Description
Verfahren zur Abscheidung der im Eigelb enthaltenen phosphor- und eisenhaltigen Kerne. Seitdem M i e s c h e r (Med. Chem. Untersuchungen Hoppe Seylers, Heft 4 [18711, S. 5o2) auf das Vorhandensein von organisch gebundenem Phosphor in mit Alkohol und Äther extrahiertem Eigelb hingewiesen hat - er nahm an daß es sich um echte Nucleine handelte -. suchten verschiedene Forscher die phosphorhaltige Verbindung mehr oder weniger frei von Eiweiß abzuscheiden. So liegt im Bungeschen Haematogen (Zeitschr. f. physiol. Chemie, Bd.9, S.49ff. [18851) eine Verbindung vor, welche 5"9 Prozent P und o,:29 Prozent Fe enthält gegenüber 6,7 bis 7,8 Prozent P im Miescherschen Präparat. Dann versuchte A 1 t m a n n (Archiv fÜr Anat. und Phys. [18891, S. 524 ff.) nach seiner allgemeinen Darstellungsmethode der Nucleinsäuren eine ähnliche Säure aus dem Eidotter abzuscheiden. Sie enthielt rund 7 Prozent P. Nach Milroy (Zeitschr. f. physiol. Chemie, Bd.22, S.307ff.) handelte es sich aber nicht um echte Nucleinsäure. Sie gab nämlich nach Hydrolyse mit Mineralsäuren keine Xanthinbasen, zeigte eine ausgesprochene Biuret-, jedoch keine Millonsche Reaktion und- enthielt von 7,51 bis 7,94 Prozent P. Im Jahre igoi stellten L e v e n e und A 1 s b e r g (Zeitschr. f. physiol. Chemie, Bd. 31 [19011, S. 543 ff.) ihre Avivitellinsäure dar durch Behandlung-von gereinigtem Vitellin mit 25prozentigem Ammoniak in der Kälte, neutralisieren mit Essigsäure, abscheiden der Eiweißstoffe mit Hilfe von Pikrinsäure und Fällen der - obengenannten Säure mit Alkohol. Die durch Umlösen gereinigte Verbindung enthielt im Mittel 9,88 Prozent P und 0,57 Prozent Fe, Diese Autoren konnten auch beim Präparat von Milroy unter gewissen Vorsichtsmaßregeln die Millonsche Reaktion erhalten. Sie gab die Biuret- und Millonsche Reaktion. Nach Hydrolyse wurden 17,8 Prozent des Gesamtstickstoffs in Form des Arginins und 3 Prozent in Form des Histidins festgestellt. Vier Jahre später beschrieben H u g o u n e n q und M o r e 1 (Comptes rendus, Bd. i4o [igo5], S. io65) ein nach Bunge dargestelltes, aber angeblich viel weiter gereinigtes Präparat (die Reinigungsmethode wurde nicht angegeben) mit 8,7 Prozent P und 0,45 Prozent Fe.Process for separating the cores containing phosphorus and iron contained in the egg yolk. Vom Verfahren der Patentschrift 4o6 963 Kl. i:2p unterscheidet sich das vorliegende unter anderem dadurch, daß eine Vorverdauung mit Pepsinsalzsäure im Falle von Eivitellin unentbehrlich ist und daß die Endprodukte etwa doppelt so viel Phosphor enthalten, als die entsprechenden Produkte aus Casein. Der Phosphorkern &s Milchcaseins enthält auch kein organisch gebundenes Eisen. Beispiel i. io kg mit kochendem Alkohol extrahiertes, getrocknetes und fein gemahlenes Eigelb werden in 5001 Wasser aufgeschlemmt, mit 2,8 kg konzentrierter Salzsäure und ioo g Pepsin oder einer entsprechenden Menge ,eines Salzsäureauszuges der Magenschleimhaut des Schweines versetzt und bei 37 bis 4o' unter stetigem Rühren erwärmt, bis das Eigelb in Lösung gegangen ist. Nach etwa io Stunden setzt man 3,8 kg Natriumacetat oder ill, kg Natriunicarbonat und i kg Eisessig zu. Man filtriert, zweckmäßig auf der Filterpresse, wäscht mit Wasser nach und nimmt den Rückstand mit einer Lösung von i .2oo g Natriumcarbonat in 4o 1 Wasser auf, setzt ioo g fein gehackter Bauchspeicheldrüse zu und verdaut während 24 bis 48 Stunden bei 40'. Die Verdauungsflüssigkeit wird dann mit Essigsäure leicht angesäuert, filtriert und das Filtrat mit Calciumchlorid oder Calciumacetat und mit einem Volumen Alkohol gefällt. Die Fällung wird abfiltriert, mit Alkohol nachgewaschen und, getrocknet. Man erhält auf diese Weise ein gelblich weißes, geschmackloses Pulver, welches nur teilweise in Wasser löslich ist, rund 10,5 Prozent P, i Prozent Fe und 9 bis io Prozent Ca enthält. Beispiel :2. DietryptischeVerdauungsflüssigkeit, wie sie im Beispiel i erhalten wurde, wird nach Ansäuern mit Essigsäure und Abfiltrieren &s Ungelösten mit Bleizuckerlösung versetzt, solange ein Niederschlag entsteht. Die Bleifällung wird filtriert, mit Wasser ausgewaschen und mit einer 2,oprozentigen Natriumcarbonatlösung verrührt, bis die Mischung auf Phenolphthaleinpapier schwach rosa anschlägt. Die Phoshporverbindungen gehen als Natriumsalze in Lösung, das Blei bleibt als Carbon-at unlösl-ich zurück. Die Lösung der Natriumsalze wird im Vakuum stark konzentriert, mit Alkohol gefällt und im Vakuum getrocknet. Die Verbindung ist ein weißes Pulver, welches in Wasser nur teilweise löslich ist,mit Alkalien lösliche Salze gibt, mit löslichen Erdalkali- und Metallsalzen, teils lösliche, teils unlösliche Salze gibt, gegenüber kalten Säuren ziemlich beständig, gegenüber Ätzalkalien sehr unbeständig ist.
