Verfahren zur Herstellung eines Biguanid-Derivates. Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung eines Bi- guanid-Derivates, nämlich des N'-p-Chlor- phenyl - NI -isopropyl - bi, tanids, welches als Chemotherapeutikum oder als Zwischenpro dukt für die Herstellung von Chemothera- peutika nützlich ist.
Das erfindungsgemässe Verfahren ist da durch gekennzeichnet, da.ss man einen N'-p- Chlor- phenyl-N-..isopropylgtianyl-S-alkyl. - iso- thioharnstoff mit Ammoniak bzw. einem Am moniumsalz zur Umsetzung bringt.
Der als Ausgangsmaterial für das erfin dungsgemässe Verfahren verwendete N'-p- Chlor-phenyl - N;=-isopropyl-guanyl-S-alky 1-iso- thioharnstoff kann durch Alkylierung des ent sprechenden Thioliarnstoffes hergestellt wer den.
Bei der Ausführung des erfindungsgemässen Verfahrens werden die Reaktionsteilnehmer vorzugsweise zusammen erhitzt, und zwar zweckmässig in Gegenwart. eines Lösungs- bzw. Verdünnungsmittels, wie z. B. Methanol, Ätha- nol oder ss-Äthoxy-ätha.nol. Der N'-p-Chlor- phenyl-Nl-isopropy lguanyl-S-alky lisothioharn- stoff kann in freier Form oder in Form.
seiner Salze, beispielsweise des Ilydroehlorids, verwendet werden. Im letzteren Fall wird der Isothioharnstoff zweckmässig in sittt durch Zugabe einer äquivalenten Menge einer basi schen Substanz, wie z. B. Natriumäthy lat oder Natriummethylat, in Freiheit gesetzt. Das N'-p-Chlor-phenyl-N5-isopropyl-bi- Uuanid, welches nach dem erfindungsgemässen Verfahren erhalten wird, ist als Chemothera- peutikum oder als Zwischenprodukt für Chemotherapeutika nützlich.
Es ist insbeson dere als Antimalariamittel wertvoll. Es ist eine starke Base, die mit organischen und an organischen Säuren beständige Salze bildet, die in vielen Fällen in Wasser leicht löslich sind. Diese Salze können dadurch erhalten werden, dass man das Big2tanid in einer wäs serigen Lösung der Säure löst und hierauf das Wasser abdampft.. Zweckmässigerweise werden sie jedoch in trockener Form erhalten, indem man die Komponenten in einem or ganischen Lösungsmittel, beispielsweise in Aceton oder in einem Alkohol, in welchem die Salze schwer löslich sind, vermischt.
Auf diese Weise kann man leicht beispielsweise die Salze mit Essigsäure, Milchsäure, Methan sulfonsäure, 3#letliylendisalicy lsäure, Methy- len-bis-2,3-o.xyna.phthoesäure und Salzsäure herstellen.
Im folgenden Beispiel bedeuten die Teile Gewichtsteile.
Beispiel: 1 Teil N'- p - Chlor - phenyl - N=- isopropy 1- gu anyl-S- äthyl - isothioha rnstof f - hydrobromid und 7 Teile einer 18prozentigen äthanolischen Lösung von Ammoniak werden während einer Stunde bei 100 C in einem verschlossenen Gefäss erhitzt. Das Gremiseh wird hierauf ab gekühlt und zur Trockene eingedampft.
Man extrahiert den gummiartigen Rückstand mit 7prozentiger Salzsäure und klärt den sauren Extrakt mit Entfärbungskohle. Die Lösung wird dann mit Natriumhydroxyd stark alka lisch gestellt und mit Benzol extrahiert. Man dampft das Benzol ab und löst den Rück stand in trockenem Äthylenessigester. Diese Lösung wird mit 0,2 Teilen Eisessig versetzt, wobei ein NTiederschlag ausfällt. Dieser wird abfiltriert, mit Äthylessigester gewaschen und dann bei 60 C getrocknet.
Man erhält auf diese Weise Nl-p-Chlor-phenyl-NJ-isopropyl- biguanid-acetat vom Smp. 188 C. Die freie Base vom Smp. 129 C wird daraus nach einer üblichen Methode erhalten.
