Es wurde gefunden, dass man zu neuen chemotherapeutisch hoch wirksamen Verbin dungen gelangt, wenn man 4-Aminomethyl- benzolsulfonamide darstellt. Die Amino- gruppe des Sulfonamidrestes kann sich nicht nur vom Ammoniak, sondern auch von den verschiedensten primären oder sekundären organischen Aminen, zum Beispiel auch von heterocyclischen Aminen, ableiten.
Auch kön nen die Amidgruppen bezw. deren Substi- tuenten durch weitere Gruppen, wie freie oder substituierte Amino- oder Hydroxylgruppen oder Acylreste weiter besetzt sein. Verbin dungen der gekennzeichneten Art besitzen eine gegenüber dem 4-Aminobenzolsulfon- amid (mit aromatischer Aminogruppe) er höhte Wasserlöslichkeit und die Lösungen ihrer mineralsauren Salze reagieren nicht mehr kongosauer.
Bei bakteriellen Infektions krankheiten haben sich die neuen Verbindun gen als gut wirksam erwiesen.
Gegenstand des vorliegenden Patentes ist in Verfahren zur Herstellung einer Sulfon- amidverbindung, welches dadurch gekenn zeichnet ist, dass man eine Verbindung, welche sich vom 4-Methyl-benzolsulfonsäure- amid dadurch ableitet, dass in der 4-Methyl- gruppe ein zur Aminogruppe verseifbarer Rest sitzt, zum 4-Aminomethyl-benzolsulfon- säureamid verseift.
Als zur Aminogruppe verseifbare Reste können in der 4-Methyl- gruppe zum Beispiel Acylamino- und Azo- methingruppen vorhanden sein. Die Versei fung kann in üblicher Weise vorgenommen werden. Das so erhältliche 4-Aminomethyl- benzolsulfonsäureamid bildet farblose Kri stalle vom Schmelzpunkt 15d , die in war mem Wasser, verdünnter Natronlauge und Salzsäure leicht löslich sind. Das neue Produkt soll therapeutische Anwendung finden.
Ein Ausgangsstoff für das erfindungs gemässe Verfahren kann zum Beispiel wie folgt erhalten werden: 50 g Acetylbenzyl- amin werden unter Rühren in 150 cm' Chlor- sulfonsäure eingetragen, wobei die Tempera- tur durch Aussenkühlung unter 40 gehalten wird. Nach mehrstündigem Stehen bei ge wöhnlicher Temperatur wird eine Stunde im siedenden Wasserbad erwärmt und nach dem Abkühlen auf Eis gegossen. Hierbei fällt das 4-Acetylaminomethyl-benzolsulfonsäure- chlorid zunächst ölig aus, erstarrt jedoch nach kurzem Rühren kristallinisch.
Das ab gesaugte und mit kaltem Wasser aus gewaschene Produkt wird in eine 1.0 T ige wässrige Ammoniaklösung eingetragen. Es tritt dabei Lösung unter Erwärmen ein, und nach kurzer Zeit fällt das 4-Acetvlamino- methyl-benzolsulfonsäurea.mid kristallin aus. Nach Erwärmen auf 70 während 30 Mi nuten wird abgekühlt, abgesaugt und aus gewaschen. Das Produkt wird aus Wasser oder verdünntem Alkohol umkristallisiert in schönen farblosen Kristallen vorn F. 177 erhalten.
Es ist in warmem Wasser gtit, ii! verdünnter Natronlauge äussert leicht lös- lieh.
Beispiel: 23 g 4-Acetylaminomethyl-benzolstilfon- säureamid werden in 60 cm- 20 % iger Natron lauge eine Stunde lang zum Kochen erhitzt. Die entstandene Lösung wird mit Tierkohle behandelt und nach dem Absaugen aus dem Filtrat das 4-Aminomethyl-benzolsulfonsäure- amid mit Ammoniumchlorid gefällt. Die Mi schung wird gut abgekühlt, abgesaugt und der Niederschlag mit Eiswasser gewaschen. Aus Wasser oder i@lethylalkohol erhält nian die neue Verbindung.
