Es wurde: gefunden, dass die Anwesen heit des Cinnamylrestes in dieser Verbindung stark an der bakteriziden Wirkung beteiligt ist, was nicht ohne weiteres zu erwarten war.
In wässeriger Lösung stellt die Verbin dung ein wertvolles Antiseptikum und Ger micid dar. Man kann sie zum Beispiel als Mundwasser und auch für lokale Anwendung benutzen. Infolge der ungewöhnlich starken Wirksamkeit der Verbindung kann man sehr verdünnte wässerige Lösungen anwenden, und diese besitzen demzufolge den ausgesprochenen Vorteil, keinen starken. Geschmack oder CTe- rueh aufzuweisen.
Die Verbindung wird erfindungsgemäss dadurch hergestellt., dass man a-Amino-dode- eylbenzol mittels Formaldehyd und Aineisen- sä.ure inet.hyliert und das erhaltene tertiäre Amin mit Cinnamylehlorid quaternisiert.
Man geht zum Beispiel wie folgt vor. Zu etwa 5 Mol 90prozentiger Ameisen säure gibt man 1 Mol des primären Amins tropfenweise zu, wobei sieh das Formiat des primären Amins bildet, und fügt dann im Verlaufe von etwa einer Minute etwa 2,2 Mol Formaldehyd (36prozentig) zu. Dabei wird das Reaktionsgefäss in ein kaltes Wasserbad getaucht und die Mischung fortwährend ge rührt. Man lässt die Mischung ohne Erhitzen weitere 15 Minuten reagieren und erwärmt dann vorsichtig, bis eine heftige Reaktion, einsetzt, die sich durch beträchtliches Schäu men infolge Kohlendioxydentwicklung kund tut.
Hernach wird die Temperatur so ge steuert, dass ein schwacher Rückfluss statt findet, indem man je nach Bedarf kühlt oder Wärme zuführt. Die heftige Reaktion nimmt nach 15 Minuten ab, doch sollte weiter ge rührt und das Reaktionsgefäss" während wei teren sieben Stunden auf einem Dampfbad erhitzt werden.
Das tertiäre Amin kann dann nach üb lichen Methoden isoliert werden, zum Bei spiel indem man die Mischung mit Wasser verdünnt und mit einem L'berschuss an Am monium- oder Natriumhy droxy dlösung stark alkalisch stellt, das ausgeschiedene ()1 in Benzol löst, die Benzollösung gut mit, Was ser auswäscht, das Benzol abdestilliert und den Rückstand- einer fraktionierten Vakuum destillation unterwirft.
Zur Quaternisierung wird das tertiäre Amin vorzugsweise bei Zimmertemperatur mit. der äquimolekularen Menge Cinnamylchlorid umsetzt, bis vollständige Wasserlöslichkeit eingetreten ist. Das quaternä,re Ammonium salz kann durch Umkristallisieren gereinigt werden.
Die Verbindung wurde dem Prüfverfah ren für antiseptische und Desinfektionsmittel der United States Food and Drug Admini stration unterworfen, wie sie im Zirkular Nr.198 vom Dezember 1931 auf Seite 2, bis zur Mitte der Seite 10 beschrieben ist, und zwar unter Verwendung einer Kultur von Staphylococcus aureus F. D. A. Nr. 209-A bei 3i C. Man bestimmt dabei die Verdün nung, bei welcher die zu untersuchende Ver bindung den Organismus in 10 Minuten tötet, in 5 Minuten jedoch noch nicht.
Dieser Ver dünnungsgrad beträgt im Fall des erfindungs gemäss erhaltenen quaternären Salzes 1:192000. Ähnlicbe Verbindungen, welche jedoch den Cinnamylrest nicht enthalten, sind beträeht- lieli -weniger wirksam. So beträgt der in glei- eher Weise ermittelte Verdünnungsgrad im Fall von Phenyldodeeyl-(a.)-dimethyl-allv1- ammoniumehlorid 1:
36 000 und im Fall von Phenyldeev 1- (a) -dimetlipl-benzv1-Ja.min oniuni- elilorid 1 : ?5 000.
