Description
Verfahren zur Herstellung eines Gallensäurederivates. Dehydrocholsäure als solche oder in Form ihres Natriumsalze s .ist bereits in der Thera pie verwendet worden.
Es wurde nun gefunden; dass bisher un- bekannte, therapeutisch wertvolle basische Ester von getogallensäuren der Formel R-C00-(CH_) n-NR,R2 dadurch gewonnen werden können, dass auf das Alkalisalz einer getogallensäure das Halogenid eines basischen Alkohols oder auf das,
Halo.genid einer getogallensäure ein basischer Alkohol einwirken gelassen oder die freie Säure mit einer Mischung des Alkohols und .Schwefelsäure oder einem andern Vereiterungskatalysator erhitzt wird. In der vorstehenden Formel 'bedeuten R-C00- den Rest einer getogall'ensäure (z.
B. Dehydrocholsäure oder Dehydrodes- oxycholsäure) ; R, und Rz Alkylgruppen, die auch unter sich verbunden sein können (z. B. Piperidina- oder Morghohnogruppie- rungen), und n eine niedere Zahl grösser als 1.
Die neuen Verbindungen besitzen wert- volle therapeutische Eigenschaften; sie sollen als Arzneimittel verwendet werden.
Gegenstand des, vorliegenden Patentest iss ein Verfahren zur Herstellung eines basisollen Esters einer gMogallensäure, welches da durch gekennzeichnet ist, dass man Dehydro- cholsäure mit ss-Diäthylamino-äthanol ver- estert. Es ist zweckmässig,
dabei ein Ver- esterungsmittel, wieg z. B. konzentrierte Schwefelsäure, zuzusetzen.
Die neue Verbindung, das Diäthylamino- äthyl-dehydrocholat, ist kristallin, löst sich in einer Mischung von Petroläther und Ben zol und hat einen Schmelzpunkt von 194 bis 195 C.
Beispiel: 1 Gewichtsteil Dehydrocholsäure in 4,5 Raumteilen konzentrierter Schwefelsäure wird mit 25 Raumteilen ss-Diäthylamino- äthanol während 3 Stunden auf 160 C er hitzt.
Hierauf wird die kalte Reaktions- mischung auf Eis gegossen, nach dem Ab filtrieren von unlöslichen Bestandteilen das Filtrat mit Natronlauge gegen Phenol- phtalein alkalisch gemacht und mit Äther extrahiert. Das. Diäthylaminoäthyl-dehydro- cholat wird durch AbdesUllieren des, Äthers erhalten und aus,
Benzol und Petroläther umkristallisiert. Es hat einen Schmelzpunkt von 194-195 C.

The new compounds have valuable therapeutic properties; they are intended to be used as medicines.
The subject matter of the present patent test is a process for the production of a basic ester of a gMogallenic acid, which is characterized in that dehydrocholic acid is esterified with ß-diethylamino-ethanol. It is useful
