Description

  

  Verfahren zur Herstellung eines     Biguanidderivates.       Die vorliegende Erfindung betrifft ein  Verfahren zur Herstellung von     Nl-3-Chlor-4-          bromphenyl-NS-n-propylbiguanid,    welches ein  wertvolles chemotherapeutisches Mittel ist oder  als Zwischenprodukt für die Herstellung von  chemotherapeutischen Mitteln verwendet wer  den kann. Es ist insbesondere ein wertvolles       Antimalariamittel.     



  Erfindungsgemäss wird die besagte neue  Verbindung,     nämlichNl-3-Chlor-4-bromphenyl-          NS-n-bropylbiguanid,    dadurch erhalten, dass  man     Na-n-Propyldicyandiamid    mit     3-Chlor-4-          bromanilin    umsetzt.  



  Die Umsetzung erfolgt zweckmässig durch  Erhitzen eines Salzes des Amins mit dem       substituierten        Dieyandiamid    in Gegenwart  eines     Lösungsmittels,    wie z. B. Wasser oder       ss-Äthoxyäthanol.     



  Das     Nl-3-Chlor-4-bromphenyl-NS-n-propyl-          biguanid    stellt eine starke Base dar, welche  mit organischen und anorganischen Säuren  beständige Salze ergibt, die in manchen Fäl  len in Wasser leicht löslich sind. Die Salze  lassen sich dadurch herstellen, dass die Base  in     wässrigen    Lösungen der Säure gelöst und  hierauf das Wasser verdampft wird, doch  können sie in trockener Form bequemer durch.  Vermischen der Komponenten in einem orga  nischen Lösungsmittel, wie z. B. Aceton, oder  in einem     Alkohol,    in welchem die Salze spär  lich löslich sind, hergestellt werden.

   Auf diese  Weise kann man beispielsweise die Salze mit  Essigsäure, Milchsäure,     ',#lethanstllfonsäure,            Methylendisalicylsäure,        Methylen-bis-ss-oxy-          naphthoesäure    und Salzsäure bequem her  stellen.  



  Das folgende Beispiel diene zur Erläute  rung der Erfindung.  



       Beispiel:     Ein Gemisch von 126 Teilen     N3-n-Pro-          pyldicyandiamid    und 24,3 Teilen     3-Chlor-4-          bromanilin-chlorhydrat    in 60 Teilen     ss-Äthoxy-          äthanol    wird während 3     Stunderi.    unter Rück  fluss zum Sieden erhitzt.     Hierauj'    lässt man  das Gemisch abkühlen, worauf     abfiltriert     wird. Der feste Rückstand wird mit kaltem       Äthylacetat    gewaschen und aus Wasser um  kristallisiert.

   Auf diese Weise erhält man     Nl-          3-.Chlor-4-brom.phenyl-N5-n-propylbiguanid    in  Form seines     Monohydroehlorids,    welches bei       217     C schmilzt.



  Process for the preparation of a biguanide derivative. The present invention relates to a process for the preparation of Nl-3-chloro-4-bromophenyl-NS-n-propylbiguanide, which is a valuable chemotherapeutic agent or can be used as an intermediate for the preparation of chemotherapeutic agents. It is a particularly valuable antimalarial drug.



  According to the invention, said new compound, namely Nl-3-chloro-4-bromophenyl-NS-n-bropylbiguanide, is obtained by reacting Na-n-propyldicyandiamide with 3-chloro-4-bromoaniline.



  The reaction is conveniently carried out by heating a salt of the amine with the substituted dieyandiamide in the presence of a solvent, such as. B. water or ss-ethoxyethanol.



  The Nl-3-chloro-4-bromophenyl-NS-n-propyl-biguanid is a strong base which, with organic and inorganic acids, results in stable salts, which in some cases are easily soluble in water. The salts can be prepared by dissolving the base in aqueous solutions of the acid and then evaporating the water, but they can pass through more conveniently in dry form. Mixing the components in an orga African solvent, such as. B. acetone, or in an alcohol in which the salts are sparse Lich soluble, are produced.

   In this way, for example, the salts with acetic acid, lactic acid, lethane-sulfonic acid, methylenedisalicylic acid, methylene-bis-β-oxynaphthoic acid and hydrochloric acid can be conveniently prepared.



  The following example serves to explain the invention.



       Example: A mixture of 126 parts of N3-n-propyldicyandiamide and 24.3 parts of 3-chloro-4-bromaniline chlorohydrate in 60 parts of ß-ethoxy ethanol is used for 3 hours. heated to boiling under reflux. The mixture is allowed to cool down and filtered. The solid residue is washed with cold ethyl acetate and recrystallized from water.

   In this way, Nl-3-chloro-4-bromo-phenyl-N5-n-propylbiguanide is obtained in the form of its monohydrochloride, which melts at 217 ° C.
