Verfahren zur Herstellung einer Benzolsulfonsäureamidverbindung. .
Process for producing a benzenesulfonic acid amide compound. . Benzalsulfonic acid amides with a p-position, substituted by halogen in a-position alkyl group, are hitherto unknown.
Attempts to bring the p-alkylbenzene sulfonic acid amides to reaction in the alkyl group by halogenating agents failed due to the reaction capability of the hydrogen atoms of the sulfonic acid amide group against these agents. The reaction between the two components proceeds with great violence and does not lead to the desired α-haloalkyl substitution products.
In contrast, it has been found that the p- (a-haloalkyl) -benzoIsulfonsäurebalogenide can be converted very smoothly with ammonia or any primary or secondary amines to p- (a-halo: genalkyl) -benzenesulfonic acid amides, advantageously at temperatures under 60 works.
The flawless course of the reaction in the sense of a single conversion: of the halogen in the sulfonic acid halide group: e was very surprising in view of the likewise very high reactivity: of the halogen in a-haloalkyl groups. The reaction can be carried out neat: or in a solvent or diluent, e.g. B. alcohol, acetone, benzene. Or water, who .den.
In addition to ammonia, aliphatic, cycloaliphatic, aromatic, aliphatic-aromatic or heterocyclic, optionally substituted amines can be used for the reaction. The isulphonic acid amides identified above are produced in almost quantitative yield.
The p - (a - haloalkyl) benzenesulfonic acid amides obtained in this way are valuable starting materials for the organic chemical industry.
The subject of the present patent is a process for the preparation of a 4-bromomethyl-benzenesulfonic acid amide, which is characterized in that a 4-bromomethyl-benzenesulfonic acid halide is reacted with ammonia.
The new connection should bezw as medicament tel. find use as an intermediate product in the construction of drugs.
Example: In the solution of 27 g of 4-bromomethylbenzenesulfonic acid chloride in 75 cm 'of alcohol, 20 cm' of 20% aqueous ammonia solution are added dropwise with cooling and stirring. The temperature is kept below 20 ge and, after the ammonia has been added, stirring is continued for 10-15 minutes. A thick crystal slurry of 4-bromomethyl-benzenesulphonic acid amide is formed, which is thoroughly washed out with water after suctioning off and obtained in glossy colorless crystals with a melting point of 197 after recrystallization from alcohol.
It only has a weak benzyl chloride odor and is easily soluble in cold, dilute sodium hydroxide solution.
