Verfahren zur Darstellung einer Calciumverbindung eines Phosphor enthaltenden Eiweissspaltproduktes. Im Patent Nr. 123060 und dessen Zusatz Nr. 124689 ist ein Verfahren zur Darstellung eines Gemisches von Verbindungen beschrie ben, welches den gesamten Phosphor und das Eisen der im Eigelb enthaltenen Proteide gebunden enthält. Dieses Gemisch besteht aus Verbindungen, die von den genannten Elementen zum Teil nur das erstere, zum Teil aber beide nebeneinander aufweisen.
Es wurde nun gefunden, dass man eine Calciumverbindurrg eines Phosphor enthalten den, eisenfreien Eiweissspaltproduktes erhält, wenn man entfettetes Eigelb zunächst in Gegenwart einer Mineralsäure einer peptischen Verdauung unterwirft, bis nach erfolgter Auf lösung des Eigelbes ein unlöslicher Nieder schlag entsteht, der durch Zusatz eines zur Abstumpfung der Mineralsäure dienenden Salzes einer organischen Säure vervollstän digt wird, die hierbei entstandene Fällung abfiltriert und einer tryptischen Verdauung in schwach alkalischem Medium unterwirft, die erhaltene Verdauungsflüssigkeit schwach ansäuert, filtriert,
aus dem Filtrat durch Be handlung mit Scbwermetallsalzen ein Gemisch von Sebwermetallverbindungen des Phosphor-, sowie des Phosphor- und eisenhaltigen Ei weissspaltproduktes abscheidet, dieses Gemisch mit solchen Natriumverbindungen zersetzt, deren Anionen mit dem betreffenden Schwer metall unlösliche Verbindungen bilden, den entstandenen Niederschlag abfiltriert, aus dem Filtrat, gegebenenfalls nach Einengung im Vakuum, mit Alkohol das Natriumsalz des Phosphor- und eisenhaltigen Eiweissspaltpro duktes ausfällt,
die Fällung filtriert und aus dem Filtrat nach Abdestillieren des Sprits mit einer löslichen Calciumverbindung das Calciumsalz des phosphorhaltigen, eisenfreien Eiweissspaltproduktes ausfällt.
Das Calciumsalz des phosphorhaltigen, eisenfreien Eiweissspaltproduktes stellt ein weisses, in Wasser unlösliches, geschmack loses Pulver dar. Durch Behandlung mit solchen Alkaliverbindungen, deren Anionen mit Calcium schwerlösliche Verbindungpn bilden, lässt sich dasselbe in ein wasserlös liches Alkalisalz überführen. Mit salzsäure haltigem Alkohol kann man aus letzterem das phosphorhaltige Eiweissspaltprodukt in Form der freien Säure gewinnen, welche in Wasser nur wenig, in mit Mineralsäuren an gesäuerten Mineralsalzen ganz unlöslich ist. Ihr Phosphorgehalt bewegt sich zwischen 12,5-13,5 %.
Sie zeigt schöne Biuretreak- tion; die Millonsche und Molischsche Reak tion sind negativ.
Die neue Verbindung soll zu therapeu tischen Zwecken verwendet werden. Beispiel: 10 kg des mit kochendem Sprit extra hierten, getrockneten und fein gemahlenen Eigelbs werden in 500 Liter Wasser auf geschlemmt, mit 2,8 kg konzentrierter Salz säure und<B>100</B> gr Pepsin oder einer ent sprechenden Menge eines Salzsäureauszuges der Magenschleimhaut des Schweines ver setzt und bei 37-40 o unter stetigem Rühren erwärmt, bis das Eigelb in Lösung gegangen ist. Nach zirka 10 Stunden setzt man 3,8 kg Natriumacetat oder 11/2 kg Natriumkarbonat und 1 kg Eisessig zu.
Man filtriert, zweck mässig auf der Filterpresse, wäscht mit Wasser nach und nimmt den Rückstand mit einer Lösung von 1200 gr Soda in 40 Liter Wasser auf, setzt 100 gr fein gehackter Bauchspei cheldrüse zu und verdaut während 24 bis 48 Stunden bei 40 0. Die Verdauungsflüssig keit wird dann mit Essigsäure leicht ange säuert, filtriert und das Filtrat mit Blei zuckerlösung versetzt, solange ein Nieder schlag entsteht. Die Bleifällung wird filtriert, mit Wasser ausgewaschen und mit einer 20 %igen Sodalösung verrührt, bis die Mi schung auf Phenolphtaleinpapier schwach rosa anschlägt.
Die Phosphorverbindungen gehen als Natriumsalze in Lösung, das Blei bleibt als Karbonat unlöslich zurück. Die Lösung der Natriumsalze wird langsam unter Rühren mit 212 bis 3 Liter Alkohol versetzt, wobei sich eine flockige Fällung bildet, welche ab filtriert wird. Das alkoholische Filtrat wird durch Destillation im Vakuum vom Alkohol befreit und mit einer Calciumchlorid- oder Calciumacetatlösung versetzt, worauf das Calciumsalz des phosphorhaltigen, eisenfreien Eiweissspaltproduktes ausfällt. Man filtriert, wäscht mit Wasser nach und tro^krlet.
Durch Umsetzung mit Natriumoxalat kann man das Calciumsalz in das Natriumsalz überführen, das in Wasser leicht löslich ist und von dem aus man durch Umsetzung mit löslichen Erdalkali- oder Metallsalzen zu den entsprechenden Erdalkali- oder Schwermetall salzen; z. B. dem Magnesium-, Eisen- oder Quecksilbersalz und dergleichen gelangen kann. Durch Eingiessen der Lösung des Natrium salzes in mit Salzsäure angesäuerten Alkohol erhält man das phosphorhaltige, eisenfreie Eiweissspaltprodukt in Form der freien Säure.

The calcium salt of the phosphorus-containing, iron-free protein breakdown product is a white, water-insoluble, tasteless powder. By treatment with such alkali compounds, the anions of which form poorly soluble compounds with calcium, the same can be converted into a water-soluble alkali salt. With alcohol containing hydrochloric acid, the phosphorus-containing protein breakdown product can be obtained from the latter in the form of the free acid, which is only slightly insoluble in water and completely insoluble in mineral salts acidified with mineral acids. Their phosphorus content ranges between 12.5-13.5%.
It shows a beautiful biuretension; the Millon and Molisch reactions are negative.
The new compound is intended to be used for therapeutic purposes. By reacting with sodium oxalate, the calcium salt can be converted into the sodium salt, which is readily soluble in water and from which it is salted by reaction with soluble alkaline earth or metal salts to give the corresponding alkaline earth or heavy metal; z. B. the magnesium, iron or mercury salt and the like can arrive. By pouring the solution of the sodium salt into alcohol acidified with hydrochloric acid, the phosphorus-containing, iron-free protein breakdown product is obtained in the form of the free acid.
