Verfahren zur Herstellung neuer Sulfonamide
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 1, 4-Endoalkylen cyclohexan-2-sulfonsäureamiden, deren Endoalkylengruppe die Methylen-oder Athylengruppe ist, das heisst von SuKonsäureamiden mit dem Kern der Formel

in der n 1 oder 2 bedeutet. Die neuen Verbindungen können weiter substituiert sein, insbesondere am Stickstoff der Sulfonsäureamidgruppe, der z. B. nie deraliphatische oder cycloaliphatische Substituenten tragen oder acyliert sein kann.
Vorzugsweise ist der Stickstoff der Sulfonamidgruppe unsubstituiert oder ist durch Kohlenwasserstoffreste, wie insbesondere eine oder zwei Niederalkylgruppen, beispielsweise Methyl, Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, Pentyl, Cycloalkylgruppen, wie Cyclopentyl und Cyclohexyl, substituiert, oder er trägt eine Alkylenkette, wie Pentamethylen, oder ist acyliert, beispielsweise durch einen Carbaminsäurerest, wie den N-n-Butylcarbaminsäurerest.
Die neuen Verbindungen können auch weiter substituiert sein, wie z. B. an Ringstellung 3, durch beispielsweise Niederalkyl.
Die am 1, 4-Endoalkylen-cyclohexan-System haftenden Substituenten, insbesondere die Sulfonsäureamidgruppe in 2-Stellung, können die endooder die exo-Konfiguration besitzen.
Die neuen Verbindungen und ihre Salze besitzen wertvolle pharmakologische Eigenschaften und kön- nen deshalb als Heilmittel oder als Zwischenprodukte zu deren Herstellung verwendet werden. So zeigen sie zentralerregende Eigenschaften.

wobei n 1 oder 2 und R ein Wasserstoffatom oder einen Methylrest bedeutet, eine analeptische Wirkung und können als Analeptica verwendet werden.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man in einem gegebenenfalls im Ring und/oder am Sulfonamidstickstoffatom substituierten 1, 4-Endo alkylen-zp-cyclohexen-2-sulfonsäureamid, dessen Endoalkylengruppe die Methylen-oder Äthylengruppe ist, die 5, 6-Doppelbindung mit Wasserstoff absättigt.
Die Absättigung der Doppelbindung erfolgt vorzugsweise mit katalytisch erregtem Wasserstoff ; als Katalysatoren eignen sich Edelmetalle mit oder ohne Träger, beispielsweise Palladiumkohle, oder Nickel.
Die Reaktion kann in üblicher Weise erfolgen, vorzugsweise in Anwesenheit eines geeigneten Lösungs- mittels.
Als Salze der neuen Verbindungen sind insbesondere solche derjenigen Sulfonsäureamidderivate zu nennen, die am Stickstoffatom ein freies Wasserstoffatom tragen, das gegen ein Kation, wie z. B. das Kation eines Alkalimetalls, beispielsweise Natrium, ausgetauscht werden kann.
Erhaltene Sulfonsäureamidderivate lassen sich am Stickstoff weiter substituieren. Insbesondere kann man zur Salzbildung mit Basen befähigte Sulfonsäurederivate mit elektrophilen Reagenzien umsetzen.
Elektrophile Reagenzien sind z. B. reaktionsfÏhige Ester von Alkoholen, wie Halogenide oder Sulfonsäureester, oder ungesättigte Verbindungen, wie Isocyanate. Beispielsweise gelangt man durch Umsatz des Natriumsalzes von 1, 4-Endomethylen-cyclo hexan-2-sulfonsäureamid mit n-Butylisocyanat zum entsprechenden Harnstoffderivat.
Die Substitution kann in an sich bekannter Weise in An-oder Abwesenheit von Verdünnungsmitteln und/oder Kondensationsmitteln und/oder Katalysatoren, bei gewöhnlicher oder erhöhter Temperatur, im offenen oder im geschlossenen Gefäss unter Druck durchgeführt werden.
Die Ausgangsstoffe sind bekannt oder lassen sich nach bekannten Methoden herstellen. Zweckmässig verwendet man solche, dass die eingangs als bevorzugt geschilderten Verbindungen entstehen.

In den nachfolgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
0, 72 g 1, 4-Endomethylen-I)-cyclohexen-2-sul- fonsäureamid vom F. 110-112- (stareochemisch einheitlich endo-oder exo-Form) werden in 20 cnf, Feinsprit mit 0, 2 g 10/'o Palladiumkohle als Katalysator hydriert, wobei 1 Mol Wasserstoff aufgenom- men werden. Die filtrierte Hydrierlösung wird zur Trockne eingedampft, und man erhält Kristalle vom F. 131-132 , die nach einmaligem Umlösen aus 15 cnr Wasser bei 132-133 schmelzen.
Das so erhaltene Produkt ist das eine der beiden möglichen Isomeren (endo oder exo) von 1, 4-Endo methylen-cyclohexan-2-sulfonsäureamid der Formel

Zur Lösung von 1 g Natrium in 50 cm3 Methanol gibt man 6, 2 g 1, 4-Endomethylen-cyclohexan-2sulfonsäureamid und tropft darauf unter Rühren und Kühlen eine Lösung von 7, 5 g Methyljodid in 20 cmi Methanol zu. tuber Nacht wird gerührt und anschliessend 1 Stunde am Rückfluss gekocht. Die klare Lösung wird darauf zur Trockne eingedampft.

Die Ausgangsstoffe lassen sich beispielsweise wie folgt herstellen :
26, 8 g Vinyl-sulfonsäuredimethylamid und 27 cm : Cyclopentadien werden im Glasbombenrohr über Nacht auf 150 erhitzt. Die dunkle Suspension wird nach dem Abkühlen mit Methanol herausgespült.
Nach dem Eindampfen dieser Lösung wird der Rückstand destilliert, und man erhält 23, 2 g 1, 4 Endomethylen-X5-cyclohexen-2-sulfonsäuredimethyl- amid als schwach gelbliches 01 vom Sdp. 90-100 (0, 04 mm), das spontan kristallisiert und nach Umlösen aus Isopropanol bei 54-57 schmilzt.
Auf dieselbe Weise erhält man aus Vinyl-sulfon säurediäthylamid und Cyclopentadien das 1, 4-Endo methylen-Sa-cyclohexen-2-sulfonsäurediäthylamid, ein farbloses Ol vom Sdp. 95 (0, 05 mm).
Beispiel 4
17, 5 g 1, 4-Endomethylen-cyclohexan-2-sulfonsäureamid werden in 100 cm3 Acetanhydrid und
10 cm3 Pyridin gelöst und während 3 Stunden am Rückfluss gekocht. Darauf wird die Lösung im Vakuum zur Trockne eingedampft, der Rückstand in Chloroform aufgenommen und diese Lösung mit verdünnter Salzsäure und Wasser gewaschen.
Nach Trocknen und Eindampfen erhält man aus dem Chloroform 19, 5 g kristallines 1, 4-Endo methylen-cycl'ohexan-2-sulfonsäure-N-acetylamid, das nach dem Umlösen aus Äther bei 92-98 schmilzt und durch folgende Formel veranschaulicht werden kann :

8, 75 g 1, 4-Endomethylen-cyclohexan-2-sulfon- säureamid löst man in 30 cm3 abs. Pyridin. Unter Rühren und Kühlen tropft man zu dieser Lösung 7, 5 g Benzoylchlorid, rührt darauf während 1 Stunde bei Raumtemperatur und während 2 Stunden bei 60 bis 70 . Man giesst die Lösung darauf in ein Gemisch von Chl'oroform und verdünnter Salzsäure und erhält aus dem Chloroform nach Waschen mit Salzsäure und Wasser 15, 0 g hochviskoses Ol. Dieses wird in 60 cm3 In Natronlauge gelöst.
Nach zweimaligem Waschen mit Ather dampft man diewässrige Lösung im Vakuum zur Trockne ein und kristallisiert den Rückstand aus Alkohol um. Man erhält so das Natriumsalz von 1, 4-Endomethylen-cyclo- hexan-2-sulfonsäure-N-benzylamid der Formel

12 g 1, 4-Endomethylen-cyclohexan-2-sulfonsäureamid werden in einem Gemisch von 75 cm3 In Natronlauge und 200 cm3 Aceton gelöst und tropfenweise unter Rühren bei Raumtemperatur mit einer Lösung von 12, 5 g n-Butylisocyanat in 50 cm3 Aceton versetzt. Das klare Reaktionsgemisch wird nun während 1 Stunde bei Raumtemperatur und während 2 Stunden bei 60-70 ger hrt. Durch Lösen in Chloroform, Waschen dieser Lösung mit Salzsäure und Wasser, Trocknen und Eindampfen, erhält man 9, 6 g Kristalle, die aus Methanol umgelöst werden.
Das so erhaltene 1, 4 Endomethylen-3-phenyl-cyclohexan-2-sulfonsäure-N- benzoylamid schmilzt bei 196-198 und kann durch folgende Formel veranschaulicht werden :
