Process for the preparation of acid addition salts of amines derived from naphthalene It has been found that the naphthalene derivatives of general formula (1) have valuable antimalarial activity against blood forms of the species Plasmodium, and that they are several times more active than quinine. These compounds have the additional advantage that they act quickly, that they can be prepared simply and inexpensively and that they are almost tasteless.

In this formula, one of the groups Rl and R ,, and one of the groups R3 and R4 is a hydroxy group, the other of the groups Rl and R2 and of the groups R, and R4 is a residue of the formula - CH, NHR,;, where R ,; is an alkyl group having 1 to 8 carbon atoms, straight chain or branched, or a cycloalkyl group having 5 to 7 carbon atoms in the ring.
These compounds can be administered in the form of their salts by addition of acids. In accordance with the process of the invention, these salts are prepared by treating with an acid, in the presence of water, an oxazine of formula

in which X and Y represent the group -CH2-NR5-CH2-O-, the reaction being accompanied by the departure of formaldehyde.
The starting oxazines can be prepared by reacting the appropriate naphthalene diol with formaldehyde and the appropriate amine R5NH2, in a suitable solvent such as methanol; these ingredients react on contact or can be heated together.
By way of example, the following series of formulas illustrate the preparation of the compound 2,5-bis- (cyclohexylamino-methyl) -naphthalene-1,6-diol

Other valuable compounds can be prepared starting from naphthalene-1,5-diol or naphthalene-2,6-diol.
The opening of the oxazine ring in the compound of formula (II) can be effected by heating this compound in acidic solution in a suitable solvent such as aqueous propanol or aqueous isopropanol. 10% hydrochloric acid or 2N sulfuric acid are suitable for these purposes.
The reaction can also be carried out in the cold in the presence of a compound such as 2,4-dinitro-phenylhydrazine which reacts with formaldehyde as it is formed; this prevents losses of this substance by polymerization.
In the following examples, all temperatures are given in degrees centigrade. Example 1 (a) 2,9-dicyclohexyl-1,2,3,4,7,8,9,10-octahydro-2,9-diaza-4,7-dioxa-chrysene departure.
To a mixture of cyclohexylamine (22.8 cc) and formaldehyde (37 cc) in methanol (100 cc), maintained at -50 C, was added a solution of naphthalene-1,6-diol (16 g ) in methanol (150 cc). After standing overnight, the deposited oily oxazine was stirred with a mixture of ether and water and was filtered.
It had a melting point of 130 after crystallization from ethanol or dioxane. (b) 2,5-bis- (cyclohexylamino-methyl) -naphthalene-1,6-diol dihydrochloride.
The starting oxazine base (5 g) prepared according to (a) was suspended in a mixture of isopropanol (50 cc) and concentrated hydrochloric acid (2 cc) and the whole mixture was subjected to the distillation until no more formaldehyde can be detected in the distillate (one to three hours), isopropanol being added to keep the volume constant. The mixture was cooled then filtered, and the solid was washed with cold isopropanol and ether.
It was crystallized by dissolving it in 80% ethanol and adding ether; a gray solid product was obtained, melting point 214-215.
