Process for the production of a bile acid derivative. Dehydrocholic acid as such or in the form of its sodium salt has already been used in the therapy.
It has now been found that previously unknown, therapeutically valuable basic esters of ketogallic acids of the formula R-C00- (CH ,:
) .NR, R2 can be obtained by letting the halide of a basic alcohol act on the alkali salt of a togallenic acid or by allowing a basic alcohol to act on the halide of a togallenic acid, or by heating the free acid with a mixture of alcohol and sulfuric acid or another esterification catalyst.
In the above formula, R-COO- denote the residue of a ketogallenic acid (for example dehydrocholic acid or dehydrodesoxycholic acid); R, _ and R2 are alkyl groups that can also be linked to one another (e.g. piperidino; or morpholino groups), and n is a lower number greater than 1.
The new compounds have valuable therapeutic properties; they are intended to be used as medicines.
The present patent relates to a process for the preparation of a basic ester of a ketogallic acid, which is characterized in that de hydrogholic acid chloride is reacted with ß-diethylaminoethano1.
The new compound, diethylamino-ethyl-dehydrocholat, is crystalline, dissolves in a mixture of petroleum ether and benzene and has a melting point of 194 to 195 C.
Examples 15 parts by weight of dehydrocholic acid are added to 20 parts by volume of thionyl chloride in 200 parts by volume of benzene and the: reaction mixture for 80.
Heated under reflux for minutes until all the acid has dissolved. After the excess thionyl chloride and the benzene have been evaporated off under reduced pressure, the remaining dehydrocholic acid chloride crystallizes on cooling.
80 parts by volume of ß-diethylamino-ethanol are then slowly added while cooling with ice, the mixture is heated at 100 ° C. for 10 minutes and left to stand for one hour at room temperature.
The reaction product is poured onto ice and sulfuric acid, the solid constituents are filtered off, the filtrate is made strongly alkaline against phenolphthalein with the aid of sodium hydroxide solution and then extracted with benzene; upon addition of petroleum ether, the diethylaminoethyldehydrocholate crystallizes out.
Another crystallization from benzene and petroleum ether gives the pure ester with a melting point of 194-195 C.
