et enfin Y représente de l'hydrogène ou du chlore ou un radical méthoxy, méthyle, nitro ou phényle se trouvant soit en position 2, soit en posi tion 3.
Conformément au procédé selon l'invention, ces dérivés de l'acridine sont obtenus, en condensant un 10-oxyde de la 6,9-dichloro-acridine de formule

en présence d'un solvant organique anhydre.
La condensation s'opère d'une manière satisfai sante par la mise en oeuvre de quantités pratique ment équimoléculaires des réactifs dans un solvant organique anhydre. Quelques solvants appropriés à cet emploi sont, par exemple :
des phénols, des alcools aliphatiques tels que l'éthanol, l'isopropanol, le pentanol et l'octanol, des cétanes aliphatiques tel les que la méthyléthylcétone et la di-isobutylcétone, des éthers aliphatiques tels que l'éther dibutylique et les monoéthers aliphatiques de l'éthylène-glycol, des éthers cycliques tels que le dioxane et des hydrocar bures tels que le benzène, le toluène et les xylènes.
On utilise de préférence le phénol. On peut, si on le désire, utiliser un excès de l'un des deux réactifs, mais il est préférable que l'aminoalcoylamine soit en excès, car elle se montre dans bien des cas un sol vant particulièrement approprié. La réaction est en général favorisée par des températures dépassant 750C. On l'effectue de préférence dans l'intervalle de température compris entre 950 et 1400C environ.
Toutefois, il est préférable d'utiliser une température inférieure à 200()C environ, car le produit final pré sente une tendance à la décomposition aux tempé ratures élevées, ce qui entraine une perte de rende ment. Le produit peut être isolé du mélange résultant de la réaction sous forme de base libre ou, si on le désire, sous forme d'un sel d'addition d'acide formé par l'introduction d'un acide approprié. On utilise de préférence à cet effet l'acide chlorhydrique, quoi- que des acides organiques ou d'autres acides miné raux puissent également être employés.
L'un des avantages du procédé est qu'il conduit aux produits finaux sans exiger la fabrication préli minaire d'un composé intermédiaire tel qu'un 9-phénoxyde.
Les produits obtenus au moyen de ce procédé présentent des propriétés antibactériennes et anti- parasitiques caractéristiques. Ils sont particulièrement utiles pour combattre les plasmodia de la malaria, les helminthes et l'Endomoeba histolytica, agent spé cifique de l'amibiase.
A ce point de vue, les chlor- hydrates des composés suivants, dérivés du 10-oxyde d'acridine, sont d'un intérêt particulier : le 10-oxyde de la 6 - chloro - 9 - (4 - diéthylamino -1- méthyl-butyl- am no)-2-méthoxy-acridine ; le 10-oxyde de la 3,6- dichloro-9-(3-pipéridino-propylamino)-acridine ;
les 10-oxydes des 3,6-dichloro- et 6-chloro-2-méthoxy- 9-(5-pipéridino-pentylamino)-acridines ; le 10-oxyde de la 6-chloro-9-(3-isopropylamino-propylamino)-2- méthoxy-acridine et le 10-oxyde du 2-[3-(6-chloro-2- méthoxy-9-acridinylamino)-propylamino]-éthanol.
Les exemples suivants montrent comment le pro cédé selon l'invention peut être exécuté Exemple 1 Dichlorhydrate <I>du</I> 10-oxyde <I>de la 6-chloro-</I> 9-(4-diéthylamino-1-méthyl-butylarrino)-2- méthoxy-acridine Un mélange de 14,8 g du 10-oxyde de la 6,9- dichloro-2-méthoxy-acridine, de 45 g de phénol et de 9 g de 5-diéthylamino-2-amino-pentane est agité et chauffé au bain-marie pendant deux heures (en le protégeant de l'humidité au moyen (fun tube à chlo rure de calcium).
On laisse refroidir à 50 le liquide de couleur rouge sombre qui résulte de l'opération et on, le verse lentement dans un mélange formé d'un litre d'acétone et de 10 ce d'acide chlorhydrique concentré. Un précipité jaune qui se sépare est recueilli par filtration, lavé à fond avec de l'acétone froid, séché sous vide et recristallisé dans l'alcool éthylique absolu. P.F. :

Le produit de départ, qui est le 10-oxyde de la 6,9-dichlo,ro-2-méthoxy-acridine, peut être obtenu de la manière suivante:
on traite à température ordi naire une solution de 278 g de 6,9-dichloro-2- méthoxy-acridine dans 51 de chloroforme avec une solution froide de 160g d'acide perbenzoïque dans 31 de chloroforme. On laisse reposer à température ordinaire pendant 36 heures, puis on ajoute de la feuille de platine, le mélange est alors concentré sous vide jusqu'à ce que son volume soit réduit à 1,5l, puis refroidi dans un bain de glace.
Le précipité formé est séparé par filtration et lavé à fond à l'éther; P.F. : 226-2280C. On obtient de cette manière le 10-oxyde de la 6,9-dichloro-2-méthoxy- acridine.
Exemple 2: Dichlorhydrate <I>de</I> 10-oxyde <I>de la</I> 3,6-dichloro- 9-(4-diéthylamino-1-rné thyl-butylarnino)- acridine On ajoute 20,9 g du 10-oxyde de. la 3,6,9-tri- chloro-acridine à un mélange de 11,1 g de 5-diéthyl- amino-2-amino-pentane et de 45 g de phénol.
Le mélange réactionnel est agité et chauffé à 110 - 11511C pendant deux heures, après quoi il est refroidi et versé lentement dans un mélange de 300 cm; d'acétone et de 10 ems d'acide chlorhydrique con centré.
Le mélange résultant est alors étendu à un volume de 1 1 par l'addition d'un mélange d'acétone et d'éther et le précipité formé est séparé par filtra tion et recristallisé à partir d'une solution dans un mélange méthanol-acétone ; P.F. :

Exemple 3 Dichlorhydrnte <I>du</I> 10-oxyde <I>de la 6-chloro-</I> 9-(4-déithylamino-1-méthyl-butylamino)- 3-rnéthyl-acridine Un mélange de 8,7 g de 5-diéthylamino-2- amino-pentane, de 50 g de phénol et de 13,9 g du 10-oxyde de la 3,9-dichloro-6-méthyl-acridine est agité et chauffé au bain-marie pendant 2 heures.
On laisse refroidir jusqu'à une température de 40CC le mélange de couleur rouge sombre qui résulte de la réaction et on le verse lentement dans un mélange formé de 1 1 d'acétone et de 10 cm; d'acide chlor hydrique concentré. Le précipité jaune formé est séparé par filtration, lavé à fond avec de l'éther, séché sous vide et recristallisé à partir de sa solution dans l'éthanol.
Le produit, qui est le dichlorhydrate du 10-oxyde de la 6-chloro-9-(4-diéthylamino-1- méthyl-butylamino) - 3 -méthyl- acridine, possède la formule

Le 10-oxyde de la 3,9-dichloro-6-méthyl-acri- dine, utilisé comme produit de départ dans le pré sent exemple, peut être préparé comme suit:
on mélange 191 g d'acide 2,4-dichloro-benzoïque, 134 g de m-toluidine, 138 g de carbonate de potas sium anhydre, 5 g de poudre de cuivre et 750 cm; d'alcool n-amylique sec, on agite et l'on fait bouillir à reflux pendant 5 heures. Le mélange est refroidi, traité au moyen de 70g de potasse caustique en solution dans l00 cm; d'eau, et soumis à l'entraî nement par la vapeur d'eau. Le résidu aqueux est filtré à chaud, acidulé légèrement avec de l'acide chlorhydrique concentré, et refroidi.
L'acide préci pité est séparé par filtration, lavé à l'eau, puis à l'éther de pétrole (point d'ébullition: 30-600C). On fait alors bouillir le précipité filtré avec 300 ce d'éthanol, on refroidit rapidement, filtre et lave à l'éther de pétrole (point d'ébullition: 30-60 C).
La recristallisation du produit à partir de sa solution dans un mélange de monochlorobenzène et ligroïne (point d'ébullition 60 à 800C) (noir animal) conduit à l'acide 4-chloro-N-(m-toluyl-anthranilique, sous forme d'aiguilles, presque incolores, fondant à 197 1990C.
Un mélange formé de 64 g de cet acide et de 200 cm- d'oxychlorure de phosphore est chauffé à feu doux jusqu'à ce que la réaction exothermique prenne fin ; on le fait bouillir ensuite à reflux pen dant deux heures.
On chasse une partie de l'excès de l'oxychlorure de phosphore par le vide, on dissout le résidu dans le chloroforme sec et l'on verse la solution chloroformique lentement et en remuant dans un excès d'ammoniaque contenant de la glace. On ajoute une seconde portion. de chloroforme et les solutions chloroformiques additionnées sont séchées sur du carbonate de potassium anhydre.
On chasse le chloroforme par le vide et le résidu est soumis à une cristallisation fractionnée à partir de sa solution dans un mélange benzène et acétone, ce qui permet d'obtenir la 3,9-dichloro-6-méthyl-acridine, P.F. 2060C. On soumet alors ce produit à l'action de l'acide perbenzoïque,
selon la méthode décrite dans l'exemple 1 en vue de la préparation du 10-oxyde de la 6,9-dichloro-2-méthyl-acridine ; P.F. : 172-1740C.
D'autres 10-oxydes de 9-aminoalcoylamino-acri- dinë peuvent être préparés de manière semblable à partir d'aminoalcoylamines et de 10-oxydes de di- chloro-acridine, comme indiqué dans le tableau suivant

In accordance with the process according to the invention, these acridine derivatives are obtained by condensing a 10-oxide of 6,9-dichloro-acridine of formula

The condensation is carried out satisfactorily by employing substantially equimolecular amounts of the reactants in an anhydrous organic solvent. Some solvents suitable for this use are, for example:
phenols, aliphatic alcohols such as ethanol, isopropanol, pentanol and octanol, aliphatic ketanes such as methyl ethyl ketone and di-isobutyl ketone, aliphatic ethers such as dibutyl ether and aliphatic monoethers ethylene glycol, cyclic ethers such as dioxane and hydrocarbons such as benzene, toluene and xylenes.
Preferably, phenol is used. An excess of one of the two reagents can, if desired, be used, but it is preferable that the aminoalkylamine is in excess, since it is in many cases a particularly suitable solvent. The reaction is generally favored by temperatures exceeding 750C. It is preferably carried out in the temperature range of between 950 and 1400C approximately.
However, it is preferable to use a temperature below about 200 () C, since the end product exhibits a tendency to decompose at elevated temperatures, resulting in loss of yield. The product can be isolated from the mixture resulting from the reaction as the free base or, if desired, as an acid addition salt formed by the introduction of a suitable acid. Hydrochloric acid is preferably used for this purpose, although organic acids or other mineral acids can also be used.
One of the advantages of the process is that it leads to the end products without requiring the preliminary manufacture of an intermediate compound such as a 9-phenoxide.
The products obtained by means of this process exhibit characteristic antibacterial and anti-parasitic properties. They are particularly useful for combating malaria plasmodia, helminths and Endomoeba histolytica, a specific agent for amoebiasis.
From this point of view, the hydrochlorides of the following compounds, derived from acridine 10-oxide, are of particular interest: 6 - chloro - 9 - (4 - diethylamino -1-methyl -butyl-am no) -2-methoxy-acridine; 3,6-Dichloro-9- (3-piperidino-propylamino) -acridine 10-oxide;
the 10-oxides of 3,6-dichloro- and 6-chloro-2-methoxy-9- (5-piperidino-pentylamino) -acridines; 6-chloro-9- (3-isopropylamino-propylamino) -2-methoxy-acridine 10-oxide and 2- [3- (6-chloro-2-methoxy-9-acridinylamino) 10-oxide - propylamino] -ethanol.
The following examples show how the process according to the invention can be carried out. Example 1 6-chloro- </I> 9- (4-diethylamino-1) <I> </I> 10-oxide <I> dihydrochloride -methyl-butylarrino) -2- methoxy-acridine A mixture of 14.8 g of 6,9-dichloro-2-methoxy-acridine 10-oxide, 45 g of phenol and 9 g of 5-diethylamino- 2-Amino-pentane is stirred and heated in a water bath for two hours (protecting it from humidity by means of (fun calcium chloride tube).
The dark red liquid which results from the operation is allowed to cool to 50 and is slowly poured into a mixture of 1 liter of acetone and 10 cc of concentrated hydrochloric acid. A yellow precipitate which separates is collected by filtration, washed thoroughly with cold acetone, dried in vacuo and recrystallized from absolute ethyl alcohol. P.F .:

The starting material, which is 6,9-dichlo, ro-2-methoxy-acridine 10-oxide, can be obtained as follows:
a solution of 278 g of 6,9-dichloro-2-methoxy-acridine in 51 of chloroform is treated at ordinary temperature with a cold solution of 160 g of perbenzoic acid in 31 of chloroform. Allowed to stand at room temperature for 36 hours, then platinum foil is added, the mixture is then concentrated in vacuo until its volume is reduced to 1.5l, then cooled in an ice bath.
The precipitate formed is separated by filtration and washed thoroughly with ether; M.p .: 226-2280C. In this way, the 10-oxide of 6,9-dichloro-2-methoxy-acridine is obtained.

The reaction mixture is stirred and heated at 110 - 11511C for two hours, after which it is cooled and slowly poured into a mixture of 300 cm; acetone and 10 ems of concentrated hydrochloric acid.

Example 3 <I> </I> 10-oxide <I> 6-chloro- </I> 9- (4-deithylamino-1-methyl-butylamino) - 3-methyl-acridine dihydrochloride A mixture of 8 , 7 g of 5-diethylamino-2-amino-pentane, 50 g of phenol and 13.9 g of 10-oxide of 3,9-dichloro-6-methyl-acridine is stirred and heated in a water bath during 2 hours.
The dark red mixture which results from the reaction is allowed to cool to a temperature of 40 ° C. and is slowly poured into a mixture formed of 1 l of acetone and 10 cm; concentrated hydrochloric acid. The yellow precipitate formed is separated by filtration, washed thoroughly with ether, dried under vacuum and recrystallized from its solution in ethanol.
The product, which is 6-chloro-9- (4-diethylamino-1-methyl-butylamino) - 3-methyl-acridine 10-oxide dihydrochloride, has the formula

The 3,9-dichloro-6-methyl-acridine 10-oxide, used as the starting material in the present example, can be prepared as follows:
191 g of 2,4-dichloro-benzoic acid, 134 g of m-toluidine, 138 g of anhydrous potassium carbonate, 5 g of copper powder and 750 cm 3 are mixed; of dry n-amyl alcohol, stirred and boiled under reflux for 5 hours. The mixture is cooled, treated with 70 g of caustic potash in solution in 100 cm; water, and subjected to entrainment by water vapor. The aqueous residue is filtered while hot, slightly acidified with concentrated hydrochloric acid, and cooled.
The precipitated acid is separated by filtration, washed with water and then with petroleum ether (boiling point: 30-600C). The filtered precipitate is then boiled with 300 cc of ethanol, cooled rapidly, filtered and washed with petroleum ether (boiling point: 30-60 ° C.).
Recrystallization of the product from its solution in a mixture of monochlorobenzene and ligroin (boiling point 60 to 800C) (animal black) leads to 4-chloro-N- (m-toluyl-anthranilic acid, in the form of 'needles, almost colorless, melting at 197 1990C.
A mixture formed of 64 g of this acid and 200 cm 3 of phosphorus oxychloride is heated over low heat until the exothermic reaction comes to an end; it is then boiled under reflux for two hours.
Part of the excess phosphorus oxychloride is removed by vacuum, the residue dissolved in dry chloroform and the chloroform solution poured slowly and with stirring into excess ammonia containing ice. Add a second portion. of chloroform and the added chloroform solutions are dried over anhydrous potassium carbonate.
The chloroform is removed by vacuum and the residue is subjected to fractional crystallization from its solution in a mixture of benzene and acetone, which gives 3,9-dichloro-6-methyl-acridine, m.p. 2060C. This product is then subjected to the action of perbenzoic acid,
according to the method described in Example 1 for the preparation of 6,9-dichloro-2-methyl-acridine 10-oxide; M.p .: 172-1740C.
Other 9-aminoalkyllamino-acridine 10-oxides can be prepared in a similar manner from aminoalkyllamines and 10-oxides of dichloro-acridine, as shown in the following table.
