Zu einer Lösung von 62 g 3-Oxy-thiophen-2-carbonsäure-n-butylester (farblose Flüssigkeit vom Kp.7 130 bis 1320C) in 155 ecm konzentrierter Schwefelsäure läßt man bei —5 bis 00C eine Mischung aus 15,4 ecm rauchender Salpetersäure (D = 1,52) und 78 ecm konzentrierter Schwefelsäure zutropfen und entfernt dann das Kältebad, worauf die Temperatur ansteigt. Durch leichtes Kühlen bzw. Erwärmen hält man die Temperatur der Reaktionsmischung 1 Stunde bei 400C, gießt das Gemisch dann auf Eis und nimmt das ausgeschiedene dunkle Öl mit Methylenchlorid auf. Nach dem Abtrennen der organischen Schicht wäscht man diese einmal mit Wasser, trocknet sie über Natriumsulfat und verdampft schließlich das Lö- ^5 sungsmittel vorsichtig bei 20 bis 30° C im Vakuum. Der Rückstand wird mit Wasserdampf destilliert und das übergegangene Öl nach schwachem Ansäuern des Destillates mit Methylenchlorid aufgenommen. Eventuell nicht verbrauchtes Ausgangsmaterial ist in den ersten Anteilen der Wasserdampfdestillation enthalten (Nachweis durch Papierchromatographie), die zweckmäßigerweise verworfen werden. Die Methylenchloridlösung wird über Natriumsulfat getrocknet und dann im Vakuum bei 20 bis 30° C vollkommen eingedampft. 72 g 3-Oxy-thiophen-2-carbonsäure-n-amylester (schwachgelbliche Flüssigkeit vom Kp.0,7 92 bis 94° C) werden bei —5 bis 00C in 168 ecm konzentrierter Schwefelsäure gelöst, und bei gleicher Temperatur wird ein Gemisch aus 16,7 ecm rauchender Salpetersäure (D = 1,52) und 84 ecm konzentrierter Schwefelsäure zugetropft. Anschließend entfernt man das Kältebad, wobei sich die Reaktionsmischung von selbst erwärmt. Durch zeitweiliges Rühren bzw. Erwärmen hält man die Temperatur der Reaktionsmischung noch 1 Stunde bei 40 bis 500C und gießt dann das Gemisch auf Eis. Hierbei scheidet sich ein dunkles Produkt aus, das mit Methylenchlorid aufgenommen wird. Die erhaltene Lösung wird nach einmaligem Waschen mit Wasser und Trocknen über Natriumsulfat im Vakuum bei 20 bis 30° C eingedampft. Der erhaltene Rückstand wird mit Wasserdampf destilliert und das übergegangene Öl nach schwachem Ansäuern des Destillates mit Methylenchlorid aufgenommen. Die Methylenchloridlösung wird über Natriumsulfat getrocknet und das Lösungsmittel im Vakuum bei 20 bis 300C vollkommen abdestilliert. Dabei bleiben als Rohprodukt 14 g eines nicht destillierbaren Gemisches von 4-Nitro-3-oxy-thiophen-2-carbonsäure-n-amylester und S-Nitro-S-oxy-thiophen^-carbonsäure-n-amylester als gelbgefärbte Flüssigkeit zurück.72 g 3-oxy-thiophene-2-carboxylic acid n-amyl ester (pale yellow liquid from Kp.0,7 92 to 94 ° C) is dissolved at -5 to 0 0 C in 168 cc of concentrated sulfuric acid, and is at the same temperature a mixture of 16.7 ecm fuming nitric acid (D = 1.52) and 84 ecm concentrated sulfuric acid was added dropwise. Zu einer Lösung von 19,6 g 3-Oxy-thiophen-2-carbonsäuremethylester in 60 ecm konzentrierter Schwefelsäure wird bei —5°C eine Mischung aus 9,5 ecm konzentrierter Salpetersäure (68%ig) und 40 ecm konzentrierter Schwefelsäure im Laufe von 90 Minuten unter Rühren zugetropft. Man rührt das Gemisch noch 1 Stunde bei der gleichen Temperatur nach, gießt dann das Reaktionsprodukt auf Eis und destilliert das Ganze anschließend mit Wasserdampf. Beim Abkühlen kristallisieren aus dem Destillat 3,3 g 4-Nitro-3-oxy-thiophen-2-carbonsäuremethylester aus.
