Verfahren zur Darstellung eines Gemisches von Verbindungen, welches den gesamten Phosphor und das Eisen der im Eigelb enthaltenen Proteide gebunden enthält. Seitdem Miescher (Med. Chem. Untersu chungen Hoppe Seylers, Heft 4, 1871, Seite 502) auf die Existenz von organisch gebunde nem Phosphor im mit Alkohol und Äther ex trahierten Eigelb hingewiesen hat - er nahm an, dass es sich um echte Nucleine handelte --,
suchten verschiedene Forscher die phos phorhaltige Verbindung mehr oder weniger frei. von Eiweiss zu isolieren. So liegt im Bungeschen Hämatogen (Zeitschrift für phy siel. Chemie, Band 9, Seite 49, 1885) eine Substanz vor, welche 5,19 % P und 0,29 % Fe enthält, gegenüber 6,7 bis 7,8 % P im Miescherschen Präparat. Dann versuchte Altmann (Archiv für Anat. und Physiol. <B>1889)</B> nach seiner allgemeinen Darstellungs methode der Nucleinsäuren eine ähnliche Säure aus dem Eidotter abzuscheiden.
Sie enthielt rund 7 % P. Nach Milroy (Zeit schrift für physiol. Chemie, Band 22, Seite 307) handelte es sich aber nicht um echte Nucleinsäure. Sie gab nämlich nach Hydro lyse mit Mineralsäuren keine Xa.nthinbasen, zeigte eine ausgesprochene Biuret-, jedoch keine Millonsche Reaktion und enthielt von 7,51 bis 7,94 % P.
Im Jahre 1901 stellten Levene und Alsberg (Zeitschrift für phy- siol. Chemie, Band 31, 1901, Seite 543) ihre Avivitellinsäure dar durch Behandlung von gereinigten Vitellin mit 25 %igem Ammo niak in der Kälte, Neutralisieren mit Essig säure, Abscheiden der Eiweissstoffe mit Hilfe von Pikrinsäure und Fällen der oben ge nannten Säure mit Alkohol. Die .durch Um lösen gereinigte Substanz enthielt im Mittel 9;88 % P und 0,57 % Fe.
Diese .'Autoren konnten auch beim Präparat von Milroy un ter gewissen Krautelen die Millonsche Reak tion erhalten. Sie gab die Biuret- und Mil- lonsche Reaktion. Nach Hydrolyse wurden 17,8 /a des Totalstickstoffes in Form des Ar- ginins und 3 % in Form des Histidins fest gestellt.
Vier Jahre später beschrieben Hu- gounenq und Morel (Comptes rendus 140, Seite 1065, [190y) ein nach Bunge darge stelltes, aber angeblich viel weiter gereinig tes Präparat (die Reinigungsmethode wurde nicht angegeben) mit 8,7 % P und 0,45 % Fe.
Aus diesen Literaturangaben erhellt, dass es noch nie gelungen ist, die Phosphor und Eisen .gebunden enthaltenden Spaltprodukte der Eigelbproteide getrennt von phosphor- und eisenfreien Eiweissabbauprodul@ten zti gewinnen.
Gegenstand vorliegender Erfindung ist nun ein Verfahren zur Darstellung eines aus schliesslich aus Phosphor- und eisenhaltigen Eiweissspaltproduktnatriumsalzen bestehen den Gemisches von Verbindungen, welches den gesamten Phosphor und das Eisen der im Eigelb enthaltenen Proteide gebunden enthält, dadurch gekennzeichnet, dass man entfettetes Eigelb zunächst in Gegenwart einer Mineralsäure einer peptischen Verdau ung unterwirft, bis nach erfolgter Auflösung ein unlöslicher Niederschlag entstellt,
der durch Zusatz eines zur Abstumpfung der Mineralsäure dienenden Salzeis einer organi schen Säure, vorzugsweise Natriumacetat, vervollständigt wird, die hierbei entstandene Fällung abfiltriert und einer tryptischen Verdauung in schwach alkalischem Medium unterwirft, .die erhaltene @erdauungsf@itäsig- keit hernach schwach ansäuert, filtriert, aus dem Filtrat durch Behandlung mit löslichen Schwermetallverbindungen, zum Beispiel Blei-, Kupfer-,
Eisen- und @uechsilbersalzen, das Gemisch der Schvrermetallsalzp der Phos phor- und eisenhaltigen Eiweissspaltprodukte abscheidet, diese mit solchen Natriumverbin- dungen zersetzt, deren Anionen mit dem be treffenden Schwermetall unlösliche Verbin dungen bilden, wie zum Beispiel Natrium carbonat, und dadurch das Gemisch der Na triumsalze der Phosphor- und eisenhaltigen Eiweissspaltprodukte gewinnt.
Das Gemisch der Natriumsalze der Phos phor- und eisenhaltigen Eiweissspaltprodukte stellt ein in Wasser lösliches, gelbliches Pul ver dar, welches eine schöne Biuretreaktion, keine Millonsche, \dagegen eine Reaktion nach '@Zolisch gibt. Es enthält etwas über 10 @,1 P, ca. 1 /" Fe und 10 /" Na.
Durch doppelte Umsetzung mit Erdalkalisalzen und Fällen mit Alkohol, oder durch Behandlung mit löslichen Schwermetallverbindungen er hält man aus dem Gemisch der Alkalisalze das (-ntsprechende Erdalkali- oder Schwer- ]Durch Fällen.
