Process for the preparation of 2-methyl-2-heptenone- (6) The present invention relates to a process for the preparation of 2-methyl-2-heptenone- (6).
This process is characterized in that acetone is reacted with a ganometallic complex obtained by the action of an active metal on an acetal of a 5-halo-pentanone-2, to form a carbinol tertiary comprising the group
EMI0001.0007
in which the terminal oxygen atoms are linked to a hydrocarbon residue so as to form a simple or cyclic acetal and in that said carbinol is converted, by dehydration and hydrolysis of the acetal group, into 2-methyl -2-heptenone- (6).
The 2-methyl-2-heptenone- (6) thus obtained, of formula
EMI0001.0013
is a known substance serving as an intermediate product for the synthesis of geraniol and farnesol which are widely used in perfumery. The acetals which can be used can be prepared by reacting a 5-halogeno-2-pentanone with an alkyl-orthoformate in the presence of an alcohol, or by reacting a 5-halogeno-2-pentanone with a 1, 2- or 1,3-glycol.
Such simple or cyclic acetals react with their halogen atom in position 5 with an active metal to form an organometallic complex.
This behavior of the acetals of 5-halogeno-2-pentanones or of their organo-metallic derivatives is abnormal if one refers to the work of Arens and Van Dorp which showed that the halogenated cyclic acetal
EMI0001.0027
does not form an organo-halo-metallic compound with an active metal such as magnesium or other comparable metals (Rec. trav. chem. 65, 729 (1946)), and in the works of Willimann and Schinz,
Helv. Chim. Acta, 32, 2158 (1949) which indicate for acetal cyclic
EMI0001.0037
and its organometallic derivatives from other reaction processes.
In a preferred embodiment of the invention, an active metal, such as Li, Mg, Zn, Cd, Hg, is reacted with an alkylenocyclic (eg ethylene-cyclic) acetal of a 5-halogeno. 2-pentanone, such as, for example, 2- (gamma-halopropyl) -2-methyl-1,3-dioxolane, to form an organohalometallic compound of the formula
EMI0002.0009
where X is a halogen atom, M is the active metal and R "is an alkylene group (ethylene, propylene, butylene ...).
This organohalometallic compound then reacts with acetone to form tertiary alcohol (carbinol).
The organometallic compound can also be prepared from a simple acetal of a 5-halo-2-pentanone, of the type
EMI0002.0015
where X can be any halogen (chlorine and bromine being however preferable) and where the alkyl groups are advantageously lower alkyl radicals, comprising from one to four carbon atoms.
The preferred tertiary carbinol formed during the reaction is that of formula
EMI0002.0017
which is preferably obtained from 5-chloro-pentanone-2. The reaction between the organometallic complex and acetone results in a significantly longer chain carbinol
EMI0002.0021
This reaction is easily carried out by adding a solution of acetone to a solution of the organometallic compound in a solvent such as anhydrous ether. An exothermic reaction immediately takes place and continues to completion with good yield.
It is then sufficient to dilute the mixture obtained with water to decompose the metal complex and thus obtain the desired tertiary carbinol.
This tertiary alcohol is then converted into the ketone which contains one more isopentane unit than the starting carbonyl compound (acetone). In this transformation, the alcohol is subjected to dehydration and hydrolysis to form 2-methyl-2-heptenone-6 (unsaturated ketone), which reactions can be carried out in any order. The dehydration of tertiary alcohol is easily carried out according to known dehydration methods, for example by heating it in the presence of an acidic dehydrating agent, such as an acid or an acid salt or other agent. dehydration analog.
Conventional dehydrating agents suitable for the operation include mineral acids, organic acids such as oxalic acid and paratoluenesulfonic acid, anhydrides, such as acetic anhydride, and acidic compounds, such as sodium chloride. zinc, phosphorus trichloride, oxalyl chloride, phosphorus oxychloride, & c.
The hydrolysis of the dehydrated acetal to form the corresponding 2-methyl-heptenone is carried out simply by an acidic aqueous solution or by other well-known methods of hydrolysis, the aqueous solutions of dilute mineral acids being particularly re orderable.
In the process according to the invention, therefore, acetone is first reacted with an organometallic compound according to the equation above. Preferably, an organometallic complex is used in which the halogen X is chlorine and the metal M is magnesium. The tertiary carbinol obtained is then converted by dehydration into the corresponding unsaturated cyclic acetal, by heating with an acid reagent, such as phosphorus oxychloride in pyridine, or any other dehydrating agent, according to the equation
EMI0003.0004
This unsaturated cyclic acetal is then hydrolyzed with dilute acid and provides the corresponding unsaturated ketone (2-methyl-2-heptenone-6)
EMI0003.0007
In this way, the acetone chain is extended by an isopentene group. The methylheptenone thus obtained can easily be converted into geraniol by reaction with a halo-acetate, which gives an ester of geranic acid,
which ester is then subjected to reduction to geraniol. 2-Methyl-2-heptenone-6 can also be used for the synthesis of geranyl-acetone and farnesol.
The process according to the invention is illustrated by the following example A, an ethereal solution of the ganometallic gold complex of formula
EMI0003.0020
prepared from 7.8 g Mg, 49.5 g of 2- (y-chloropropyl) -2-methyl-1,3-dioxolane, 1 cm3 CH; iJ and 250 cm3 of anhydrous ether, a solution is added of 17.4 g of acetone in 100 cm 3 of anhydrous ether until no more heat was given off.
After decomposition of the reaction mixture with ice and ammonium chloride, the ether was removed by distillation and the residue was heated to 100 () C under a pressure of 1 mm Hg, to remove a volatile side product. The residue consisting of the desired tertiary carbinol weighs 26.2 g.
Its infrared spectrum has a band of 2.9, u. which is characteristic for the carbinol group
EMI0003.0031
18.4 g of phosphorus oxychloride are then added to 55 cm3 of dry pyridine in a closed container fitted with a stirrer, a tap-type funnel and a reflux condenser fitted with a tube filled with chloride. of cal cium. 18.8 g of said tertiary carbinol dissolved in 25 cm3 of anhydrous benzene are added to this mixture dropwise.
After adding the total amount of the carbinol, the mixture is stirred for one hour, then heated to reflux while continuing to stir for one hour. After cooling, poured onto 400 g of ice containing 60 cm3 of concentrated hydrochloric acid, and extracted with ether. The ethereal solution was washed with dilute hydrochloric acid, then with water, then with NaHCO3 solution and water, and dried. After evaporation of the ether, 12.9 g of dehydrated product (unsaturated cyclic acetal) are obtained. The infrared spectrum of this product does not show a band at 2.9 P.
Then stirred for 2 hours at a temperature of 500 ° C., a mixture of 10 g of this unsaturated acetal, 60 cm3 of methanol, 20 cm3 of water and 2 cm3 of concentrated sulfuric acid. Then 600 cm3 of water are added and the mixture is subjected to water vapor distillation. The distillate is saturated with sodium chloride and extracted with ether. The ether is removed by distillation through a column of Vigreux and the liquid residue is distilled which has a boiling point of 170-173. The yield is 6.1 g.
The product thus obtained is 2-methyl-2-heptenone-6, the semicarbazone of which melts at 135-137o.
