Procédé de préparation de nouveaux esters d'hormones corticosurrénales Certaines hormones corticosurrénales, no tamment la cortisone (11- déhydro -17a - hy - droxy-corticostérone), l'hydrocortisone (17a- hydroxy-corticostérone) et la 11-désoxy-corti- costérone, ont été commercialisées jusqu'ici principalement sous forme de 21-acétates dont l'action n'est pas très prolongée.

Le prix élevé de ces substances et la néces sité de les administrer fréquemment, si elles sont employées sous leur forme actuelle, rendent désirable une forme dont l'action soit plus prolongée. On a découvert que les hexahydrobenzoates en 21, exercent cette action.
Les hexahydrobenzoates permettent d'es pacer les injections et d'éviter la surcharge hormonale de l'organisme en réalisant une absorption lente et continue de l'hormone chez l'homme ou en médecine vétérinaire.
Ce résultat est tout à fait inattendu, car les 21-benzoates des mêmes hormones ne présentent pas cette action prolongée et rien ne pouvait laisser prévoir que la saturation des doubles liaisons cycliques de l'agent d'esté rification permettrait d'obtenir des esters à action très prolongée. Les nouveaux esters peuvent être employés en solution dans les huiles végétales neutres additionnées ou non d'un glycol et d'un sel organique tel que le 2-éthylcaproate d'aluminium ou en solution dans d'autres solvants injectables, ou encore sous forme de suspensions microcristallines aqueuses, ou d'émulsions dans l'eau de solu tions dans un tiers solvant en présence d'un agent émulsionnant neutre et non irritant.
Le procédé suivant la présente invention pour la préparation de 21-hexahydrobenzoates d'hormones corticosurrénales de formule

consiste à faire agir de l'acide hexahydroben- zoïque ou un dérivé fonctionnel de celui-ci sur une hormone corticosurrénale de formule cor respondante et à purifier le produit réactionnel.
Un mode de mise en aeuvre préféré de ce procédé consiste à faire agir un halogénure d'hexahydrobenzoyle, par exemple le chlorure ou le bromure, sur l'hormone libre en présence d'une base tertiaire comme la 2-méthyl-5-éthyl- pyridine, la pyridine ou la triéthylamine. On dilue de préférence le milieu réactionnel à l'aide d'un solvant.
Pour isoler et purifier le produit réactionnel, on peut procéder comme suit : on reprend par un solvant non miscible à l'eau, comme l'éther, le benzène, etc., lave avec un agent acide pour éliminer l'excès de base tertiaire employée, puis avec un agent alcalin pour enlever l'excès d'acide et l'acide qui n'a pas réagi, et finalement à l'eau. On distille le solvant après avoir séché la solution et recristallise l'ester dans un solvant approprié. Le produit est alors prêt pour l'emploi.
Selon une autre variante d'exécution du présent procédé, on peut également employer l'anhydride hexahydrobenzoïque, au lieu de l'halogénure d'acide, en présence d'une base tertiaire, les rendements étant toutefois infé rieurs.
On peut aussi, bien entendu, estérifier les hormones corticosurrénales en question par l'acide hexahydrobenzoïque libre en présence d'un catalyseur d'estérification, mais les ren dements sont alors moins bons que lorsqu'on utilise le chlorure ou le bromure d'hexahydro- benzoyle.
Si on désire préparer une suspension micro- cristalline de l'hexahydrobenzoate, on opère selon les procédés connus pour transformer les cristaux obtenus après purification en micro- cristaux, c'est-à-dire qu'on peut effectuer par exemple le broyage d'un mélange d'hexahydro- benzoate, d'alcool benzylique, de monooléate de polyoxyéthylène-sorbitol, de carboxyméthyl- cellulose, de chlorobutanol,
de chlorure de sodium et d'eau dans un broyeur à boulets sous atmosphère inerte.
Les hormones libres servant de matières premières peuvent être obtenues, soit par sapo nification de leurs acétates du commerce, soit encore directement par synthèse. Quant au chlorure d'hexahydrobenzoyle, on peut l'obtenir commodément par action du chlorure de thio- nyle sur l'acide hexahydrobenzoïque, selon Darzens et Rost (C. R. 1911,<I>153,</I> 773). Le bromure peut être obtenu très facilement par action du pentabromure de phosphore sur l'acide hexahydrobenzoïque.
Les nouveaux produits excellent par leur grande solubilité en chloroforme. Les exemples suivants illustrent l'invention.

A cette solution, on ajoute 3 g d'anhydride hexahydrobenzoïque, F. 250 C, préparé selon Bartlett et Rylander (J. Am. Chem. Soc., 1951, 73, 4275). Après un repos de seize heures, le mélange réactionnel est chauffé au bain-marie pendant trois heures pour compléter la réaction. On refroidit, verse dans l'eau et extrait au benzène. Les extraits benzéniques réunis sont lavés à l'acide chlor hydrique dilué, à la soude puis à l'eau. La solution benzénique ainsi traitée est distillée à sec.
Après recristallisation en méthanol, on obtient 1,5 g de 21-hexahydrobenzoate d'hydro- cortisone, soit un rendement de 38 %. Le composé a les mêmes constantes que celui obtenu selon l'exemple 1.
<I><U>Exemple 5:</U></I> On traite selon la technique de l'exemple 4, la cortisone (3,3 g) par l'anhydride hexahydro- benzoïque. Le rendement final en composé identique à celui obtenu d'après l'exemple 2 est de 2 g, soit 47 % de la théorie.
<I><U>Exemple 6:</U></I> En partant de 3,8 g de 11-désoxy-corti- costérone, la technique de l'exemple 4 fournit 1 g de 21-hexahydrobenzoate de 11-désoxy- corticostérone, F. 11311- 114o C, identique au composé obtenu selon l'exemple 3.

The high cost of these substances and the need to administer them frequently, if used in their present form, make a more prolonged form desirable. It has been found that the hexahydrobenzoates at 21 exert this action.
The hexahydrobenzoates make it possible to space the injections and to avoid the hormonal overload of the organism by carrying out a slow and continuous absorption of the hormone in humans or in veterinary medicine.
This result is completely unexpected, because the 21-benzoates of the same hormones do not exhibit this prolonged action and nothing could suggest that the saturation of the cyclic double bonds of the esterifying agent would make it possible to obtain esters at very prolonged action. The new esters can be used in solution in neutral vegetable oils with or without a glycol and an organic salt such as aluminum 2-ethyl caproate or in solution in other injectable solvents, or in the form of aqueous microcrystalline suspensions, or emulsions in water of solutions in a third solvent in the presence of a neutral and non-irritating emulsifying agent.
The process according to the present invention for the preparation of 21-hexahydrobenzoates of adrenocortical hormones of formula

consists in causing hexahydrobenzoic acid or a functional derivative thereof to act on an adrenal cortex hormone of the corresponding formula and in purifying the reaction product.
A preferred embodiment of this process consists in causing a hexahydrobenzoyl halide, for example chloride or bromide, to act on the free hormone in the presence of a tertiary base such as 2-methyl-5-ethyl- pyridine, pyridine or triethylamine. The reaction medium is preferably diluted with a solvent.
To isolate and purify the reaction product, one can proceed as follows: it is taken up in a solvent immiscible with water, such as ether, benzene, etc., washed with an acidic agent to remove the excess of tertiary base used, then with an alkaline agent to remove excess acid and unreacted acid, and finally with water. The solvent is distilled off after the solution has been dried and the ester is recrystallized from a suitable solvent. The product is then ready for use.
According to another variant embodiment of the present process, it is also possible to use hexahydrobenzoic anhydride, instead of the acid halide, in the presence of a tertiary base, the yields being, however, lower.
It is of course also possible to esterify the adrenal cortex hormones in question with free hexahydrobenzoic acid in the presence of an esterification catalyst, but the yields are then less good than when hexahydro chloride or bromide is used. - benzoyl.
