Description
verfahren zur Herstellung einer Nueleotidverbindung eines Faktors des Vitamin-B-Komplexes. Im Hauptpatent Nr. 290136 ist ein Verfah ren zur Gewinnung von therapeutisch wert vollen Verbindungen der Adenylsäure mit Faktoren des Vitamin-B-Komplexes beschrie ben.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung einer neuen Verbindung der Adenylsäure mit einem Fak- tQr des Vitamin-B-Komplexes, welches da durch gekennzeichnet ist, dass 1 Mol La.etofla- vin bzw. eines Salzes desselben mit. 1 Mol Aclenylsäure bzw. eines Salzes derselben zur Reaktion gebracht wird.
.Man kann zum Beispiel ein Salz des Lacto- flavins mit einem Salz der Adenylsäure um setzen, doch wird vorgezogen, die freie Base mit der freien Säure, vorzugsweise in einem wässerigen Medium unter Wärmezufuhr, zur Reaktion zu bringen. Die erhaltene wässerige Lösung kann allenfalls ohne weitere Behand lung per os verabreicht werden. Für Injek tionen wird sie zweckmässig mit Natronlauge auf den. geeigneten p,1-Wert eingestellt. Durch Eindampfen der Lösung kann ein Trocken präparat gewonnen werden, welches in Form @-on Pulver oder von Granulen verabreicht werden kann.
Es wurde gefunden, dass bei einer Dosie rung von 2 bis 5 mg, ein- bis dreimal täglich, eine sichere Wirkung erzielt wird. (0,001 Mol) Lactoflavin zugesetzt. Nach Er hitzen zum Siedepunkt trat vollständige Lö sung ein. Die orangegelbe Lösung wurde im Vakuum eingedampft, wobei das Lactoflavin- monoadenylat als goldgelbes kristallines Pro dukt vom Sehmp. 174 bis 175 C erhalten wurde.
Die vergrösserte Wasserlöslichkeit der Adenylsäur e bei Gegenwart von Lactoflavin zeigte, dass eine chemische Vereinigung der beiden Reaktionskomponenten, und zwar eine Salzbildung, erfolgt. Wässerige Lösungen von Lactoflavin-monoadenylat haben eine fast un beschränkte Haltbarkeit. Die therapeutische Wirksamkeit des Laetoflavins wird durch Überführung in das Monoadenylat beträcht lich verbessert.

The subject of the present patent is a process for the production of a new compound of adenylic acid with a factor of the vitamin B complex, which is characterized in that 1 mol of La.etoflavin or a salt thereof with. 1 mol of aclenic acid or a salt thereof is reacted.
For example, a salt of lactoflavin can be reacted with a salt of adenylic acid, but it is preferred to react the free base with the free acid, preferably in an aqueous medium with the supply of heat. The aqueous solution obtained can at best be administered orally without further treatment. For injections, it is best to apply caustic soda to the. appropriate p, 1 value set. By evaporating the solution, a dry preparation can be obtained, which can be administered in the form of a powder or granules.
It has been found that a safe effect is achieved at a dosage of 2 to 5 mg, one to three times a day. (0.001 mole) lactoflavin added. After heating to the boiling point, complete solution occurred. The orange-yellow solution was evaporated in vacuo, the lactoflavin monoadenylate as a golden yellow crystalline product from the Sehmp. 174 to 175 C was obtained.
The increased water solubility of adenylic acid in the presence of lactoflavin showed that a chemical combination of the two reaction components, namely salt formation, takes place. Aqueous solutions of lactoflavin monoadenylate have an almost unlimited shelf life. The therapeutic effectiveness of laetoflavin is improved considerably by converting it into the monoadenylate.
