Process for the preparation of an oestrone isomer. The present invention relates to a process for the preparation of an () estrone isomer.
It has been found that a highly unknown oestrone isomer can be obtained if "1-oxo-2-methyl-7-inetlioxr -.1, 2, 3.
4, 9,10, 1..1.12 - octaliy drophen, - @ intlii-en-2-earboilsäureiethylester from F.87 üis 89, a haloacetic acid methyl ester and zinc can act on each other, the reaction product hydrolyzed, on the so obtained oxyester water splits off, saturates the double bond formed with hydrogen,
in the methyl 7-1lethyl-marrianolate formed the carbomethoxy group in the 1-position side chain '# - the half-ester melting at 132 to 134.5 according to the reaction of Arndt-Eistert, followed by saponification, via the acid chloride and the diazoketone into the 7-:
Converts 1lethyl-homomarrianoleic acid from mp 201 to 203, treats this compound with a cyclizing agent and converts the estron methyl ether obtained from mp 146 to 147 by hydrolysis into the free oestrone isomer from mp 230 to 232.
The synthesis can be illustrated, for example, by the following reaction scheme
EMI0001.0042
EMI0002.0001
The intermediate products obtained during the synthesis are also new. The 1-oxo-2-methyl-7-methoxy-1,2,3, -1, 9,10,11,12-octa-hydrophenanthrene-2-carboxylic acid methyl ester of F. 8 7 to 8911 can be used as starting material Procedure according to Switzerland. Patent No. 263974 will be obtained.
The reaction of Arndt-Eistert is the process described in B.68, 200 (1935) for converting a carboxylic acid into the next higher homologue.
Example: 15 parts by weight of 1-oxo-2-methyl-7-methoxy-1, 2, 3, 4, 9, 10, 11, 12 -'octahy drophenanthrei @ - 2-carboxylic acid methyl ester from F. .

EMI0002.0023
earth with 15 parts by volume of methyl bromoacetate, 45 parts by weight of zinc and 1.5 parts by weight of iodine in 250 parts by volume of benzene and 250 parts by volume of ether heated to 60 to 70 "for 18 hours.
After cooling, dilute ice-cold acetic acid is added, the slightly colored benzene-ether solution is separated off and it is shaken with dilute aqueous amnionia solution until the latter is no longer colored. becomes. After washing with water and drying, the organic solvent is evaporated and the residue is recrystallized from methanol.
The l.-oxy-7-methyl-marrianolsäure-diinetliyl- ester of formula
EMI0002.0040
melts at 86-87 after further redissolution from methanol.
16 parts by weight of this oxy ester of F. 86-871 'are heated with. 16 parts by volume of phosphorus oxychloride and 160 parts by volume of pyridine j / 1 hour to the boil. After about 45 minutes the mixture is cooled and poured. on ice, add excess hydrochloric acid and take the precipitated reaction product. in ether. The ethereal solution, washed with hydrochloric acid and water, is then dried and the ether is evaporated.
The unsaturated 7-3lethyl-marrianolic acid dimethyl ester obtained in this way is an oil.
13.7 parts by weight of this oil are dissolved in 500 parts by volume of glacial acetic acid and hydrogenated in the presence of a palladium catalyst at about 400, the hydrogenation coming to a standstill after the calculated amount of hydrogen has been taken up. The hydrogenation product is an oil.
For the purpose of semi-saponification, the above oil (7-l1ethylmarrianolsäure-dimethylester) is warmed up with the calculated amount of 0.7 N aqueous potassium hydroxide and liethanol on a water bath. The mixture of the half esters of the formula thus obtained
EMI0003.0006
is separated by fractional crystallization from .leeton into a fraction melting at 126-1400 and a fraction melting at 166-171.0 .
Repeated dissolving gives the fraction from F. 126-14011 the pure half ester of F. 132-134.5 ". The further purification, the fraction from F. 166-17111 leads to the isomerically pure half ester of F. 173 to 175 ,.
1 part by weight of the above half-ester with a melting point of 132-134.5 is slurried in 1.0 part by volume of benzene to guide the liver into the acid chloride and, after adding 1 part by volume of oxalyl chloride, left to stand overnight at room temperature. Evaporation of the solvent gives the acid chloride, which is dissolved in benzene without further purification and mixed with ice-cold, ethereal diazomethane solution; becomes.
The yellow colored diazoketone obtained after evaporation of the solvent is taken up in 50 parts by volume of methanol and the solution, after adding 2 parts by weight of freshly precipitated and dried silver oxide, is boiled until the elimination of nitrogen has stopped.
The reaction product obtained from the filtered methanol solution is then heated in an open vessel to 160 to 17011 in a mixture of 10 parts by volume of alcohol, 1 part by volume of water and 3 parts by weight of potassium hydroxide for the purpose of saponifying the two carbomethoxy groups. The potassium salt which has precipitated solid after evaporation of the solvent is taken up in water and the alkaline solution is acidified after shaking with ether.
The acid which melts at 201-9-03 "after redissolving from methanol is the 7-llethyl-homomarrianolsaltre of the formula
EMI0003.0052
0.5 part by weight of this dicarboxylic acid is mixed ver with 0.5 part by weight of lead carbonate, heated until the reaction occurs and then distilled in a high vacuum.
The distillate, which soon solidifies, is dissolved in methanol, decolorized with animal charcoal and, after concentration, the oestrone methyl ether of F.146-147 is obtained. 0.5 part by weight of it is mixed ver with 5 parts by weight of pyridine hydrochloride and 4 hours for about. Heated in 18011. By treating the cooled melt with hydrochloric acid and ether, shaking the ether layer with aqueous potassium hydroxide solution and acidifying the alkaline extracts, the oestrone isomer is obtained, which after dissolving from methanol at 230-9-320 melts.
Its benzoate is melting. at 134-136.
