarbonat- lösung auf neutrale Reaktion gewaschen.
Nach dem Abtreiben von Benzol erhält man Aminomalon- esterxylidid. Dieses wird mit einer äquivalenten Menge Natriumethylat behandelt sowie mit der dop pelten theoretischen Menge von Tetramethylenbromid in absolutem Alkohol gekocht.
Nach 6 Stunden Ko- chens wird das gebildete Natriumbromid abgeschie den und die Mischung wasserdampfdestilliert, um den Überschuss an Tetramethylenbromid zu enffer- nen. Das zurückbleibende Öl, das hauptsächlich aus Brombutylaminomalonesterxylidid besteht, wird vom Wasser getrennt und während 3 Stunden mit 3 Teilen konzentrierter Salzsäure gekocht. Danach wird mit Kohle filtriert und bis zur Trockne im Vakuum ab gedampft.
Der Rest wird in Wasser gelöst, und der pH-Wert wird mit Lauge auf 5,5 eingestellt. Die Lö sung wird zweimal mit Äther extrahiert, und das Wasser wird mit Lauge stark alkalisch gemacht. Das Öl fällt aus und kristallisiert nach einiger Zeit. Die Kristalle werden abgeschieden und im Vakuum getrocknet. Das erhaltene Pipekolylxylidid wird während 3 Stunden mit 2 Gewichtsteilen Diäthyl- sulfat im Wasserbad erwärmt. Es wird Wasser zuge setzt, und die Lösung wird mit Äther extrahiert. Lauge wird auf p$<I>5,5</I> zugesetzt, und die Lösung wird gekohlt. Die Base wird mit Lauge gefällt, ab gesaugt und getrocknet.
Die erhaltene Base besteht aus N-Äthylpipekolyl-2,6-xylidid und ist zur Her stellung von Salzen genügend rein.
<I>Beispiel 2</I> 250 Gewichtsteile Aminomalonesterxylidid, das gemäss Beispiel 1 hergestellt wurde, werden mit einer äquivalenten Menge Natriumäthylat behandelt sowie 6 Stunden mit der doppelten theoretischen Menge Trimethylenbromid in absolutem Alkohol gekocht. Das Natriumbromid wird abgeschieden, und die Mi schung wird wasserdampfdestilliert, um den Über schuss an Trimethylenbromid zu entfernen.
Das zu rückgebliebene Öl, das aus Brompropylaminomalon- esterxylidid besteht, wird abgeschieden und während 3 Stunden mit 3 Teilen konzentrierter Salzsäure ge kocht. Danach wird filtriert und im Vakuum ein gedampft. Der Rückstand wird in Wasser gelöst und der pH-Wert mit Lauge auf 5,5 justiert. Die Lösung wird einige Male mit Äther extrahiert. Die Base wird mit Lauge aus der Wasserlösung gefällt und mit Äther extrahiert. Nach Abtreiben des Äthers erhält man a-Pyrrolidincarbonsäurexylidid als ein Öl.
Das Öl wird mit einem Teil Dimethylsulfat während 5 Stunden im Wasserbad erhitzt, in Wasser gelöst und bei p. 5,5 gekohlt. Die Base wird mit Lauge als Kristalle gefällt, welche abgesaugt und mit Wasser gewaschen werden. Das erhaltene (N-Methyl-pyrro- lidin-a-carbonsäure)-2,6-xylidid kann zur Herstel lung von Salzen verwendet werden.

It is then cooled to 80 and 500 parts of alcohol are added. After cooling, the dixylidide is suctioned off and the alcohol solution is poured with the malonic ester monoxylidide into 2000 parts of water. The monoxidide precipitates, is suction filtered, washed with water and recrystallized in dilute alcohol.
The nitrosation then takes place by dissolving the dried monoxylidide in chloroform and adding the nitrosyl chloride at 0 until the nitration is finished. The isonitrosomalone ester xylidide is filtered off with suction and dried. The reduction is then done with zinc powder and formic acid at 90 to 100. The formic acid is distilled off, the remainder is dissolved in warm benzene and washed with bicarbonate solution until it reacts neutral.
After stripping off benzene, aminomalone ester xylidide is obtained. This is treated with an equivalent amount of sodium ethylate and boiled with double the theoretical amount of tetramethylene bromide in absolute alcohol.
After 6 hours of boiling, the sodium bromide formed is separated out and the mixture is steam distilled in order to remove the excess of tetramethylene bromide. The remaining oil, which consists mainly of bromobutylaminomalone ester xylidide, is separated from the water and boiled with 3 parts of concentrated hydrochloric acid for 3 hours. It is then filtered with charcoal and evaporated to dryness in vacuo.
The remainder is dissolved in water and the pH is adjusted to 5.5 with lye. The solution is extracted twice with ether and the water is made strongly alkaline with lye. The oil precipitates and crystallizes after a while. The crystals are deposited and dried in vacuo. The resulting Pipekolylxylidid is heated for 3 hours with 2 parts by weight of diethyl sulfate in a water bath. Water is added and the solution is extracted with ether. Lye is added to p $ <I> 5.5 </I> and the solution is carbonized. The base is precipitated with lye, sucked off and dried.
The base obtained consists of N-ethylpipekolyl-2,6-xylidide and is sufficiently pure for the production of salts.
<I> Example 2 </I> 250 parts by weight of aminomalone ester xylidide, which was prepared according to Example 1, are treated with an equivalent amount of sodium ethylate and boiled in absolute alcohol for 6 hours with twice the theoretical amount of trimethylene bromide. The sodium bromide is separated and the mixture is steam distilled to remove the excess trimethylene bromide.
The remaining oil, which consists of bromopropylaminomalone ester xylidide, is separated off and boiled with 3 parts of concentrated hydrochloric acid for 3 hours. It is then filtered and evaporated in vacuo. The residue is dissolved in water and the pH is adjusted to 5.5 with alkali. The solution is extracted a few times with ether. The base is precipitated from the water solution with lye and extracted with ether. After the ether has been stripped off, α-pyrrolidinecarboxylic acid xylidide is obtained as an oil.
The oil is heated with a part of dimethyl sulfate for 5 hours in a water bath, dissolved in water and at p. 5.5 charred. The base is precipitated as crystals with lye, which are filtered off with suction and washed with water. The (N-methyl-pyrrolidine-a-carboxylic acid) -2,6-xylidide obtained can be used for the production of salts.
