Process for preparing 1-chloro-4-methylthiaxanthone. As is well known, 1.-fi-diethylaminoethyliamino-4-methylthiaxantlione, the structural formula of which is given under (I), as well as its salts, is active against schistosomal infections.
EMI0001.0006
In the known method of synthesizing this substance, thiosalicylic acid (II) and p-chlorotoluene <B> (</B> III) are condensed to obtain a mixture of the two isomeric compounds 1-methyl-4 -chlorothiaxanthone (IV)
and 1-chloro-4-methylthiaxanthone (V). The mixture is then reacted with β-di-ethylaminoethylamine, and compound (V) reacts to form the desired therapeutically active derivative (I), while compound (IV) does not react.
EMI0001.0017
The disadvantage of this method is that the two isomers (IV) and (V) are produced in approximately equal amounts in a non-separable mixture, and that separation is possible only after the. reaction with β-diethylaminoethylamine, leaving compound (IV) as a waste product; in this way, there is therefore a waste of about 50% on the starting material. The object of the present invention is to remedy this drawback by synthesizing the intermediate 1-ehloro-4-methylthiaxanthone (compound V) by a process which avoids the simultaneous production of isomeric and undesirable thiaxanthones.
This process, which therefore forms the object of the invention, is characterized in that the elements of the water are removed from 2- (5'-chloro \ '' - methylphenyl-thio) -benzoic acid to form l.-Ehloro-4-methylthiaxanthone (V). This reaction takes place with efficiency. high in the desired product without the formation of large amounts of unnecessary substances.
2- (5'-Ehlox-o-2'-meth -,> lphenyl-tllio) - benzoic acid (VIII) is a new substance, and can be conveniently synthetically prepared by condensation of 5-chloro? - methylthiophenol (VI) with o-chlorobenzoic acid (VII).
This reaction is preferably carried out in the presence of an acid scavenger, such as sodium or potassium carbonate; as a suitable solvent, there may be mentioned benzyl alcohol, and in this case preferably two equivalents of said agent are employed.
Suitable agents for converting 2- (5'-ehloro-2'-methylphenyl-thio) -benzoic acid to 1-eh] oro-4-methyl-thiaxanthone are those already known for the production. eyelic ketones from carboxylic acids, such as strong sulfuric acid, or phosphorus halide and aluminum chloride.
Thus, the preparation of the starting material, its transformation into 1-chloro-4-methylthiaxanthone and the manufacture, from the latter, of 1-fl-diethylaminoethi-1-amino-4-methylthiaxanthone, can be represented by the following diagram
EMI0002.0036
Another method for preparing 2- (5'-Chloro-2'-methylphenyl-thio) -benzoic acid (VIII) comprises.
condensation of 5-chloro-2-methylthiophenol (IX) with diazotized anthranilic acid (X), in the formula in which X is halogen. The reaction is carried out as follows:
EMI0003.0006
The invention is described more fully with the aid of the following example, in which all temperatures are given in degrees centigrade.
The starting 2- (5'-chloro-2'-methylphenyl-thio) - benzoic acid was prepared as follows 7.9 - (1 / 2c mole) of 5-chloro-2-methylthiophenol, 7, 7 g (1 / 2o mol) of o-chlorobenzoic acid, 6.92 g (2 equivalents) of anhydrous potassium carbonate, 79 em3 of benzyl alcohol, 0.5 g of powdered copper and 1 g of potassium iodide have been mixed well,
mechanically stirred and heated continuously under reflux in a sand bath for five hours. The reaction mixture was cooled somewhat and filtered to remove some brown material, diluted with about 300 cc of ether and extracted several times with 10/0 aqueous caustic soda.
The combined alkaline extracts were decolored with animal charcoal, filtered hot, and acidified with 50% sulfuric acid. The precipitate was filtered off without cooling; after drying, it weighed 1.2.28 g (yield 87.77 / o); it had a melting point of 182-1850. This substance can be purified, although that. is not necessary, by crystallization from toluene, and then melts in 195-1970.
It is important to use the indicated amount of potassium carbonate to avoid the formation of the benzyl ester.
The crude 2- (5'-chloro-2'-methylphenyl-thio) -benzoic acid (8.2 g) thus prepared was heated in a water bath for 75 minutes with 25 cms of strong sulfuric acid (98 0 / 0) to form a light brown red solution. After cooling, the product was poured with stirring onto about 250 g of crushed ice. A yellow precipitate formed which was filtered, washed with water and stirred without drying it with 200 cm3 of water and enough ammonia to make the. alkaline liquor. The solid 1-chloro-4-methylthia.xanthone was filtered off, washed and dried.
The yield was 6.92 g, which is equivalent to 90.2 Vo of the theoretical yield; the product melted at 135 to 1370; it is pure enough to be able to be converted into the product (I), but can be recrystallized from alcohol and then melts at 143 to 1450.
2- (5'-Chloro-2'-methylphenyl-thio) benzoic acid can also be prepared as follows: Anthranilic acid is dinitrogenated by the so-called invert method, as follows:
2.74 g of anthranilic acid are dissolved by heating in 8.5 ml of a 10% solution of caustic soda; 1.45 g of solid sodium nitrite are added, and the. The solution is cooled to approximately -f- 50 C. The cooled solution is then added slowly to 22 ml of 2N hydrochloric acid cooled to 00 C, the temperature being maintained below + 50 C during this addition.
The excess nitrous acid is then removed by adding urea and the cold diazo solution is added to a solution. of 3.16 g of 5-chloro-2-methylthiophenol in 9.2 ml of ammonia (P. S. 0.885) diluted with an equal volume of water, the temperature being kept below about 20 ° C. during this addition.
(It is advantageous to run the reaction in an inert atmosphere in order to minimize oxidation of the thiophenol.) When the diazo solution is added, a yellow precipitate is formed which is believed to be the iazonium-mercaptide. . The mixture is. stirred for about an hour, during which the yellow precipitate is decomposed with evolution of nitrogen. An orange solution is then formed containing a small amount of a white solid.
This solid body, which is a bis- (5-chloro-2-methylphenyl) - disulfide, is removed by filtration, and the clear wire is extracted with ether to remove traces of impurities, and. is acidified. The precipitate slightly. brown, which consists of 2- (5'-chloro 2'-methylphenyl-thio) -benzoic acid, est. separated by filtration and crystallized for example at. from toluene. The acid melts at 195-197 C and is identical to the substance described in Example 1.
