Description
Verfahren zur Herstellung von 4-lllethyl-5-ogyäthylthiazol. Das Hauptpatent betrifft ein Verfahren zur Herstellung von 4-Methyl-5-oxyäthylthia- zol. Gegenstand des vorliegenden Patentes ist eine Abänderung jenes Verfahrens zur Herstellung des gleichen 4-Methyl-5-oxyäthyl- thiazols. Das Verfahren ist dadurch gekenn zeichnet, dass man das Methylpropylketon,
dessen Propylrest an dem zur .getogruppe a-ständigen gohlenstoffatom einen austausch fähigen Substituenten und endständig eine in Hydroxyl umwandelbare Carbonsäureester- gruppe enthält, mit Salzen der Rhodanwas- serstoffsäure zu dem 2-Oxy-4-methylthiazol, das in 5-Stellung einen die oben gekenn zeichnete Carbonsäureestergruppe enthalten den Äthylrest aufweist,
umsetzt und dass man alsdann die Hydroxylgruppe in 2-Stel- lung durch Einwirken eines Phosphorhalo genids durch Halogen und letzteres durch Behandeln mit einem Reduktionsmittel durch Wasserstoff ersetzt und dass man die an dem Äthylrest stehende Carbonsäureestergruppe durch Einwirkung von Ammoniak in die Carbonsäureamidgruppe, diese durch Einwir ken von Alkalihypohalogenit in die Amino- gruppe umwandelt und letztere durch Ein wirken von salpetriger Säure durch die Hydroxylgruppe ersetzt.
Geeignete Ketone der bezeichneten Art sind die r-Halogen-r- acetobuttersäureester. Von den Salzen der Rhodanwasserstoffsäure kommen vorzugsweise die Alkali- und Erdalkalisalze für die Kon densation in Betracht. Die bei der Um setzung mit den Salzen der Rhodanwasser- stoffsäure zunächst entstehende Rhodanver- bindung kann durch Behandeln mit Mineral säure zum 2-Oxy-4-methylthiazol-(5)-propion- säureester kondensiert werden.
Für den Er satz der Hydroxylgruppe in 2-Stellung durch Halogen hat sich als Phosphorhalogenid be sonders Phosphoroxychlorid als geeignet er wiesen; der Ersatz des in 2-Stellung ein geführten Halogenatoms durch Wasserstoff wird zweckmässig durch Behandeln mit Zink staub und Eisessig bewirkt.
In dem so er hältlichen 4-Methylthiazol-(5)-propionsäure- ester wird die Carbonsäureestergruppe in an sich bekannter Weise in die Aminogruppe übergeführt, indem man die Carbonsäureester- gruppe durch Behandeln mit Ammoniak in die Carbonsäureamidgruppe überführt und die Carbonsäureamidgruppe durch Einwirken von Alkalihypohalogenit (nach der Methode von Hoffmann) in die Aminogruppe umwandelt.
Die Aminogruppe wird schliesslich in an sich bekannter Weise durch Einwirken von sal petriger Säure in die Hydroxylgruppe über geführt. Das so erhältliche 4-Methyl-5-oxy- äthylthiazol zeigt die gleichen Eigenschaften wie das im Hauptpatent beschriebene Produkt. Beispiel: 158 g r-Acetobuttersäureäthylester (vgl.
"Helv. Chim. aetall 11 (19191 Seite 152) wer den mit 160 g Brom in 300 cin3 absolutem Äther bromiert und durch 200 g Barium- rhodanid in den rohen Rhodan-r-acetobutter- säureäthylester übergeführt. Der Ringschluss zum Thiazol erfolgt durch zweistündiges Erhitzen mit alkoholischer, etwa 10%iger Schwefelsäure.
40 Gewichtsteile des 2-Oxy-4-methylthia- zolyl-5-propiorisäureätliylesters werden mit 160 g Phosphoroxychlorid Z Stunden unter Rückfluss zum Sieden erhitzt. Nach Ab dampfen des überschüssigen Phosphoroxy- chlorids im Vaeuum wird mit kaltem Wasser und Natriumcarbonat unter Kühlen behan delt und der abgeschiedene 2-Chloi--4-methyl- thiazolyl-5-propionsäureäthylester durch De stillation gereinigt. Er siedet unter 7 mm Druck bei 148-1500.
Das Chloratom in 2-Stellung wird durch Behandeln mit Zinkstaub und Eisessig durch Wasserstoff ersetzt. Der dabei erhaltene 4- Methylthiazolyl-5-propionsäureäthyleste- sie det unter 7 mm Druck bei 130-132 0.
19,9 Gewichtsteile dieses Produktes wer den mit 250 eins bei 0 0 gesättigtem inethyl- alkoholischem Ammoniak 12 Stunden im Autoklaven auf 120-130 0 erhitzt. Nach dem Erkalten wird der Methylalkohol ver dampft. Der Rückstand wird durch Ver reiben mit Äther kristallin. Man erhält farb- lose Kristalle des 4-Dletliy l-5-thiazolylpropion- säureaniids vom F.960 nach dein L'mkristalli- sieren aus Alkohol/Ätlier.
7,7 Gewichtsteile dieser Verbindung wer den fein gepulvert und mit einer kalten Lö sung von 7,3 Gewichtsteilen Brom in 100 Gewichtsteilen 2,5 normaler wässeriger Ka- liurnhydroxy dlösung übergossen und bis zur erfolgten Lösung gerührt. Dann werden 15 Gewichtsteile Kaliumhydroxyd hinzugefügt und 2 Stunden auf 90--100' erhitzt. Die Lösung wird mit Kaliumcarbonat gesättigt und mit Äther wiederholt extrahiert. Nach dem Trocknen über geglühtem Kaliumcar- bonat wird das Lösemittel verdampft und destilliert.
Das 4-3letliyl-5-aminoäthylthiazol siedet unter 7 mm Druck bei 1030. Sein salzsaures Salz bildet farblose Nadeln vom F.246'. Sein Pikrat schmilzt bei<B>2270.</B>
14,3 Gewielitsteile 4-Methyl-5-aminoäthyl thiazol werden in 120 Gewichtsteilen 15-n Schwefelsäure gelöst und kalt mit einer kon zentrierten Lösung von 7,5 Gewichtsteilen Natriumnitrit versetzt. Nach Stehen über Nacht wird mit Kaliumearbonat übersättigt und das gebildete 4-llethy 1-5-oxyätliy lthiazol mit Äther extrahiert. Es zeigt die im Haupt patent angegebenen Eigenschaften.
Process for the preparation of 4-IIIethyl-5-ogyäthylthiazole. The main patent relates to a process for the production of 4-methyl-5-oxyäthylthiazole. The subject of the present patent is a modification of that process for the preparation of the same 4-methyl-5-oxyethyl thiazole. The process is characterized in that the methyl propyl ketone,
whose propyl radical contains an exchangeable substituent on the carbon atom in the a-to-geto group and a carboxylic ester group which can be converted into hydroxyl at the end, with salts of rhodanic acid to form 2-oxy-4-methylthiazole, which in the 5-position has one of the above marked carboxylic acid ester group contains the ethyl radical,
and that the hydroxyl group in the 2-position is then replaced by the action of a phosphorus halide by halogen and the latter by treatment with a reducing agent by hydrogen and that the carboxylic acid ester group on the ethyl radical is converted into the carboxamide group by the action of ammonia, this by action The alkali metal hypohalite is converted into the amino group and the latter is replaced by the hydroxyl group through the action of nitrous acid.
Suitable ketones of the type indicated are the r-halo-r-acetobutyric acid esters. For the replacement of the hydroxyl group in the 2-position by halogen, phosphorus oxychloride has proven to be particularly suitable as phosphorus halide; the replacement of the halogen atom introduced in the 2-position by hydrogen is conveniently effected by treatment with zinc dust and glacial acetic acid.
In the 4-methylthiazole- (5) -propionic acid ester available in this way, the carboxylic acid ester group is converted into the amino group in a manner known per se by converting the carboxylic acid ester group into the carboxamide group by treatment with ammonia and the carboxamide group by the action of alkali metal hypohalite (according to Hoffmann's method) converted into the amino group.
The amino group is finally converted into the hydroxyl group in a manner known per se by the action of saline acid. The 4-methyl-5-oxyethylthiazole thus obtainable shows the same properties as the product described in the main patent. Example: 158 g of ethyl r-acetobutyrate (cf.

14.3 parts by weight of 4-methyl-5-aminoethyl thiazole are dissolved in 120 parts by weight of 15N sulfuric acid and a concentrated solution of 7.5 parts by weight of sodium nitrite is added while cold. After standing overnight, it is oversaturated with potassium carbonate and the 4-llethy 1-5-oxyätliy lthiazole formed is extracted with ether. It shows the properties specified in the main patent.
