Procédé de préparation du 3a,21-diacétoxy-11,20-dicéto-12a,15,21-tribromo-d"-prégnène. La présente invention se rapporte à un procédé de préparation du 3 a, 21-diacétoxy- 11,20-dicéto-12 a,15,21-tribromo-d 16-prégnène (II), avec de bons rendements, à partir du 3 a, 21-diacétoxy-1.1,20-dicéto-12 a, 21-dibr omo- prégnane (I).
Ce procédé est caractérisé en ce qu'on traite du 3 a, 21-diacétoxy-11,20-di- c6to-12a,21-dibromo-prégnane (I) au moyen de brome au sein d'un solvant polaire, dans des conditions pratiquement anhydres et en présence d'acide bromhydrique.

Comme solvant polaire, on peut employer un acide aliphatique inférieur, de préférence de l'acide acétique, ou du dioxane.
Le composé obtenu est nouveau;' il peut être utilisé pour préparer la cortisone, ainsi que l'illustre ce qui suit: Les atomes de brome des positions 15 et 21 du composé II peuvent être facilement enlevés, par exemple par traitement au moyen d'acide iodhydrique, avec formation de 3 a, 21- diacétoxy-11,20-dicéto-12a-bromo-dis-prégnène, qui peut être transformé en cortisone par di verses voies, comportant l'époxylation de la double liaison en position 16-17, la transfor mation du composé 16,17-époxy en composé 16-bromo-17-hydroxy, le bromage en position 4,
suivi du déshydrobromage pour introduire une double liaison en position 4-5 et l'élimi nation des atomes de brome des positions 12 et 16. Ces opérations peuvent être effectuées dans tout ordre approprié et de diverses ma nières.
Exemple: A 1200 cms d'acide acétique, on ajoute 20 cm3 de bromure d'acétyle et on élimine sous pression réduite la majeure partie de l'acide bromhydrique. Cette élimination n'est toutefois pas totale, du fait de la réaction qui se poursuit lentement entre les deux réactifs, de sorte que le mélange réactionnel contient, après avoir été traité sous pression réduite, une quantité faible, mais suffisante, d'acide bromhydrique pour catalyser le bromage. On constate toutefois que la réaction a lieu phis facilement lorsque la. plus grande partie de l'acide bromhydriqiïe a été éliminée.
On ajoute ensuite 116 g de 3a,21-diacétoxy- 11,20-dicéto-12a,21-dibromo-prégnane (I) et 10,2 cms de brome. Quand tout le brome a été utilisé, on en rajoute 10,2 cm3, et, après que cette nouvelle quantité a été utilisée, on en rajoute encore 10,2 cms et on aban donne an repos pendant 7 jours. On sé pare par filtration les cristaux de composé II, on les lave au moyen d'acide acétique et d'éther sec. On concentre la liqueur mère à un faible volume, on ajoute du benzène et on l'élimine sous pression réduite.
L'addition d'éther donne une nouvelle quantité du com posé (II), le 3a,21-diacétoxy-11,20-dicéto- 12a,15,21-tribromo-drs-prégnène, qui pré sente les caractéristiques suivantes: Point de fusion: 231-232 C; aD =-62 (CHC13); s 235 mu = 8700 (éther).
La nouvelle substance obtenue par le pro cédé selon l'invention se décompose graduelle ment, même en absence d'air. La nature des produits de décomposition n'est pas eonnue.

116 g of 3a, 21-diacetoxy-11,20-diketo-12a, 21-dibromo-pregnan (I) and 10.2 cms of bromine are then added. When all the bromine has been used, 10.2 cm3 is added, and after this new quantity has been used, another 10.2 cm3 is added and left to stand for 7 days. The crystals of compound II are separated by filtration, washed with acetic acid and dry ether. The mother liquor is concentrated to a small volume, benzene is added and removed under reduced pressure.
The addition of ether gives a new amount of the compound (II), 3a, 21-diacetoxy-11,20-diketo-12a, 15,21-tribromo-drs-pregnene, which has the following characteristics: Point melting point: 231-232 C; aD = -62 (CHCl3); s 235 mu = 8700 (ether).
The new substance obtained by the process according to the invention decomposes gradually, even in the absence of air. The nature of the decomposition products is not known.
