En exécutant la première partie du procédé, il est avantageux, quoique non essentiel, d'em ployer un solvant inerte tel que l'eau, un alcool aliphatique inférieur ou un mélange de tels al cools, l'éther, le benzène, le toluène ou sem- blables, pour faciliter la réaction de l'acide ou de l'anhydride succiniques avec l'ammoniac ou la méthylamine. Il n'est pas nécessaire d'em ployer les réactifs en proportions particulières exactes ;

Process for preparation of snccinimides The invention relates to a process for the preparation of succinimides having a chemotherapeutic activity, namely the o-hydroxyphenylsuccinimides of general formula

in which R is hydrogen or a methyl radical.
According to the invention, the above compounds are prepared by reacting a- (o-hydroxyphenyl) -succinic acid or a- (o-hydroxyphenil) -succinic anhydride with at least one chemical equivalent of ammonia, usually in the form of ammonium hydroxide, or methylamine, and by heating the intermediate products thus formed to a temperature between 1001 and 3500 C.
In carrying out the first part of the process, it is advantageous, although not essential, to employ an inert solvent such as water, a lower aliphatic alcohol or a mixture of such alcohols, ether, benzene, toluene. or the like, to facilitate the reaction of succinic acid or anhydride with ammonia or methylamine. It is not necessary to employ the reagents in exact particular proportions;
however, from the point of view of economy, it is advisable to employ several equivalents of ammonia or methylamine per equivalent of a - (o -hydroxyphenyl) - succinic acid or anhydride. The intermediate products are different depending on which of the two above-mentioned raw materials is used, and also on the amount of ammonia or methylamine used. When using a- (o-hydroxyphenyl) - succinic anhydride with 1 equivalent of ammonia or methylamine,
the intermediate product is the semi-amide, tan say that if two or more equivalents of ammonia or methylamine are used, the intermediate product is the ammonium or methylamine salt of the semi-amide of the acid a - (o-hydroxyphenyl) -succinic. In the case where a- (o-hydroxyphenyl) -suckinic acid is used as the starting material, the intermediate product is the mono- or di-ammonium or -methylamine salt of a- (o -hydroxy-phenyl)
-succinic. These intermediates can be converted to the desired succinimides by heating them to a temperature between 100 and 3500 C. The preferred temperature for this conversion is in the region of 1900 to 2100 C, at which temperature the reaction is complete. after about 1 hour.
The products obtained by the process according to the invention have valuable therapeutic properties, and are particularly useful for the treatment of the petit mal type of epi lepsy.
The invention is illustrated by the following examples Example 1 Gradually. 10 g of a- (o-hydroxyphenyl) -succinic acid is added to 10 cc of concentrated ammonium hydroxide and distilled off. tion water and excess ammonia. The temperature of the residue is raised and maintained at 2000 ° C. until there is no further elimination of ammonia. The residue is dissolved in dry ether; the solution is then decolorized with animal charcoal and the ether is distilled off. The solid product is recrystallized from water, and the expensive a- (o-hydroxyphenyl) -succinimide is obtained; P. F. 207-209 C.
Yield: 44 0 / ro. Example 2 10 g of a- (o-hydroxyphenyl) -succinic acid is gradually added to 20 g of aqueous methylamine and the water and excess methylamine are distilled off. The temperature of the residue is raised and maintained at 200 ° C. until no more evolution of methylamine. The residue is dissolved in dry ether, the solution is decolorized with ani mal charcoal and the ether is distilled off.
The residue is then distilled in vacuo, and the desired N-rnethyl- a- (o-hydroxyphenyl) -succinimide is obtained; Mp. 136-138 C. Yield: 57%.
