Description
Verfahren zur Herstellung einer Verbindung des Theobromins. Gegenstand des Hauptpatentes ist ein Ver fahren zur Herstellung einer Verbindung von Theobromin mit einer organischen Säure, wo bei Theobromin mit Natriumhydroxyd zu Theobrominnatrium umgesetzt und dieses mit dem Natriumsalz der Isovaleriansäure zur Re aktion gebracht wird.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung einer weiteren therapeutisch wertvollen Doppelverbindung aus Theobromin. Das Verfahren ist dadurch ge kennzeichnet, dass Theobromin mit Lithium- hydroxyd zu Theobrominlithium umgesetzt und dieses mit dem Lithiumsalz der Iso- valeriansäure zur Reaktion gebracht wird, wo bei eine farblose, leicht wasserlösliche Ver bindung entsteht.
Die neue Verbindung hat neben blutdrucksenkender und diurethischer Wirkung auch ausgezeichnete sedative und krampflösende Wirkung, ohne die unerwünsch ten Begleiterscheinungen bekannter Präparate zu zeigen. Sie eignet sich vorzüglich zu Dauer behandlungen, zum Beispiel bei Arterioskle- rose, klimakterischen Beschwerden und Be schwerden, hervorgerufen durch. hohen Blut druck.
Die Umsetzung der Reaktionskomponen ten erfolgt vorteilhaft in wässerigem oder alkalisch wässerigem Medium, derart, dass man molekulare Mengen des Theobromin- lithiums und des Lithiumsalzes der Isovale- riansäure in einem Lösungsmittel zur Um setzung bringt und das Reaktionsgemisch zweckmässig zur Trockene einengt. <I>Beispiel:</I> 11,97 Teile Lithiumhydrogyd löst man in 200 Teilen destilliertem Wasser und setzt hierauf eine konzentrierte Lösung von 51 Teilen Isovaleriansäure zu.
Zu dieser Lösung wird eine alkoholische Lösung, hergestellt aus 99 Teilen Theobromin und 11,97 Teilen Lithium- hydroxyd, zugesetzt und hierauf wird das Reaktionsgemisch auf dem Wasserbade, zweck mässig unter vermindertem Druck, bis zur Trockene eingedampft. Es verbleibt ein weisses, kristallines, leicht wasserlösliches Produkt, welches rote Flammenfärbung und Murezid- reaktion gibt.

The new compound has an antihypertensive and diuretic effect as well as an excellent sedative and antispasmodic effect without showing the undesirable side effects of known preparations. It is particularly suitable for long-term treatments, for example for arteriosclerosis, climacteric complaints and complaints caused by. high blood pressure.
The reaction components are advantageously reacted in an aqueous or alkaline aqueous medium in such a way that molecular amounts of theobromonithium and the lithium salt of isovaleric acid are reacted in a solvent and the reaction mixture is expediently concentrated to dryness. <I> Example: </I> 11.97 parts of lithium hydrogen are dissolved in 200 parts of distilled water and a concentrated solution of 51 parts of isovaleric acid is then added.
An alcoholic solution, prepared from 99 parts of theobromine and 11.97 parts of lithium hydroxide, is added to this solution, and the reaction mixture is then evaporated to dryness on a water bath, advantageously under reduced pressure. A white, crystalline, easily water-soluble product remains, which gives a red flame color and a mureocide reaction.
