Procédé de préparation d'un. nouvel antibiotique L'invention a pour objet un procédé de prépara tion d'un nouvel antibiotique, qui est caractérisé en ce que l'on cultive le<I>Streptomyces</I> caespitosus <I>NRRL-2564</I> dans un milieu nutritif contenant des sources assimilables de carbone et d'azote et des sels inorganiques, en aérobic, jusqu'à ce qu'une quantité substantielle d'antibiotique soit produite dans le mi lieu de culture, et en ce que l'on isole l'antibiotique dudit milieu de culture.
Comme sources, d'azote pour la culture, on peut employer la peptone, l'extrait de viande, la liqueur de corn-steep, la farine de soja, la levure, l'urée, le sulfate d'ammonium, le nitrate d'ammonium et autres corps similaires,; comme sources de carbone, on peut employer le glucose, l'amidon, le glycérol, l'huile de soja, le maltose, la dextrine, etc. ;
comme sels inor ganiques, on peut employer le chlorure de sodium, le chlorure de potassium, le carbonate de calcium, le phosphate de potassium, le sulfate ferreux, le sul fate de magnésium, le sulfate de zinc et les corps similaires. Tous ces corps peuvent être employés dans un milieu liquide, par exemple aqueux, ayant un pH d'environ 7,0, suivant toutes combinaisons appropriées.
Cependant, le glucose, l'amidon et le glycérol sont préférés comme sources de carbone ; la levure, l'extrait de viande, la farine de soja, la liqueur de corn-steep et le sulfate d'ammonium sont préférés comme sources d'azote ; et le chlorure de sodium et le carbonate de calcium, sont préférés comme sels inorganiques.
Un milieu de culture liquide ainsi pré paré est inoculé avec une suspension de spores de <I>Streptomyces</I> caespitosus (NRRL No 2564). La cul- turc en aérobie submergée est préférée. En général, lorsqu'on effectue la fermentation à 26-320C pen dant 2 à 6 jours, on obtient la formation pratique ment maximum d'antibiotique.
Le mycélium est alors rapidement séparé par fil trage du liquide nutritif résultant, après que celui-ci a été refroidi à une température de, par exemple, 0 à 5()C. Le nouvel antibiotique est ensuite isolé à par tir dudit liquide (bouillon de culture), comme décrit ci-après.
Selon une première manière de procéder, 1e nou vel antibiotique peut être adsorbé sur le charbon actif à partir du bouillon de culture ayant un pH compris entre 6 et 9, ou un pH un peu supérieur ou un peu inférieur. Ainsi, par exemple, si l'anti biotique est adsorbé sur 1 'o/o de charbon actif pen dant 30 minutes, le pH étant compris dans ledit intervalle, et que l'on sépare par filtrage le char bon actif, le filtrat ne contient pratiquement pas d'antibiotique et n'a pas d'activité antibactérienne.
Pour éluer le nouvel antibiotique à partir du charbon actif sur lequel il est adsorbé, on emploie un solvant approprié tel que l'acétone, le cyclo- hexanone, la méthyl-isobutyl-cétone, le butanol ou le chloroforme, ou un mélange de deux ou plusieurs de ces corps. L'acétone est celui qui convient le mieux.
Lorsqu'on emploie de l'acétone pour l'élution, l'eau contenue dans le gâteau de charbon actif entre dans l'éluat, de sorte que ce dernier contient une quantité considérable d'eau.
Cette teneur en eau peut être évitée en lavant le charbon actif avec du mé- thanol pur avant l'élution. La quantité de nouvel antibiotique qui est éluée sans, profit par le métha nol à partir du charbon actif est seulement de 5 0/0 environ. Après le lavage avec le méthanol, le char bon actif est ensuite élué avec de l'acétone, et lors que l'extrait est concentré sous pression réduite, on obtient un mélange complexe concentré contenant l'antibiotique.
Le nouvel antibiotique peut aussi être extrait du filtrat du bouillon de culture par des solvants organi- ques non miscibles à l'eau. Des solvants non misci bles à l'eau pouvant être utilisés dans ce but sont, par exemple, les alcools tels que le butanol, etc., les esters tels que l'acétate de butyle, etc., les cétones telles que la méthyl-isobutyl-cétone, le cyclohexanone, etc., le chloroforme et les corps similaires.
Le taux d'extraction est le plus favorable et pratiquement in variable si le pH est maintenu dans l'intervalle de 6 à 9. Le taux d'extraction peut être augmenté par l'utilisation simultanée d'agents salants, tels que le sel commun (NaCl), le sulfate d'ammonium ou corps similaires, dans, la couche aqueuse.
En ajoutant di verses quantités (en poids) de sel commun, l'extrac tion par le chloroforme à un pH de 7,0 a lieu avec les taux d'extraction suivants

Afin d'isoler sélectivement le nouvel antibiotique désiré à partir du mélange complexe susmentionné contenant cet antibiotique, le mélange peut être sou mis à la chromatographie sur une colonne d'alumine, ou bien l'on peut employer un. procédé de distribu tion à contre-courant.
Le concentré du mélange complexe obtenu par l'adsorption sur charbon actif ou par extraction par solvant organique est dissous dans le chloroforme. Lorsque le chloroforme est employé pour l'extrac tion, l'extrait est concentré à un degré approprié et l'on emploie un tel concentré. On fait ensuite passer la solution au chloroforme à travers une tour garnie d'alumine active. Le nouvel antibiotique très adsor- bable est adsorbé dans. la partie supérieure de la tour, pendant que le chloroforme s'écoule vers le bas.
On fait ensuite passer le chloroforme qui contient 1 % de méthanol à travers la tour pendant un cer- tain temps, de sorte que le nouvel antibiotique désiré est séparé en une couche supérieure ayant une cou leur pourpre distinctive.
En continuant l'élution avec le solvant de développement, suivie par l'élution avec le chloroforme qui contient 2 à 2,9 % de méthanol, on élimine les matières indésirables.
La couche supé rieure susmentionnée contenant le nouvel antibioti que est ensuite séparée avec le chloroforme qui con- tient 3 à 6 % de méthanol. Le méthanol contenu dans le solvant de déve loppement peut être remplacé par de l'éthanol ou du butanol. L'acétone peut aussi être employée, bien que le rendement puisse être un peu réduit.
Selon une variante, la couche d'adsorption du nouvel antibiotique peut être séparée en même temps que l'alumine sur laquelle elle est adsorbée, et peut être éluée par le méthanol. Ensuite, l'éluat peut être concentré sous pression réduite pour obtenir les cris taux désirés du nouvel antibiotique.
Le nouvel antibiotique obtenu comme ci-dessus est dissous dans le chloroforme et la solution de chloroforme est concentrée jusqu'à un certain degré de dessiccation. Lorsqu'une petite quantité d'acétone y est ajoutée, on obtient des cristaux pourpre foncé.
En isolant le nouvel antibiotique désiré par dis tribution à contre-courant, le chloroforme peut, par exemple, être utilisé comme phase fixe, tandis que la phase mobile (qui contient le filtrat du bouillon de culture) peut être constituée par un tampon de phosphate aqueux de pH 6,0 contenant 10 % de sel commun (NaCI). Si la séparation est effectuée dans une machine à 30 tubes,
le nouvel antibiotique désiré sera récolté dans les tubes de rang 20 à 25. Le con tenu de ces tubes est concentré et fournit la nouvelle substance désirée.
Le nouvel antibiotique obtenu par l'un quelcon que des moyens décrits ci-dessus se présente sous la forme de cristaux en aiguilles de couleur pourpre foncé et ne fondant pas,, même à 3600C. L'analyse quantitative de cet antibiotique donne: C 53,84 % ;
H 5,14 % et N 15,49 0/0.
Sur le dessin annexé la fig. 1 est un spectre d'absorption infrarouge du nouvel antibiotique dans la pâte de Nujol , Nujol étant une marque d'huile de paraffine lourde, et la fi-. 2 est un spectre d'absorption ultraviolet du nouvel antibiotique dans l'eau distillée.
Comme clairement montré sur la fig. 2, on ob serve des pointes sur le spectre d'absorption ultra violet à 216 m#t ; El @# = 742, et à 360 mi. ; Ei #m = 742. Comme clairement montré sur la fig. 1, les pointes du spectre d'absorption infrarouge sont ob servées aux fréquences suivantes (cm-') 2930, 2857, 1700, 1640, 1596, 1545, 1448,<B>1</B>370, 1333, 1230,<B>1</B>170, 1155, 1125, 1060, 1025, 1002, 969, 952, 919, 895, 854, 824, 780, 701.
Le nouvel antibiotique présente une faible solu bilité en général, mais est relativement facilement soluble dans l'eau, le méthanol, l'éthanol, le chloro forme, etc. Il est légèrement soluble dans le tétra chlorure de carbone, le benzène, l'éther éthylique, etc., et insoluble dans l'éther de pétrole.
Il donne des réactions positives dans les réactions suivantes Réaction de Fehling, réaction d'Ehrlich, réaction de Liebermann, réaction du biuret, réaction avec l'hydroxylamine, réaction, avec le chlorure ferrique, et réaction avec l'acide nitreux.
Les réactions suivantes furent négatives Réaction de Benedict, réaction de Tollens, ré action de Million, réaction de Raymond, réaction de Legal, réaction avec le sulfite de fuchsine, réaction avec la ninhydrine, réaction avec le xanthogène, et réaction avec le cyanure de. potassium.
La solution aqueuse du nouvel antibiotique est stable à un pH neutre et l'on n'observe pratiquement pas d'abaissement de son pouvoir même après un chauffage d'une heure à 800C. Cependant, si le pH est inférieur à 5 ou supérieur à 11, l'antibiotique est rapidement détruit. En outre, il est beaucoup moins stable dans une solution d'un. solvant organique que dans une solution aqueuse ; en, particulier lorsqu'il est irradié avec de la lumière visible dans un solvant organique, il est détruit très rapidement. Par exemple, si on laisse reposer sa solution dans le chloroforme à la température ordinaire de jour dans une cham bre, presque tout son pouvoir est perdu après six heures, mais dans un endroit sombre, son pouvoir n'est pratiquement pas réduit.
Les cristaux du nouvel antibiotique sont extrê mement stables à la température ordinaire (20 à 30 C).

<SEP> = <SEP> point <SEP> de <SEP> fusion L'antibiotique obtenu conformément à la pré sente invention est utile comme antiseptique<I>in vitro,</I> et comme agent thérapeutique topique et interne pour combattre les bactéries pathogènes, par exemple dans le cas de staphylodermatite, pneumonie bactérienne, leptospirose, rickettsiose et salmonellose, etc.
Le nouvel antibiotique est aussi efficace en provoquant la palliation et des rémissions cliniques plus ou moins prolongées dans le cas, d'états morbides tels que certaines leucémies, par exemple la maladie de Hodgkm. Il est aussi efficace pour la thérapie pallia tive combinée dans, de tels cas.
La nouvelle substance a une tolérance mar quée. La toxicité aiguë de la substance est d'environ LDso = 5 mg/kg chez la souris.
Les exemples suivants se rapportent à des for mes d'exécution préférées de l'invention et à l'iso lement et à la purification de l'antibiotique. Dans ces exemples, comme dans le texte qui précède, les pourcentages sont en poids. Les essais pour déter miner le pouvoir de l'antibiotique sont basés sur le procédé de dilution, en employant le Bacillus subtilis (PCI 219) comme organisme-test.
<I>Exemple 1</I> Un bouillon formé de 2 % de glucose, 1,0 % de farine de soja, 0,3 % d'extrait de viande, 0,
5 % de chlorure de sodium, 0,5 a/o de levure, 0,3 % de carbonate de calcium, et le reste d'eau, et ayant un pH d'environ 7,2,
est stérilisé par de la vapeur à 1200C. Le bouillon stérilisé est ensuite inoculé avec une suspension de spores de<I>Streptomyces</I> caespitosus (NRRL No 2564), et la culture est effectuée en aé rant pendant 72 heures à 280C. A la fin de cette période, le bouillon est filtré pour éliminer les my céliums.
10 kg de charbon actif sont ajoutés à 10001 du filtrat de bouillon 3000 u/ml ainsi obtenu et, après avoir agité pendant 30 minutes, le produit est filtré avec addition de 5 kg du produit marque Dicalite (produit favorisant le filtrage).
Le charbon actif ré colté est déshydraté par lavage avec 301 de métha nol, puis est élué trois fois de suite avec 301 d'acé tone pure. L'éluat est concentré sous. pression réduite, et l'on obtient 45 g d'un résidu concentré de couleur pourpre. Son pouvoir est de 9000 u/mg. <I>Exemple 2</I> 10 kg de charbon actif sont ajoutés à 10001 de filtrat de bouillon 3500 u/ml (obtenu comme dans l'exemple 1) et, après 30 minutes d'agitation, le pro duit est filtré avec addition de 5 kg de Dicalite .
Le charbon actif récolté est immédiatement élué trois fois de suite avec 301 d'acétone pure. L'éluat est concentré sous pression réduite jusqu'à ce qu'il ne reste pratiquement pas. d'acétone, et l'on obtient 81 d'une solution aqueuse. Environ 3 kg de sel commun y sont ajoutés et dissous dans la solution,, et ensuite on procède à une extraction deux fois de suite avec 81 de chloroforme. Lorsque l'extrait a été concentré, on obtient 20 g de résidu concentré de couleur pour pre. Son pouvoir est de 15 000 u/mg.
<I>Exemple 3:</I> Le résidu concentré obtenu dans l'exemple 1 est dissous dans 31 de chloroforme et des cristaux blancs insolubles sont séparés par filtrage. On fait passer le filtrat à travers une tour garnie de 1500 g d'alumine active (diamètre 7 cm, hauteur 40 cm).
Après lavage avec 500 ml de chloroforme, on fait passer le chloroforme qui contient 1 % de méthanol à travers la tour pour l'élution. Pendant l'élution, la teneur en méthanol augmente progressivement de 1 à 6 0/0. Le volume total du chloroforme traversant l'alumine est d'environ 8 à 101.
La matière éluée avec le chloroforme contenant 3 à 6'% de méthanol est concentrée jusqu'à environ 100 ml sous pression réduite et filtrée, puis est concentrée encore davan tage jusqu'à dessiccation presque complète et l'on ajoute 5 à 10 ml d'acétone, après quoi l'on obtient 1,
5 g de cristaux pourpres. Le pouvoir du produit obtenu est de 100 000 u/mg. <I>Exemple 4:</I> Le résidu concentré obtenu dans l'exemple 2 est dissous dans 21 de chloroforme et les matières inso lubles sont séparées par filtrage. On fait passer le filtrat à travers, une tour garnie de 1 kg d'alumine active (diamètre 7 cm, hauteur 26 cm).
On fait pas- ser 41 de chloroforme qui contient 0,5 % de métha- nol à travers la tour et, après le lavage et le déve loppement, la partie supérieure de l'alumine portant la couche d'adsorption pourpre de l'antibiotique dé siré est prélevée et sortie de la tour.

As sources of nitrogen for cultivation, peptone, meat extract, corn steep liquor, soy flour, yeast, urea, ammonium sulfate, nitrate can be employed. ammonium and other similar substances; as carbon sources, glucose, starch, glycerol, soybean oil, maltose, dextrin, etc. can be used. ;
as inorganic salts, sodium chloride, potassium chloride, calcium carbonate, potassium phosphate, ferrous sulfate, magnesium sulfate, zinc sulfate and the like can be employed. All of these bodies can be employed in a liquid medium, for example aqueous, having a pH of about 7.0, in any suitable combination.
However, glucose, starch and glycerol are preferred as carbon sources; yeast, meat extract, soybean meal, corn steep liquor and ammonium sulfate are preferred as nitrogen sources; and sodium chloride and calcium carbonate, are preferred as inorganic salts.
A liquid culture medium thus prepared is inoculated with a suspension of <I> Streptomyces </I> caespitosus spores (NRRL No 2564). Submerged aerobic culture is preferred. In general, when fermentation is carried out at 26-320C for 2-6 days, the practically maximum antibiotic formation is obtained.
The mycelium is then rapidly separated by filtration from the resulting nutrient liquid, after the latter has been cooled to a temperature of, for example, 0 to 5 () C. The new antibiotic is then isolated by shooting said liquid (culture broth), as described below.
According to a first way of proceeding, the new antibiotic can be adsorbed onto the activated carbon from the culture broth having a pH between 6 and 9, or a slightly higher or a little lower pH. Thus, for example, if the anti-biotic is adsorbed on 1% of activated carbon for 30 minutes, the pH being within the said range, and the activated charcoal is filtered off, the filtrate does not contains practically no antibiotic and has no antibacterial activity.
To elute the new antibiotic from the activated charcoal on which it is adsorbed, an appropriate solvent such as acetone, cyclohexanone, methyl-isobutyl-ketone, butanol or chloroform, or a mixture of two or more of these bodies. Acetone is the best fit.
When acetone is used for the elution, the water contained in the activated carbon cake enters the eluate so that the latter contains a considerable amount of water.
This water content can be avoided by washing the activated carbon with pure methanol before elution. The amount of new antibiotic which is eluted without benefit by methanol from the activated charcoal is only about 5%. After washing with methanol, the active char is then eluted with acetone, and when the extract is concentrated under reduced pressure, a concentrated complex mixture is obtained containing the antibiotic.
The novel antibiotic can also be extracted from the filtrate of the culture broth by organic solvents immiscible with water. Water-immiscible solvents which can be used for this purpose are, for example, alcohols such as butanol, etc., esters such as butyl acetate, etc., ketones such as methyl- isobutyl ketone, cyclohexanone, etc., chloroform and the like.
The extraction rate is the most favorable and practically in variable if the pH is maintained in the range of 6 to 9. The extraction rate can be increased by the simultaneous use of salting agents, such as common salt. (NaCl), ammonium sulfate or the like in the aqueous layer.

The chloroform solution is then passed through a tower packed with active alumina. The new highly adsorbable antibiotic is adsorbed into. the top of the tower, while the chloroform is flowing down.
The chloroform which contains 1% methanol is then passed through the tower for a period of time so that the desired new antibiotic is separated into an upper layer having a distinctive purple color.
By continuing the elution with the developing solvent, followed by the elution with chloroform which contains 2 to 2.9% methanol, unwanted material is removed.
The above-mentioned top layer containing the new antibiotic is then separated with chloroform which contains 3 to 6% methanol. The methanol contained in the development solvent can be replaced by ethanol or butanol. Acetone can also be used, although the yield may be somewhat reduced.
According to a variant, the adsorption layer of the new antibiotic can be separated at the same time as the alumina on which it is adsorbed, and can be eluted with methanol. Then, the eluate can be concentrated under reduced pressure to obtain the desired levels of the new antibiotic.
The new antibiotic obtained as above is dissolved in chloroform and the chloroform solution is concentrated to a certain degree of desiccation. When a small amount of acetone is added to it, dark purple crystals are obtained.
By isolating the desired new antibiotic by countercurrent distribution, the chloroform can, for example, be used as a fixed phase, while the mobile phase (which contains the filtrate from the culture broth) can be constituted by a phosphate buffer. aqueous pH 6.0 containing 10% common salt (NaCl). <SEP> = <SEP> melting point <SEP> The antibiotic obtained in accordance with the present invention is useful as an antiseptic <I> in vitro, </I> and as a topical and internal therapeutic agent for combating pathogenic bacteria, for example in the case of staphylodermatitis, bacterial pneumonia, leptospirosis, rickettsiosis and salmonellosis, etc.
The new antibiotic is also effective in causing palliation and more or less prolonged clinical remissions in the case of morbid conditions such as certain leukaemias, for example Hodgkm's disease. It is also effective for combined palliative therapy in such cases.
The new substance has marked tolerance. The acute toxicity of the substance is approximately LD 50 = 5 mg / kg in mice.
The following examples relate to preferred embodiments of the invention and to the isolation and purification of the antibiotic. In these examples, as in the preceding text, the percentages are by weight. The tests to determine the potency of the antibiotic are based on the dilution method, using Bacillus subtilis (PCI 219) as the test organism.
<I> Example 1 </I> A broth formed from 2% glucose, 1.0% soybean meal, 0.3% meat extract, 0,
5% sodium chloride, 0.5 a / o yeast, 0.3% calcium carbonate, and the rest of water, and having a pH of approximately 7.2,
is sterilized by steam at 1200C. The sterilized broth is then inoculated with a suspension of <I> Streptomyces </I> caespitosus spores (NRRL No 2564), and the culture is carried out in air for 72 hours at 280C. At the end of this period, the broth is filtered to remove the mycelium.
10 kg of activated charcoal are added to 10001 of the 3000 u / ml broth filtrate thus obtained and, after stirring for 30 minutes, the product is filtered with the addition of 5 kg of the Dicalite brand product (product which promotes filtering).
The collected activated carbon is dehydrated by washing with 301 of methanol, then is eluted three times in a row with 301 of pure acetone. The eluate is concentrated under. reduced pressure, and 45 g of a concentrated residue of purple color are obtained. Its power is 9000 u / mg. <I> Example 2 </I> 10 kg of activated charcoal are added to 10001 of 3500 u / ml broth filtrate (obtained as in example 1) and, after 30 minutes of stirring, the product is filtered with addition of 5 kg of Dicalite.
The collected activated carbon is immediately eluted three times in a row with 301 of pure acetone. The eluate is concentrated under reduced pressure until hardly any remains. of acetone, and 81 of an aqueous solution is obtained. About 3 kg of common salt is added thereto and dissolved in the solution, and then extraction is carried out twice in succession with 81 chloroform. When the extract has been concentrated, 20 g of concentrated pre-colored residue are obtained. Its potency is 15,000 u / mg.
<I> Example 3: </I> The concentrated residue obtained in Example 1 is dissolved in 31 chloroform and insoluble white crystals are filtered off. The filtrate is passed through a tower packed with 1500 g of active alumina (diameter 7 cm, height 40 cm).
After washing with 500 ml of chloroform, the chloroform which contains 1% methanol is passed through the tower for elution. During the elution, the methanol content gradually increases from 1 to 6%. The total volume of chloroform passing through the alumina is about 8-101.
The material eluted with chloroform containing 3-6% methanol is concentrated to about 100 ml under reduced pressure and filtered, then is further concentrated until almost complete drying and 5-10 ml of water is added. 'acetone, after which we obtain 1,
5 g of purple crystals. The potency of the product obtained is 100,000 u / mg. <I> Example 4: </I> The concentrated residue obtained in Example 2 is dissolved in 21 chloroform and the insoluble matters are separated by filtering. The filtrate is passed through a tower packed with 1 kg of active alumina (diameter 7 cm, height 26 cm).

<I> Streptomyces </I> caespitosus is a currently known organism, which was first isolated in 1954 by Toju Hata et al, from a soil sample collected in Yoochima- chi, Shibuya-ku , Tokyo, Japan. Live cultures of this organism have been deposited in the Permanent Microorganism Collection of the Northern Regional Research Laboratory in Peoria, Illinois, where they have been assigned the following identifying designation: NRRL No. 2564.
