wertvolle therapeutische Eigenschaften be sitzen.

have valuable therapeutic properties.
In the above formula, R, and R, each denote a hydrogen atom, a methyl or a methoxy group, n denotes an integer between 2 and 6 including an integer of at least and Am denotes the radical of a secondary aliphatic, eyeloaliphatic or heteroeyelic amine .
Under secondary, heterocyclic amines should in this definition heteroeyeles with endocyclic amino group of the type z. B. of piperidine or morpholine to be understood. C ", I3_", in addition to the tetra methylene radical, can also mean alkyl-substituted tetra methylene radicals.
As has been found, the basic esters of the formula I are obtained by. Condensation of 1-aryl-cyelopentane-1-carboxylic acids of the formula 1I

or their reactive, functional derivatives, with amino alcohols of the formula III HO-C "II _" - Am III or their reactive, functional derivatives, one of the two components being in the form of a reactive derivative with hydrogen sulfide (e.g.
Compound II as --L # lonothio-carboxylic acid, monothiocarboxylic acid ester or compound III as mercaptane) must be present.
Either a 1-aryl-cyclopentane-1-carboxylic acid (II) or a reactive, functional derivative thereof can be reacted with a basic substituted mercaptan of the formula IV 11-SC “H_” - Ain IV or with a metal salt thereof zen, or a monothiocarboxylic acid of the formula V
EMI0001.0051
their esters or their salts, with a compound of the formula VI X-C "H =" - to implement on VI.
X denotes a halogen atom, a hydroxyl or a sulfhydryl group.
A few possibilities are listed for a more detailed explanation: The acid chloride of a carbonate of the formula II is reacted with an amino-substituted mercaptan of the formula IV. Instead of the acid chloride, another acid halide, the anhydride or the free acid, the latter advantageously with the addition of esterification catalysts, can be reacted with the mercaptan. Instead of the free mercaptan, its metal salts can be used.
There is a preferred embodiment of this procedure. in bringing the 1-aryl-eyelopentane-1-carboxylic acid chloride together with the equimolecular amount of the basic mercaptan (IV), in the presence or absence of inert organic solvents such as ether, benzene, dioxane or acetone. The implementation usually takes place at room temperature; faster, however, at higher temperatures. It supplies the hydrochloride of the new bases.
Furthermore, esters of carboxylic acids of the formula II with alcohols, phenols, mercaptans and thiophenols can be expediently removed by heating with basic substituted merapans of the formula IV in the presence of catalysts such as e.g. B. the corresponding sodium mercaptides, transesterify.
In addition, 1-aryl-ey clopentane-1-monothio-carboxylic acids (V) and their salts with haloalkylamines of the formula VII Hal-C “H_” - Am VII can be set to form esters of the formula I and I.
The same esters are finally also obtained by esterifying the I-aryl-cyclopentane-1-monothio-carboxylic acids with amino alcohols of the formula III and by transesterifying l-aryl-cyelopentane-1-.inonothio-carboxylic acid-alkyl- or aryl esters with amino alcohols of the formula III.
The new esters of the formula I form mostly water-soluble salts with inorganic acids and with organic acids; With reactive esters of aliphatic and araliphatic alcohols, such as with alkyl halides, dialkyl sulfates or aralkyl halides, they can be converted into quaternary salts.
Subject of the present patent is. a process for the preparation of a new basic ester of a 1-aryl-cyclopentane-1-monothio-carboxylic acid. The process is characterized in that one has a compound of the formula
EMI0002.0062
with a compound of the formula
EMI0002.0063
wherein X and F are two, with the exception of a sulfur atom contained in one of them, see radicals which split off during the reaction, converts.
In the execution of the method you can, for. B.1-Pheny l-ey elopentan-l-carboxylic acid, its ester, the anhydride or the acid halide with your mercaptan or with a metal salt of the formula 'V. CII .. CH,.,. Let N (C @ H;) _ react.
However, 1-phenyl-eyelopentane-1-inonothio-carboxylic acid, its esters or its salts can also be reacted with f3-diethylaminoethanol or its hydrogen halide ester.
The new base obtained, the 1-phenyl-ey clopentane-1-monot.liio-carboxylic acid-ß-diethyl-ainino-ethyl ester, boils. under 0.05 mm of pressure. at 122 to 123. She should. find therapeutic use.
Example 1: To a solution of 26.6 parts (3-diet.liyl-ainino-ethyl mercaptan in 120 parts of absolute benzene, 20.85 parts of 1- pheny 1- ey elopentan - 1. Acid chloride in 120 parts of absolute benzene was added dropwise, the temperature rising to 30 to 40.
Then it is boiled for a few minutes on reflux, cooled with ice, suctioned off and with. washed with absolute benzene. The combined benzene solutions are washed with water, dried and evaporated. The residue boils at 122 to 123 under 0.05 mm pressure and represents 1-phenyl-cyclopentane -1-monothio-carboxylic acid - ss - diethylamino-ethyl ester.
The water-soluble hydrochloride of the base melts at 138 to 1-10; it also forms water-soluble salts with sulfuric acid, phosphoric acid, acetic acid, citric acid and maleic acid. By heating with excess ethyl bromide, the quaternary salt is obtained in the form of colorless crystals which are easily soluble in water.
EXAMPLE 20.6 parts of 1-phenyl-cyclopentane-1-monothio-carboxylic acid, obtainable from Plienyl-cyclopen-tan-carboxylic acid chloride in pyridine with hydrogen sulfide, are mixed with 15 parts of β-diethylaminoethyl chloride and 20 parts of potash (anhydrous ) heated to boiling in 65 parts of dry acetone 1.8 hours. After the cold is filtered, the solvent is distilled off and the residue is distilled.
The distillate represents the 1-phenyl-cyclopentane-1-monotliio-carboxylic acid, 8-diethylamino-ethyl ester with the boiling point. 122 'under 0.05 mm pressure.
